Asthma is a complex pulmonary disorder with multiple pathological mechanisms. A key pathological feature of chronic asthma is airway remodeling, which is largely attributed to airway smooth muscle (ASM) hyperplasia that contributes to thickening of the airway wall and further drives asthma pathology. The cellular processes that mediate ASM cell proliferation are not completely elucidated. Using multiple approaches, we demonstrate that the adapter protein Abi1 (Abelson interactor 1) is upregulated in ∼50% of ASM cell cultures derived from patients with asthma. Loss-of-function studies demonstrate that Abi1 regulates the activation of Jak2 (Janus kinase 2) and STAT3 (signal transducers and activators …

Rheumatoid arthritis (RA)-associated interstitial lung disease (RA-ILD) is the most common pulmonary complication of RA, increasing morbidity and mortality. Anti-citrullinated protein antibodies have been associated with the development and progression of both RA and fibrotic lung disease; however, the role of protein citrullination in RA-ILD remains unclear. Here, we demonstrate that the expression of peptidylarginine deiminase 2 (PAD2), an enzyme that catalyzes protein citrullination, is increased in lung homogenates from subjects with RA-ILD and their lung fibroblasts. Chemical inhibition or genetic knockdown of PAD2 in RA-ILD fibroblasts attenuated their activation, marked by decreased myofibroblast differentiation, gel contraction, and extracellular matrix …

Omega-3 polyunsaturated fatty acids (n-3 PUFAs), obtained from diet and dietary supplements, have been tested in clinical trials for the prevention or treatment of several diseases. n-3 PUFAs exert their effects by activation of free fatty acid (FFA) receptors. FFA1 receptor, expressed in the pancreas and brain, is activated by medium- to long-chain fatty acids. Despite some beneficial effects on cognition, the effects of n-3 PUFAs on the blood-brain barrier (BBB) are not clearly understood. We examined the effects of FFA1 activation on BBB permeability in vitro, using rat brain microvascular endothelial cells (RBMVEC), and in vivo, by assessing Evans …

Adrenergic Modulation Of Precursor Cells Of Ovarian Cancer, Sweta Dash Feb 2022

Adrenergic Modulation Of Precursor Cells Of Ovarian Cancer, Sweta Dash

USF Tampa Graduate Theses and Dissertations

Ovarian cancer remains one of the most lethal gynecological cancer and ranks eighth in cancer-related mortality among women. The high mortality rate can be attributed to majority of the cases being diagnosed at advanced stages of the disease when the survival rate and overall prognosis are poor. There is lack of effective screening modalities to detect ovarian cancer at early stages. Hence, a better understanding of the molecular mechanisms leading to ovarian cancer initiation is required.

Epidemiological studies have reported that conditions which cause chronic behavioral stress are associated with a higher risk of developing ovarian cancer. The activation of …

Proposed hydropower dams at more than 350 sites throughout the Amazon require strategic evaluation of trade-offs between the numerous ecosystem services provided by Earth's largest and most biodiverse river basin. These services are spatially variable, hence collective impacts of newly built dams depend strongly on their configuration. We use multiobjective optimization to identify portfolios of sites that simultaneously minimize impacts on river flow, river connectivity, sediment transport, fish diversity, and greenhouse gas emissions while achieving energy production goals. We find that uncoordinated, dam-by-dam hydropower expansion has resulted in forgone ecosystem service benefits. Minimizing further damage from hydropower development requires considering …

BACKGROUND: An abrupt lung deflation in rodents results in lung injury through vascular mechanisms. Ventilator disconnections during endo-tracheal suctioning in humans often cause cardio-respiratory instability. Whether repeated disconnections or lung deflations cause lung injury or oedema is not known and was tested here in a porcine large animal model.

METHODS: Yorkshire pigs (~ 12 weeks) were studied in three series. First, we compared PEEP abruptly deflated from 26 cmH

RESULTS: As compared to PEEP 5 cmH

CONCLUSION: Single abrupt deflation from high PEEP, and repeated short deflations from moderate PEEP cause pulmonary oedema, impaired oxygenation, and increased PVR, in this …

During the Covid pandemic, the importance of wearing mask has been noted globally. Additionally, crowded human clusters facilitated the transmission of the virus, which brings up the need for new systems for monitoring such situations. To address such issues, this research proposes an object recognition visual system based on deep learning to monitor the mask-wearing in a certain space and the control of the number of people indoors as an important tool during an epidemic. This research mainly investigates two types of identification. The first is to monitor whether people entering the site wear a mask at the entrance and …

When “There Is” A Black: Levinas And Fanon On Ethics, Politics, And Responsibility, Leswin Laubscher Feb 2022

When “There Is” A Black: Levinas And Fanon On Ethics, Politics, And Responsibility, Leswin Laubscher

Middle Voices

This paper examines some of the ways in which the Black other, by Frantz Fanon’s articulation, complicates and challenges Emmanuel Levinas’s philosophies of ethics and justice. Additionally, it brings Levinas’s notion of the il y a, or “there is,” and Fanon’s “zone of nonbeing” into critical conversation with respect to the body and being of the Black other.

Ethics Of Interaction: Levinas And Enactivism On Affectivity, Responsibility, And Signification, Edward A. Lenzo Feb 2022

Ethics Of Interaction: Levinas And Enactivism On Affectivity, Responsibility, And Signification, Edward A. Lenzo

Middle Voices

In recent years, there have been a number of attempts to connect enactivism with the work of Emmanuel Levinas. This essay is such an attempt. Its major theme is the relationship between affectivity and ethics. My touchstones in enactivist thought are Giovanna Colombetti and Steve Torrances’ “Emotion and Ethics: an (inter-)enactive account” (2009) and the influential concept of participatory sense-making developed by Hanne De Jaegher and Ezequiel Di Paolo (2007). With respect to Levinas, I deploy major insights from Totality and Infinity and Otherwise than Being. I first show that enactivist thought (thus represented) and Levinas roughly agree on …

Ethics In The Breakdown: Levinas, Winnicott, And Schizoid Phenomena, Matthew J. Devine Feb 2022

Ethics In The Breakdown: Levinas, Winnicott, And Schizoid Phenomena, Matthew J. Devine

Middle Voices

This article addresses the common concern that Emmanuel Levinas’ ethics amounts to a life-denying, moral masochism. To the contrary, I demonstrate close resonances between Levinas’ project and that of the psychoanalyst D.W. Winnicott, for whom the purpose of therapy is to feel alive. In the first section, I trace the Levinasian subject’s coming to be out of the impersonal Il y a. Exploiting the object-relations undertones, I emphasize that the Levinasian subject comes to be as fastened, riveted, or bound to existence, and thereafter seeks to loosen its bond to its existence. In the second section, I discuss Winnicott’s …

King, Levinas And The Interruption Of Love: The Alchemy Of The Fire Fable, Claire S. Lebeau, Kaleb Sinclair

Middle Voices

Levinas begins Totality and Infinity with a haunting allusion from the 19th century French poet Arthur Rimbaud, “though the true life is absent”, we are in the world. This lamentation is a fitting beginning for his exposition of a radical reformulation of an Ethics that precedes all thought, language, or systematic attempts to cast morality as a Truth. Similarly, Martin Luther King Jr. presented a lamentation for a dream of a world where the transcendence of race, creed, or classification of any kind could allow children to grow up to be first ethical human beings in relation to one …

Toward A Crip Provenance: Centering Disability In Archives Through Its Absence, Gracen M. Brilmyer Feb 2022

Toward A Crip Provenance: Centering Disability In Archives Through Its Absence, Gracen M. Brilmyer

Journal of Contemporary Archival Studies

Using the records that document the 1893 World’s Columbian Exposition as a case study, this article discusses the messiness and unknowability of provenance. Drawing attention to how the concept of provenance can emphasize the reconstruction of a fonds when records have been moved, rearranged, and dispersed, this article draws attention to the ‘curative’ and ‘rehabilitative’ orientations of established notions of provenance. Put in conversation with disability studies scholarship, which critiques rehabilitating, curing, and restoring, this article outlines the theoretical scaffolding of a crip provenance: a disability-centered framework of resisting the desire to restore and instead meets records where they are …

An Ecophisiological Proposal To Manage Natural Grasslands: A Long Term Trial, F. L. F. De Quadros, D. Roberti, L. Marin, B. Kuinchtner, P. L. Nascimento

IGC Proceedings (1977-2023)

Natural grasslands on Southern Brazil comprise the so called “Rio de La Plata Grasslands” in South America. They are an important fodder source for ruminant pastoral systems and contribute to regional ecosystem services. Strength of these grasslands is its floristic diversity that poses a dilemma to farmers: how to choose management protocols that could be applied for hundreds of species. We propose to use a functional ecophysiological approach based on groups of grasses, the most abundant on aerial biomass of this natural grasslands. We clustered the most frequent grasses in two groups based on its leaf traits (leaf dry matter …

IGC Proceedings (1977-2023)

On pseudogamy process, pollen fertility affects endosperm development in Brachiaria apomictic hybrids. Also, it was well-known that self-incompatibility could affect the fertilization in cross pollination species. Evaluation of pollen viability and percentage of filled seed, and the demonstration of pollen induction technique for increasing filled seed percentage in Brachiaria spp. are presented in this paper. In the first experiment, 8 cultivars/breeding lines of Brachiaria, including 2 open-pollinated cultivars, an apomictic cultivar and 5 hybrids, were planted in randomized completely block design, with 3 replications at northeast Thailand. Each experimental unit consisted of 20 plants, 6 central plants were evaluated. …

Fluid and solute transporters of the retinal pigment epithelium (RPE) are core components of the outer blood-retinal barrier. Characterizing these transporters and their role in retinal homeostasis may provide insights into ocular function and disease. Here, we describe RPE defects in tvrm77 mice, which exhibit hypopigmented patches in the central retina. Mapping and nucleotide sequencing of tvrm77 mice revealed a disrupted 5' splice donor sequence in Slc4a5, a sodium bicarbonate cotransporter gene. Slc4a5 expression was reduced 19.7-fold in tvrm77 RPE relative to controls, and alternative splice variants were detected. SLC4A5 was localized to the Golgi apparatus of cultured human …

It has become evident that somatic mutations in cancer-associated genes accumulate in the normal endometrium, but spatiotemporal understanding of the evolution and expansion of mutant clones is limited. To elucidate the timing and mechanism of the clonal expansion of somatic mutations in cancer-associated genes in the normal endometrium, we sequence 1311 endometrial glands from 37 women. By collecting endometrial glands from different parts of the endometrium, we show that multiple glands with the same somatic mutations occupy substantial areas of the endometrium. We demonstrate that "rhizome structures", in which the basal glands run horizontally along the muscular layer and multiple …

There is a current need for enhancing our insight in the effects of antimicrobial treatment on the composition of human microbiota. Also, the spontaneous restoration of the microbiota after antimicrobial treatment requires better understanding. This is best addressed in well-defined animal models. We here present a model in which immune-competent or neutropenic mice were administered piperacillin-tazobactam (TZP) according to human treatment schedules. Before, during and after the TZP treatment, fecal specimens were longitudinally collected at established intervals over several weeks. Gut microbial taxonomic distribution and abundance were assessed through culture and molecular means during all periods. Non-targeted metabolomics analyses of …
