Nf04-598 Rabies In Nebraska, Annette K. Bredthauer, David R. Smith Jan 2004

Nf04-598 Rabies In Nebraska, Annette K. Bredthauer, David R. Smith

University of Nebraska-Lincoln Extension: Historical Materials

Rabies is a deadly virus infection that can attack the central nervous system of most warm blooded animals, including humans. Rabies is a very old disease. The first recorded description of the disease dates from the 23rd century BC in Babylon. It was rampant in Europe in the 19th century and was carried to the Americas by animals on the early ships.

This NebFact describes rabies in animals and explains how people can protect themselves, as well as their pets or livestock from rabies exposure.

Effect Of Antisense Oligodeoxynucleotides For Icam-1 On Renal Ischaemia–Reperfusion Injury In The Anaesthetised Rat, L.V. Kiew, M. Abdul Sattar, C.H. Law, N.A. Abdullah, N. Abdul Rahman, K. Khalifah Sidik, E.J. Edward J. Johns

Research Publications (2000 to 2005)

An antisense oligodeoxynucleotide (As-ODN) to the 3’ untranslated region of the mRNA sequence expressing the intracellular adhesion molecule-1 (ICAM-1) was employed to determine ICAM-1’s role in renal ischaemia–reperfusion injury in the rat. Wistar-Kyoto rats receiving I.V. either lipofectin–As-ODN (As-ODN group), lipofectin–reverse ODN (Rv-ODN group) or lipofectin (ischaemia control group) 8 h prior to study were anaesthetized and subjected to 30 min of renal artery occlusion. Renal haemodynamic and excretory parameters were monitored before and after renal ischaemia. On termination of the study renal tissue was subjected to histological and Western blot analysis. Renal blood flow decreased in the 3 h …

Optimal Beaver Population Management Using Reduced Order Distributed Parameter Model And Single Network Adaptive Critics, Radhakant Padhi, S. N. Balakrishnan Jan 2004

Optimal Beaver Population Management Using Reduced Order Distributed Parameter Model And Single Network Adaptive Critics, Radhakant Padhi, S. N. Balakrishnan

Mechanical and Aerospace Engineering Faculty Research & Creative Works

Using a distributed parameter model for beaver population that accounts for their spatial and temporal behavior, an optimal control for a desired distribution of the animals is presented. Optimal solutions are obtained through a "single network adaptive critic" (SNAC) neural network architecture. The objective of this research is to design an "optimal" beaver harvesting scheme for a region of interest.

Quantitative Analysis Of Hemocyte Morphological Abnormalities Associated With Campoletis Sonorensis Parasitization, Matthew W. Turnbull, Stacy B. Martin, Bruce A. Webb

Entomology Faculty Publications

Endoparasitoids of arthropods evoke host cellular immune responses that result in hemocytic encapsulation of the endoparasitoid, unless these responses are disrupted by the parasite. Our interest has focused on mutualistic viruses found in some hymenopteran endoparasitoids that disrupt hemocyte function and prevent encapsulation. Specifically, the Campoletis sonorensis polydnavirus interacts with wasp factors to suppress immunity via expression of intracellular and secreted viral proteins. To study the roles of specific parasitization-associated factors on immunocyte morphology, fluorescence microscopy was used to visualize the actin cytoskeleton in infected and uninfected cells, or after treatment with C. sonorensis ovarian proteins or plasma from infected …

Dartmouth Scholarship

Quorum sensing triggers virulence factor expression in medically important bacterial pathogens in response to a density-dependent increase in one or more autoinducing pheromones. Here, we show that phagocyte-derived oxidants target these autoinducers for inactivation as an innate defense mechanism of the host. In a skin infection model, expression of phagocyte NADPH oxidase, myeloperoxidase, or inducible nitric oxide synthase was critical for defense against a quorum-sensing pathogen, Staphylococcus aureus, but not for defense against a quorum sensing-deficient mutant. A virulence-inducing peptide of S. aureus was inactivated in vitro and in vivo by reactive oxygen and nitrogen intermediates, including HOCl and ONOO(-). …

Dartmouth Scholarship

The stem–loop binding protein (SLBP) binds the 3′ end of histone mRNA and is present both in nucleus, and in the cytoplasm on the polyribosomes. SLBP participates in the processing of the histone pre-mRNA and in translation of the mature message. Histone mRNAs are rapidly degraded when cells are treated with inhibitors of DNA replication and are stabilized by inhibitors of translation, resulting in an increase in histone mRNA levels. Here, we show that SLBP is a component of the histone messenger ribonucleoprotein particle (mRNP). Histone mRNA from polyribosomes is immunoprecipitated with anti-SLBP. Most of the SLBP in cycloheximide-treated cells …

A Continuing Whimsical Search For The True Meaning Of The Term “Product” In Products Liability Litigation, Charles E. Cantú Jan 2004

A Continuing Whimsical Search For The True Meaning Of The Term “Product” In Products Liability Litigation, Charles E. Cantú

Faculty Articles

More than a decade has elapsed since an initial attempt was made to discern the true meaning of the term product in products liability litigation. At the time, a brief history of events leading up to the adoption of Section 402A of the Restatement (Second) of Torts was outlined, and it was emphasized that what had at first seemed so simple subsequently proved to be somewhat complex.

An examination of cases involving the sales/service transaction, as well as those involving real estate, blood, electricity, component parts, water, computer software, and ideas, sometimes held that what was involved was a product. …

Dogs, Domestication, And The Ego, Gary Shapiro Jan 2004

Dogs, Domestication, And The Ego, Gary Shapiro

In Zarathustra's "On the Vision and the Riddle," three animals-a spider, a snake, and a dog-make significant appearances, as do three human or quasihuman figures-Zarathustra himself, the dwarf known as the Spirit of Gravity, and the shepherd who must bite off the head of the snake. Of these animals, it is the dog who receives the most extended attention. Here, in the passage that along with "The Convalescent" (with its eagle and serpent) is usually and rightly taken to be Nietzsche's most articulate and yet highly veiled approach to explaining the teaching of eternal recurrence, the riddling vision involves animals. …

Regulation Of Development By Rx Genes, Travis J. Bailey, Heithiem El-Hodiri, Li Zhang, Rina Shah, Peter H. Mathers, Milan Jamrich

Biology

The paired-like homeobox-containing gene Rx has a critical role in the eye development of several vertebrate species including Xenopus, mouse, chicken, medaka, zebrafish and human. Rx is initially expressed in the anterior neural region of developing embryos, and later in the retina and ventral hypothalamus. Abnormal regulation or function of Rx results in severe abnormalities of eye formation. Overexpression of Rx in Xenopus and zebrafish embryos leads to overproliteration of retinal cells. A targeted elimination of Rx in mice results in a lack of eye formation. Mutations in Rx genes are the cause of the mouse mutation eyeless (ey1), the …

Etoposide Differentially Affects Bone Marrow And Dermal Derived Endothelial Cells, S. W. Adams, Lin Wang, J. Fortney, Laura F. Gibson

Faculty & Staff Scholarship

Chemotherapy alteration of the bone marrow microenvironment has the potential to influence hematopoietic recovery following transplantation. To discern the effect of specific drugs on components of the complex marrow microenvironment, in vitro models have significant utility. In the current study we sought to determine whether dermal (HMEC-1) and marrow derived endothelial cells (BMEC-1) respond differently to identical chemotherapy exposure. BMEC-1 cells were consistently more sensitive to etoposide exposure than HMEC-1 cells, measured as reduced viability. BMEC-1 also had reduced focal adhesion kinase (FAK) and VCAM-1 protein expression following chemotherapy, in contrast to dermal derived endothelial cells in which neither protein …

Creation Of Non-Human Primate Neurogenetic Disease Models By Gene Targeting And Nuclear Transfer, Robert B. Norgren Jan 2004

Creation Of Non-Human Primate Neurogenetic Disease Models By Gene Targeting And Nuclear Transfer, Robert B. Norgren

Journal Articles: Genetics, Cell Biology & Anatomy

Genetically modified rhesus macaques are necessary because mouse models are not suitable for a number of important neurogenetic disorders; for example, Kallmann's syndrome, Lesch-Nyhan's disease and Ataxia-Telangiectasia. Mouse models may not be suitable because there may be no mouse ortholog of the human gene of interest, as is the case for Kallmann's syndrome, or because mutant mice do not exhibit the same phenotype observed in humans, as is the the case for Lesch-Nyhan's disease and Ataxia-Telangiectasia. Non-human primate models of neurogenetic diseases are expected to more closely resemble human diseases than existing mouse models. Genetically modified rhesus macaques can be …

B lymphocytes express phospholipase C-gamma(1) (PLC-gamma(1)) and phospholipase C-gamma(2) (PLC-gamma(2)) isozymes. However, the relative importance of these two isozymes in B cell signaling is not known. We report here the identification and analysis of a B cell line deficient in PLC-gamma(1). Mature splenic B lymphocytes and a panel of cell lines representing pre-B, immature and mature B cell stages expressed phospholipase C-gamma (PLC-gamma), but not the beta or delta isoforms of phospholipase C (PLC). While all the tested B cell lines and primary splenic B cells expressed PLC-gamma(1) and PLC-gamma(2) isozymes, the L1.2 B cell line exclusively expressed PLC-gamma(2), but …

Journal Articles: Regenerative Medicine

The SDF-1alpha/CXCR4 ligand/chemokine receptor pair is required for appropriate patterning during ontogeny and stimulates the growth and differentiation of critical cell types. Here, we demonstrate SDF-1alpha and CXCR4 expression in fetal pancreas. We have found that SDF-1alpha and its receptor CXCR4 are expressed in islets, also CXCR4 is expressed in and around the proliferating duct epithelium of the regenerating pancreas of the interferon (IFN) gamma-nonobese diabetic mouse. We show that SDF-1alpha stimulates the phosphorylation of Akt, mitogen-activated protein kinase, and Src in pancreatic duct cells. Furthermore, migration assays indicate a stimulatory effect of SDF-1alpha on ductal cell migration. Importantly, blocking …

In Vitro Analog Of Classical Conditioning Of Feeding Behavior In Aplysia, Riccardo Mozzachiodi, Hilde A. Lechner, Douglas A. Baxter, John H. Byrne

Faculty, Staff and Student Publications

The feeding behavior of Aplysia californica can be classically conditioned using tactile stimulation of the lips as a conditioned stimulus (CS) and food as an unconditioned stimulus (US). Moreover, several neural correlates of classical conditioning have been identified. The present study extended previous work by developing an in vitro analog of classical conditioning and by investigating pairing-specific changes in neuronal and synaptic properties. The preparation consisted of the isolated cerebral and buccal ganglia. Electrical stimulation of a lip nerve (AT4) and a branch of the esophageal nerve (En2) served as the CS and US, respectively. Three protocols were used: paired, …

Il-23 Produced By Cns-Resident Cells Controls T Cell Encephalitogenicity During The Effector Phase Of Experimental Autoimmune Encephalomyelitis, Burkhard Becher, Brigit G. Durell, Randolph J. Noelle Oct 2003

Il-23 Produced By Cns-Resident Cells Controls T Cell Encephalitogenicity During The Effector Phase Of Experimental Autoimmune Encephalomyelitis, Burkhard Becher, Brigit G. Durell, Randolph J. Noelle

Dartmouth Scholarship

CNS-resident cells, in particular microglia and macrophages, are a source of inflammatory cytokines during inflammation within the CNS. Expression of IL-23, a recently discovered cytokine, has been shown to be critical for the development of experimental autoimmune encephalomyelitis (EAE) in mice. Expression of the p40 subunit of IL-12 and IL-23 by microglia has been shown in situ and in vitro, but direct evidence for a functional significance of p40 expression by CNS cells during an immune response in vivo is still lacking. Here we report that p40 plays a critical role in maintaining encephalitogenicity during the disease course. By using …

Transcriptional Inhibition Of Type I Collagen Gene Expression In Scleroderma Fibroblasts By The Antineoplastic Drug Ecteinascidin 743., Natalia Louneva, Biagio Saitta, David J Herrick, Sergio A. Jimenez Oct 2003

Transcriptional Inhibition Of Type I Collagen Gene Expression In Scleroderma Fibroblasts By The Antineoplastic Drug Ecteinascidin 743., Natalia Louneva, Biagio Saitta, David J Herrick, Sergio A. Jimenez

Department of Medicine Faculty Papers

We previously showed that COL1A1 expression is up-regulated at the transcriptional level in systemic sclerosis (SSc) fibroblasts and that the CCAAT-binding factor (CBF) is involved in this increased expression. Ecteinascidin 743 (ET-743) is a chemotherapeutic agent that binds with sequence specificity to the minor groove of DNA and inhibits CBF-mediated transcriptional activation of numerous genes. Therefore, we examined the effects of ET-743 on the increased COL1A1 expression in SSc fibroblasts. The drug caused a potent and dose-dependent inhibition of type I collagen biosynthesis, which reached 70-90% at 700 pM without affecting cell viability. The same drug concentration caused 60-80% reduction …

Aspidoderid Nematodes From Bolivian Armadillos, With The Description Of A New Species Of Lauroia (Heterakoidea: Aspidoderidae), F Agustín Jiménez-Ruiz, Scott L Gardner

Publications

One nine-banded armadillo (Dasypus novemcinctus) and 1 yellow armadillo (Euphractus sexcinctus) were necropsied in the field during an expedition to collect parasites of mammals in Bolivia. A total of 205 Aspidodera binansata Railliet and Henry, 1913 (Heterakoidea: Aspidoderidae), and 40 specimens of Lauroia bolivari n. sp. (Heterakoidea: Aspidoderidae) were recovered from the cecum and large intestines of D. novemcinctus and E. sexcinctus. Aspidodera esperanzae Fujita et al., 1995, is proposed as a junior synonym of A. binansata based on the structure of the cordons on the hood. Lauroia bolivari n. sp. has an undercut cephalic …

Il-7 Enhances Peripheral T Cell Reconstitution After Allogeneic Hematopoietic Stem Cell Transplantation., Onder Alpdogan, Stephanie J Muriglan, Jeffrey M Eng, Lucy M Willis, Andrew S Greenberg, Barry J Kappel, Marcel R M Van Den Brink

Department of Medical Oncology Faculty Papers

We used clinically relevant murine allogeneic bone marrow transplantation (BMT) models to study the mechanisms by which IL-7 administration can improve posttransplant peripheral T cell reconstitution. After transplant we could distinguish two populations of mature donor T cells: (a) alloreactive T cells with decreased expression of CD127 (IL-7 receptor alpha chain) and (b) nonalloreactive T cells, which express CD127 and undergo homeostatic proliferation. IL-7 administration increased the homeostatic proliferation of nonalloreactive T cells, but had no effect on alloreactive T cells and the development of graft-versus-host disease. Allogeneic transplant of purified hematopoietic stem cells and adoptive transfer of thymocytes into …

A Null Mutation For Tissue Inhibitor Of Metalloproteinases-3 (Timp-3) Impairs Murine Bronchiole Branching Morphogenesis., Sean E Gill, M Cynthia Pape, Rama Khokha, Andrew J Watson, Kevin J Leco

Obstetrics & Gynaecology Publications

Tissue inhibitors of metalloproteinases (TIMPs) regulate extracellular matrix (ECM) degradation by matrix metalloproteinases (MMPs). We have examined the role of TIMP-3 on ECM homeostasis and bronchiole branching morphogenesis during murine embryogenesis. Employing an in vitro organ culture system, we found decreased bronchiolar branching in null lungs when compared with wild type (WT) counterparts after 2 days in culture. When a synthetic inhibitor of MMPs at low dose was added to the culture system, branching was augmented regardless of genotype. Gelatin and in situ zymography revealed that null lungs exhibited enhanced activation of MMPs throughout lung development. We analysed the impact …

Quantitation Of Contacts Among Sensory, Motor, And Serotonergic Neurons In The Pedal Ganglion Of Aplysia, Han Zhang, Marcy Wainwright, John H. Byrne, Leonard J. Cleary

Faculty, Staff and Student Publications

Present models of long-term sensitization in Aplysia californica indicate that the enhanced behavioral response is due, at least in part, to outgrowth of sensory neurons mediating defensive withdrawal reflexes. Presumably, this outgrowth strengthens pre-existing connections by formation of new synapses with follower neurons. However, the relationship between the number of sensorimotor contacts and the physiological strength of the connection has never been examined in intact ganglia. As a first step in addressing this issue, we used confocal microscopy to examine sites of contact between sensory and motor neurons in naive animals. Our results revealed relatively few contacts between physiologically connected …
