Stress-induced illnesses, such as major depression and anxiety, are significant global causes of disability. This highlights the need for reliable animal models that accurately mimic social stress-induced maladaptations, thus, allowing the experimental study of the etiology of such disorders. Prairie voles (Microtus ochrogaster), known for their human-like bi-parental behavior and monogamous partner preferences, are a relevant model for the study of stress-induced psychopathologies. For this reason, in this dissertation, I examined whether exposing voles to the social defeat stress (SDS) paradigm, an experimental approach that captures bullying behavior, would result in both depression and anxiety-related profiles. G protein-coupled receptors (GPCR) can bind multiple ligands, leading to differential signaling events termed biased signaling. C-C chemokine receptor 7 (CCR7) is a GPCR known to have biased signaling between the two ligands it binds to, CCL19 and CCL21. Binding of C-C Chemokine Ligand 19 (CCL19) promotes a high level of CCR7 phosphorylation, while CCL21 leads to a much lower level of CCR7 phosphorylation; findings observed in HEK and dendritic cells. GPCR kinases (GRKs), which can control the phosphorylation patterns of a GPCR, are hypothesized to regulate biased signaling of a receptor. GRK expression differs between cell lines and cell …

Generational trauma, internalized racism, and family dysfunction permeated her psyche and crippled her self-worth sending her into the fantasy worlds she created in her head. The 21st century ushered in major social, political, and cultural changes in the United States, including a new era of homeland security and new forms of domestic terrorism. Little is known about how such shifts may have impacted women’s participation in terrorism plots. In response, data from the American Terrorism Study (ATS) are used to comparatively examine the involvement of women in homegrown and domestic terrorism plots before and after the 9/11 terrorist attacks. Findings of similarities and differences in women’s involvement in terrorism across time periods are presented and contextualized.

When low energy levels are detected, Ghrelin, a peptide hormone, is produced in the fundus of the stomach. Referred to as the "hunger hormone", Ghrelin is traditionally associated with increased food intake and blood sugar levels. A key modulator in the gut-brain interaction, Ghrelin's growth hormone secretagogue receptor can be found widely across the cortico-striosomal circuit. Critical for decision-making, Ghrelin-based manipulation of this circuit can directly affect reward and cost valuation. While Ghrelin is uniformly considered to increase the valuation of food-based rewards, little research has explored its impact on non-food-based rewards or its effects on cost valuation. Introduction: Opioid drugs (e.g., morphine) are used medicinally for pain relief but can also lead to opioid use disorder (OUD). These medications are prescribed at a higher rate among individuals with obesity (who might consume high fat diets) than the general population. Further, previous work from our lab and others has demonstrated that eating a high fat diet enhances sensitivity of rats to other addictive substances, including stimulants. However, it is not known if dietary manipulations impact individual sensitivity to the effects of opioids. Introduction. Testing is one of the most effective methods for reducing the spread of COVID-19. COVID-19 antigen tests can detect proteins from the COVID-19 virus called antigens and produce results within minutes, making them suitable for at-home use. Several factors have been identified as barriers to self-administering a COVID-19 antigen test, including the language of the testing instructions, the cost of testing, and access to tests. Increased expression of the cyclin-dependent kinase inhibitor p16Ink4a (p16) is detected in neurons of human Alzheimer's disease (AD) brains and during normal aging. Importantly, selective eliminating p16-expressing cells in AD mouse models attenuates tau pathologies and improves cognition. But whether and how p16 contributes to AD pathogenesis remains unclear. To address this question, we tested whether induction of p16 expression in neurons exacerbates AD pathologies. We created a doxycycline-inducible system to trigger p16 up-regulation in human-induced pluripotent stem cells (iPSCs) and neurons differentiated from iPSCs. Acne vulgaris remains one of the most prevalent dermatologic conditions worldwide, yet its management has become increasingly influenced by pop culture trends and consumer marketing. This review critically compares the American Academy of Dermatology (AAD) guidelines for acne treatment with the most popular skincare practices of the 2020s, including natural remedies, light therapy, and multi-step routines popularized through platforms like TikTok and Instagram. Genome-wide association studies (GWASs) in Alzheimer disease (AD) have uncovered over 70 loci significantly associated with AD risk, but identifying the true causal gene(s) at these loci requires systematic functional validation that is rarely performed due to limitations of time and cost. Here, we integrate transcriptome-wide association study (TWAS) with colocalization analysis, fine-mapping, and additional annotation of AD GWAS variants to identify 123 genes at known and suggestive AD risk loci. A comparison with human AD brain transcriptome data confirmed that many of these candidate genes are dysregulated in human AD and correlate with neuropathology. We then tested all available …

Over the last 20 years, research into the Meibomian glands of the eyelids has increased exponentially and is now widely recognized as a field of research. It is all the more astonishing that knowledge about another type of gland in the eyelids, the Moll glands or ciliary glands, has almost stagnated and there has been little to almost no progress, even though this type of gland as a whole takes up a relatively large volume in the upper and lower eyelids. Metabolism is fundamental to organism physiology and pathology. From the intricate network of metabolic reactions, diverse chemical molecules, collectively termed metabolites, are produced. In multicellular organisms, metabolite communication between different tissues is vital for maintaining homeostasis and adaptation. However, the molecular mechanisms mediating these metabolite communications remain poorly understood. Here, we focus on nucleosides and nucleotides, essential metabolites involved in multiple cellular processes, and report the pivotal role of the SLC29A family of transporters in mediating nucleoside coordination between the soma and the germline. Background
Head and neck squamous cell carcinoma (HNSCC) presents significant treatment challenges, particularly in cases unrelated to human papillomavirus (HPV). The chemokine receptor CXCR4, interacting with its ligand CXCL12, plays a crucial role in tumor proliferation, metastasis, and treatment resistance. This study explores the therapeutic potential of engineered monomeric and dimerized CXCL12 variants (CXCL121 and CXCL122, respectively) in HNSCC and evaluates potential additive effects when combined with radiation therapy.
Methods

Adipocyte differentiation plays an important role in bone remodeling due to secretory factors that can directly modulate osteoblast and osteoclast, thus affecting overall bone mass and skeletal integrity. Excessive adipocyte differentiation within the bone marrow microenvironment can lead to decreased bone mass, eventually causing osteoporosis. The mechanical microenvironment of bone marrow, including fluid shear, maintains the balance of adipocyte and osteoblast differentiation during bone remodeling. The Alzheimer's Disease Research Centers (ADRCs) consortium represents a critical locus of research on Alzheimer's disease and related disorders (ADRD) prevention, diagnosis, and intervention. Through the National Alzheimer's Coordinating Center's (NACC) standardized protocol, the Uniform Data Set (UDS), ADRCs have collected rich, harmonizable clinical and cognitive data. However, the collection of social data has been sparse and Center specific, constraining ADRD science that addresses research priorities on social determinants of health (SDOH) and health equity. We are surrounded, in both indoor and outdoor environments, by air containing particles of biological origin (bioaerosols). We constantly inhale them, and, depending upon their size, they deposit in different parts of our airways. Despite their ubiquitous nature and our constant exposure, bioaerosol diversity and composition of the environment are not well characterized, and we understand little about which bioaerosols we are exposed to and how this impacts our health, either positively or negatively. Vertebrate photoreceptors have been studied for well over a century, but a fixed nomenclature for referring to orthologous cell types across diverse species has been lacking. Instead, photoreceptors have been variably-and often confusingly-named according to morphology, presence/absence of 'rhodopsin', spectral sensitivity, chromophore usage, and/or the gene family of the opsin(s) they express. Initially identified as a regulator of complement activation on host cells, the known roles of CD46 (membrane cofactor protein [MCP]) have expanded. We now know that this ancient molecule is expressed on almost all nucleated cells as a family of four predominant isoforms. It also is involved in human reproduction, modulation of T cell activation and immunoinflammatory effector functions, autophagy, and the newly identified intracellular complement system (complosome). There is a sex bias in the incidence and progression of many kidney diseases. To better understand such sexual dimorphism, we integrated data from six platforms, characterizing 76 kidney samples from 68 mice at six developmental and adult time points, creating a molecular atlas of the mouse kidney across the lifespan for both sexes. We show that proximal tubules have the most sex-biased differentially expressed genes emerging after 3 weeks of age and are associated with hormonal regulations. Huntington's disease (HD) is a debilitating neurodegenerative disorder affecting an individual's cognitive and motor abilities. HD is caused by a mutation in the huntingtin gene producing a toxic polyglutamine-expanded protein (mHTT) and leading to degeneration in the striatum and cortex. Yet, the molecular signatures that underlie tissue-specific vulnerabilities remain unclear. Here, we investigate this aspect by leveraging multi-epitope protein interaction assays, subcellular fractionation, thermal proteome profiling, and genetic modifier assays. The use of human cell, mouse, and fly models afforded capture of distinct subcellular pools of epitope-enriched and tissue-dependent interactions linked to dysregulated cellular pathways and disease relevance. A brief roadmap of this article may be helpful. Part I describes the emergence of the Child Wellbeing framework and its powerful role in shaping twenty-first-century youth justice reforms captured by the Restatement. Part II shows how this framework has begun to have a far broader influence on the law affecting children and how the framework and the developmental approach embodied in the Restatement can guide courts and shape the trajectory of the law in the future. Part III explores potential challenges to the Restatement’s effectiveness in attaining a fully integrated system based on the Child Wellbeing framework.

This thesis aims to elucidate the intersections between the portrayal of tuberculosis in the Romantic era and the Enlightenment, evangelism, and the Industrial Revolution. This study aims to do so by locating and analyzing depictions in the primary source representations of the disease, then considering the effects of the three factors mentioned above on its representation. The Enlightenment, evangelism, and the Industrial Revolution interacted in a variety of ways, and the influence of all three factors was found within the representations of tuberculosis, albeit in often indirect ways. In Texas, approximately 1.6 million acres support an equine population exceeding 748,000 horses. This study assessed the effects of organic and inorganic fertilizer treatments on improved Bermudagrass (BG) and other vegetation (OTH) in overgrazed equine pastures at the Stephen F. Austin State University Equine Center. Treatments included equine manure (H), broiler poultry litter (P), and a commercial 13-13-13 fertilizer (C), with application rates based on pasture requirements.

Understanding their spatial distribution and relative abundance in streams draining suburbanizing watersheds is crucial for effective watershed and water quality management. This study investigates how the concentrations and composition of PFAS in surface water and sediment are impacted by urbanization. Viscosity plays an important role in the food industry as it affects the quality, processing stability, and shelf-life of food and impacts physiological responses such as satiation and satiety. Greater viscosity reduces food intake by increasing the perception of satiety, primarily by delaying gastric emptying and increasing tactile stimulation. Starch is commonly added to food products as a viscosifying ingredient, which often involves chemically modifying the starch via cross-linking, to enhance its resistance to high shear, temperature and acidity. Macrophages are a subset of immune cells with a variety of functions that are abundant in numerous tumor types such as colorectal and breast cancer. It has been suggested that macrophages may serve an important function as immune mediators in tumor behavior; however, macrophage activity is complex and remains not completely understood.
