Alzheimer's disease is characterized by complex molecular and cellular heterogeneity, which complicates efforts to identify consistent biomarkers and therapeutic targets. To better characterize the heterogeneity, we applied latent factor modeling to RNA sequencing data from approximately 2,500 human Alzheimer's disease brain samples, uncovering underlying patterns in gene expression. These transcriptional groups demonstrated unique gene expression profiles related to synaptic and neuronal pathways, vasculature development, and protein folding and antigen processing. Notably, this latent factor reflects variation in spatial sampling. Adjusting for the latent factor improved the identification of differentially expressed genes in disease samples. This finding suggests that spatial heterogeneity …

Single nucleotide variants (SNVs) and copy-number variant (CNV) deletions involving TBX4 have been associated with pulmonary arterial hypertension, ischiocoxopodopatellar syndrome, and lethal lung developmental disorders (LLDDs). Thus far, all large CNV deletions encompassing entire TBX4 have been found to have arisen de novo. Here, we present a three-generation family with three neonate siblings who died within 35-66 days due to histopathologically diagnosed LLDD. Whole-genome sequencing identified an ~108-kb CNV deletion encompassing TBX4 in all three infants. The deletion was also found in their mother with a history of pneumonia and persistent thick upper airway secretions and in the maternal grandfather …

The anterior cingulate cortex (ACC) is functionally closely related with the insula and the ventral lateral prefrontal cortex (vlPFC). The ACC and insula are the main hubs of the salience network, a set of functionally related brain regions involved in detecting salient stimuli and coordinating inter-network communication, while communication between ACC and vlPFC is closely linked to inhibitory control. However, despite strong indirect evidence from nonhuman primate (NHP) tract tracing, the structural connections between the ACC and the insula/vlPFC remain poorly understood in the human brain, in part because standard approaches fail to capture this projection. In this study, we …

Demographic Response Of Rana Uenoi To Land Use Near Breeding Sites: A Case Study From The Gayasan Region, South Korea, Hyun Kim, Hyun Tae Kim, Na Yeong Kim, Soo Dong Lee, Jared Wolfe

Michigan Tech Publications

Amphibians are highly sensitive to habitat disturbance because of their permeable skin and reliance on both aquatic and terrestrial environments. To evaluate how human-driven land-use change affects a forest-stream amphibian, we analysed long-term population dynamics of Rana uenoi in the Gayasan region of South Korea, with a focus on breeding habitats. Between 2007 and 2023, annual egg-mass surveys were conducted at 14 breeding sites, and land-use composition was extracted each year within a 500 m buffer from national geospatial databases. Land-use categories were reclassified into ecologically relevant groups (breeding, high-quality, low-quality and non-living/high-mortality habitats) and their areas were quantified for …

Faculty Perceptions Of Engaging Generation Z Students Through Positive Education In The Classroom, Ashley D. Ragsdale

Electronic Theses and Dissertations

This qualitative study examined the engagement gap between Generation Z students and faculty in higher education, emphasizing how positive education approaches can help faculty become more confident and effective instructors. Generation Z students prefer instant internet access and digital learning, have shorter attention spans, and tend to be disengaged when instructors rely on traditional teaching methods. After the COVID-19 pandemic, this engagement gap became more evident. This basic qualitative study used semistructured interviews, reflexive journaling, and nonparticipatory observations. Participants included full-time health science faculty at a private university in central Texas. Each had at least 2 years of teaching experience …

A virus-derived serpin, Serp-1, has proven efficacy in treating inflammatory and coagulation disorders in preclinical and clinical studies. Serp-1 evolved over millions of years to block host immune responses, targeting serine proteases in immune and coagulation pathways. Treatment with PEGylated Serp-1 (PEGSerp-1) protein reduced lung injury in both lupus lung hemorrhage and SARS-CoV-2 models. Here, PEGSerp-1 effects on immune-coagulopathic responses is examined in a mouse colitis model. Inflammatory bowel disease (IBD) is associated with life-threatening complications with severe inflammation, bleeding, vasculitis, cancer and toxic megacolon. Serine protease cascades activate coagulation and complement pathways throughout the human body and are regulated …

Mir-302 Regulates Pancreatic Progenitor Pool And Pancreatic Size, Ziyue Z Yang, Caroline G Snider, Ronald J Parchem

Faculty, Staff and Students Publications

Disruptions in pancreatic development can lead to health issues such as pancreatic agenesis and congenital diabetes mellitus. Understanding pancreatic organogenesis is critical for elucidating disease mechanisms and developing regenerative therapies. The pancreas consists of endocrine and exocrine cells, both of which are derived from multipotent progenitor cells (MPCs). MPC proliferation and differentiation are tightly controlled by multiple mechanisms, including post-transcriptional regulation by miRNAs. However, these regulatory factors are not fully understood. Here, we profiled miRNA expression in MPCs and identified that mir-302 was highly enriched during the earliest stages of pancreatic development. Loss of mir-302 resulted in reduced pancreatic size …

Faculty, Staff and Students Publications

Introduction: Erythropoiesis-stimulating agents (ESAs) and iron supplementation are standard treatments for chronic kidney disease (CKD)-related anemia. Targeting higher hemoglobin values in CKD increases cardiovascular risk. Whether the increased risk is from higher ESA doses or higher hemoglobin levels is uncertain, but alternative therapies are sought for patients requiring high ESA doses. Phase 3 INNO2VATE trials in patients with dialysis-dependent CKD (DD-CKD) demonstrated vadadustat's noninferiority compared with darbepoetin alfa. To determine vadadustat's potential to treat anemia, including in patients requiring high ESA doses, its safety and efficacy were compared with those of darbepoetin alfa across prespecified baseline ESA dose subgroups in …

Gallbladder Decompression And Hydrodissection As Adjuncts For Ablation Of Pericholecystic Hepatocellular Carcinomas, Taylor Hoffman, Arta Kasaeian, Johnathon Chung, Ashkan Berenji, David Wynne, Mohammad Ghasemi-Rad, David Léon

Faculty, Staff and Students Publications

Microwave ablation is an effective treatment for hepatocellular carcinoma, but tumors adjacent to extrahepatic organs carry a higher risk for thermal injury. Thermoprotective techniques such as hydrodissection and gallbladder decompression can reduce this risk and expand the range of lesions amenable to percutaneous treatment. We present a case of a 68-year-old woman with hepatocellular carcinoma in hepatic segment V, with partial treatment response from prior transarterial chemoembolization. Due to the lesion's proximity to the gallbladder and transverse colon, microwave ablation was performed with concurrent ultrasound- and CT-guided gallbladder decompression and hydrodissection. The ablation was technically successful without post-procedural complications. At …

Spinning-Induced Exertional Rhabdomyolysis Without Acute Kidney Injury, Yolanda V. Gutierrez, Ronald Shaju, Alyssa L. Sepulveda, Joaquin Rivera, Aamir Ahmad

School of Medicine Publications

Rhabdomyolysis is a condition with a myriad of causes, including both traumatic and non-traumatic. Traumatic conditions are observed in crush injuries, compartment syndrome, and fractures, while non-traumatic rhabdomyolysis is commonly seen due to exertional heat stroke, extreme physical exercise, and dehydration. This case report explores two cases of extreme non-traumatic rhabdomyolysis caused by indoor cycling, also known as spinning. A common complication of severe rhabdomyolysis is acute kidney injury (AKI), which increases the risk of renal failure. Rapid interventions such as aggressive intravenous fluids are necessary to prevent kidney function loss. In this case report, two sisters, on two separate …
