Current area-based management systems regulate only about 1.18% of the high seas, leaving highly migratory fish species at risk of overexploitation. As a result, new legal mechanisms are essential for protecting and managing high seas fisheries. In recent decades, stakeholders have debated how to balance competing interests while ensuring equitable and sustainable access to areas beyond national jurisdiction. One proposed solution is the establishment of Marine Protected Areas (MPAs). However, creating and managing high seas MPAs is a complex process. Removal models have long been used to estimate population abundance by progressively capturing and removing individuals from a closed population. These models provide a valuable tool for ecological monitoring, but their accuracy depends heavily on assumptions about detection probability, which may decline over successive sampling passes. Traditional removal models assume constant detection probabilities, an assumption that is often violated in real-world applications. This thesis aims to advance hierarchical Bayesian models by accounting for variable detection probabilities, improving the reliability of abundance estimates and trend detection. Purpose: This study examined the effectiveness of laser utilization in stabilizing periodontal disease over a one-year period. This research focused on the results following scaling and root planing with adjunctive laser-assisted periodontal therapy and laser bacterial reduction at subsequent periodontal maintenance visits. While short-term advantages are documented in the literature, research on sustained effectiveness is limited. Methods: A retrospective chart review of 30 adult patients who received the above treatment was conducted. Periodontal charting was recorded at three visits over one year. Since stalking laws were enacted in the 1990s, they have been historically difficult to enforce. As less and less of our lives become private due to our ever-present devices, stalkers frankly have more access to their targets than ever before. The tenuous nature of stalking laws and the growing resources for stalkers place targets of stalking in a very precarious situation: the legal system has never been adequately equipped to handle stalking cases, and local justice systems do not have the resources to handle tech-based crimes. Discrimination has profoundly shaped the lives of individuals within disadvantaged communities, but its impact on mental and physical health has often been overlooked. In this project, I review literature on perceived discrimination based on immigrant status and its effects on psychological and physical health and examine a potential mediator and moderator of these relationships. Specifically, the relationships between immigration-based discrimination and both mental health and physical health may be mediated by perceived stress, meaning that discrimination may lead to higher stress levels, which results in poorer health outcomes. Water scarcity and rising global temperatures are two of the greatest current and future threats to poultry sustainability. Therefore, selection for water efficiency (WE) and heat resilience are of vital importance. Additionally, intestinal integrity is of critical importance under challenging conditions to maintain nutrient absorption and therefore, growth and performance of broilers. Here, we examined the effect of chronic cyclic heat stress (HS) on the ileal expression profile of tight-junction, gap-junction, adherens, and desmosome genes in the fourth generation of divergently selected low (LWE)- and high water efficient (HWE)-chicken lines. Bacteria utilize sophisticated secretion systems to inject effectors into host cells in order to facilitate their survival and replication in the host. Some pathogenic species of Gram-negative bacteria use a conserved contact-dependent T3SS to inject a wide array of effectors. While the effectors diverge in their structures, functions and cell-localization, the T3SS is well conserved. Several effectors discussed in this thesis target key host inflammasome responses. The focus of this work is how the Yersinia effector YopM inhibits the pyrin inflammasome. During Yersinia infection, two effectors, YopE and YopT inadvertently activate the pyrin inflammasome while targeting RhoA to avoid phagocytosis. …

Priming CD8+ T cells against tumors or viral pathogens results largely from cross-presentation of exogenous antigens by type 1 conventional dendritic cells (cDC1s). Although monocyte-derived DCs and cDC2s can cross-present in vitro, their physiological relevance remains unclear. Here, we used genetic models to evaluate the role of cDC subsets in presentation of cell-associated and immune complex antigens to CD4+ and CD8+ T cells in vivo. For cell-associated antigens, cDC1s were necessary and sufficient to prime both CD4+ and CD8+ T cells. Microglia impact brain development, homeostasis, and pathology. One important microglial function in Alzheimer's disease (AD) is to contain proteotoxic amyloid-β (Aβ) plaques. Recent studies reported the involvement of autophagy-related (ATG) proteins in this process. Here, we found that microglia-specific deletion of Atg7 in an AD mouse model impaired microglia coverage of Aβ plaques, increasing plaque diffusion and neurotoxicity. Single-cell RNA sequencing, biochemical, and immunofluorescence analyses revealed that Atg7 deficiency reduces unfolded protein response (UPR) while increasing oxidative stress. This thesis reviews the development of narrative and systematic methods of argumentative legitimation in the Western tradition of philosophy from its conception until the present day. Utilizing both primary sources from central figures in key movements within this development process and supplementary secondary sources, this analysis notes which systems were used as appeals to higher authority and the trends in the adoption and transformation of new models. It is demonstrated that the Enlightenment era punctuated a progression begun in the premodern era from simple, cultural narratives to individualized, subjective, and self-perpetuating Grand Narrative systems, and that it generated the preconditions …

  1. We aimed to elucidate potential methylation, proteomic, and metabolomic mechanisms by which APOL1 variants may be linked to kidney disease.

  2. We report distinct methylation profiling between APOL1 risk allele carriers and noncarriers, many near APOL gene family.

  3. We report higher APOL1 protein and lower C18:1 cholesteryl ester in two risk allele carriers.

Background: The APOL1 high-risk haplotype has been associated with CKD and the deterioration of kidney function, particularly in populations with West African ancestry. Around the time it was passed, the Maine Indian Claims Settlement Act of 1980 was considered by many to be a great victory for the Wabanaki Nations in Maine. But in the decades since, the Act has substantially hindered the Wabanaki Nations’ self-determination efforts. Frequent litigation between the Nations and the state of Maine, narrow interpretation by courts, and the state of Maine’s opposition to remedying the Act’s most harmful provisions have constrained the sovereignty of the Wabanaki Nations. Background: Methamphetamine (MA) use has been linked to engaging in sexual risk behaviors (SRBs) that are associated with HIV/STIs, particularly among men who have sex with men (MSM) and men who have sex with men and women (MSMW; hereafter MSM/W). The objectives of this analysis were to determine whether reduced MA is associated with decreases in SRBs in a sample of MSM/W.

Motion sickness is associated with thermoregulation and metabolic control, but the underlying neural circuitry remains largely unknown. Here we show that neurons in the medial vestibular nuclei parvocellular part (MVePC) mediate the hypothermic responses induced by motion. Reactivation of motion-sensitive MVePC neurons recapitulates motion sickness in mice. We show that motion-activated neurons in the MVePC are glutamatergic (MVePCGlu), and that optogenetic stimulation of MVePCGlu neurons mimics motion-induced hypothermia by signalling to the lateral parabrachial nucleus (LPBN). Acute inhibition of MVePC-LPBN circuitry abrogates motion-induced hypothermia. Finally, we show that chronic inhibition of MVePCGlu neurons prevents diet-induced obesity and improves glucose homeostasis …

Borrelia burgdorferi (Bb) causes Lyme disease (LD), one of the most common vector-borne diseases in the Northern Hemisphere. Here, we solve the crystal structure of a mutated Bb vaccine antigen, CspZ-YA that lacks the ability to bind to host complement factor H (FH). We generate point mutants of CspZ-YA and identify CspZ-YAI183Y and CspZ-YAC187S to trigger more robust bactericidal responses. Compared to CspZ-YA, these CspZ-YA mutants require a lower immunization frequency to protect mice from LD-associated inflammation and bacterial colonization. People who regularly used Roundup have brought civil lawsuits against its manufacturer, Monsanto (now owned by Bayer), claiming Roundup caused their cancer diagnoses. Juries, particularly those in state courts, have awarded some plaintiffs massive damages awards. But jury trials elude most of the glyphosate plaintiffs who find themselves in federal court. Vaccine hesitancy is a term that has entered the spotlight since the beginning of the COVID-19 pandemic. With concerns about efficacy, safety, and individual choice, many people have developed a hesitancy toward receiving any vaccine, creating concerns for public and global health. To thrive in extreme conditions, organisms have evolved a diverse arsenal of adaptations that confer resilience. These species, their traits, and the mechanisms underlying them comprise a valuable resource that can be mined for numerous conceptual insights and applied objectives. One of the most dramatic adaptations to water limitation is desiccation tolerance. Understanding the mechanisms underlying desiccation tolerance has important potential implications for medicine, biotechnology, agriculture, and conservation. Freshwater snails are widely distributed in the rice field ecosystem. Freshwater snails’ feeding and metabolic activities can potentially increase the complexity of freshwater ecosystems, particularly through the nutrient cycle. Cipangopaludina chinensis, Physa acuta, and Semisulcospira libertina are commonly found in Japan's paddy field ecosystems. Spirodela polyrhiza is expected to support biodiversity, improve soil fertility, and reduce greenhouse gas emissions in paddy field ecosystems, while providing food and shelter for the snails. Weeds are managed by herbicides, but this can reduce the abundance of soil bacteria. This research aimed to determine the effect of active compounds of herbicides on weeds, the abundance of soil bacteria, growth and yield of sweet corn. The experiment was conducted from June to October 2023 in the fields and Laboratory of Agronomy and Horticulture, Universitas Jenderal Soedirman, Purwokerto. Ultimately this thesis will examine how Mean Girls melodramas moralize gender and sexuality. The Mean Girl is not an embodiment of moral hegemony. She may look like the perfect subject, but she is imperfect, because she tries to be perfect. She is a failure of femininity, because her attempts to be feminine reveal the masquerade. Womaniliness is not inherent; it is not real. The Mean Girl is attractive to the sapphic spectator because she exposes the alienation and artifice of gender and heterosexuality. The stereotypical image of the Mean Girl is plastic perfection, but she is not.

This thesis aims to demonstrate that relationships between students and nature can be improved through teaching young learners about music and bugs together, potentially leading to a greater overall love for and willingness to protect nature.
