Post-Fire Riverscapes: Exploring The Ecological And Behavioral Responses Of Stream Communities To Wildfire In Boreal Streams, Elizabeth Geraldine Hinkle

Wildfire is the dominant disturbance in boreal forest ecosystems, and its frequency and severity are increasing with climate change. This dissertation examines how wildfire affects boreal stream habitats, food webs, and fish movement across spatial and temporal scales in interior Alaska. In Chapter 2, I compared burned and unburned streams 10 to 15 years post-fire using habitat survey, water chemistry, and aquatic community composition data. Burned sites had marginally significant lower canopy cover (-19.6%, p = 0.095), coarser substrate (+23.7 mm, p = 0.102), and higher macroinvertebrate abundance (626 ± 361 vs. 331 ± 349 individuals/m2, p = 0.03); fish …

Living with a rare disorder, such as Ehlers–Danlos Syndrome (EDS), presents unique psychosocial challenges. This autobiographical case study explores the psychological, social, and professional impacts of navigating life with hypermobile EDS (hEDS), a condition marked by chronic pain, joint instability, and diagnostic uncertainty. Placing personal experiences into the context of clinical findings and research, this study highlights both the physical and emotional toll of the syndrome, including stigma, isolation, and medical gaslighting often accompanying rare conditions. Attribution studies have shown that warming Arctic temperatures have led to a substantial loss of sea ice and an increase in large wildfire seasons. With wildfire and sea ice decline impacting everyday life by disrupting transportation or damaging infrastructure, for example, a clear need for skillful forecasts at seasonal timescales has emerged. This dissertation addresses critical challenges in seasonal prediction for wildfire and sea ice in Alaska. The first study evaluates outlooks of Buildup Index derived from three seasonal forecasts to assess their skill in predicting fire conditions. This longitudinal study explored entry route- and age-related fitness differences between cadet (CR) and general population (GR) firefighter recruits. Fitness data were collected from eight separate firefighter recruit academy cohorts (N = 317; 122 CR, 195 GR; 27.17 ± 7.58 yrs, 177.30 ± 8.72 cm, 88.65 ± 17.78 kg). Soil salinity stress, intensified by extreme weather patterns, significantly threatens global watermelon [Citrullus lanatus (Thunb.) Matsum & Nakai] production. Watermelon, a moderately salt-sensitive crop, exhibits reduced germination, stunted growth, and impaired fruit yield and quality under saline conditions. As freshwater resources decline and agriculture’s dependency on irrigation leads to soil salinization, we need sustainable mitigation strategies for food security. Recent advances highlight the potential of using salt-tolerant rootstocks and breeding salt-resistant watermelon varieties as long-term genetic solutions for salinity. Movement of leukocytes from the circulatory system into a site of inflammation is a highly complex process. The migration of neutrophils into the lumen of the mammary gland during mastitis is no exception. There is information about the involvement of bacterial-produced products during this process, but less is known regarding the role of host products. Two bovine cell lines, a primary mammary gland endothelial cell line and an immortalized bovine mammary gland epithelial cell line (Mac-T), along with freshly isolated bovine neutrophils, were used to study this further. Management paradigms for newly-diagnosed acute myeloid leukemia (ND-AML) in patients considered unfit to receive intensive chemotherapy have evolved with improved understanding of disease biology. In this setting, management requires clear delineation of goals of therapy that should include preservation of quality-of-life (QoL). Rare variants affecting the epigenetic regulator KDM2B cause a recently delineated neurodevelopmental disorder. Interestingly, we previously identified both a general KDM2B-associated episignature and a subsignature specific to variants in the DNA-binding CxxC domain. In light of the existence of a distinct subsignature, we set out to determine if KDM2B CxxC variants are associated with a unique phenotype and disease mechanism. We recruited individuals with heterozygous CxxC variants and assessed the variants' effect on protein expression and DNA-binding ability. This study investigates the ways in which the artists and artworks depicted throughout their K-12 art education influence students’ personal sociocultural identity development. By utilizing a self-reporting survey and individual interviews, this study explores the perspectives of current university students; their exposure to diversity in their K-12 art education; and the curricular perspectives that influence their identity. The artists and the artwork that art educators show in the classroom are only one aspect of what makes up a student’s art curriculum; this research project demonstrates the impact those artists can have on students’ identity development. Broken tools indicate a broken system. This new study reveals how medical equipment failures destroy Ghana's public hospitals, turning life-saving technologies into silent murderers that disproportionately affect rural areas. Moving beyond technical diagnostics, we study the cascading systemic failures, chronic underfunding, critical staff shortages, and fragmented supply chains that leave 40% of vital equipment, such as ventilators, MRI scanners, and dialysis machines, inoperable at any given time. We quantify the devastating human toll through rigorous mixed-methods analysis of equipment downtime records across ten regional hospitals (2018-2023) and profound narratives from 50 frontline stakeholders, doctors forced to improvise without working incubators, …

This study describes the development of a three-dimensional (3D) oral mucosal model (OMM) to investigate how oral tissues respond to masticatory forces. The OMMs replicated key features of human oral mucosa, such as stratified keratinocyte telomerase-immortalized gingival keratinocytes (TIGK) layers and fibroblast-populated collagen matrices. Cyclical mechanical forces (0-10 N) for 2 h applied to the model caused force-dependent changes in the histological structure, including thinning of the epithelium and collagen matrix and cell displacement at higher forces. Lactate dehydrogenase (LDH) cytotoxicity assays revealed that 10 N forces led to significant cell damage (about 50% cell death) in TIGK monolayers, whereas …

Harmful algal blooms (HABs) impact livestock producers. Most Extension agents have limited knowledge about HAB management. Our study goal was to assess agent readiness and training on HABs. We used data from in-person surveys of South Carolina (SC) Extension personnel. We examined how training influenced HAB knowledge using pre/post-tests. Training improved agent knowledge of HAB management, but water resource agents learned more. The results of this study can be used to identify Extension training shortfalls and drive policy to better prepare agents throughout the state. Further research should involve prioritizing additional extension topics and assessing knowledge in a similar manner.

Implementation of soil health management systems, such as reduced tillage and cover cropping, can reduce soil erosion, increase water infiltration, and improve soil productivity. Adoption of these practices can also improve net farm income, but effects vary based on region, commodity, and production methods. A partial budget analysis approach was used to evaluate impacts of adoption of soil health management systems on profitability from a regional, on-farm, perspective. The approach provides farmers with a resource to support decisions when considering the adoption of soil health management systems.

Creating partnerships with private landowners will require agencies to identify and remove barriers to solidarity through targeted approaches to social interactions. This study provides a heuristic to segment landowners according to the decision-making the level of shared responsibility they associate with conservation outcomes on private lands. I segmented landowners at Texas Parks and Wildlife Department workshops and conducted interviews with participants to better understand the outcomes of the segmentation event. The results of the case study revealed that the segmentation heuristic effectively distinguishes between landowner types and that value alignment with landowners can supplement perceptions of interdependence.

This study aimed to establish whether a laboratory (lab) based 3-minute all-out (3MT) protocol and a field-based 3MT protocol would yield similar peak power (Pmax), critical power (CP), and curvature constant (W’) profiles and the implications of parameter estimation for informing a 40-km time trial (TT) performance. The study aimed to determine the Post-Traumatic Stress Disorder Symptom Scale and the Coping Mechanisms utilized by the Internally Displaced Persons of Typhoon Yolanda. It sought to identify the profile, risk factors of the respondents, the Post-Traumatic Stress Disorder (PTSD) symptoms manifested and coping mechanisms utilized. The study also distinguished the differences in PTSD and Coping mechanisms when respondents are grouped according to their profile and death of family members and examined the relationship of PTSD and Coping.
