Theses and Dissertations

Reactive Oxygen Species (ROS) are radicals of great concern to biologists. Their role in several diseases--such as neurodegenerative disease, diabetes, premature aging and cancer--has been intensively investigated during the last decade. Since a major focus in cancer research is to better understand how it is induced and therefore how it can be cured, the study of the cytotoxic effects of ROS production within cancer cells is vital. Nanotechnology is an emerging field of science that promises great improvements in a number of disciplines. Nano medicine is one of its daughter fields. Various nanomaterials are used for diagnosis and disease detection, …

The Effects Of Physicochemical Properties Of Ceo2 Nanoparticles On Toxicity To Soil Denitrification Processes, Jessica Dahle

All Theses

Due to the rise of nanotechnology in industry, the concentrations of engineered metal oxide nanoparticles being released to the environment are increasing daily. Cerium-based compounds such as nanoparticulate ceria (NP CeO2) have received much attention in the last several years due to their popular applications (e.g., mischmetal, pharmaceuticals, nanotechnology) in industrial and commercial uses. Like many emerging pollutants, NP CeO2 is often released to the environment, where its toxicological effects are not well known. Though several environmental exposure pathways have been identified for CeO2 NPs, a major route of NP introduction to the environment is via biosolids application to agricultural …

Generation Of Intracellular Reactive Oxygen Species And Genotixity Effect To Exposure Of Nanosized Polyamidoamine (Pamam) Dendrimers In Plhc-1 Cells In Vitro, Pratap Naha, Hugh Byrne

Articles

Polyamidoamine (PAMAM) dendrimers elicit systematically variable cyto- and eco-toxic responses, promising the basis for structure activity relationships governing nanotoxicological responses. Increased production of intracellular reactive oxygen species (ROS), genotoxicity and apoptosis due to in-vitro exposure of hepatocellular carcinoma cells to dendrimer generations G-4, G-5 and G-6 is demonstrated. A generation dependent increase in ROS and genotoxicity was observed, consistent with our previous studies. The responses correlate well with the number of surface amino groups per generation. Although ROS production initially increases approximately linearly, it saturates at higher doses. Notably, normalised to molar dose of surface amino groups, the dose-dependent ROS …

United States Environmental Protection Agency: Publications

Chronic exposure to inorganic arsenic (iAs) is carcinogenic to the human urinary bladder. It produces urothelial cytotoxicity and proliferation in rats and mice. DMAV, a major methylated urinary metabolite of iAs, is a rat bladder carcinogen, but without effects on the mouse urothelium. DMAIII was shown to be the likely urinary metabolite of DMAV inducing urothelial changes and is also postulated to be one of the active metabolites of iAs. To evaluate potential DMAIII-induced urothelial effects, it was administered to As3mt knockout mice which cannot methylate arsenicals. Female C57BL/6 wild type and As3mt knockout …

The Assessment Of An In-Vitro Model For Evaluating The Role Of Parp In Ethanol-Mediated Hepatotoxicity, Jayme Coyle

USF Tampa Graduate Theses and Dissertations

This investigation assesses the role of poly(ADP-ribose) polymerase in ethanol-mediated hepatotoxicity using the untransfected HepG2 hepatocellular carcinoma line, an established, well-characterized toxicological model. HepG2 cells were treated with ethanol at concentrations between 100 mM and 800 mM, and assessed for markers of cytotoxicity. PARP-1 activity in total cell protein lysates was quantified as a proxy of apoptotic induction at six hours. Our results demonstrated a 1.43-fold AST activity increase in culture medium isolates of cells exposed to 800 mM without significant effect on cellular viability. PARP-1 activity varied greatly and results for enzyme activity remained inconclusive. The results suggest a …

Inhibitory Effect Of Il-17 On Neural Stem Cell Proliferation And Neural Cell Differentiation., Zichen Li, Ke Li, Lin Zhu, Quancheng Kan, Yaping Yan, Priyanka Kumar, Hui Xu, Abdolmohamad Rostami, Guang-Xian Zhang

Department of Neurology Faculty Papers

BACKGROUND: IL-17, a Th17 cell-derived proinflammatory molecule, has been found to play an important role in the pathogenesis of autoimmune diseases, including multiple sclerosis (MS) and its animal model, experimental autoimmune encephalomyelitis (EAE). While IL-17 receptor (IL-17R) is expressed in many immune-related cells, microglia, and astrocytes, it is not known whether IL-17 exerts a direct effect on neural stem cells (NSCs) and oligodendrocytes, thus inducing inflammatory demyelination in the central nervous system.

METHODS: We first detected IL-17 receptor expression in NSCs with immunostaining and real time PCR. We then cultured NSCs with IL-17 and determined NSC proliferation by neurosphere formation …

Two Modes Of Cell Death Caused By Exposure To Nanosecond Pulsed Electric Field, Olga N. Pakhomova, Betsy W. Gregory, Iurii Semenov, Andrei G. Pakhomov

Bioelectrics Publications

High-amplitude electric pulses of nanosecond duration, also known as nanosecond pulsed electric field (nsPEF), are a novel modality with promising applications for cell stimulation and tissue ablation. However, key mechanisms responsible for the cytotoxicity of nsPEF have not been established. We show that the principal cause of cell death induced by 60- or 300-ns pulses in U937 cells is the loss of the plasma membrane integrity (‘‘nanoelectroporation’’), leading to water uptake, cell swelling, and eventual membrane rupture. Most of this early necrotic death occurs within 1–2 hr after nsPEF exposure. The uptake of water is driven by the presence of …

Novel Copper (Ii) Mediated Transfection Nano-Systems And Biological Properties Of Pyridinium Surfactants, Sarit Pal

Open Access Theses & Dissertations

Efficacy of gene therapy is relied upon the improvement of gene delivery vectors into mammalian cells. Although viral-based polynucleotide carriers are still considered as the most common method for gene delivery but the cost effectiveness, nonspecific lethal immune response has evoked the renaissance of non-viral synthetic delivery systems. An initial discovery by Felgnar et al showed Cationic Liposomes (CLs) (having an overall positive charge) when mixed with DNA to form CL-DNA complexes can enhance the transfer of polynucleotides into cells followed by its expression. Presently, a flurry of experimental works is taking place for the better understanding of structural characterization …

Cytotoxic Activities Of Some Pseudomonas Aeruginosa Isolates: Possible Mechanisms And Approaches For Inhibition, Ghadir Saeed El-Housseiny, Mohammad Mabrouk Aboulwafa, Nadia Abdelhaleim Hassouna

Turkish Journal of Biology

The cytotoxicity of 6 highly cytotoxic Pseudomonas aeruginosa isolates was tested against Vero cells. Cytotoxicity was found to be cell-associated, high cytotoxicity was observed at a multiplicity of infection of 200:1, and this activity gradually decreased as the bacterial count decreased. This cytotoxicity caused rounding and detachment of the Vero monolayer as early as 1 h after infection. Moreover, viable metabolically active bacteria were necessary for Vero cell killing. These results imply that one or more of the type III secretion system effector proteins (ExoU, ExoS, ExoT, or ExoY) were responsible for the observed cytotoxic effects. This cytotoxicity was affected …

Turkish Journal of Veterinary & Animal Sciences

In the present work, the biocompatibility of orthodontic devices was tested by the propagation of human osteoblasts. A different methodological approach was applied. Since the materials used in human and veterinary orthodontics (not in implantology) had been tested, osteoblasts were not implanted on the surface of the biomaterial, but have been cultured in the medium containing the products of corrosion. In the present work, osteoblasts were propagated in the medium spiked with Ni(II) salt versus the control. Osteoblast cells (live, dead, and total) were counted under light microscope. The statistical significance of the differences between the experimental and control groups …

In Vitro Cytotoxicity And Skin Irritation Testing Of Antimicrobial Conjugated Electrolytes : Interactions With Mammalian Cells, Kristin N. Wilde

Chemical and Biological Engineering ETDs

An estimated 19,000 deaths and $3-4 billion in health care costs per year in the U.S. are attributed to methicillin-resistant Staphylococcus aureus (MRSA) infections. Certain cationic phenylene ethynylene (CPE)-based polymers (PPEs) and oligomers (OPEs) have been demonstrated to exhibit dark and light-activated antimicrobial activity. Envisioned applications of these CPEs include the fabrication of antimicrobial surfaces to reduce or prevent the spread of potentially untreatable strains of bacteria. Until recently, it was unknown if the polymers or oligomers would exhibit similar biocidal activity toward mammalian cells. This work examines the toxicity of CPEs to mammalian cells at three levels: cytoxicity testing …

Did Mineral Surface Chemistry And Toxicity Contribute To Evolution Of Microbial Extracellular Polymeric Substances?, Jie Xu, Jay M. Campbell, Nianli Zhang, William J. Hickey, Nita Sahai

College of Polymer Science and Polymer Engineering

Modern ecological niches are teeming with an astonishing diversity of microbial life in biofilms closely associated with mineral surfaces, which highlights the remarkable success of microorganisms in conquering the challenges and capitalizing on the benefits presented by the mineral-water interface. Biofilm formation capability likely evolved on early Earth because biofilms provide crucial cell survival functions. The potential toxicity of mineral surfaces toward cells and the complexities of the mineral-water-cell interface in determining the toxicity mechanisms, however, have not been fully appreciated. Here, we report a previously unrecognized role for extracellular polymeric substances (EPS), which form biofilms in shielding cells against …

Design, Synthesis And Biological Evaluation Of New-Generation Taxoid-Based Tumor-Targeting Drug Conjugates, Edison S. Zuniga

Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)

Over the past decades, chemotherapy has transitioned to an age of "targeted therapy." The development of Gleevec (Novartis) and Tarceva (Genentech and OSI Pharmaceuticals) has revolutionized the treatment of certain cancers, such as chronic myelogenous leukemia (CML), non-small cell lung cancer (NSCLC) and pancreatic cancer. Gleevec specifically targets the cancer cell-overexpressed Abl tyrosine kinase, and Tarceva targets the epidermal growth factor receptor that is mutated in different types of cancers. However, the use of these modern targeted-therapies is limited to specific cancer types. Therefore, considerable efforts have been made to find cancer-specific markers which can be exploited for tumor-specific delivery …
