The MAPK pathway can drive resistance in highly aggressive breast cancers. Our previous work showed that the MEK inhibitor (MEKi) AZD6244 (selumetinib) prevented lung metastasis in a breast cancer xenograft model. In clinical studies, MEKis as single agents have had only modest activity against solid tumors due to the onset of resistance. Using synthetic lethality siRNA screening, we identified myeloid cell leukemia-1 (MCL-1) as a potential contributor to AZD6244 resistance. We hypothesized that MCL-1 promotes MEKi resistance in highly aggressive breast cancers and that MCL-1 inhibition overcomes AZD6244 resistance. The ataxia telangiectasia and RAD3-related (ATR) kinase functions with ataxia telangiectasia-mutated (ATM) kinase as a modulator of DNA damage response (DDR). We assessed the antitumor effects of the ATR inhibitor elimusertib (BAY-1895344) in patient-derived xenograft (PDX) models with DDR alterations. Antitumor activity was assessed by change in tumor volume (TV) from baseline. Responses were categorized as follows: partial response (PR), ≥30% decrease in TV; ≥20% increase in TV, progressive disease; and non-PR/progressive disease, stable disease (SD). Event-free survival was defined as time for tumor doubling (EFS-2). Of 21 PDX models tested, 11 had significant prolongation of EFS-2 with elimusertib monotherapy. …

The outflow of 'dirty' brain fluids from the glymphatic system drains via the meningeal lymphatic vessels to the lymph nodes in the neck, primarily the deep cervical lymph nodes (dcLN). However, it is unclear whether dcLN drainage is essential for normal cerebral homeostasis. Using dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) and computational fluid dynamics, we studied the impact of long-term mechanical stress from compromised dcLN drainage on brain solute and fluid outflow in anesthetized rats. We found that in young, but not middle-aged, rats, impairment of dcLN drainage was linked to moderately increased intracranial pressure and the emergence of extracranial …

University Of Plymouth, Plymouth, UKPeople living with obesity have a greater chance of developingcomplications during pregnancy and are over-represented inmaternal mortality data relating to both direct and indirect causesduring pregnancy and up to a year after the birth of their child. Inaddition, epigenetic influences result in metabolic changes inoffspring of people living with obesity leading to a greater chanceof them experiencing long-term health consequences. In view ofthese health concerns, people living with obesity are advised to loseweight in the preconception period, before becoming pregnant.Sustained weight loss, however, can be difficult to achieve, andpeople do not always seek preconception advice. Purpose: While PSMA-targeted radioligand therapy (RLT) has shown remarkable efficacy for treating end-stage prostate cancer, the α-emitting RLT often results in severe salivary gland toxicity, limiting its use. Various strategies to mitigate this side effect have been attempted with limited success. Accordingly, this study introduced a new PSMA-targeting ligand with more favorable binding characteristics than the existing ligands. Procedures: The binding affinity of PSMA-1-DOTA to PSMA was compared with that of PSMA-11 and PSMA I&T. The ubiquitous fungal pathogen Candida albicans has the potential to either asymptomatically colonize the gastrointestinal (GI) tract or become an invasive pathogen through mechanisms that remain incompletely understood. Here we explored the fungal, host, and environmental factors that influence the ability of C. albicans to colonize the mouse GI tract using a representative clinical strain, CLCA10. After a single gavage challenge (5 × 106 CFU C. albicans), specific pathogen-free (SPF) mice remained colonized with C. albicans strain CLCA10, but not other Candida species, for at least 58 days with the fungus confined largely to the gut luminal …

Background: Mitis group non-pneumococcal streptococci (MGNPS), specifically Streptococcus mitis, Streptococcus infantis and Streptococcus oralis, have recently been shown to cause pneumonia and/or bacteremia. These organisms often have capsular (cps) operons resembling those in pneumococci, and some express cps-generated polysaccharides that antigenically cross-react with pneumococcal capsular serotypes. But, to date, a series of MGNPS isolates has not been studied by electron microscopy (EM) for the presence of a capsule.
Methods: We studied 21 MGNPS; 11 were isolated from sputum and determined to have caused pneumonia, 3 were isolated from blood, and 7 were commensal isolates cultured …

The Onondaga Nation is petitioning the Organization of American States (OAS) for land rights to Onondaga Lake, a notoriously polluted body of water in Central New York State. The Onondaga Nation is one of six nations of the Haudenosaunee Confederacy (Iroquois), and Onondaga Lake is the sacred site where Gayanashagowa, the Great Law of Peace, was established. As Keepers of the Central Fire, the Onondaga Nation bears the responsibility of maintaining unity and peace within the Haudenosaunee Confederacy. In 2021 during the global Covid-19 lockdowns, Billy and Maria ran an IRB-exempted online survey, looking to hear from fans who attend anime conventions. Conventions had been shut down as non-essential services that drew large crowds, and we hoped to capture a screenshot of this moment, to better learn from it in the future. And the resulting data collection went quite well – we were able to partner with a major anime organization to share the survey, and our 1000+ respondents had a lot to say about the conventions they were missing during lockdowns.

Make It Pink: Superman, Pink Kryptonite, And Fandom, Tom Ue, Kristofer Starzomski-Wilson

This paper explores several representations of pink kryptonite. In Doug Murphy’s animated short film True Colors (2017), Superman (Jason J. Lewis) transforms into a woman when he’s exposed to this substance. This short, as we have shown elsewhere (2025), offers a useful lens for examining the perpetuation and the critique of gender stereotypes in superhero media: Superman may be equally capable regardless of his sex, but the film pokes fun at, rather than celebrates, his transformation. Pink kryptonite rarely appears in the canon. A single panel in Supergirl (2003) has received perhaps the most sustained attention. Tom Ue’s archival work …

This study investigates whether German-language fan fictions featuring lesbian and gay characters within the anime and manga domain can be regarded as genuinely queer. It situates these fan works within the specific traditions and practices of German fandom. Employing qualitative content analysis, the research examines two prominent fandoms, Naruto and Sailor Moon, focusing on the presence of heteronormative narrative patterns in fan fiction with queer protagonists. The study analyzes which aspects of the original media are adapted and how these adaptations shape the portrayal of queer identities. The sensory environment plays a critical role in shaping learners’ attention, engagement, and retention. This thesis investigates how varying levels of sensory input in virtual reality (VR) classrooms influence learning for two age groups—children and young adults—and explores how individual differences in sensory processing moderate these effects. Using immersive VR technology to simulate classroom settings, two experimental studies were conducted. Study One involved 60 children aged 6–15 years, recruited primarily through a university-run summer camp. Post-Finasteride Syndrome (PFS) is a controversial illness associated with a host of distressing symptoms—most notably sexual dysfunctions—reported to persist long beyond the intake of finasteride itself. It appears the PFS population consists exclusively of younger patients with alopecia, even though millions take a higher (if pharmacologically similar) dosage of finasteride for benign prostatic hyperplasia. This dichotomy is not what one would expect to observe if finasteride caused PFS. No medical explanation of this one-sided syndrome exists. The surge of cases of PFS following a publicity spike in 2011 and 2012 suggests that PFS owes much nonbiological factors. The present analysis …

The U.S. Environmental Protection Agency (EPA) regulates pesticide use in the United States. The EPA is charged by the Federal Insecticide, Fungicide, and Rodenticide Act (FIFRA) with ensuring that a pesticide will not cause unreasonable adverse effects on the environment. Incident reports (documentation of exposure and injury from pesticide applications) can serve as a reality check on the pesticide registration decisions made by the EPA scientists and risk managers. The EPA collects incident reports on human, domestic animal, and ecological injury. Each winter in Florida, thousands of manatees gather in the warm water outflows of power plants. These refuges play a crucial role for the Florida manatee: when ocean temperatures drop, manatees shelter at these artificial sources to survive. But in recent years, thousands of manatees wintering at the Florida Power & Light Company’s Cape Canaveral Clean Energy Center have died. The reason? Seagrass beds––a primary food source for manatees––are vanishing, killed off by pollution from agricultural, industrial, and residential sources.

Hybridization is common among minnows and shiners in the family Leuciscidae, and mitonuclear discordance can reveal evidence of historical hybridization and introgression events that have shaped extant species and populations. We have identified a notable case of serial mitogenome replacement in populations of two shiner species, Luxilus zonatus and L. chrysocephalus, which are syntopic in drainages throughout the northern and eastern Ozark Interior Highlands of North America. These mitogenome replacement events involved L. zonatus acquiring the mitogenome of L. chrysocephalus, and populations of L. chrysocephalus acquiring the mitogenome of an allopatric congeneric species, L. cornutus. More areas across the globe are becoming affected by water stress or water scarcity. The decrease in available freshwater due to overconsumption reveals the direct need to consider other sources for necessities such as drinking water or food production. Agricultural irrigation uses 70% of the world’s freshwater, making the focus on safe and reliable reclaimed water for agricultural reuse imperative. However, contaminants of emerging concern (CECs), such as antibiotics and their resulting antibiotic resistance genes (ARGs), can spread and affect human health if reclaimed water is not treated to the proper level. This syllabus is for a survey course that combines common themes in American history since 1865 with a specific focus on civic engagement and voting rights. It is centered around the stories of everyday Americans who participated in the Progressive Movement, the Civil Rights Movement, labor rights, and other turning points in American history related to civic engagement. The syllabus and course assignments highlight the role of oral history in preserving stories and introduce students to the methods and impact of historical advocates for justice.
