Lafora disease (LD) is a fatal childhood progressive myoclonus epilepsy and glycogen storage disease that is caused by recessive mutations in either EPM2A or EPM2B. The hallmarks of LD are cytoplasmic, aberrant glycogen-like aggregates, called Lafora bodies (LBs), that drive disease progression. The 9th Annual Lafora Science Symposium was held in San Diego, California and brought together over 70 researchers, clinicians, academic trainees, and friends and family members of patients with LD and 80 attendees joined virtually. Phospholipase C isozymes (PLCs) hydrolyze phosphatidylinositol 4,5-bisphosphate (PIP2) into inositol 1,4,5-trisphosphate (IP3) and diacylglycerol (DAG), important signaling molecules involved in many cellular processes including Ca2+ release from the endoplasmic reticulum (ER). PLCG1 encodes the PLCγ1 isozyme that is broadly expressed. Hyperactive somatic mutations of PLCG1 are observed in multiple cancers, but only one germline variant has been reported. Here, we describe seven individuals with heterozygous missense variants in PLCG1 [p.(Asp1019Gly), p.(His380Arg), p.(Asp1165Gly), and p.(Leu597Phe)] who present with hearing impairment (5/7), ocular pathology (4/7), cardiac septal defects (3/6), and various immunological issues (5/7). Acute lymphoblastic leukemia in children is a bone marrow and blood cancer. Healthy blood cells begin to change and grow out of control when leukemia initially occurs. Acute lymphoblastic leukemia (ALL) is an immune system-related malignancy of the lymphocytes, a subtype of white blood cells. The most common form of cancer affecting children is ALL. In this work, the topological indices are derived from the structure of a chemical graph, and predict the physical, chemical, and biological characteristics of several anticancer drugs. This study aimed to determine the input layer neurons for the Generalized Regression Neural Network (GRNN) model by using various classical methods, namely 1) Partial Autocorrelation Function (PACF), 2) frequency-based method, 3) frequency and Forward Selection (FS), 4) frequency and Backward Elimination (BE), 5) frequency and step-based methods, and 6) frequency method combined with the Least Absolute Shrinkage and Selection Operator (LASSO). These classical methods were combined with various parameters within GRNN, including smoothing parameters, forecasting strategies, and transformations. Glaucoma, a chronic eye disease characterized by progressive optic nerve damage, remains a significant public health concern worldwide. In the diagnosis of skin cancer, dermoscopy images have to be evaluated for feature extraction. In this paper, the authors design a new method to enhance the skin dermoscopy image segmentation accuracy by eliminating artifacts and hair using photometric quasi-invariants, followed by a unique approach for skin lesion segmentation using histogram-based feature fusion. Neurofibromin/NF1 is a RAS (rat sarcoma virus) GTPase activating protein and estrogen receptor (ER) transcriptional corepressor. NF1low status, identified by copy number loss or low mRNA/protein expression is associated with endocrine therapy resistance in approximately 20% of ER+/HER2− (human epidermal growth factor receptor 2) early-stage breast cancers. The identification of targeted treatments for NF1low ER+/HER2− breast cancer is therefore a priority. In this study proteogenomic analysis of ER+/HER2− breast cancer demonstrated that NF1low tumors exhibited elevated cyclin-dependent kinase 4/6 (CDK4/6) activity. Introduction: Research indicates a strong correlation between obesity and the risk of dementia, both are linked to steroid receptor coactivator-1 (SRC-1), a transcriptional coactivator.
Methods: We used RNA sequencing analysis (RNA-Seq) to investigate the transcriptome of SRC-1-KO mice, and identified S100 calcium-binding protein A6 (S100A6), an AD associated gene, as one target of SRC-1. We tested cognitive behaviors in SRC-1-KO mice and mice with a humanized SRC-1 mutation (SRC-1L1376P), and performed promoter luciferase assays on S100A6.
Results: Loss of SRC-1 caused alterations in gene signatures that are commonly associated with neurodegenerative diseases, including AD, and diminished the neural plasticity of …

This study aimed to evaluate market opportunities and formulate strategic pathways for the sustainable growth of Betong chicken, an indigenous premium poultry breed in southern Thailand known for its distinctive meat quality. Recent declines in the Betong chicken population and productivity have created a supply–demand imbalance, threatening the Betong chicken’s long-term economic viability. Using a mixed-methods design, data were collected from 280 stakeholders—farmers, processors, retailers, and consumers—in Pattani, Yala, and Narathiwat provinces. The 2007 excavation at the Knights Hotel Site yielded about 170 metal finds, mostly iron or copper alloy, though silver and gilded items were also found. Some objects associated with warfare may have been linked to the final battle of ‘Akko in 1291 CE, but most of the finds relate to daily life, encompassing activities such as horse keeping, dress, foodways, medicine and construction. The dominance of Eastern-type arrowheads indicates that local traditions were sometimes adopted, certainly for the arrows if not the bows. The Crusader-period ceramic assemblage unearthed in the Knights’ Hotel site in ‘Akko contains a large variety of daily wares. The assemblage consists of local and imported wares from various regions, some of which were identified through petrographic analysis. The imported wares indicate trade connections with Mediterranean port cities, and the petrographic analysis identified a new group of vessels from Egypt’s northern Nile region. The article outlines the pottery typology and quantification, and presents the ceramic distribution within the excavated urban area. The findings indicate changes in land use, as some residential and commercial areas were converted into dumps and cesspits. …

This report details the second season of excavations at the Knights’ Hotel site, carried out in 2007, covering three areas (A–C) not excavated in the first season (1995). The excavations revealed domestic and public(?) architecture dating from the thirteenth century CE. An excavation southwest of Tel Gamma exposed two Iron Age II updraft pottery kilns and a refuse pit. The eastern kiln was only partially preserved; its fill contained workshop debris and storage-jar wasters. The better-preserved western kiln retained an almost intact, elongated combustion chamber with two sets of mud-brick flues and evidence of periodic upkeep. An activity layer was identified between and around the kilns, from which a mixed Iron IIA–C ceramic assemblage and a few stone artifacts were collected. A Middle Bronze Age IIA pottery production site was uncovered on the northern bank of Naḥal Soreq, near Yavne-Yam. Excavations revealed installations, including pits, an oven, a stone pavement and a two-story pottery kiln. The discovery of a potter’s wheel and petrographic analysis confirm local ceramic production using nearby ḥamra and kurkar materials. Stone anchors found on-site suggest the transport of vessels via small boats along the stream, connecting the site with nearby settlements such as Nabi Rubin and Yavne-Yam.

The rural site of Naḥal Rimmonim yielded 421 identifiable animal bones that originated in well-stratified loci from five strata dating to the Intermediate and Middle Bronze Ages. The site’s economy seems to have been independent, or production-based, with residents providing most of their own needs while also trading with other groups. The economy was based on animal husbandry, with some hunting, mostly of gazelles, alongside the production of secondary products, such as milk, wool and labor, with a preference for wool. Stratified remains—including domestic structures, installations, a pottery workshop and burials—demonstrate diachronic shifts in site function and organization. The Intermediate Bronze Age strata exhibit substantial domestic activity with evidence of localized adaptation to marshy conditions. Three Intermediate Bronze Age wicks discovered in the Newe Efrayim cemetery constitute some of the earliest known archaeological evidence of wick use, an essential object for lighting, yet lacking distinctive features, as it was intended to be burned, making it a rare find. The exceptional preservation of the wicks sheds light on both preservation issues and the technological aspects of wick production in antiquity. The Late Chalcolithic period in the Southern Levant, mainly the Ghassulian culture (mid-fifth–mid-fourth millennia BCE), heralded a new and unique trajectory in craft specialization, with ivory objects, including figurines, tools and vessels, among the outstanding ones. Signaling by norepinephrine (NE) via adrenergic receptors (ARs) mediates attention, yet the underlying molecular mechanisms are largely unknown. AMPA receptors (AMPARs) form a complex with β2ARs, Gs, adenylyl cyclase, and protein kinase A (PKA) to augment AMPAR phosphorylation and, thereby, surface expression. We show that signaling by intracellular β2ARs is required for these effects and two different forms of long-term potentiation (LTP) that depend on β2AR signaling and phosphorylation of the AMPAR GluA1 subunit on S845. Oocyte vitrification success is highly dependent on cryoprotectant agent (CPA) choice as well as possible additives that could help preserve mitochondrial membrane potential (mMP), reactive oxygen species (ROS) production, and calcium homeostasis within the cell. It is a delicate balance between levels needed for proper oocyte function and activation and levels needed to induce apoptotic pathways. Heart failure is a leading cause of death worldwide, and even with current treatments, the 5-y transplant-free survival rate is only ~50 to 70%. As such, there is a need to develop new treatments for patients that improve survival and quality of life. Recently, there have been efforts to develop small molecules for heart failure that directly target components of the sarcomere, including cardiac myosin.
