Laozi’s “Daodejing”: A New Translation With Environmentalist Commentary offers a new translation and fresh interpretation of the eighty-one-verse Daodejing, one of the central texts of Eastern philosophy. Likely written during the late Zhou Dynasty between 600–400 BCE, this foundational work is generally attributed to an individual named Laozi, although it is unlikely that any such person actually existed. he invention of pottery played a crucial role in human history, promoting human settlement and the transition to an agricultural society [1,2]. Pottery first emerged in East Asia during the Late Pleistocene/Paleolithic across a diverse range of environmental settings, becoming more widespread with the climatic warming of the Holocene/Neolithic [3]. The relatively cold Mongolian Plateau was a key region for early human adaptation. Chimeric antigen receptor (CAR)-engineered invariant NK T cells (CAR-NKT) are a novel cell platform for cancer immunotherapy. Unlike conventional T cells, NKTs are characterized by innate antitumor properties, minimal alloreactivity, and a unique ability to modulate the tumor microenvironment. This article provides a comprehensive overview of preclinical and early clinical studies evaluating CAR-NKTs in both autologous and allogeneic clinical settings. We discuss the contributions of CAR signaling domains, cytokine coexpression, and other functional measures that correlate with CAR-NKT persistence, function, and metabolic fitness. Mitochondrial metabolism orchestrates T cell functions, yet the role of specific mitochondrial components in distinct T cell subsets remains poorly understood. Here, we explored the role of mitochondrial complex II (MC II), the only complex from the electron transport chain (ETC) that plays a role in both ETC and metabolism, in regulating T cell functions. Surprisingly, MC II exerts divergent effects on CD4+ and CD8+ T cell activation and function. Using T cell-specific MC II subunit, succinate dehydrogenase A-deficient (SDHA-deficient) mice, we integrated single-cell RNA-seq and metabolic profiling, with in vitro and in vivo T cell functional assays to illuminate …

The novel long non-coding RNA (lncRNA) Leat1 is extraordinarily conserved in both its location (syntenic with EfnB2, an essential gene in anogenital patterning) and sequence. Here we show that Leat1 is upregulated following the production of testosterone from the developing testis in mice and interacts with EfnB2, positively regulating its expression. Leat1 expression is suppressed by estrogen, which in turn suppresses the expression of EfnB2. Moreover, the loss of Leat1 leads to reduced EfnB2, resulting in a severe hypospadias phenotype. The human LEAT1 gene is also co-expressed with EFNB2 in the developing human penis, suggesting a conserved function for this …

Fisheries managers work with the three components of fisheries—biota, environment, and people - to achieve recreational, stewardship, and socioeconomic goals. Although manipulation of biota and the aquatic environment remains popular (e.g. harvest regulation, protecting or enhancing habitats, and guarding water quality), the human dimension of fisheries must also be considered throughout the fisheries management process. In this study, we explored the intersection of performing arts and entrepreneurship through an autoethnographic study of a new arts venture, theLAB*, which participated in the Von Allmen Center for Entrepreneurship's accelerator program at the University of Kentucky. In creating theLAB*, we investigated the feasibility of establishing a dance-forward performing arts presenting organization within a profit-driven entrepreneurial ecosystem. As a multi-generational team of arts managers, we analyzed the experience utilizing scenario-planning and the DIEP (Describe, Interpret, Evaluate, Plan) model of reflection to assess the challenges and benefits of participating in this environment. Spatial omics (SO) produces high-definition mapping of subcellular molecules within tissue samples. Mapping transcripts to anatomical regions requires segmentation, but this remains challenging in tissue cross-sections with tubular structures like axons in peripheral nerve or spinal cord. Neural networks could address misidentification but are hindered by the need for extensive human annotations. We present SiDoLa-NS (Simulate, Don't Label-Nervous System), an image-driven (top-down) approach to SO analysis in the nervous system. We utilize biophysical properties of tissue architectures to design synthetic images of tissue samples, eliminating reliance on manual annotation and enabling scalable training data generation. Although 3D face generation is extensively studied in computer vision, most existing methods prioritize reconstructing 3D geometry from available 2D or 3D inputs rather than generating novel faces directly from latent representations. To bridge this gap, we present the application of Adversarial Volumetric Convolutional Neural Networks (AVCNN), a tailored adaptation of the vanilla 3D Generative Adversarial Network (3D-GAN), to 3D face generation using latent space Gaussian embeddings. We first assemble a custom 3D facial dataset to provide the requisite facial characteristics and to ensure sufficient coverage of geometric variation across identities. Improving measurement of sexual violence has contributed to major policy changes in the United States. However, research on sexual violence largely overlooks those exposed to conflict and displacement: a population especially at risk. Assessment measures often ignore contextual features and use phrasing that is vulnerable to underreporting. This community-based participatory research project collaborated with cultural experts from Mexico/Central America and Afghanistan to develop a preliminary item list for a self-report measure of prior sexual violence. As cannabis use has increased steadily over the past decade, there is critical need to evaluate early cannabis use as a potential risk factor for the development of psychosis. Neurobehavioral mechanisms underlying novelty salience could clarify how cannabis exposure may influence subclinical psychotic-like experiences (PLEs). The present study leveraged dynamic functional connectivity and task-fMRI to assess the role of novelty processing in cannabis use and PLEs. A group of brain regions dynamically tied to the salience network while viewing short films were found to increase their activation as a function of stimulus novelty during a novelty bandit task. This thesis explores Alexis de Tocqueville's theory of democratic armies through the lens of the Mexican-American War (1846-1848), testing his predictions about how democratic social conditions influence military institutions and performance. Tocqueville argued that armies in democratic societies would face tensions between egalitarian ideals and military discipline, with citizen-soldiers resisting hierarchy while professional officers, often isolated from civil society, grew dangerously ambitious for war. He predicted that democratic armies would show energetic initiative but struggle with weak formal discipline, and that equality would fundamentally change the relationship between the military and the broader society. In this MA Thesis, I lay out a synthetic overview of polemical French writer Guillaume Dustan's (1965-2005) political philosophy. He is criticized first as irresponsible and dangerous for his defense of barebacking (sex without condoms) in the midst of the AIDS epidemic, and then, after his death, as a neoliberal/anarcho-capitalist for his focus on individual liberty. My overview touches on Dustan's Freudian belief that repression is the root of all social ills, his functional ethics of self-responsibility, the revaluation of heteronormative Judeo-Christian morality towards 'virtusex,' and his social contract that minimizes repression. Intermittent streams are increasingly recognized as vital components of river networks by supporting dynamic fish communities shaped by variable hydrology and habitat connectivity. They are becoming increasingly vulnerable to fragmentation by anthropogenic activities, namely road crossings, dams, and climate shifts that intensify seasonal drying. In response, I conducted two studies in the Illinois Bayou watershed in Arkansas to assess the effects of fragmentation on fish community dynamics and morphology in intermittent streams.

Patient-Derived Ovarian Cancer Models Provide Insight Into Cancer Cell-Intrinsic And Immune Cell-Dependent Effects On Therapeutic Response, Parisa Nikeghbal

Ovarian cancer is the most lethal gynecologic malignancy, largely due to late diagnosis and resistance to chemotherapy. This dissertation applies patient-derived organoids (PDOs), xenograft-derived organoids (PDXOs), and humanized xenografts (huPDX) to investigate therapeutic responses and immune interactions. Organoids were shown to retain critical features of the original malignancy. Across platinum-sensitive, -resistant, and -refractory cases, both PDOs and PDXOs exhibited drug sensitivities that aligned with patients’ clinical classifications. Building on this foundation, a drug screen revealed novel strategies. Auranofin restored platinum sensitivity in Notch3-active models, afatinib broadly reduced viability, and adavosertib–gemcitabine and sorafenib–topotecan combinations produced synergy. Injured ligaments are often repaired with tendon grafts which may over or under constrain the joint; there is a need for an artificial graft option with the mechanical properties of a native ligament. Crimped fibers within ligaments allow for recruitment which means the viscoelastic properties of stress relaxation and creep are not inversely related. A feasibility study using ACLs and large-scale crimp models to study viscoelasticity, led to an in-depth analysis of the scapholunate interosseous ligament. This document explores the intersection between birdsong and piano music using four cases studies from the piano repertoire in the Western classical tradition. The Dynamic Admittance Parameterization of Non-Prehensile Multi-Robot Trans- port with Optimal Coordinated Planning (DYNAMO) architecture offers a practical framework for cooperative payload transportation using two robots equipped with nonholonomic mobile bases and four-degree-of-freedom manipulators. Coordinated mobile manipulation is a difficult problem in robotics, and the non-prehensile case is even more challenging than its prehensile counterpart because the robot bases and the payload are dynamically coupled. DYNAMO adapts arm motion in response to interaction forces and generates coordinated base trajectories that account for this coupling. Pulmonary hypertension (PH) is characterized by increased pulmonary arterial constriction, pulmonary arterial remodeling, and right ventricular dysfunction. While metabolic and mitochondrial derangements are increasingly recognized as central pathogenic features, the underlying mechanisms and effectors are not yet fully understood. Acid-sensing ion channel 1a (ASIC1a) is a proton-gated cation channel involved in the development of chronic hypoxia (CH)-induced PH. ASIC1a represents a potential candidate involved in metabolic-mitochondrial dysfunction, as metabolic byproducts can act as ligands for ASIC1a.
