ea level rise is having major impacts on estuaries due to salinity intrusion. These changes in stress profiles have ripple effects in ecosystems, including altering the invasibility of these wetlands depending on the salt tolerance of the invading species. Alternanthera philoxeroides Mart. (Griseb.) (alligator weed), native to South America and long recognized as one of the world’s worst freshwater aquatic weeds, recently invaded tidal wetlands in California’s San Francisco Bay–Delta Estuary. Generally considered a freshwater-limited glycophyte, observations suggested this invasive macrophyte may have some degree of salinity tolerance, though its degree of tolerance and capacity to spread with increased salinity …

Investigating Chronic Calcium Leak In The Aging Brain, Elizabeth K. Woo Jan 2025

Investigating Chronic Calcium Leak In The Aging Brain, Elizabeth K. Woo

Yale Graduate School of Arts and Sciences Dissertations

Alzheimer’s disease (AD) is a devastating, progressive neurodegenerative disease, characterized by amyloid plaques and tau tangles, with synapse loss, calcium dysregulation, and accumulation of advanced glycation end products (AGEs). These pathologies develop in a region-specific manner, targeting regions responsible for higher-order integrative processing, such as the prefrontal cortex (PFC) and entorhinal cortex. Calcium signaling, through cAMP-PKA-calcium signaling, is critical to support the persistent firing needed for working memory. However, under various conditions, e.g., aging, inflammation, chronic stress, these circuits lose regulation of cAMP-PKA-calcium signaling, leading to calcium leak from the smooth endoplasmic reticulum (SER) and chronically elevated levels of cytosolic …

The Assessment Of Morphological, Anatomical, And Phylogenetic Relationships Of Three Helicid Snails In Egypt, Sheriene E. Ali, Amina M. Ibrahim, Eman T. Hussien, Irene S. Gamil, Reda M. Mansour, Mostafa Y. Morad

Trends in advanced sciences and technology

Land snails provide valuable models for eco-genetic research because of their morphology and banding variability. Snails have shown unusually high intraspecific variation in anatomical traits leading to confusion for taxonomical studies, although their taxonomic resolution is low specifically in Egypt. This study aims to investigate the morphological, anatomical, and phylogenetic relationships for the first time among the snails Eobania vermiculata, Eremina desertorum, and Helix aspersa within the family Helicidae. The morphological indices revealed similarities among E. vermiculata, E. desertorum, and H. aspersa, However, SEM examinations indicated that the marginal teeth of the radulae in E. vermiculata and H. aspersa were …

Molecular Docking And Adme Calculations Of Structurally Similar Compounds To Anastrozole, Habiba Abd-Rabbo, Somaia Abdel-Rahim, Maha Bakr, Manar Mohammed, Ola Nosseir

Trends in advanced sciences and technology

Aromatase inhibitors prevent the conversion of androgens into estrogens, leading to decreased levels of circulating estrogens. Thus, they are significantly used for the treatment of breast cancer, particularly, in postmenopausal women. Anastrozole, an approved aromatase inhibitor, has gained interest due to its remarkable activity and pharmacokinetics. In this work, small molecules of structural similarity to anastrozole were identified, their ADME-related properties were calculated, and their binding modes to the human aromatase enzyme were evaluated. The calculated properties of the compounds with the highest similarity scores were comparable to those of anastrozole. Six compounds were also predicted to exhibit higher binding …

Effects Of Single-Cell Dietary Protein Sources On Feeding, Growth, And Resource Allocation In The Juvenile Sea Urchin Lytechinus Variegatus, Darrien Jamila De Jesus Jan 2025

Effects Of Single-Cell Dietary Protein Sources On Feeding, Growth, And Resource Allocation In The Juvenile Sea Urchin Lytechinus Variegatus, Darrien Jamila De Jesus

All ETDs from UAB

An understanding of nutrition can improve culture of sea urchins, whether as a research model or in the production of seafood products. Macronutrients, including dietary protein sources, can affect growth and reproductive profiles and alter resource allocation to somatic and gametic tissues. Although sea urchins in culture can be fed natural products, diet formulations using various ingredients can be developed to reliably produce desired outcomes. Single-cell protein sources represent a potential alternative to current industry standards of animal-derived protein sources such as fish and squid meal, which are becoming increasingly overfished and expensive. In the present study, we evaluated the …

Optimizing Recruitment Strategies, Outcome Measures, And Intervention In Clinical Research With Adults With Multiple Sclerosis: Insights From The Tele-Exercise And Multiple Sclerosis (Teams) Study, Tracy Flemming Tracy

All ETDs from UAB

Background: Recruiting people with multiple sclerosis (PwMS) into research can be a significant challenge. This is often due to limited awareness of the research objectives, lack of accessibility, and restrictive inclusion criteria. In addition to a general reluctance to participate, assessing multiple sclerosis (MS) and assigning appropriate interventions can be challenging due to the heterogeneity of symptoms. Identifying outcome measures that accurately reflect a participant’s perception and functional performance, while remaining sensitive to change across different levels of disability, poses a challenge. Methods: Data from the Tele-Exercise and Multiple Sclerosis (TEAMS) trial was analyzed to identify effective recruitment strategies (Study …

Effect Of Pre-Treatment Lab-Based Comorbidities On Treatment Related Outcomes Among Patients With Cancer, Jeffrey Alan Franks Jan 2025

Effect Of Pre-Treatment Lab-Based Comorbidities On Treatment Related Outcomes Among Patients With Cancer, Jeffrey Alan Franks

All ETDs from UAB

Cancer is one of the leading causes of mortality and morbidity in the United States. In addition to their cancer, nearly 50% of individuals have a comorbid condition. This adds complexity to the treatment of these patients as it may delay cancer diagnosis or treatment or prevent treatment all together. Lab-values are an important tool to measure comorbidities and end-organ damage as they are routinely captured as part of standard of care and the basis of clinician decision making. However, despite the widespread multimorbidity and how crucial they are to decision making, few studies assess the association of lab abnormalities …

Elucidating Non-Canonical Interactions Of Mcl1 With The P53 Family Of Tumor Suppressors, Alexus Marie Acton Jan 2025

Elucidating Non-Canonical Interactions Of Mcl1 With The P53 Family Of Tumor Suppressors, Alexus Marie Acton

All ETDs from UAB

Various signaling mechanisms contribute to a cancers cell’s ability to survive through the stressors of tumorigenesis. Canonically, when a cell is exposed to irreversible stress, the induction to apoptosis is regulated by the Bcl-2 family of apoptotic regulators. However, to accommodate for the stressors of tumor progression, many solid tumors over express one of the pro-survival proteins (e.g., BCL2, BCLxL, MCL1) allowing for persistent growth without the induction to cell death. Over the past 10 years, researchers have investigated how over expression of these pro-survival proteins contribute to chemoresistance to standard of care therapies. To sensitize cells to these apoptotic …

Targeting The Thyroid-Stimulating Hormone Receptor (Tshr) In Thyroid Cancer, Grayson Robert Gimblet Jan 2025

Targeting The Thyroid-Stimulating Hormone Receptor (Tshr) In Thyroid Cancer, Grayson Robert Gimblet

All ETDs from UAB

Radioactive iodine (RAI) has been effective in the treatment of thyroid cancer, the most common endocrine neoplasia, since its introduction into clinical use over eighty years ago. However, not all patients benefit from RAI. Up to 20% of patients with well-differentiated thyroid cancer (WDTC) become radioactive iodine refractory (RAI-R). These RAI-R patients have 5-year survival rates of 50% and 10-year survival rates of 10%. In addition, more aggressive subtypes including oncocytic thyroid carcinoma (OC), poorly differentiated thyroid carcinoma (PDTC), and anaplastic thyroid carcinoma (ATC) are less responsive to RAI therapy. Despite accounting for less than 5% of annual thyroid cancer …

Electrophysiological Integration Of Engineered Tissue Graft With The Host Swine Myocardium, Bijay Guragain Jan 2025

Electrophysiological Integration Of Engineered Tissue Graft With The Host Swine Myocardium, Bijay Guragain

All ETDs from UAB

The adult mammalian heart lacks regenerative capacity to replenish cardiomyocytes (CMs) lost due to myocardial infarction (MI). Instead, the damaged heart undergoes extensive remodeling that significantly impairs normal cardiac function and may eventually lead to heart failure. Recent therapies that implant engineered tissues fabricated from human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs) have shown promising results with improved cardiac function and structural integration. However, the extent of electrophysiological coupling between engineered and host tissue remains unclear. This is a critical factor for functional integration of the implant as well as for arrhythmia risk arising from the implant. Our study aims …

Development Of A Resistive Force Paradigm For The Treatment Of Hemiparesis, Jonathan Daniel Hill Jan 2025

Development Of A Resistive Force Paradigm For The Treatment Of Hemiparesis, Jonathan Daniel Hill

All ETDs from UAB

Hemiparesis is weakening of one side of the body as a result of a stroke that contributes to a decrease in propelling forces (i.e. propulsion) while walking. Existing research to improve propulsion of individuals post-stroke has implemented single-belt and split-belt treadmills, various walking speeds, asynchronous belt speeds (on split-belt treadmills), and a variety of methods for inducing resistive force upon a paretic limb in an attempt to strengthen the paretic limb and improve overall gait symmetry. However, despite this concerted effort of clinicians and scientists, gait deficits remain. Novel methods are required to improve reduced propulsion of individuals post-stroke. Recently, …

The Role Of Aquaporin-3 Lysine 282 Acetylation In Renal Water Balance, Nha Van Huynh Jan 2025

The Role Of Aquaporin-3 Lysine 282 Acetylation In Renal Water Balance, Nha Van Huynh

All ETDs from UAB

Aquaporin-3 (AQP3) mediates basolateral water transport in the kidney collecting duct principal cells, contributing to vasopressin-mediated urine concentration. We previously identified a novel post-translational modification of AQP3, the acetylation of lysine 282 (K282), which we hypothesized as a positive regulator of AQP3 water permeability. First, we found that collecting duct K282 acetylated AQP3 increased upon water deprivation, suggesting an involvement in renal water balance. To decipher the functional role of this modification, we utilized AQP3 acetylation (K282Q or Q) and deacetylation (K282R or R) mimetic mutant mice models generated using CRISPR/Cas9 knock-in technology. Both male and female wild-type (WT) and …

The Impact Of Transportation Insecurity On No-Show Appointments And Acute Care Utilization In Pediatric Sickle Cell Patients, Seabrook Hannah Jeffcoat Jan 2025

The Impact Of Transportation Insecurity On No-Show Appointments And Acute Care Utilization In Pediatric Sickle Cell Patients, Seabrook Hannah Jeffcoat

All ETDs from UAB

Sickle cell disease (SCD) patients experience high levels of transportation insecurity. Missed or delayed medical care due to transportation barriers can result in worsened clinical outcomes and increased hospital admissions and emergency department (ED) visits. We hypothesized that the relationship between transportation insecurity and acute care utilization is partially mediated by no-show appointments. This retrospective cohort study included 101 SCD patients aged ≤ 21 treated through the Pediatric Sickle Cell Clinic at Children’s of Alabama (COA) hospital from 2023 to 2024 and completed a social determinants of health (SDoH) questionnaire. Negative binomial or Poisson regression was used to model ED …

Mechanoactivation Of Vegfr-2 Promotes Angiogenesis In The Tumor Microenvironment, Bronte Johnson Jan 2025

Mechanoactivation Of Vegfr-2 Promotes Angiogenesis In The Tumor Microenvironment, Bronte Johnson

All ETDs from UAB

Cancer progression relies on blood vessel growth; without vasculature, tumors would be limited to less than 200µm in diameter. With this reasoning, several cancer therapies have been developed that target the process of angiogenesis, or the growth of vessels off pre-existing vasculature. These treatments target the major promoters of angiogenesis, vascular endothelial growth factor (VEGF) and VEGF receptor 2 (VEGFR-2); however, anti-angiogenic therapies are not effective by themselves. This is likely due to mechanoactivation of VEGFR-2 through physical alterations in the tumor microenvironment (TME). One prevalent mechanical change is increased strain on surrounding cells and matrix due to highly contractile …
