The gene BIN1 is the second-largest genetic risk factor for late-onset Alzheimer’s disease (LOAD). It is expressed in neurons and glia in the brain as cell-type specific and ubiquitous isoforms. BIN1 is an adaptor protein that regulates membrane dynamics in many cell types. Previously, we reported that BIN1 predominantly localizes to presynaptic terminals in neurons and regulates presynaptic vesicular release. However, the function of neuronal BIN1 in relation to LOAD is not yet fully understood. Background: Osteosarcoma (OS) lung metastases remain a significant therapeutic challenge. Innate immune activation is a promising therapeutic approach. Innate immune agonists can modulate the tumor immune microenvironment and improve therapeutic response.

Methods: Using an experimental syngeneic OS lung metastasis BALB/c mouse model with K7M3-luc OS cells, we evaluated the antitumor effects of yeast-derived particulate β-glucan in prevention and therapeutic settings. We then assessed whether the CD40 agonist (CD40a) in combination with β-glucan increased therapeutic response in two different immune-competent mouse models of OS lung tumor burden.

Singing A Wide Heart: Timbre And Sung Prayer In Senegal’S Layène Community, Margaret L. Rowley

Layène sung prayer (called sikkar) is embedded within the religious practices of the Layène community, a minority Sufi order in Dakar, Senegal. This article explores the functions of Layène sung prayer, focusing especially on its loud, vibrational timbre. Both men and women vocalists in call-and-response sing loudly, using wide vibrato and electronic amplification to ensure that the sikkar reaches the inhabitants—both human and non-human—in the surrounding community. This article demonstrates that the distinctive timbre of Layène sung prayer serves the spiritual functions of binding the community together in spiritual and social cohesion, increasing individual and collective generosity, and demarcating …

This article is a media history and contemporary case study of The Angelus Temple in Los Angeles, founded by Aimee Semple McPherson in 1923. My analysis of the Angelus insists upon the centrality of a hyperaudible paradigm of worship that uses media to maximize the legibility of a Christian message, and, more than this, imagines that media itself as a living soul in need of redemption. This essay argues that within the socio-cultural context of North American Christianity, the timbral qualities of electric guitar, with its long lineage as a site of cultural values conflict and identity negotiation, serves a theological function in the emerging and cohering sonority of digital worship. Using as comparative case studies the timbral qualities of delay and ambience as promoted and sold by Worship Tutorials as representing normative sonic values for these Christians, on the one hand, and the dialectic and schismatic deployment of distortion by the band My Epic on the other, we suggest that these timbral qualities become important …

How might attending to timbre shift the boundaries of the experiences and practices that come under the analytical purview of religion? What timbral qualities render prayer efficacious and invite divine encounter? What cosmological claims and social relations are forwarded through sounds invoked in religious rituals and gatherings? How do marginalized voices and instruments shape worshipping communities? On the other hand, how are dominant identities and attendant structures reified through religiously marked timbre? Such questions about the character of sounds and their implications for religion animate this issue of the Yale Journal of Music & Religion.

Infanticide is one of the key factors in explaining changes in human populations since the antiquity. This paper examines dynamics of infanticide practices among the Ngoni of Tanzania from the late pre- colonial to the end of German colonial rule in 1916. Relying on primary and secondary sources, it argues that while infanticide remained a critical problem in Songea from the 19th century to the end of German colonialism in 1916, the context in which it was practiced and the interventions devised to alleviate it changed over time. Cardiac inflammation and hypertrophy develop as a pathologic response to an array of insults, such as myocardial infarctions, chronic systemic hypertension, and valvular defects. Due to the high prevalence of such conditions, there is an increasing need to prevent and halt cardiac hypertrophy. Because cardiac damage and subsequent remodeling can lead to arrhythmias, heart failure, and even sudden cardiac death, inhibition of cardiac hypertrophy is key to reducing cardiovascular-related mortality. To determine if olfactory primes have an influence on risky behavioral intentions, a study was conducted on college students. Participants (N=64) were primed with the smell of either beer, rubbing alcohol, or a control condition followed by a survey that asked questions related to risk-taking behavior intentions with 5 subscales (ethical, financial, health/safety, recreational, social) as well as sexual risks. It was predicted that college students who are exposed to the smell of beer are more likely to report intentions of engaging in risky behaviors, including unprotected sexual activity. Genetic Engineering Of Esophageal Organoids: Crispr-Based Knock-In For Cell Lineage Tracing, Kyung-Pil Ko, Jae-Il Park

Esophageal organoids serve as powerful systems to study epithelial lineage hierarchies and cancer biology. Gene manipulation in these organoids has traditionally involved overexpression or knockout strategies. However, CRISPR-/Cas9-based knock-in (KI) approaches now enable precise cell lineage tracing and live imaging. Here, we describe protocols to generate fluorescent KI organoids from murine esophageal epithelium by tagging Krt13 (BFP) and Sox2 (mNeon). These dual-reporter organoids allow direct monitoring of growth dynamics and differentiation trajectories. We outline CRISPR/Cas9 design, donor construction using homology-independent approaches (CRISPaint), delivery into organoid cells, enrichment and single-clone isolation, and validation by fluorescence. Blastocystis is the most ubiquitous intestinal protozoan parasite found in humans and animals worldwide. Commonly, ST7 and ST6 are the major Blastocystis subtypes reported in birds, and their frequencies fluctuate greatly with various factors. However, little literature is available on the prevalence and subtype characteristics of this parasite in free-living wild birds in the same city. For this purpose, a total of 114 fresh fecal specimens were collected from wild free-living birds in urban districts of Xinxiang city and tested for Blastocystis using PCR of the barcoding region of the small subunit ribosomal RNA (SSU rRNA) gene. This paper compares various codes and standards for the behavior of “Concrete-Filled Steel Tubes” (CFST) columns under fire and axial load. The codes evaluated include European standards (EC4.2 and EC3.2), the American standard (AISC 360), and the Australian standard (AS/NZS 2327). The fire resistance of Concrete-Filled Steel Tubular (CFST) and steel hollow (SH) columns was assessed using a computer program developed with Visual Studio .NET. The program's results were compared with previous experimental data to evaluate the accuracy of various codes and standards in predicting the behavior of these columns under fire conditions. Eleanor Arnason’s Fantastic Mitochondrial Humor

In this essay I explore Brian Attebery’s description of mitochondrial fantasy to explain how humor works in Eleanor Arnason’s short fantastic fiction. Mitochondria are the energy powerhouses in cells. They are symbiotic creatures that once were separate but became an evolutionary part of a female egg cell, providing a history that can be traced to a Mitochondrial Eve. In Fantasy: How It Works, Attebery uses mitochondria metaphorically to explain how fantastic ideas evolve. Attebery describes fantastic mitochondria as “a cellular dynamo that lives within the tissues” of a story (91). He demonstrates how stories lead …

Some interpreters of The Wind in the Willows have suggested that Ratty’s beloved river should be understood as a symbol of the imagination. Whether this is true or not, we may benefit from thinking of Grahame’s own imagination as a river that derives much of its force from both Classical and Romantic tributaries. In what follows, I will explore the lower reaches of these tributaries, not only to contextualize and illuminate The Wind in the Willows, but also to address several persistent questions about its composition and reception. Welsh writer Saunders Lewis was, just as his almost exact contemporary J.R.R.Tolkien, a politically conservative Catholic with a deep interest in medieval myths. Both authors wrote fantasy based on Celtic tales, such as Lewis's play Blodeuwedd and Tolkien's poem The Lay of Aotrou and Itroun. While the latter follows the dicta of Tolkien's own "On Fairy-stories," Lewis created a more modernist work. Despite biographical and ideological similarities, the two approached their sources differently. Tolkien looked for unifying aesthetic principles that he called "unhazy unromantic momentariness," while Lewis used the traditional material to escape the constraints of romantic and realistic …

Tolkien’s portrayal of Lothlórien is layered with complexities that reflect broader tensions between the feminine landscape and the masculine impulse to understand, name, and rationalize the world. This article examines the ‘fight’ over Lothlórien and its entitlement to truth from both female and male perspectives. It combines a close reading of passages from The Fellowship of the Ring with theories of name-giving, power, and mapping. The article concludes by evaluating these insights through the lens of Kristeva’s concept of the “abject” and discussing what ‘queer’ might mean in the Tolkien oeuvre.

While the River-daughter is a prevalent poetic name for Goldberry, Tom introduces her as the River-woman’s daughter when singing and skipping along the banks of the Withywindle. The River-woman variant occurs only once in Goldberry’s narrative but is not easily dismissed. The contrast between the two variant names raises several questions on how to read Goldberry’s enigma.
