Background and Aim The present study aims to evaluate the protective role of Moringa olifera (MO) extract against Zinc Oxide Nanoparticles (ZnO NPs) induced immunotoxicity in male albino rats. Adolescence is a critical developmental period during which behavioral and physical health outcomes are shaped by social and family determinants of health; yet limited research has examined how these determinants operate among adolescents with intellectual and developmental disabilities (IDD) within a multilevel ecological framework. To address this gap, this thesis used data from Years 9 and 15 of the Future of Families and Child Wellbeing Study (FFCWS), with a final analytic sample of 3,176 adolescents at Year 15, including 150 with IDD. In this study, three EEG datasets, created via different levels of pre-processing, were used to train VO2 prediction models whose performance was tested and compared. Understanding the link between EEG and VO2 during walking is important when working in rehabilitation or studying metabolic energy optimization. Common methods of measuring these two bio-signals tend to be bulky, uncomfortable, and inefficient, so finding a way to predict VO2 from EEG via 1D CNN model training is a worthy pursuit.  It was hypothesized that, due to a CNN needing more data to find patterns for prediction, a minimally processed EEG dataset would have …

Vitamin B12 deficiency is classically associated with megaloblastic anaemia and neurological manifestations; however, atypical presentations continue to pose diagnostic challenges. Fever in association with thrombocytopenia often prompts evaluation for infectious aetiologies such as dengue, malaria, or sepsis, particularly in tropical settings. We report a case of vitamin B12 deficiency presenting with pyrexia and isolated thrombocytopenia, initially suspected to be an infectious illness. Extensive evaluation failed to reveal an infective cause, and subsequent haematological workup demonstrated megaloblastic changes with low serum vitamin B12 levels. Remarkably, the patient showed rapid defervescence and normalisation of platelet counts following parenteral vitamin B12 therapy. This …

Latinos continue to experience many logistical and systemic barriers to professional mental health services (Derr, 2016) despite being the second-largest racial-ethnic group in the United States (U.S.). Background and Objectives: Immigrants in the United States undergo acculturation — a process of sociocultural and behavioral adaptation to the host country. Increasingly, behavioral adaptations have been associated with cardiometabolic health and chronic disease risk. Physical frailty is a prevalent geriatric syndrome characterized by age-related decline across multiple physiological systems. Physical frailty is associated with an elevated risk of adverse health outcomes, including chronic disease progression and all-cause mortality, representing a significant and growing public health concern. Disrupted circadian rest-activity rhythms (RARs) are increasingly recognized as clinically meaningful markers of physical frailty in older adults. Background/objectives: Phenylketonuria (PKU) is an autosomal recessive inborn error of metabolism. If untreated, elevated blood phenylalanine (Phe) levels lead to neurological and behavioral impairments. Phe levels can be managed through a lifelong Phe-restricted diet; however, this can be challenging to maintain. Sepiapterin is an adjunct therapy for PKU that has shown efficacy in reducing blood Phe levels in both tetrahydrobiopterin (BH4)-responsive and BH4-non-responsive people with PKU in clinical trials. Practical guidance is needed for dietitians and other healthcare professionals supporting the dietary management of people with PKU who are initiating or receiving sepiapterin.

Background: The ReCePI randomized, controlled trial compared amustaline/glutathione pathogen-reduced (PR) and conventional red blood cell (RBC) transfusions in cardiac surgery. Study design and methods: We performed a post-hoc analysis of hemoglobin (Hb) utilization and safety outcomes. Hb content of study RBCs was measured at manufacture. Subject Hb levels, estimated blood loss and blood product use were recorded during and for 7 days post-surgery. Acute kidney injury (AKI) incidence was graded over 7 days, and adverse events were assessed for 28 days. Results: PR-RBCs contained 5.2% less Hb (mean [SD] Test 58.1 [6.5] g; Control 61.5 [6.8] g Hb, p <.001) due to manufacturing volume losses. Four hundred fifty-six PR- and 524 conventional RBCs were transfused to 159 Test and 162 Control subjects, respectively. Mean subject baseline and post-surgery Hb levels, and blood loss were not different between groups. Test subjects tended to require fewer total RBCs (3.5 [2.7] Test vs. 4.0 [3.5] Control, p =.08) resulting in 17.6% significantly less mean total Hb transfused (Test 203.2 g; Control 246.8 g Hb, p =.01). The proportions of subjects transfused with, and the number of platelet and cryoprecipitate units transfused were not different. Test subjects received less plasma (2.3 [1.8] units Test vs. 3.5 [3.8] Control, p =.03). Overall incidence of adverse events and AKI were not clinically different. Imbalances in AKI severity (Grade III Test 15/159 [9.4%]; Control 7/162 [4.3%], p =.07) may suggest under-transfusion in the highest-risk Test subjects. Discussion: Test patients required less transfused Hb than control subjects to maintain equivalent daily hemoglobin levels following similar blood loss.

Background – Sepsis survivors commonly suffer from chronic Post-Sepsis Syndrome (PSS). To understand the mechanisms of PSS, it is critical to utilize appropriate animal models. While cecal ligation and puncture (CLP) is a widely used model of sepsis, a concern exists regarding its suitability for studying long-term outcomes. This thesis investigates symbolic logistic regression for interval-valued predictors through simulation studies and a real health data application. Classical logistic regression assumes exact predictor values, whereas in many practical settings variables are available only in interval form due to coarsening or reporting uncertainty. Two simulation studies examine the impact of interval uncertainty under asymmetric intervals and measurement error. Symbolic models based on midpoint and midpoint-plus-width representations are compared with the classical approach. Results show that midpoint modeling captures the general relationship but introduces bias under asymmetry, while incorporating width reduces this distortion. Previous research has consistently shown that experiencing maltreatment has significant, negative effects on youth’s psychological health; however, research on the psychological impact of maltreatment is often limited by challenges in accurately capturing children’s maltreatment experiences, as most children who are exposed to maltreatment experience multiple types of maltreatment. The present study utilized latent class analyses (LCAs), a person-centered, data-driven approach to empirically identify patterns in children’s maltreatment experiences at ages 3, 5, and 9. Paper spray mass spectrometry (PS-MS) is a novel ambient ionization method that allows chemical and biological samples to be rapidly analyzed with little preparation being required. In this paper, PS-MS was set up and streamlined as an acetylcholinesterase (AChE)-monitoring platform in addition to screening an existing and newly produced AChE-inhibitors. The detection of the substrate, acetylcholine (m/z 146), and the AChE catalysis product, choline (m/z 104) was performed without applying chromatographic separation or derivatization to the dried paper substrates. Assay conditions were optimized in a systematic way, to develop a robust MS-compatible enzymatic workflow.

This dissertation argues that Milton’s creative use of the three disparate concepts of consuming and digesting knowledge, chaos, and the night sky allows him to respond to Lucretian atomism, to address questions regarding the mortality of the soul, and to navigate the scientific discoveries of the seventeenth century, especially the astronomical discoveries of Galileo Galilei. Skeletal muscle is a highly plastic tissue with a robust capacity for regeneration, largely driven by resident satellite cells. Muscular dystrophies comprise a heterogeneous group of inherited disorders characterized by progressive muscle degeneration, chronic inflammation, and impaired regenerative capacity. Despite well-defined genetic etiologies, effective disease-modifying therapies for these disorders, as well as many acquired myopathies, remain limited. Emerging evidence identifies nuclear receptors (NRs) as key regulators of skeletal muscle homeostasis, integrating hormonal, metabolic, and environmental signals to control transcriptional programs governing mitochondrial function, metabolism, inflammation, and myogenesis. This course portfolio describes the design and development of a new course focused on data analysis and visualization in animal research at the University of Nebraska – Lincoln. The course was developed to meet the needs of students enrolled in Master of Science and Doctor of Philosophy graduate programs in the Department of Animal Science as well as those in the Master of Applied Science in Beef Cattle Production program.
