Infertility is one of the problems facing women and men. Obesity, lipid level disorders, and insulin resistance are among the risk factors that cause infertility, in addition to other relevant factors. Children represent a highly complex and variable population for treatment, including interindividual differences in drug dose-exposure. Midazolam has been used as a sedative for hospitalized children on- and off-label; however, factors affecting interindividual variability (IIV) in observed clearance for this population are not fully understood and can result in extreme under- or overexposure. Obesity has been described as a significant influence on midazolam in adolescents, which could potentially alter drug exposure. Obesity and type 2 diabetes are interconnected global health challenges, contributing significantly to morbidity, mortality, and healthcare costs. This literature review compares the efficacy, safety, and applicability of glucagon-like peptide-1 (GLP-1) receptor agonists and bariatric surgery as treatment options. A literature search with defined inclusion and exclusion criteria was conducted using PubMed and EBSCOhost from May to July 2024, resulting in 14 randomized controlled trials for evaluation. Despite extensive research, the mechanisms underlying excessive alcohol and food consumption remain incompletely understood. Emerging evidence suggests that neuro-immune crosstalk plays a pivotal role in feeding and alcohol-induced behaviors, with inflammation as a key pathological hallmark. The objective of this literature review is to explore the relationship between early-life stress and the development of disordered or dysregulated eating in adulthood. A search was conducted across PubMed, CINAHL, Embase, and PsycINFO databases. Results from each study were compared and synthesized. Twelve studies were analyzed including case reports, systematic reviews, cohort studies, and cross-sectional studies. Research indicates that childhood emotional abuse, often more prevalent than other forms of maltreatment, is a significant risk factor for eating pathology, with studies investigating psychological mediators such as anger, anxiety, and dissociation in this relationship. Maternal mortality is an increasing concern in the United States. According to the WHO, from 2018-2022 the Maternal Mortality Rate (MMR) was 23.3 deaths per every 100,000 live births. New Jersey's MMR exceeded this average at 26 per 100,000 during the same period, with 91% of cases deemed preventable. The hypothalamic leptin-proopiomelanocortin (POMC) pathway is critical for regulating metabolism. POMC neurons in the arcuate nucleus respond to leptin and play a pivotal role in mediating energy and glucose balance. However, during diet-induced obesity (DIO), these neurons often develop resistance to exogenous leptin. Recently, the small GTPase Rap1 has been implicated as an inhibitor of neuronal leptin signaling; however, its specific role within POMC neurons remains unexplored. We generated tamoxifen-inducible, POMC neuron-specific Rap1 knockout mice to selectively delete both Rap1a and Rap1b isoforms in POMC neurons. Overweight and obesity (OW/OB) are significant concerns among individuals with Autism Spectrum Disorder (ASD), yet the underlying causes remain unclear. Emerging evidence points to metabolic and hormonal factors—particularly leptin, a hormone involved in appetite regulation—as potential contributors. This scoping review examined five key studies from 2003 to 2024 that investigated the relationship between leptin and ASD in overweight populations. Across studies, individuals with ASD consistently exhibited elevated leptin levels compared to controls. While some findings also noted changes in other adipokines, such as lower resistin or progranulin, leptin was the most consistently dysregulated. To better understand and address this issue, the Monterey County Office of Education Early Learning Program (MCOE) is a government agency dedicated to supporting early childhood education and healthy lifestyles. Using a quantitative approach, a study was conducted to analyze the Body Mass Index (BMI) of children enrolled in the program for the 2023-2024 school year. Obesity, the excessive accumulation of body fat, increases risk for chronic disease and affects 34% of U.S. adults (10). Lifestyle interventions are effective but heavily rely on calorie tracking, while adherence to this behavioral strategy is low (6). An alternative to calorie tracking is to simply track instances of overeating. In a pilot trial, obese individuals used Habit app, a mobile app that assists users in tracking overeating episodes. The rates of obesity and suicidal behavior have continued to increase over the years among young adults in the United States. Previous research has indicated a relationship between obesity and suicidal behavior, primarily among women, yet minimal research has been done to examine factors that explain this relationship (i.e., mediators). Additives such as antibiotic residues, GMOs, pesticide residues, anti-caking agents, emulsifiers, artificial sweeteners, colorants, flavor enhancers, and preservatives. Most of these additives seem to shift the gut microbiome towards more pro-inflammatory signals. When all these additives are considered as an amalgam, such as with the modern processed diet, the ramifications on the gut microbiome are concerning. Obesity, which is characterized by low-level chronic inflammation, is likely exacerbated by these food additives. The correlation analysis, conducted using Pearson's test with a significance level of 95% (α<0.05), revealed significant regional disparities in the prevalence of NCDs, dietary consumption, and lifestyle factors across Indonesia. Moreover, the findings reveal a statistically significant positive correlation between obesity, high-risk food consumption, and lifestyle risk factors with the prevalence of several NCDs, including diabetes mellitus, heart disease, hypertension, and chronic kidney disease. These results underscore the potential impact of our research on public health in Indonesia. The results highlight also the urgent need for targeted public health initiatives, such as policies promoting healthier food choices, controlling the food environment, encouraging physical activity, and supporting smoking cessation to mitigate the growing burden of NCDs in Indonesia.

Background/Objectives: Time spent in sedentary pursuits is associated with adverse metabolic profiles. Adolescents spend 65-75% of their day in sedentary pursuits; however, evidence among youth is less conclusive. This study examined the effects of an acute 4 h bout of sedentary behaviors on cardiometabolic outcomes and energy expenditure in Hispanic adolescents (12-16 years old) with obesity (BMI% ≥ 95th).
