The advent of transcriptome-wide association studies (TWAS) has expanded the classical genome-wide association study (GWAS) framework by integrating gene expression with genetic variation to identify trait-associated variants. While multi-tissue TWAS approaches improve statistical power over single-tissue models, existing methods often lose information during result aggregation and require intensive computation. Here, we present TWAS-CTL (Cross-Tissue Learner), a novel framework that leverages heterogeneous gene expression across tissues by adaptively reweighting and optimizing multiple single-tissue learners. Simulations demonstrate that TWAS-CTL achieves higher statistical power than the leading method, UTMOST, while maintaining proper type I error control and reducing computational time by over half. …

This dissertation explores how spatial presence in Virtual Reality (VR) creates awe and appreciation through the role of perceived vastness. 92 participants played a custom-built VR game (VR Nature Photographer) in three environments designed to be low, medium, and high in vastness. A repeated-measures design measured spatial presence, vastness, awe, and appreciation across conditions. Results showed higher vastness increased all four outcomes, with awe and vastness having the largest effect sizes and presence and appreciation medium sized. Mediation analyses showed perceived vastness fully mediated the effect of spatial presence on awe in all vastness conditions and partially mediated its effect …

This dissertation examines how German industrial trade unions and workers in the automobile and coal industries navigate the profound challenges of the green transition. Workers’ experiences are situated within a broader political-economic context shaped partly by market-led decarbonization efforts, welfare retrenchment/austerity, erosion of progressive institutions in society, and global competition, as well as a global restructuring of value accumulation. The analysis critically engages with unions’ current responses to a market-led transition and their implications for labor’s future. Colloidal semiconductor nanocrystals feature unique properties like quantum confinement, tunable band gaps, and high surface-to-volume ratio; and have been proven as prominent materials in applications such as light-emitting diodes, sensors, lasers, solar cells, etc. Optoelectronic properties of semiconductor nanocrystals are highly associated with their sizes, morphologies, and compositions. Understanding the growth mechanism can lead to the ability to control the size, morphology, and composition of nanocrystals. In this dissertation, we are interested in a study of the minimizers to the $L^p$-mean distortion among Sobolev homeomorphisms $f:\mathbb T_*\to \mathbb T$ of finite distortion between embedded tori $\mathbb T = \mathbb T(R,r), \mathbb T_* = \mathbb T(R_*,r_*)$. For such mappings, the $L^p$-mean distortion for $1\le p < \infty$ is defined as \[ \int_{\mathbb T_*} K_f^p. \] We will show that the minimizer within a specific homotopy class to the $L^p$-mean distortion between embedded tori always exists. We will use free Lagrangians, whose integral only depends on the homotopy type of the function.

Deep learning for 3D point cloud analysis has made significant progress, yet several critical challenges, including the following, remain underexplored: (1) traditional max-pooling operations discard a substantial portion of learned features, resulting in information loss and inefficient use of computational resources; (2) existing few-shot point cloud classification models lack robustness when faced with occlusion, missing points, and limited training data; (3) semantic segmentation methods often fail to fully exploit background–foreground interactions, leading to reduced accuracy. However, their increased connectivity and sophistication introduce new vulnerabilities, making security a paramount concern. This dissertation presents a comprehensive pipeline for enhancing CPS security, focusing on accurate diagnosis of trustworthy time frames and subsystems, followed by system resilience measures to restore safe states. The foundation of system resilience lies in an advanced checkpointing protocol for real-time multi-process systems. This thesis explores sparse network inference from high-dimensional, noisy, and underdetermined data—a fundamental challenge in many scientific domains. We focus on the development of evolutionary computation methods for discovering underlying network structures that are both interpretable and biologically plausible. Our methods are applied to the domain of Gene Regulatory Network (GRN) inference, where sparsity, indirect interactions, and limited observations pose significant hurdles. The framework models both steady-state and time-series gene expression data, with particular emphasis on biological sparsity and regulatory dynamics. The idea that the activity of an active bath can act as an effective temperature and alter system behavior has been theorized and demonstrated for micron-scale systems, but it has not yet been shown for nanoscale systems. Previous studies have suggested that, although passive and active systems can be combined at the nanoscale, activity from the active system typically disrupts and destroys the passive system. In this work, I attempt to create an enzyme-driven active bath to control the liquid–liquid phase separation (LLPS) and condensation of a protein. With five core-collapse supernova candidates within 1 kiloparsec from Earth, extracting information from the neutrino flux will be important to understanding the explosion mechanism of the star (and its composition). The Deep Underground Neutrino Experiment, (DUNE), will be capable of detecting neutrinos from supernovae, primarily from charged-current interactions due to the electron neutrino flavor. However, considering neutrino oscillations in the presence of matter (MSW effects), the muon neutrinos and tau neutrinos will be undetectable if not for the neutral current channel. This dissertation contains two chapters related to housing choices of low-income families, neighborhood quality, and the remediation of urban blight. The first chapter examines the effectiveness of Detroit's 2014–2022 residential demolition program in addressing neighborhood blight and vacancy in a shrinking city context. Using a stylized model of durable housing with downward price frictions, I combine parcel-level data on vacancies, blight, and demolitions to estimate the causal effects of demolition exposure on vacancy rates for non-demolished, single-family homes. Employing a difference-in-differences design, I find that the program as a whole did not reduce vacancies, and instead slightly increased them by …

This platform offers high gate speeds and can make use of readily scalable complementary metal-oxide semiconductor (CMOS) fabrication techniques. However, superconducting qubits are susceptible to a variety of losses that arise in systems with superconductors, such as due to ubiquitous two-level systems, quasiparticles, and more. However, designing solutions that balance robustness while meeting the practical constraints of diverse applications, such as limited resources and data, remains a challenge. Post-translational modifications (PTMs) are essential for the diversification of the human proteome. Prenylation is a PTM defined by the addition of a 15-carbon or 20 carbon isoprenoid group by FTase and GGTase-I, respectively, onto a cysteine near the C-terminus of proteins. This modification is the first step within the prenylation pathway where proteolysis and carboxymethylation occur afterwards, aiding in proteins localization to the cellular membrane. These prenyltransferases canonically recognize a “CaaX” box motif which has been determined through biochemical and structural studies. This dissertation investigates the problem of change-point detection in segmented logistic regression models, where the total number of change-points is unknown. We consider both unconstrained models, which allow discontinuities at change-points, and constrained models, which impose continuity across change-points. The central challenge is to determine the number of change-points and to accurately estimate their locations, while ensuring valid statistical inference for the associated regression coefficients. We begin by introducing a set of regularity assumptions that underlie the theoretical analysis throughout this work. Abstract In this dissertation, I present three studies that examine how different forms of inconsistency—arising from timing disruptions, eligibility structures, and administrative churn—shape the functioning and effectiveness of the Supplemental Nutrition Assistance Program (SNAP). Although SNAP is designed to provide a reliable source of food assistance, access to benefits is not always predictable, continuous, or evenly accessible. Optimizing beamforming and network slicing is critical for enhancing spectral efficiency, energy efficiency, and resource distribution fairness in dense urban 5G networks. This paper proposes a hybrid genetic algorithm particle swarm optimization (GA-PSO) method to jointly optimize beamforming weights, bandwidth allocation, and power distribution, balancing computational efficiency with near optimal performance. The hybrid approach uses GA for global exploration and PSO for fast convergence, overcoming the limitations of standalone heuristic and exact optimization methods. In response to the ongoing challenges posed by the scarcity of reliable fresh-water sources in Lukobe Ward, Morogoro Municipality, Tanzania, this study applied an integrated approach that combines geospatial and geophysical datasets to identify previously undocumented aquifers. The datasets utilized include an apparent density map generated from high-resolution GGMPlus satellite gravity data, a geological map (Quarter Degree Sheet 183), a water drainage map, and a slope map created from a digital elevation model (DEM). Forest degradation is one of the leading sources of carbon emission globally. In Africa, particularly in Tanzania, forest degradation is driven by selective logging and forest fires. Monitoring degradation over large areas is very challenging due to the lack of accurate and reliable methods. The study investigated the in vitro oxidative-stress-mitigating effect and anti- inflammatory potential of the soluble protein of the arils of Blighia unijugata in order to establish possible use in inflammatory therapy. Air-dried Blighia unijugata aril was pulverized into powder, defatted using petroleum ether, and homogenized in Phosphate Buffer Saline. The homogenate was centrifuged, and the supernatant was subjected to ammonium sulphate fractionation. The precipitate was thoroughly dialyzed, dialysate was freeze-dried, and termed partially purified Blighia unijugata aril protein extract (ppBUAPE). The increased sales of locally packaged roasted cashew kernels (RCK) in Tanzania reflect growing consumer demand for healthy, convenient snacks. Despite this popularity and frequently reported cases of gastrointestinal illness, the microbial safety of these plastic- bagged products is not well-documented, posing a potential public health concern. To address this, a study was performed to evaluate packaging hygiene, package integrity, and microbial contamination of randomly selected packages of RCK (small: n=99; medium: n=87; large: n=118) from vendors in Coast and Morogoro regions, Tanzania. Genetic diversity of Manihot esculenta associated virus type 1 (MEaV-1) species suspected to cause the most destructive Brown Streak of Cassava (CBSD) in Tanzania, has remained elusive since the year 2020 when MY-6mb-4 virus isolate was first identified in Chambezi, Coast region. Seagrasses are highly productive coastal ecosystems that harbor diverse bacterial communities, including both culturable and unculturable taxa. These bacteria play vital roles in ecological, biogeochemical, and biotechnological processes. In this study, a metagenomic approach using Pacific Biosciences next-generation sequencing was employed to investigate bacterial diversity associated with Thalassia hemprichii and Syringodium isoetifolium collected from the Mjimwema coast, Dar es Salaam, Tanzania. This study investigates the dynamic interactions of a prey-predator ecosystem infected with Canine Distemper Virus (CDV), focusing on predation and disease transmission. A deterministic model, coupled with Monte Carlo simulations (MCS), employed to capture both stochastic variability and environmental fluctuations. The findings indicate that classical prey-predator cycles provide the baseline for population dynamics, but the introduction of CDV and environmental noise fundamentally alters system behavior. Initially, populations exhibit oscillations that gradually stabilize into dynamic equilibria. Fluctuations in wholesale gasoline prices significantly impact the economy of import-dependent countries like Tanzania, making accurate forecasting essential for policymakers and market participants. This study forecasts monthly wholesale gasoline prices in Dar es Salaam by comparing the Autoregressive Integrated Moving Average (ARIMA) model with a Neural Network Autoregressive (NNAR) model. Using data from January 2015 to December 2024 from the Energy and Water Utilities Regulatory Authority (EWURA), the models were evaluated. This study thoroughly investigates the physicochemical properties, microbiological profiles, and nutrient dynamics of the Temi River in Tanzania. The sources and implications of fluctuations in dissolved organic carbon (DOC) and nutrients during the wet and dry seasons are carefully analysed. Water samples were collected from various geographically referenced points along the river, and analyses were performed using the APHA Standard Methods for Water and Wastewater Testing.
