Iron And The Liver: Acute Effects Of Iron-Loading On Hepatic Heme Synthesis Of Rats. Role Of Decreased Activity Of 5-Aminolevulinate Dehydrase., H L. Bonkowsky, J F. Healey, P R. Sinclair, J F. Sinclair, S I. Shedlofsky, G H. Elder

Dartmouth Scholarship

Acute iron loading of rats, by intraperitoneal administration of iron-dextran (500 mg Fe/kg body wt 18-20 h before killing) decreased by 30% the rate of conversion of 5-amino-[14C]levulinate ([14C]ALA) into heme as measured with a recently described procedure for liver homogenates (1981. Biochem. J. 198: 595-604). The decrease in conversion of labeled ALA into heme caused by iron loading was shown to be due to a 70-80% decrease in activity of ALA dehydrase. The decrease in activity of ALA dehydrase caused by iron loading was not associated with a decrease in hepatic concentrations of GSH, nor could it be reversed …

Evidence For Hydrated Spermidine-Calf Thymus Dna Toruses Organized By Circumferential Dna Wrapping, Kenneth A. Marx, George C. Ruben Mar 1983

Evidence For Hydrated Spermidine-Calf Thymus Dna Toruses Organized By Circumferential Dna Wrapping, Kenneth A. Marx, George C. Ruben

Dartmouth Scholarship

In spermidine-condensed calf thymus DNA preparations, torus–shaped condensates were shown by transmission electron microscopy to exist under the hydrated conditions of the freeze fracture experiment. Using extremely low Pt metal deposition levels (9 A Pt/C) high–contrast replicas of the spermidine–DNA toruses were obtained that showed circumferential wrapping of single DNA double helix–size surface fibres. Stereoscopic analysis of high magnification stereomicrographs established some details of the three-dimensional organization of two DNA double helix sections winding circumferentially on the inner surface of one such torus. These measurements demonstrate the usefulness of stereoscopic analysis of these high macromolecular organization magnification. Measurements on a …

Historical Trends In American Animal Use And Perception, Stephen R. Kellert, Miriam O. Westervelt Jan 1983

Historical Trends In American Animal Use And Perception, Stephen R. Kellert, Miriam O. Westervelt

Attitudes Towards Animals Collection

Changes in American attitudes and behaviors toward animals from 1900-1976 will be examined. The data are derived from an empirical analysis of 4,873 animalrelated newspaper articles. Four newspapers were used in this analysis- the Los Angeles Times; Hartford Courant; Buffalo, Wyoming Bulletin; and the Dawson, Georgia News. A content analysis procedure was employed to record animal-related information in the articles, and these data were subjected to a variety of statistical analyses. A comparison of the results with a 1978 national survey of American attitudes and behaviors is briefly attempted. Finally, some policy implications of the data are considered.

Attitudes Toward Animals In Greco-Roman Antiquity, Liliane Bodson Jan 1983

Attitudes Toward Animals In Greco-Roman Antiquity, Liliane Bodson

Attitudes Towards Animals Collection

Both wild and domesticated animals had a direct and wide-ranging role in the life of the ancient Greeks and Romans. The bond between humans and animals which first originated in the economic needs went far beyond strictly practical matters. It did influence and enrich the Classical culture in its major aspects from literature and arts to philosophy and ethics. It also induced people to analyze the main implications of their relationship with "subhuman" creatures. The present paper aims to survey the range of the attitudes they developed about animals. It also examines to what extent they were concerned with the …

Monoclonal Antibodies To Novel Myeloid Antigens Reveal Human Neutrophil Heterogeneity., Edward D. Ball, Robert F. Graziano, Li Shen, Michael W. Fanger Sep 1982

Monoclonal Antibodies To Novel Myeloid Antigens Reveal Human Neutrophil Heterogeneity., Edward D. Ball, Robert F. Graziano, Li Shen, Michael W. Fanger

Dartmouth Scholarship

Three cytotoxic murine monoclonal antibodies that recognize myeloid-specific antigens have been produced by immunization with normal human neutrophils or myeloblasts from a patient with acute myelomonocytic leukemia. Two of these, PMN 6 and PMN 29, are specific for neutrophils; the third monoclonal antibody, AML-2-23, is reactive with the majority of normal monocytes as well as a subpopulation of mature neutrophils. Although neutrophils from all individuals tested expressed these antigens, cytofluorographic analysis revealed that the percentage of cells bearing the PMN 6 and AML-2-23 antigens varied among individuals. Significant additional heterogeneity in the density of each antigen among antigen-bearing cells was …

The Cytoskeletal System Of Nucleated Erythrocytes. I. Composition And Function Of Major Elements, William D. Cohen, Diana Bartelt, Richard Jaeger, George M. Langford, Iris Nemhauser Jun 1982

The Cytoskeletal System Of Nucleated Erythrocytes. I. Composition And Function Of Major Elements, William D. Cohen, Diana Bartelt, Richard Jaeger, George M. Langford, Iris Nemhauser

Biology - All Scholarship

We have studied the dogfish erythrocyte cytoskeletal system, which consists of a marginal band of microtubules (MB) and trans-marginal band material (TBM). The TBM appeared in whole mounts as a rough irregular network and in thin sections as a surface-delimiting layer completely enclosing nucleus and MB. In cells incubated at 0 degrees C for 30 min or more, the MB disappeared but the TBM remained. MB reassembly occurred with rewarming, and was inhibited by colchicine. Flattened elliptical erythrocyte morphology was retained even when MBs were absent. Total solubilization of MB and TBM at low pH, or dissolution of whole anucleate …

Hair As An Indicator Of Excessive Aluminum Exposure, Robert A. Yokel Apr 1982

Hair As An Indicator Of Excessive Aluminum Exposure, Robert A. Yokel

Pharmaceutical Sciences Faculty Publications

To determine if excessive systemic exposure to aluminum would be reflected in increased aluminum concentration in hair, rabbits were given a series of aluminum lactate injections. Hair was collected before the aluminum lactate administration from the site of injections and twice after the injections from this site as well as from an area adjoining the injection site. Aluminum was determined by flameless atomic absorption analysis of acid-digested samples. The concentration of aluminum in the hair increased after the injections in samples taken at both times from both sites. Considerable variability in hair aluminum was found before excessive exposure, as has …

Proteins Antigenically Related To The Human Erythrocyte Glucose Transporter In Normal And Rous Sarcoma Virus-Transformed Chicken Embryo Fibroblasts., Donald W. Salter, Stephen A. Baldwin, Gustav E. Lienhard, Michael J. Weber Mar 1982

Proteins Antigenically Related To The Human Erythrocyte Glucose Transporter In Normal And Rous Sarcoma Virus-Transformed Chicken Embryo Fibroblasts., Donald W. Salter, Stephen A. Baldwin, Gustav E. Lienhard, Michael J. Weber

Dartmouth Scholarship

Antibody raised against the purified human erythrocyte glucose transporter specifically precipitated four proteins from normal and Rous sarcoma virus-transformed chicken embryo cells: a major protein of Mr 41,000 and minor proteins of Mr 68,000, 73,000, and 82,000. The Mr 41,000 and 82,000 proteins were found only in a membrane fraction, not in the soluble fraction, and displayed a heterogeneous mobility on NaDodSO4/polyacrylamide gel electrophoresis, suggesting glycosylation. The Mr 41,000 and 82,000 proteins were increased in amount after malignant transformation in direct proportion to the increase in hexose transport rate, and the increase was dependent on the expression of the src …

Genetically Controlled Variation Of “Acid” Beta-Galactosidase Detected In Rattus Norvegicus By Isoelectric Focusing, T C Douglas, K A Kimmel, P E Dawson Mar 1982

Genetically Controlled Variation Of “Acid” Beta-Galactosidase Detected In Rattus Norvegicus By Isoelectric Focusing, T C Douglas, K A Kimmel, P E Dawson

Faculty, Staff and Student Publications

Two genetically variant forms of rat "acid" beta-galactosidase were found to differ in isoelectric point and pH dependence, but not in thermostability or sensitivity to inhibition by p-mercuribenzoate (PMB). The results of two backcrosses and an intercross indicated that the isoelectric focusing phenotypes are controlled by two codominant alleles at a single autosomal locus, for which we propose the name Glb-1. No significant linkage between Glb-1 and albino (LG I), brown (LG II), or hooded (LG VI) was observed. Strain-specific differences in total levels of kidney beta-galactosidase were detected, but it is not yet known whether the variation is controlled …

Synthesis Of Low Molecular Weight Heat Shock Peptides Stimulated By Ecdysterone In A Cultured Drosophila Cell Line., Robert C. Ireland, Edward M. Berger Feb 1982

Synthesis Of Low Molecular Weight Heat Shock Peptides Stimulated By Ecdysterone In A Cultured Drosophila Cell Line., Robert C. Ireland, Edward M. Berger

Dartmouth Scholarship

Treatment of Schneider's line 3 Drosophila cells with the steroid hormone ecdysterone rapidly stimulated the synthesis and accumulation of the polypeptide previously designated p7 [Berger, E. M., Ireland, R. C. & Wyss, C. (1980) Somatic Cell Genet. 6, 119-129]. In this report, p7 is identified as the 23,000-dalton heat shock polypeptide (hsp23). In addition to hsp23, the synthesis of the low molecular weight heat shock polypeptides hsp22, hsp26, and hsp27 was also stimulated by ecdysterone, although to different extents. Hybridization of a nick-translated genomic clone containing the hsp23 gene to a total RNA blot showed that ecdysterone stimulation of hsp23 …

Attitudes Toward Animal Suffering: An Exploratory Study, John Braithwaite, Valerie Braithwaite Jan 1982

Attitudes Toward Animal Suffering: An Exploratory Study, John Braithwaite, Valerie Braithwaite

Attitudes Towards Animals Collection

A total of 302 undergraduates in the social sciences and the humanities, at two Australian universities, were given a questionnaire designed to explore public attitudes toward animal suffering. The results, though preliminary, strongly suggest that attitudes may be in great part supportive of animal welfare and animal rights. However, as reflected in the answers to the questionnaire, actual behavior does not always follow suit. The recommendation is made that the animal welfare/animal rights movement should perhaps place more emphasis on raising people's awareness of the inconsistencies between their attitudes toward animals and their behavior concerning them.

Unnecessary Suffering: Definition And Evidence, Frank Hurnik, Hugh Lehman Jan 1982

Unnecessary Suffering: Definition And Evidence, Frank Hurnik, Hugh Lehman

Attitudes Towards Animals Collection

Although it is possible to formulate stronger moral principles than "animals should not be made to suffer unnecessarily," there are significant grounds for doubting these stronger principles. But the principle that underlies the dictum regarding unnecessary suffering is generally recognized as valid, since denial of it implies that we can do whatever we want with animals, a conclusion that is usually considered unacceptable. A determination of whether any particular instance of suffering is necessary or unnecessary must be based on an analysis of both the seriousness of the purpose of the act that involves pain in animals, and its relative …

The Changing Concept Of Animals As Property, Vincent P. Mccarthy Jan 1982

The Changing Concept Of Animals As Property, Vincent P. Mccarthy

Attitudes Towards Animals Collection

Enforced and maintained by a legal superstructure that regulated every aspect of a black [slave's] social, political, economic, and religious life, his property status continued until the middle of the nineteenth century when Congress passed the 13th, 14th, and 15th Amendments to the Constitution, which overturned the Dred Scott decision and recognized that a black human being had legally protectible rights.

There are some signs in recent legal decisions that a similar evolution in the status of animals is taking place: judges are beginning to draw distinctions between animals and property.

But can we ever expect that the courts will …

The Problem Of Pain: What Do Animals Really Feel?, Dana H. Murphy Jan 1982

The Problem Of Pain: What Do Animals Really Feel?, Dana H. Murphy

Sentience Collection

Much of the contention and confusion that seem inevitably to arise whenever the subject of pain in animals comes up appear to stem principally from problems with the word "pain" itself. When used to describe responses in humans, "pain" can mean any subset of an incredibly broad spectrum of sensations and emotions, ranging from the instantaneous, galvanizing effect of a dentist drill hitting the nerve in a molar, to more airy notions such as the "pain" of rejection or "painfully" embarrassing situations. Humans even use concepts as abstruse as the German term, weltschmerz, or "world pain," which denotes a vaguely …

The Economics Of Farm Animal Welfare, A. J. F. Webster Jan 1982

The Economics Of Farm Animal Welfare, A. J. F. Webster

Agribusiness Collection

The number of ways that one can be nice or nasty to animals are legion. This article will consider only one very specific aspect of farm animal welfare, namely, those systems of intensive animal production in which the system itself, irrespective of the quality of the stockmanship within the system, appears to restrict the normal behavior of farm animals to an unacceptable degree. The systems that were considered by the House of Commons Select Committee on Agriculure (1981) include egg production from hens in battery cages, production of veal from calves deprived of solid food and isolated in wooden crates, …

Detergent Extraction Of A Presumptive Gating Component From The Voltage-Dependent Sodium Channel, W J. Culp, D T. Mckenzie Nov 1981

Detergent Extraction Of A Presumptive Gating Component From The Voltage-Dependent Sodium Channel, W J. Culp, D T. Mckenzie

Dartmouth Scholarship

A physiologically characterized radiolabeled neurotoxin complex obtained from venom of the scorpion Leiurus quinquestriatus has been used to identify detergent-solubilized presumptive sodium channel components in sucrose gradients. This toxin-binding component is found in extracts prepared from three sources of excitable membrane but appears to be absent from similar extracts prepared from nonexcitable membrane or from Torpedo californica membrane. Procedures that destroy the physiological activity of the Leiurus neurotoxin lead to a corresponding loss of toxin binding to the putative sodium channel component. The major component recognized by the Leiurus toxin sediments at 6.5 S. Scatchard analysis of quantitative binding experiments …

The Isolation Of Ecdysterone Inducible Genes By Hybridization Subtraction Chromatography, Michael P. Vitek, Susan G. Kreissman, Robert H. Gross Mar 1981

The Isolation Of Ecdysterone Inducible Genes By Hybridization Subtraction Chromatography, Michael P. Vitek, Susan G. Kreissman, Robert H. Gross

Dartmouth Scholarship

We have developed a procedure for selectively enriching a mRNA population for inducible sequences. Other than the induced mRNA species, the population of mRNA in control cells is approximately the same as the mRNA population in induced cells. Cytoplasmic mRNA from control cells is bound to oligo (dT)-cellulose and used as a template for reverse transcriptase, the oligo (dT) serving as a primer. After removing the template mRNAs, the cDNA-cellulose column is used to hybridize a population of mRNAs from induced cells. The non-hybridized poly A + RNAs are greatly enriched in the inducible sequences. We have used this technique …

The Limits Of Legislation In Achieving Social Change, Theodore S. Meth Jan 1981

The Limits Of Legislation In Achieving Social Change, Theodore S. Meth

Laws and Legislation Collection

This paper is about law, not laboratory animals or philosophical ethics. It proceeds from the premise that law is an appropriate, perhaps inevitable, instrument for dealing with ethical issues related to the use of research animals.
