The scope of RNA-based therapeutics continues to expand as more siRNA, mRNA, and CRISPR-based treatments are approved for various medical applications. Purification of synthetic RNA drugs is inherently challenging and requires considerable time and resources. New cost-effective and efficient purification strategies will expedite drug development process and will lower the drug costs. In this thesis I describe two generations of photolabile linkers for the non-chromatographic purification of therapeutic RNAs. This doctoral dissertation on the foundations of quantum theory tells the story of a conceptual protagonist I have called “Epistemic Constraint.” Here, epistemic constraints are the definite, intersubjectively agreeable, ordinary-language conditions under which experiments are described.

The usual formulation of the quantum measurement problem, which I call the Schrodingerian measurement problem, has the structure of an anomaly: if we take quantum theory at face value, we expect no definite values, and yet we see definite values in experiments. Extreme precipitation events in the South American Altiplano present significant hazards to communities in the Andean region, yet their future characteristics under anthropogenic climate change remain uncertain because of complex terrain and limitations of course-resolution climate models. This study evaluates the ability of South America Affinity Group Weather Research and Forecasting (SAAG-WRF) model with 4-km grid-spacing to reproduce 2000-2015 temperature, precipitation, and extreme precipitation events over the Altiplano. Examination of projected future changes is completed using the SAAG-WRF pseudo global warming (WRF PGW) simulation representing an approximately 3°C warmer global climate. This study uses atmospheric reanalysis, precipitation and snow water equivalent (SWE) analyses, mesonet observations, and streamflow records to examine the impact of an atmospheric river storm that produced heavy rainfall and snowpack ablation across the Catskill Mountains of New York on 24–25 December 2020. Earlier in December, an antecedent storm produced SWE values in the Catskills above the 90^th percentile for that point in the season. The atmospheric river event followed with strong southerly warm-air advection ahead of an anomalously deep trough, temperatures and dewpoints that exceeded 10°C, and heavy orographically enhanced rainfall and rapid snowmelt. This dissertation asks how multicultural adolescents in South Korea develop their attitudes toward crossing group boundaries in social and intimate life—whom they would accept as a close friend, a romantic partner, and a spouse—and what factors shape these attitudes. Korea’s immigrant population has grown more than seventyfold since 1990, largely through marriage migration, and the children of those unions are now reaching adulthood. Research on these adolescents has focused on how well they adjust, leaving open how they themselves come to view relationships across group lines.

Transportation agencies make critical operational decisions during hazardous weather events, including assessment of road conditions and resource allocation. In this study, machine learning models are developed to provide additional support for the New York State Department of Transportation (NYSDOT) by automatically classifying road conditions across the state. Convolutional neural networks and random forests are trained on NYSDOT roadside camera images and weather data to predict road surface conditions. Extreme rainfall is a persistent hazard in the Hawaiian Islands, where steep terrain and short catchments convert heavy rain into damaging floods with little warning. The trade wind inversion, a subsidence-driven stable layer that caps the marine boundary layer over the subtropical North Pacific, limits the depth of convection and is therefore expected to govern whether available moisture can be converted into extreme rainfall. This thesis details the development of multivalence-nanostructured vanadium oxide (VOₓ) thin films for uncooled microbolometer applications, with a systematic optimization of magnetron-sputtering parameters. The temperature coefficient of resistance (TCR), resistivity, and optical response of VOₓ thin-film sensing layers are controlled by valence composition, grain growth, and surface morphology. The primary goal was to achieve a high TCR with low resistivity to improve thermal detector performance.

This dissertation examines the persistence and spatial concentration of gender-based educational inequality in Afghanistan's public school system from 2002 to 2021–– a period spanning post-2001 reconstruction, the expansion of the Republic era, and the reversal under the second Taliban regime. Despite substantial international investment and two decades of repeated commitments to gender parity, access to and progression through public schooling remained profoundly uneven across the country’s thirty-four provinces. The Gender Parity Index values differ sharply between provinces, and those with the lowest overall enrollment tend to exhibit the widest gender gaps. One of these processes is ventilation: the injection of relatively cool, dry environmental air into the warm core of a TC that acts as “anti-fuel”, inhibiting intensification and increasing uncertainty with intensity forecasts. We analyze the underlying geometry of the solution to an IVP for a matrix diffusion PDE. We first derive the fundamental solution to the PDE. We then determine the unique solution to the IVP. From there, we begin analyzing its underlying geometry. We first observe that the geometry exhibits an ellipsoidal nature. Furthermore, we observe that it is described by the principal semi-axis geometry of the ellipsoids associated with the solution. This conclusion follows from applying the Principal Axis Theorem to the associated ellipsoids to establish their principal semi-axis geometry, as governed by the eigenstructure of the matrix. This dissertation studies the dynamic effects of economic shocks under uncertainty across three self-contained essays.

The first essay introduces a two-stage model averaging estimator for impulse response functions that flexibly combines local projection (LP) and vector autoregression (VAR) methods, as well as different statistical paradigms (Bayesian or frequentist). In the first stage, we compute weights within the LP and VAR groups. A second-stage weight then integrates the two groups by accounting for their structural differences and incorporating their respective strengths. BACKGROUND: Prior studies suggest that longer drive time (DT) to specialists and higher area deprivation index (ADI) are linked to worse outcomes in inflammatory bowel disease (IBD), but their impact on postileocolic resection (ICR) care in patients with Crohn's disease (CD) is not well defined.

OBJECTIVES: We aimed to evaluate disparities in post-ICR care based on DT and ADI.

OBJECTIVES: We evaluated Mycoplasma genitalium (MG) positivity and co-infection with Chlamydia trachomatis (CT) and/or Neisseria gonorrhoeae (NG), based on results from a trial assessing clinical performance of the Cobas® Liat CT/NG/MG point-of-care test.

METHODS: A prospective, US, multicenter, noninterventional study assessed MG positivity in prospective urine samples (male) and clinician-collected vaginal swabs (female) from symptomatic/asymptomatic patients aged ≥14 years attending various clinical settings. Participants were designated positive or negative for MG, CT, and NG based on combined results from three US Food and Drug Administration-approved assays and one laboratory-developed test. Insulin resistance accompanied by hepatic steatosis is a common complication of obesity. In an effort to identify plasma lipids that could be biomarkers or causes of insulin resistance with steatosis in people with obesity, we evaluated the plasma lipidome in three distinct groups separated by adiposity, hepatic steatosis, and insulin sensitivity, assessed by using the hyperinsulinemic-euglycemic clamp procedure: i) insulin-sensitive lean (ISL, n = 13); ii) insulin-sensitive obese (ISO, n = 14); and iii) insulin-resistant obese with hepatic steatosis (IROS, n = 13). PURPOSE: Same-day surgery (SDS) cancellations have significant implications on patients and hospitals. This study aimed to identify demographic and socioeconomic factors that impact SDS cancellations across a single pediatric health system.

METHODS: A case-control retrospective review captured patients (age < 18) who underwent SDS procedures from July 2022 to August 2024. Patients were included if they underwent any procedure at three operative sites: a tertiary pediatric academic hospital (PAH) and two outpatient surgery centers (OSC). Patients whose SDS was cancelled (n = 2,034) were compared to those whose surgeries proceeded as scheduled (n = 25,573). Patient zip codes were matched to Child Opportunity Index (COI) 2.0 data.

RESULTS: The overall SDS cancellation rate was 7.37 %, with a significantly higher rate observed at the PAH compared to OSC (11.4 % vs 53 %, p < 0.001). Patients with cancelled cases were more likely to identify as Black and have the lowest quintile COI (p < 0.001). Cancelled in preop and no shows were two reasons with the most racial disparities. In a sub-analysis comparing cancelled cases between operating sites, PAH cancelled patients tended to be Black (23.8 % vs 11.7 %, p < 0.001) and have very low COI (20.1 % vs. 10.6 %, p < 0.001).

CONCLUSION: SDS cancellation rates are significantly higher amongst patients who identify as Black and reside in low opportunity neighborhoods. Disparities in cancellation rate are more significant in patients scheduled at the urban PAH compared …

This secondary data analysis aimed to determine the nature of the relationship between obstructive sleep apnea (OSA) risk, biological age acceleration, and nonspecific chronic low back pain (CLBP). In total, 199 adults aged 18-82 years who filled both STOPBANG and pain questionnaires subsetted for secondary analysis. Based on the STOPBANG questionnaire, 104 had a low OSA risk and 95 had an intermediate or high OSA risk. Inter-brain synchrony (IBS), measured by hyperscanning, refers to the synchronization of multiple individuals' brain activities during social interactions. Traditional fNIRS-based hyperscanning suffers shortcomings like low spatial resolution and high susceptibility to superficial interference, causing imprecise estimation of IBS in complex social tasks. BACKGROUND: Vascular anomalies (VAs) are rare diseases that cause significant morbidity. Adults with VAs struggle to access expert care. Hematologists and oncologists in adult medicine are positioned to provide expert care, but no prior studies have characterized their openness to this role.

OBJECTIVES: This study characterized the experience, confidence, willingness, and motivating factors for hematologists/oncologists to care for adults with VAs.
