Variation in body fat distribution contributes to the metabolic sequelae of obesity. The genetic determinants of body fat distribution are poorly understood. The goal of this study was to gain new insights into the underlying genetics of body fat distribution by conducting sample-size weighted fixed-effects genome-wide association meta-analyses in up to 9,594 women and 8,738 men for six ectopic fat traits in European, African, Hispanic, and Chinese ancestry populations, with and without sex stratification. In total, 7 new loci were identified in association with ectopic fat traits (ATXN1, UBE2E2, EBF1, RREB1, GSDMB, GRAMD3 and ENSA; PATXN1 and UBE2E2 …
Influence Of Media Messages On Obesity And Health Perceptions Among African American Women, Carolyn Grant
Influence Of Media Messages On Obesity And Health Perceptions Among African American Women, Carolyn Grant
Walden Dissertations and Doctoral Studies
African American women have the highest prevalence of overweight and obesity in the United States, thus increasing their risks for chronic diseases. Their understanding of their health status and response to it could be triggered by messages in the media, yet few researchers have examined this topic with African American women. The purpose of this phenomenological study was to explore how advertisement and editorial content in media contributed to perceptions African American women have about overweight and obesity. Following the theoretical foundation of the social cognitive theory, the research questions explored what messages study participants encountered and which messages prompted …

“Mexico, Public Policy And Obesity In A Global Context”, Daniela Carina Bermudez
Master's Theses
Mexico has one of the most obese populations in the world. A country known for its diversity of rich flavorful food is drowning in low nutritional food products. This thesis examines Mexico’s obesity epidemic within the larger global context of international economic trade policies, public policies and Mexico’s health policies. The key research questions are 1) why is there an obesity epidemic in Mexico? and 2) what remedies should Mexico implement to control it? This thesis contributes to a viable policy strategy for the Mexican government to control and prevent the further increase of this obesity epidemic. Reviewing both the …

CONTEXT: Recent analyses of epidemiological data including the National Health and Nutrition Examination Survey (NHANES) have suggested that the harmful effects of obesity may have decreased over calendar time. The shifting BMI distribution over time coupled with the application of fixed broad BMI categories in these analyses could be a plausible "nuisance contributor" to this observed change in the obesity-associated mortality over calendar time.
OBJECTIVE: To evaluate the extent to which observed temporal changes in the obesity-mortality association may be due to a shifting population distribution for body mass index (BMI), coupled with analyses based on static, broad BMI categories. …
Spatial Distribution Of High Obesity Rates In South Carolina, Tom Bennett
Spatial Distribution Of High Obesity Rates In South Carolina, Tom Bennett
Earth and Environmental Sciences Presentations
In 2015, the rate of adult obesity in South Carolina was nearly 32%, which places South Carolina as the 13th worst state when it comes to the prevalence of adult obesity. Along with overall rates of adult obesity, South Carolina has the 8th highest high school student obesity rate with 16.3% of high school students being obese. These statistics are troubling because obesity is a primary cause of Type II Diabetes, high blood pressure, and many other negative health effects. These health care issues then affect supply and demand for health care, advertising, physical exercise, and many other market segments, …

The goal was to determine if a high fat diet given in a time restricted fashion can protect against obesity and inflammatory mediators. 12 week old C57BL/6 male mice were assigned to either a standard chow diet or a diet containing 45% fat. The high fat diet was provided ad libitum or in a time restricted fashion, either during the first of the second half of the active phase for 8 weeks. Time restricted meal intake reduced body mass gain, adiposity, glucose intolerance and cholesterol levels in mice fed a high fat diet. Mice fed a high fat diet, even …

Governmental expenditures are directed at a particular objective, but their effects have consequences far beyond the named target of the expenditures. Specific farm subsidies, for example, encourage consumption of particular foods by reducing the costs of producing these foods. To what extent do these subsidies affect the American obesity epidemic? How do the subsidies create disparate negative effects on those in poverty? Exploring these questions stimulates us to take greater care when designing legislation to take a broader look at the stakeholders affected by any particular governmental expenditure.
Harnessing Redox Regulation Of Ptp1b: A Novel Path To Therapeutic Design, Ioana Rus Rus
Harnessing Redox Regulation Of Ptp1b: A Novel Path To Therapeutic Design, Ioana Rus Rus
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
Protein tyrosine phosphatase 1B (PTP1B) is an important negative regulator of signaling by the insulin and leptin receptors. PTP1B-null mice are sensitized to insulin and resistant to diet-induced obesity, making PTP1B a prized target in the pharmaceutical industry for therapeutic intervention in type two diabetes and obesity. Efforts to develop inhibitors of PTP1B focused on the active site pocket; however, those efforts have been hindered by poor oral bioavailability of the compounds. Our lab has exploited the conformational changes associated with redox regulation of PTP1B to devise an alternative method of inhibiting the enzyme. Reactive oxygen species, generated in response …

Capstone Projects and Master's Theses
Ever since No Child Left Behind (NCLB) legislation became adopted into our school system, students’ physical health has suffered. Recently, the number of unhealthy children has grown. One in three children are classified as overweight or obese. This senior capstone project demonstrates the causes and effects of such an issue in regards to both physical education and nutritional education in elementary schools in the tri-county areas of Monterey, Salinas, and Santa Clara counties. The results from twenty-seven survey results and one interview suggest that most teachers feel that there is a need for daily physical education and nutritional education should …
Multi-Level Predictors Of Obesity And Diet In Multi-Ethnic Populations, Tali Elfassy
Multi-Level Predictors Of Obesity And Diet In Multi-Ethnic Populations, Tali Elfassy
Open Access Dissertations
The obesity epidemic in the United States is a major public health issue. Poor socio-economic conditions such as poverty or living under financial stress may adversely influence health outcomes such as obesity, and often disproportionately impact ethnic minority populations. Further, individuals living in poverty are often exposed to environments that promote unhealthy diet and obesity. Such obesogenic environments are characterized by increased exposure to foods high in sodium and low in potassium which may also be independently associated with obesity. Yet these associations have not been adequately explored, especially among ethnic minority populations. The current dissertation seeks to explore: 1) …

Overweight and obesity are particularly saient concerns for adolescents and young adults, the age group with the greatest risk of weight gain each year. The present study examined differences in behavioral economic measures of food reinforcement and delay discounting among normal weight women compared to overweight and obese adult women. Effects of symptoms of depression and ethnicity were also examined. Young adult women were given a variety of questionnaires. The current study hypothesized that overweight and obese women would rate food images as being more pleasant, discount money more steeply, have higher food-related reinforcement, and higher demand for food. Overweight …

The intestinal microbiota are integral to understanding the relationships between nutrition and health. Therefore, fecal sampling and processing protocols for metagenomic surveys should be sufficiently robust, accurate, and reliable to identify the microorganisms present. We investigated the use of different fecal preparation methods on the bacterial community structures identified in human stools. Complete stools were collected from six healthy individuals and processed according to the following methods: (i) randomly sampled fresh stool, (ii) fresh stool homogenized in a blender for 2 min, (iii) randomly sampled frozen stool, and (iv) frozen stool homogenized in a blender for 2 min, or (v) …

Assessing Feasibility And Readiness To Address Obesity Through Policy In American Indian Reservations, Valarie Blue Bird Jernigan, Gail Boe, Carolyn Noonan, Leslie Carroll, Dedra Buchwald
Journal of Health Disparities Research and Practice
The Institute of Medicine and Centers for Disease Control and Prevention (CDC) have identified policy and environmental strategies as critical to the prevention and control of obesity. However such strategies are rare in American Indian communities despite significant obesity-related disparities. Tribal policymaking processes differ by tribal nation and are often poorly understood by researchers and public health practitioners, hindering the dissemination, implementation, and successful scale-up of evidence-base obesity strategies in tribal communities. To address these gaps in knowledge we surveyed 138 diverse stakeholders in two American Indian reservations to assess the feasibility of and readiness to implement CDC-recommended obesity policy …

This paper presents the design, implementation and evaluation of a hybrid clinic plus home based intervention targeting Latino children to improve obesity outcomes. The intervention applies motivational game based learning and behavior change theories during design. Latino American children are the main target group for this study as they have significantly high obesity rates due to socio-economic conditions and lack of awareness. There have been several interventions that have targeted game based strategies in the clinic to promote health outcomes and some have even targeted obesity problems, however to our knowledge this is the first effort that adds an inhome …

Obesity and osteoporosis are two alarming health disorders prominent among middle and old age populations, and the numbers of those affected by these two disorders are increasing. It is estimated that more than 600 million adults are obese and over 200 million people have osteoporosis worldwide. Interestingly, both of these abnormalities share some common features including a genetic predisposition, and a common origin: bone marrow mesenchymal stromal cells. Obesity is characterized by the expression of leptin, adiponectin, interleukin 6 (IL-6), interleukin 10 (IL-10), monocyte chemotactic protein-1 (MCP-1), tumor necrosis factor-alpha (TNF-α), macrophage colony stimulating factor (M-CSF), growth hormone (GH), parathyroid …

Obesity is associated with the development of vascular insulin resistance; however, pathophysiological mechanisms are poorly understood. We sought to investigate the role of WNT5A-JNK in the regulation of insulin-mediated vasodilator responses in human adipose tissue arterioles prone to endothelial dysfunction. In 43 severely obese (BMI 44±11 kg/m2) and five metabolically normal non-obese (BMI 26±2 kg/m2) subjects, we isolated arterioles from subcutaneous and visceral fat during planned surgeries. Using videomicroscopy, we examined insulin-mediated, endothelium-dependent vasodilator responses and characterized adipose tissue gene and protein expression using real-time polymerase chain reaction and Western blot analyses. Immunofluorescence was used to quantify endothelial nitric oxide …

Vitamin D related research continues to expand and theorise regarding its involvement in obesity, as both hypovitaminosis D and obesity strike in pandemic proportions. Vitamin D plays an important role in immune system through Vitamin D Receptors (VDR), which are transcription factors located abundantly in the body. Due to this characteristic, it is potentially linked to obesity, which is a state of inflammation involving the release of cytokines from adipose tissue, and exerting stress on other organs in a state of positive energy balance. Research trials in the past couple of years and …

Metabolic Syndrome: Differences For Asian Americans Is In Their Percentage Of Body Fat, Patricia Alpert, Diane L. Thomason
Asian/Pacific Island Nursing Journal
Asian Americans are not frequently thought of as being obese or overweight yet some of the Asian American subgroups have a disproportionate risk for cardiovascular disease and type 2 diabetes mellitus. Although the standardized body mass index (BMI) assessment is an adequate tool for reporting secular prevalence trends for overweight/obesity across populations, it falls short in accuracy when assessing Asian Americans. In recent years more has been written about the re-evaluation of BMI cut points for normal weight, overweight, or obese Asian Americans. Additionally, the waist circumference norm was modified to indicate a smaller waist size is a risk for …

Epigenetic mechanisms, including DNA methylation, mediate the interaction between gene and environment and may play an important role in the obesity epidemic. We assessed the relationship between DNA methylation and obesity in peripheral blood mononuclear cells (PBMCs) at 485,000 CpG sites across the genome in family members (8-90 y of age) using a discovery cohort (192 individuals) and a validation cohort (1,052 individuals) of Northern European ancestry. After Bonferroni-correction (Pα=0.05 = 1.31 × 10-7) for genome-wide significance, we identified 3 loci, cg18181703 (SOCS3), cg04502490 (ZNF771), and cg02988947 (LIMD2), where methylation status was associated with body mass index percentile (BMI%), a …

The rates of diabetes and obesity in Navajo communities are higher than national rates. A healthy diet of fresh fruits and vegetables will help to prevent and reduce such rates. So the project focuses on promoting gardening to make vegetables and fruits more accessible at the comfort of one’s own home. Previous research in this area shows that gardeners consume 2 more servings of fruits and vegetables than nongardeners. Thirty participants were recruited by flyer distribution at chapter houses, senior centers, and community centers, from four Navajo targeted areas: Shiprock, NM; Tsaile, AZ; Lukachukai, AZ, and Hogback, NM. With the …

Trends in adult obesity have been used to motivate key public health policies in the United States. While these analyses provide important insights into the broad historical contours of the obesity epidemic in the U.S., they shed less light on the proximate mechanisms that have generated these changes and that will ultimately determine the long-term course and pace of change in obesity rates. We used data from the National Health and Nutrition Examination Survey (NHANES), Glenn Firebaugh's linear decomposition technique, and Kitagawa's algebraic decomposition method to decompose change in body mass index (BMI), obesity, and morbid obesity from 1971 through …
