BACKGROUND AND PURPOSE: Diffusion tensor imaging analysis along the perivascular space (DTI-ALPS) has emerged as a promising noninvasive method for evaluating water motion that may reflect glymphatic system function. However, the reliability of DTI-ALPS measurements across different region-of-interest (ROI) selection methods remains underinvestigated. This study aimed to assess the interrater reliability among three neuroradiologists in native space and compare DTI-ALPS indices derived from ROIs placed in subjects' native space versus standardized Montreal Neurological Institute (MNI) space.

Since the Reagan era, American economic policy has amounted to self-colonization. Democratic majorities have consistently supported legal regimes that have enabled corporations to extract the lion’s share of the gains from trade from the public. For example, they have supported a corporate law regime that denies the public democratic control over the behavior of corporations and instead gives dictatorial powers to shareholders and managers. The Internet has made it even easier for firms to extract surpluses from consumers through surveillance and algorithmic pricing. The SARS-CoV-2 (COVID-19) pandemic revealed both the systemic vulnerabilities of our healthcare system as well as the importance of timely, localized data for informing public health responses. Traditional testing approaches were essential but often failed to capture the full extent of viral spread, especially in those with mild or asymptomatic conditions that relied on at home testing. This underreporting hindered effective allocation of resources and made developing effective intervention strategies difficult. We also realized the potential for wastewater surveillance to be applied to tracking antimicrobial resistance gene markers. Efficient use of native rangelands and improved reproductive success are primary objectives in beef cattle production. Grazing behavior during the breeding season may be influenced by pasture characteristics, including size and water availability, as well as physiological differences due to reproductive status. This study aimed to evaluate the relationship between grazing activity and conception method in crossbred Angus cows over a 2-year period. The study was conducted during the winter months (December – February) on the Central Coast of California with a mean temperate of 53 degrees Fahrenheit. Social connections within primate groups are continuously changing, and an individual's connectedness within their social network can have important consequences on morbidity and mortality. Here, we examined the effects of early life social experiences and age on social connectedness of captive female olive baboons (Papio anubis). Subjects included 54 mother-reared (MR) and 35 nursery-reared (NR) baboons (4.03 to 19.8 years of age). We conducted four 15-min focal observations and recorded all social interactions. For every possible dyad, we calculated total amount of time in proximity to one another and total amount of time spent grooming (in each direction), then used …

The prevalence of physical aggression among women has increased, challenging traditional gender norms that portray women as nurturing and nonviolent. While most research on aggression has focused on men, children, or intimate partner violence, research is sparse concerning female aggression outside familial contexts. In this qualitative multiple-case study, the contextual antecedents of nonfamilial aggression in women arrested for aggressive behavior were explored using the general aggression model as the theoretical framework. Lagos State, Nigeria, faces male teacher retention challenges in public high schools, including destabilizing the educational environment, reducing the availability of experienced mentors, and negatively impacting student performance and outcomes. School leaders need to understand the challenges leading to male teacher turnover. The purpose of this qualitative phenomenological study was to explore the lived experiences of Lagos State male public high school teachers who left the profession, focusing particularly on their perceptions of onboarding and reasons for leaving the profession. The olfactory system is able to filter odor representations based on attention and learning. Two PLoS Biology studies reveal how short axon cells in the olfactory bulb integrate cholinergic input from the basal forebrain to dynamically regulate olfactory input.

The mammalian kidney contains numerous nephrons connected to the collecting ducts, and each nephron consists of a glomerulus, a proximal tubule, the loop of Henle (LoH), and a distal tubule. Folliculin (FLCN) is a causative gene for Birt-Hogg-Dubé syndrome, which is characterized by a variety of manifestations, including renal cysts and cancer. Although deletion of Flcn in the mouse collecting duct and distal nephron leads to cyst formation, its precise role in the entire nephron remains unclear. Decreased brain levels of coenzyme Q10 (CoQ10), an endogenously synthesized lipophilic antioxidant1,2, underpin encephalopathy in primary CoQ10 deficiencies3,4 and are associated with common neurodegenerative diseases and the ageing process5,6. CoQ10 supplementation does not increase CoQ10 pools in the brain or in other tissues. The recent discovery of the mammalian CoQ10 headgroup synthesis pathway, in which 4-hydroxyphenylpyruvate dioxygenase-like protein (HPDL) makes 4-hydroxymandelate (4-HMA) to synthesize the CoQ10 headgroup precursor 4-hydroxybenzoate (4-HB)7, offers an opportunity to pharmacologically restore CoQ10 synthesis and mechanistically treat CoQ10 deficiencies. Objective: Exome sequencing (ES) benefits the genetic work-up for fetuses with structural anomalies, but data on its utility for fetuses without anomalies and stillbirths is more limited. We report our experience with prenatal ES for all three indications.
Method: We retrospectively reviewed results from 344 trio-ES performed for fetuses with structural anomalies (N = 262), stillbirths (N = 39), and fetuses without anomalies (N = 43), many of which had a relevant family history. Background: Molecular aberrations have been incorporated into tumour classification guidelines of meningioma. TERT-promoter (TERTp) mutation is associated with worse prognosis and is designated a WHO grade 3 biomarker. However, it remains unclear whether TERTp mutation is context-dependent, with other co-occurring genetic alterations potentially driving its association with prognosis. We sought to characterise the role of TERTp mutation in meningioma and guide TERTp sequencing.

The evolution of prenatal genetic testing has transformed prenatal diagnosis into a more precise and individualized approach. Advanced tools such as chromosomal microarray analysis and exome sequencing have enabled the prenatal diagnosis of more genetic conditions, including anomalies and disorders eligible for fetal therapy. When in utero therapy is considered, accurate genetic diagnosis is essential for guiding providers' and patients' decisions regarding management and outcomes. This chapter reviews available prenatal genetic screens and tests, their indications, and counseling strategies. It also explores genetic abnormalities associated with fetal structural anomalies and their implications for decision-making in fetal interventions.

Introduction: Biomarkers are essential for monitoring the progression of frontotemporal dementia (FTD). Although dysregulated brain lipid metabolism, particularly sphingolipids enriched in the nervous system, is a key feature of neurodegeneration, plasma lipids remain underexplored as biomarkers compared to imaging and serum proteins.
Methods: We examined plasma lipidomes using liquid chromatography-tandem mass spectrometry (LC-MS/MS) from individuals carrying pathogenic variants linked to autosomal dominant FTD (GRN, C9orf72, MAPT) and non-carriers.
Results: FTD subjects exhibited increased plasma levels of gangliosides (GM3(d18:1_16:0), GM3(d18:1_24:1)), ceramide Cer(d18:1_23:0), and select polyunsaturated triacylglycerols. Post-translational modifications (PTMs) are critical regulators of protein function, and their disruption is a key mechanism by which missense variants contribute to disease. Accurate PTM site prediction using deep learning can help identify PTM-altering variants, but progress has been limited by the lack of large, high-quality training datasets. Here, we introduce PTMAtlas, a curated compendium of 397,524 PTM sites generated through systematic reprocessing of 241 public mass-spectrometry datasets, and DeepMVP, a deep learning framework trained on PTMAtlas to predict PTM sites for phosphorylation, acetylation, methylation, sumoylation, ubiquitination and N-glycosylation. Background: Rare genetic variation provided by whole genome sequence datasets has been relatively less explored for its contributions to human traits. Meta-analysis of sequencing data offers advantages by integrating larger sample sizes from diverse cohorts, thereby increasing the likelihood of discovering novel insights into complex traits. Furthermore, emerging methods in genome-wide rare variant association testing further improve power and interpretability.

A comprehensive postmortem examination is an essential component of a work-up after stillbirth. Its findings can support accurate counseling of parents about causes and risk of recurrence. It also supports providers' decisions about most appropriate testing and management plans for future pregnancy to prevent recurrence. Informing parents about fetal autopsy and obtaining their consent is challenging, and conducting a fetal autopsy requires expertise that is, not universally available. Newer non-invasive or minimally invasive methods such as postmortem MRI and targeted biopsies can replace or supplement autopsies, but one must recognize that expertise in these methods is likewise not broadly available. …

Background
Infantile epileptic spasms syndrome (IESS) is a devastating developmental epileptic encephalopathy (DEE) and patients exhibit diffuse white matter alterations and structural remodeling. However, the correlation between these structural changes and brain network properties, or their effect on the efficacy of treatment outcomes in MRI non-lesional IESS patients is not clear.
Method
This retrospective study was conducted on IESS patients using fixel-based analysis (FBA) of diffusion MRI and graph theory analysis of structural connectivity, involving 26 non-lesional IESS patients aged 2 to 12 months and 120 age-matched controls. Bioretention systems are widely used in stormwater management to mitigate nutrient pollution, particularly nitrogen and phosphorus. Over time, these systems undergo changes in nutrient retention efficiency due to organic matter accumulation and biogeochemical transformations. This study examines the distribution and long-term retention of total nitrogen (TN), total Kjeldahl nitrogen (TKN), nitrate (NO3-), total phosphorus (TP), and orthophosphate (PO43-) within a 16-year-old bioretention system. A combination of k-means clustering and non-parametric statistical tests (Kruskal-Wallis and Wilcoxon rank-sum tests) were employed to analyze nutrient concentrations across different soil depths and media types. The Ozark Cavefish is one of multiple species of conservation concern found in Cave Springs Cave and Logan Cave in Northwest Arkansas (NWA). Over the past two decades, the urbanization of NWA has negatively impacted the recharge areas of both caves, and in turn the water quality of both cave streams. The changing climate has also altered temperature and precipitation, which influence many aspects of karst groundwater. These effects are shown in the historical water quality records for both caves. Modern broiler production faces persistent challenges from metabolic and skeletal disorders, particularly ascites and bacterial chondronecrosis with osteomyelitis (BCO), which compromise animal welfare and production efficiency. This dissertation examines two key factors that contribute to these conditions: mitochondrial gene expression associated with pulmonary hypertension syndrome (PHS) and the role of intestinal barrier integrity in mitigating lameness. In the first study, the expression of mitochondrial genes COXII and ATP6 was evaluated in the breast, thigh, and lung tissues of three broiler genetic lines—ascites-susceptible (SUS), ascites-resistant (RES), and unselected relaxed (REL). Interventions to mitigate the impact of racism on health are necessary and important to reduce disease burden and improve health outcomes for people of color.
