Doctoral Dissertations and Projects
In 2020, COVID-19 impacted the world and transformed the way individuals and families operated and functioned daily. Lockdowns and restrictions forced families to stay home and find alternative ways of providing and living concerning employment, education, and family functioning. One particular area impacted by this phenomenon was how families operated and conducted family meals. During COVID-19, increases in family mealtime were seen to take place but little evidence was found regarding the quality of these mealtimes, the father’s experience, and what family functioning looked like after COVID-19. While family functioning and mealtimes during COVID-19 have been examined and analyzed over …

Bone metastasis is a major cause of cancer death; however, the epigenetic determinants driving this process remain elusive. Here, we report that histone methyltransferase ASH1L is genetically amplified and is required for bone metastasis in men with prostate cancer. ASH1L rewires histone methylations and cooperates with HIF-1α to induce pro-metastatic transcriptome in invading cancer cells, resulting in monocyte differentiation into lipid-associated macrophage (LA-TAM) and enhancing their pro-tumoral phenotype in the metastatic bone niche. We identified IGF-2 as a direct target of ASH1L/HIF-1α and mediates LA-TAMs' differentiation and phenotypic changes by reprogramming oxidative phosphorylation. Pharmacologic inhibition of the ASH1L-HIF-1α-macrophages axis elicits …

During and after the COVID-19 pandemic, teachers recognized that online learning stagnated students’ social emotional development (SED). SED is the ability to foster meaningful relationships while also being able to manage, express, and acknowledge emotions, (Office of Head Start, n.d., para.2). SED is typically fostered through in-person interactions, therefore, social-emotional learning (SEL) strategies had to be modified to address online learning. SEL, is the practice of a wide range of skills, attitudes, and behaviors that affect a student’s success in school and life, and is necessary for SED. For example, critical thinking, managing emotions, working through conflicts, decision making, and …

Abstract
The COVID-19 pandemic changed the world drastically, and its impact was monumental. COVID-19 or SARS-CoV-2, or severe acute respiratory syndrome, was the virus responsible for causing such mayhem. This contagious virus can enter the body via different routes, live on surfaces for hours, and be transmitted through the air, increasing infection risk. Once it enters the body, it can trick the body into replicating the virus. From this reaction within the body, signs and symptoms follow. While the name indicates a respiratory illness, the virus can inflict injury on other parts of the body. Due to the organ systems …

For people in marginalized communities, particularly the Black and African American community, the combined, disproportionate damage and loss from of the pandemic and a cacophony of racial injustices offer cause for labeling the phenomenon a syndemic. Syndemic is defined as “a conceptual framework for understanding diseases or health conditions that arise in populations and that are exacerbated by the social, economic, environmental, and political milieu in which a population is immersed” (Singer et al., 2017, p. 881). The purpose of this study is to explore, describe, and analyze Kentucky students’ perceptions of school climate and safety by demographic groups, racial …

Although vaccines are usually given intramuscularly, the intranasal delivery route may lead to better mucosal protection and limit the spread of respiratory virus while easing administration and improving vaccine acceptance. The challenge, however, is to achieve delivery across the selective epithelial cell barrier. Here we report on a subunit vaccine platform, in which the antigen is genetically fused to albumin to facilitate FcRn-mediated transport across the mucosal barrier in the presence of adjuvant. Intranasal delivery in conventional and transgenic mouse models induces both systemic and mucosal antigen-specific antibody responses that protect against challenge with SARS-CoV-2 or influenza A. When benchmarked …
