Mitochondria are essential for survival and as such, impairments in organelle homeostasis significantly accelerate age-related morbidity and mortality. Here, we determined the contribution of bioenergetic efficiency to life span and health span in Drosophila melanogaster utilizing the mitochondrial uncoupler BAM15. Life span was determined in flies fed a normal diet (ND) or high fat diet (HFD) supplemented with vehicle or BAM15. Locomotor function was determined by negative geotaxis assay in middle-aged flies fed vehicle or BAM15 under ND or HFD conditions. Redox capacity (high-resolution respirometry/fluorometry), citrate synthase (enzyme activity), mtDNA content (qPCR), gene expression (qPCR), and protein expression (western blot) …

Examining The Risks And Resilience Of Older Adults Living With Hiv/Aids: Qualitative Community-Based Participatory Research Studies In Southern Nevada, Brandon Ranuschio

UNLV Theses, Dissertations, Professional Papers, and Capstones

Since the height of the HIV pandemic in the 1980s, medical interventions have significantly increased the life expectancy of people living with HIV/AIDS (PLWH) and made HIV/AIDS a livable, chronic health condition. Because of this, PLWH have been living longer lives and thriving with the help of HIV sector healthcare and service providers. However, with their longer life expectancy, older PLWH have been experiencing other health challenges. One such challenge is the risk of developing HIV-Associated Neurocognitive Disorder (HAND), which is a condition that affects up to 50% of PLWH at some point in their lifetime, with middle-aged and older …

Changes In Young Adults’ Perspective On Ageism After Participating In A Virtual Nutrition Education Program With Older Adults, Julianne Hensley

Electronic Theses and Dissertations

The growing older adult population has led to increased ageist tendencies among younger generations. Previous research suggests that ageism is associated with cognitive and physical decline among older adults. This mixed-methods research project examined changes in ageist perceptions among graduate student facilitators and undergraduate nutrition ambassadors after participating in a training that included information about healthy aging, addressing ageism, and communicating with older adults. Results from a secondary data analysis of pre-test/post-test data and thematic analysis of semi-structured interviews with facilitators of Socially Nutritious, a virtual nutrition education program for older adults, indicate that ageist perceptions decreased after the …

A Comprehensive Examination Of Age-Related Lower Limb Muscle Function Asymmetries Across A Variety Of Muscle Contraction Types, Kylee Heap

All Graduate Reports and Creative Projects, Fall 2023 to Present

Previous research has found that lower limb muscle asymmetries increase with age and are linked to fall and injury risks. However, past studies lacked a wide variety of muscle function modes and measures as well as comparison to a comparable young aged group. The purpose of this study was to examine age-related lower limb muscle function asymmetries across a variety of muscle contraction types and velocities in young and old adults. Lower limb muscle balance, strength, power, and velocity were evaluated with concentric, isometric, isotonic, and eccentric muscle actions during a single-leg stance test and on single- and multi-joint dynamometers …

Factors Influencing Driving Confidence In Older Adults, Jill Linder, Beth Ann Walker, Elizabeth Moore, Anne Hegberg Apr 2024

Factors Influencing Driving Confidence In Older Adults, Jill Linder, Beth Ann Walker, Elizabeth Moore, Anne Hegberg

The Open Journal of Occupational Therapy

Background: It has been determined that both confidence and cognition impact driving performance and self-regulation. While it is known that cognitive decline impacts driving safety and that decreases in driving confidence can lead to self-driving regulation, further research is needed to determine whether cognition, demographics, and driving routines are associated with driving confidence.

Method: A non-experimental, cross-sectional design study using a convenience sample of 100 older adults was conducted to determine if individual factors are related to daytime and nighttime driving confidence.

Results: Multiple regression indicated that four variables predicted daytime driving confidence, including cognition, driving at …

Peer-Mediated Family Support Project: Evaluation Of Changes In Family Quality Of Life, Preethy S. Samuel, Elizabeth Janks, Nia S. Anderson, Michael Bray, Christina Topolewski, Sharon Milberger

Developmental Disabilities Network Journal

The Family Quality of Life (FQOL) approach represents a paradigm shift from fixing to supporting people with intellectual/developmental disabilities (I/DD) by changing the focus from the individual to the family and highlighting strengths rather than deficiencies. Aging family caregivers of individuals with I/DD often encounter obstacles, including accessibility, acceptability, and affordability of services. Little is known about best practices to support aging families of adults with I/DD. Understanding how a state-wide peer-mediated family support project implemented in this study helped improve the FQOL of aging caregivers is important in broadening participation of other caregivers in these types of programs. To …

Calcific aortic stenosis can be considered a model for geriatric cardiovascular conditions due to a confluence of factors. The remarkable technological development of transcatheter aortic valve replacement was studied initially on older adult populations with prohibitive or high-risk for surgical valve replacement. Through these trials, the cardiovascular community has recognized that stratification of these chronologically older adults can be improved incrementally by invoking the concept of frailty and other geriatric risks. Given the complexity of the aging process, stratification by chronological age should only be the initial step but is no longer sufficient to optimally quantify cardiovascular and noncardiovascular risk. …

Calorie restriction (CR) with adequate nutrient intake is a potential geroprotective intervention. To advance this concept in humans, we tested the hypothesis that moderate CR in healthy young-to-middle-aged individuals would reduce circulating biomarkers of cellular senescence, a fundamental mechanism of aging and aging-related conditions. Using plasma specimens from the Comprehensive Assessment of Long-term Effects of Reducing Intake of Energy (CALERIE™) phase 2 study, we found that CR significantly reduced the concentrations of several senescence biomarkers at 12 and 24 months compared to an ad libitum diet. Using machine learning, changes in biomarker concentrations emerged as important predictors of the change …

Dietary restriction (DR) delays aging, but the mechanism remains unclear. We identified polymorphisms in mtd, the fly homolog of OXR1, which influenced lifespan and mtd expression in response to DR. Knockdown in adulthood inhibited DR-mediated lifespan extension in female flies. We found that mtd/OXR1 expression declines with age and it interacts with the retromer, which regulates trafficking of proteins and lipids. Loss of mtd/OXR1 destabilized the retromer, causing improper protein trafficking and endolysosomal defects. Overexpression of retromer genes or pharmacological restabilization with R55 rescued lifespan and neurodegeneration in mtd-deficient flies and endolysosomal defects in fibroblasts from patients with lethal loss-of-function …

Aging entails gradual functional decline influenced by interconnected factors. Multiple hallmarks proposed as common and conserved underlying denominators of aging on the molecular, cellular and systemic levels across multiple species. Thus, understanding the function of aging hallmarks and their relationships across species can facilitate the translation of anti-aging drug development from model organisms to humans. Here, we built AgeAnnoMO (https://relab.xidian.edu.cn/AgeAnnoMO/#/), a knowledgebase of multi-omics annotation for animal aging. AgeAnnoMO encompasses an extensive collection of 136 datasets from eight modalities, encompassing 8596 samples from 50 representative species, making it a comprehensive resource for aging and longevity research. AgeAnnoMO characterizes …

Age-Related Delay In Reduced Accessibility Of Refreshed Items, Julie A. Higgins, Matthew R. Johnson, Marcia K. Johnson Jan 2024

Age-Related Delay In Reduced Accessibility Of Refreshed Items, Julie A. Higgins, Matthew R. Johnson, Marcia K. Johnson

Department of Psychology: Faculty Publications

Previously we demonstrated that in young adults briefly thinking of (i.e., refreshing) a just-seen word impairs immediate (100ms delay) perceptual processing of the word, relative to words seen but not refreshed. We suggested that such reflective-induced inhibition biases attention towards new information. Here, we investigated whether reduced accessibility of refreshed targets dissipates with a longer delay, and whether older adults would show a smaller and/or delayed effect, versus young adults. Young and older adult participants saw two words, followed by a cue to refresh one. After either a 100ms or 500ms delay, participants read a word that was the …

Strategies For Integrating Aging And Older Adulthood Into Counselor Training, Matthew C. Fullen, Jordan Westcott, Kailey Bradley, Natese R. Dockery, Jungeun Lee, Rumbidzai Mushunje, Afroze N. Shaikh

Adultspan Journal

Following recent changes to Medicare mental health law, counselors will have expanded opportunities to work with older adults. Historically, there has been a dearth of counselor training dedicated to counseling older adults, necessitating greater attention to understanding and applying best practices for preparing counselors to serve older adult clients. Looking to professions that have a longer history of serving older adults, specifically gerontology, social work, and geropsychology, we describe approaches to training counselors to work with older adults that may prove helpful for the counseling profession.

Environmentalism Among Older Adults: Investigating Predictors Of Pro-Environmental Behavior In Later Life, Ajit Singh Mann

CGU Theses & Dissertations

Human actions are largely responsible for degradation of the natural environment, and environmental threats due to climate change are increasingly prevalent. Despite the immediate need to conserve the natural environment, the role of older adults in pro-environmental initiatives has largely been ignored. I conducted two studies to understand pathways for promoting pro-environmental behavior among older adults, with the goal of shedding light on older adults’ engagement in environmental action. In the first study, I investigated the indirect effect of age on pro-environmental behavior, through personal values and environmental concern, among a general sample (N=292). I considered the role of flow …

Sex-Driven Alterations In Aging Alpha Motoneurons: Exploring Size, Type, Density, And Kv2.1 Channel Expression, Kalin R. Gerber

Browse all Theses and Dissertations

This research explores how motor neurons (MNs) and Kv2.1 clustering change with age, emphasizing sex differences and MN subtypes. We found that MN density decreases with age in both sexes, while soma size increases in male mice. FF MNs were the most affected, and old weak mice had smaller MNs than their stronger counterparts, underscoring FF MN vulnerability. Baseline studies revealed that FF and FI MNs have larger Kv2.1 clusters compared to FR and S MNs. Female mice had smaller, denser Kv2.1 clusters than male mice, suggesting less clustering in females. With age, Kv2.1 clusters grew larger but became less …

Objectives: Deprived living environments contribute to greater heart failure (HF) risk among non-Hispanic Black persons, who disproportionately occupy disadvantaged neighborhoods. The mechanisms for these effects are not fully explicated, partially attributable to an insufficient understanding of the individual factors that contribute additional risk or resilience to the impact of neighborhood disadvantage on health. The objective of this study was, therefore, to clarify the complex pathways over which such exposures act to facilitate more targeted, effective interventions. Given the evidence for a mediating role of biological age and a moderating role of individual psychosocial characteristics in the neighborhood disadvantage-HF link, we …

Trillions of microbes live symbiotically in the host, specifically in mucosal tissues such as the gut. Recent advances in metagenomics and metabolomics have revealed that the gut microbiota plays a critical role in the regulation of host immunity and metabolism, communicating through bidirectional interactions in the microbiota-gut-brain axis (MGBA). The gut microbiota regulates both gut and systemic immunity and contributes to the neurodevelopment and behaviors of the host. With aging, the composition of the microbiota changes, and emerging studies have linked these shifts in microbial populations to age-related neurological diseases (NDs). Preclinical studies have demonstrated that gut microbiota-targeted therapies can …

Urinary aging is associated with decreased tissue responsiveness, decreased control over the voiding reflex, signaling dysfunction along the …

Impact Of The Built Environment On The End-Of-Life Journey, Kelechi Akwazie Dec 2023

Impact Of The Built Environment On The End-Of-Life Journey, Kelechi Akwazie

Masters in Architecture Program: Theses

Several research studies and personal anecdotes show that home-based hospice care patients report better quality of life than their inpatient care counterparts – suggesting that the location of care/built environment is a critical component of hospice care. As a result, other research studies have attempted to provide evidence-based design recommendations for inpatient hospice facilities; however, several of these recommendations either conflict with each other or are vaguely prescribed – which may dull any attempts to implement them.

This literature review takes a unique approach to the provision of evidence-based design recommendations for inpatient hospice facilities by holistically assessing hospice care, …
