This study focuses on the effect of Environmentally Persistent Free Radicals (EPFRs) on the P450 enzymes of the CYP1 family. EPFRs are a component of particulate pollutants, that are stable in the environment, but can generate free radicals, leading to oxidative stress and subsequent toxicity of the respiratory, cardiovascular, and immune systems once they enter an organism. The results show differences in the ability of EPFRs to inhibit CYP1-dependent substrate metabolism, with CYP1B1 being inhibited to the greatest extent. There also were differences in the ability of EPFRs to disrupt the POR•CYP1 complex, with CYP1B1 being the only form where …

Cannabis and cannabinoid mixtures have been linked to a variety of health benefits including pain mitigation, suppression of nausea produced by chemotherapeutic agents, anti-inflammatory effects, and effects on energy homeostasis, glucose, and lipid metabolism. The latter properties have led to the suggestion that these products could have therapeutic effects on the development of metabolic dysfunction-associated steatohepatitis (MASH) - a severe type of liver pathology in obese and diabetic patients. However, varying agonist and antagonistic properties of different cannabinoids on the endogenous cannabinoid system make prediction regarding hepatic effects and diet interactions difficult. The current study was designed to examine hepatic …

Journal of Dental Sciences

Abstract Background/purpose Individuals with diabetes mellitus (DM) are more susceptible to periodontitis, largely due to the accumulation of advanced glycation end-products (AGEs), which drive oxidative stress and inflammaging. Inflammaging is a state of chronic low-grade inflammation and accelerated cellular aging that contributes to periodontal degradation, mediated by AGEs-induced cellular senescence and senescence-associated secretory phenotype (SASP). 2-O-methylmagnolol (2-MG), a bioactive compound with antioxidant and anti-inflammatory properties, remains underexplored in DM-associated periodontal degeneration. This study investigated the effects of 2-MG on AGE-induced oxidative stress and inflammaging in human gingival epithelial cells (HGEs) and human gingival fibroblasts (HGFs). Materials and methods The study …

Fatostatin Delayed Lip Sensory Recovery After Inferior Alveolar Nerve Transection By Inhibiting Sterol Regulatory Element-Binding Protein 1, Li Ye, Suning Mao, Zhongkai Ma, Gaowei Zhang, Pingchuan Ma, Chunjie Li

Journal of Dental Sciences

Abstract Lipid metabolism is essential for nerve repair in damaged nerves. Fatostatin, a selective inhibitor of sterol regulatory element-binding protein 1 (SREBP1), could reduce cholesterol synthesis and disturb lipid homeostasis. However, whether fatostatin would delay lip sensory recovery after inferior alveolar nerve transection remains unclear. In this preliminary study, we investigated the effects of fatostatin on lip sensory recovery in vivo and axon growth in vitro . Fatostatin significantly delayed lip sensory recovery of mice after inferior alveolar nerve transection as evidenced by quantitative sensory testing. Fatostatin also reduced the average axon length of primary trigeminal neurons. Despite SREBP1, expressions …
