Purpose: Determine if molecular classification using mismatch repair (MMR) and p53 protein expression predicts recurrence-free survival (RFS) and overall survival (OS) in endometrial cancer (EC) patients treated with chemotherapy and radiation (CRT) versus chemotherapy (CT).

Methods: GOG-0258, a phase III randomized trial (NCT00942357), compared CRT to CT. Immunohistochemistry assessed MMR and p53 status. Kaplan-Meier curves and adjusted Cox models analyzed survival outcomes by molecular subtype.

Results: ECs classified as deficient MMR (dMMR) (27 %), p53 abnormal (p53abn) (24 %), and p53 wild type (p53wt) (49 %). p53abn were more frequent in patients that were older, Black, and had …

Purpose: Preclinical studies demonstrated that dual inhibition of epidermal growth factor receptor (EGFR) and vascular endothelial growth factor (VEGF) pathways delay the emergence of resistance to EGFR tyrosine kinase inhibitors (TKIs), and in trials with first-generation EGFR TKIs, the combination of EGFR VEGF pathway inhibitors prolonged progression-free survival (PFS).

Methods: The RAMOSE trial (ClinicalTrials.gov identifier: NCT03909334, HCRN LUN-18-335) is a randomized, open-label multicenter phase II study comparing osimertinib with ramucirumab (arm A) to osimertinib (arm B) for initial treatment of metastatic EGFR-mutant non-small cell lung cancer (NSCLC) with 2:1 random assignment. Cancer research remains clinically unmet in many areas due to limited access to patient samples and the lack of reliable model systems that truly reflect human cancer biology. The emergence of patient-derived induced pluripotent stem cells and engineered human pluripotent stem cells (hPSCs) has helped overcome these challenges, offering a versatile alternative platform for advancing cancer research. These hPSCs are already proving to be valuable models for studying specific cancer driver mutations, offering insights into cancer origins, pathogenesis, tumor heterogeneity, clonal evolution, and facilitating drug discovery and testing. Non-muscle-invasive bladder cancer (NMIBC) often recurs and can progress to MIBC due to resistance to treatments like intravesical chemotherapy or Bacillus Calmette-Guérin (BCG). Therefore, we established the Gemcitabine-Resistant Cells (GRCs) to study the molecular evolution under external pressure. A 63-gene Chemoresistance-Motility (CrM) signature was created to identify stage-specific traits of GRCs. This signature was tested on 1846 samples using log-rank tests and Cox regression to evaluate clinical utility. Early and intermediate resistance stages showed increased cell motility and metastatic potential. FAK, PI3K-AKT, and TGFβ pathways were activated first, followed by MAPK signaling. Introduction: Uveal melanoma remains a disease with aggressive behavior and poor prognosis despite advances in clinical management. Because monotherapy with immune checkpoint inhibitors has led to limited improvement in response rates, combination with other agents that act on the biological basis of oncogenesis has been proposed as a possible therapeutic strategy.

Methods: We designed a phase 1b trial to test the safety and tolerability of selinexor in combination with immune checkpoint inhibitors in patients with advanced uveal melanoma. Analysis of ecosystem impacts from offshore wind (OSW) farm development has primarily focused on localized effects. However, in Eastern Boundary Current Upwelling Systems (EBUS) like the California Current, OSW farms can modify the intensity and spatial structure of wind-driven upwelling, inducing non-local (tens of kms away) changes to seawater temperature. Recent numerical modeling research determined that a hypothetical upper bound full buildout of OSW farms in central California could warm coastal waters through a reduction in upwelling. Aggressive behavior is ubiquitous across many contexts, including defense of territories, mates, and offspring. For decades, researchers have detailed the effect of aggressive behavior on physiology, but our understanding of these mechanisms in females lags behind that of males, despite the fact that female aggression is widespread, particularly in the context of maternal defense of eggs or offspring (i.e., maternal aggression). Here, we measured effects of a social challenge on brain gene expression in free-living incubating females. Diseases are ubiquitous in natural systems, with broad effects across populations, communities and ecosystems. However, the drivers of many diseases remain poorly understood, particularly in marine environments, inhibiting effective conservation and management measures. We examined biogeographical patterns of infection in the foundational seagrass Zostera marina by the parasitic protist Labyrinthula zosterae, the causative agent of seagrass wasting disease, across >20° of latitude in two ocean basins. We then identified and characterized relationships among wasting disease prevalence and a suite of host traits and environmental variables. This dissertation explores how culturally sustaining pedagogy and arts-based learning can engage multilingual learners of English as an Additional Language (EAL) in Burkina Faso. The research emphasizes the incorporation of local cultural elements and learners' linguistic backgrounds into the EAL curriculum to address challenges such as limited classroom interactions (Somé-Guiebré, 2020) and monolingual instructional strategies with inadequate cultural representation (Ndlangamandla, 2024). This qualitative study integrates culturally relevant materials, including Forum Theater, slam poetry, and griot-style storytelling, to create an engaging and inclusive learning environment. Natural selection, a cornerstone of evolutionary biology, shapes the adaptations organisms develop to survive environmental pressures. This paper explores how natural selection drives biological adaptations to radiation exposure. We examine the genetic mechanisms at play, exemplified by the enhanced DNA repair capabilities observed in bacteria like Escherichia coli (E. coli) following exposure to radiation. We then investigate adaptations in humans residing in high-background radiation areas, highlighting potential genetic variations for radiation resistance. Finally, the contemporary relevance of natural selection is discussed, emphasizing its role in the emergence of antibiotic-resistant bacteria and the need for sustainable medical practices. Background: Intramuscular injection of botulinum toxin type A is a first-line pharmacotherapy for adults with upper limb spasticity (ULS). However, reemergence of symptoms within 12 weeks of treatment is common and longer-lasting treatments are needed.

Objective: To evaluate the efficacy and safety of three doses of DaxibotulinumtoxinA for Injection (DAXI) for treatment of ULS in adults with stroke or traumatic brain injury.

Intervention: Intramuscular injections of placebo (N = 24), DAXI 250 U (N = 22), DAXI 375 U (N = 19), or DAXI 500 U (N = 18) to the suprahypertonic muscle (SMG) and other muscle groups.

Background: Most extremely preterm (EP) infants are vitamin D deficient (serum 25-hydroxyvitamin D levels below 20 ng/mL), and optimal supplementation practices for EP infants remain unknown. Our objective is to assess current vitamin D supplementation practices in U.S. neonatal intensive care units (NICU) for EP infants to provide baseline information for the design of future clinical trials.

Methods: We conducted an online survey to study vitamin D intake and supplementation practices in U.S. NICUs caring for EP infants. Descriptive statistics compared responses by affiliation and level of care.

The enhancement of predictive accuracy of the first-order integer autoregressive (INAR 1) model is the focus of the study through the effective amalgam of machine learning techniques and classical statistics. INAR (1) model structures are key elements for the prediction of nonnegative integer-valued time series processes. Conventional estimation methods include Classical Least Squares (CLS), Conditional Maximum Likelihood (CML), and Yule-Walker (YW) as compared with advanced methods like Support Vector Regression (SVR) and Genetic Algorithms (GA). On April 7, 2023, a federal judge issued a nationwide stay on the United States Food and Drug Administration (FDA) approval of the abortifacient medication mifepristone. It was instantly a landmark case, decried as the first time in over one-hundred years that a federal court nullified an FDA drug approval. A few hours later, a second federal district court enjoined FDA restrictions on mifepristone. Two federal courts substantively evaluating FDA drug approval data in one day is unprecedented. It begs the question: will courts overturn FDA drug approvals again?

In this autoethnography, I am using my personal experiences as data to examine the inextricable personal, political, social, and cultural aspects of one of the most decisive ruptures in Hungarian “progressive” activism of the second half of the 2010s. In the mid-2010s, I was part of an informal collective of women that started to outline a feminist approach critical of mainstream individualistic renditions of women’s rights as social progress, thereby diverging fundamentally from the country’s institutionalised feminism. The relationship between gender and social construction has long been discussed in feminist and gender studies scholarship. However, the current discussion on gender has run up against a blockage of misapplication of social construct theory. To untangle this blockage, my analysis applies the philosophy of social construction as explained by Ian Hacking in his 1999 book The Social Construction of What? and the framework of gender as a social structure as put forth by feminist sociologist Barbara Risman since the early 2000s. Arginine modification can be a "switch" to regulate DNA transcription and a post-translational modification via methylation of a variety of cellular targets involved in signal transduction, gene transcription, DNA repair, and mRNA alterations. This consequently can turn downstream biological effectors "on" and "off". Arginine methylation is catalyzed by protein arginine methyltransferases (PRMTs 1-9) in both the nucleus and cytoplasm, and is thought to be involved in many disease processes. Background: Partial anterior cruciate ligament (ACL) tears, particularly injuries detected with magnetic resonance imaging (MRI) but stable on ligamentous examination, appear to be recognized more frequently; however, there remains no consensus management for this complex pathology.

Purpose: To present the results of a survey of a group of international ACL experts on the management of partial ACL injuries.

The metabolome and lipidome comprise the thousands of molecular compounds in an organism. Molecular compounds consist of the upstream metabolic components of intracellular reactions or the byproducts of cellular pathways. Molecular and biochemical perturbations are associated with disorders in newborns and infants. The diagnosis of inborn errors of metabolism has relied on targeted metabolomics for several decades. Newer approaches offer the potential to identify novel biomarkers for common diseases of the newborn and infant. There are several pharmacologic agents that have been touted as guideline-directed medical therapy for heart failure with preserved ejection fraction (HFpEF). However, it is important to recognize that older adults with HFpEF also contend with an increased risk for adverse effects from medications due to age-related changes in pharmacokinetics and pharmacodynamics of medications, as well as the concurrence of geriatric conditions such as polypharmacy and frailty. Conflicted Help-Seeking: Vulnerable And Grandiose Narcissistic Traits In College Students, Daniel G. Lannin, Rachel Brenner, Jake R. Solka, Amanda Martin, Anthony Hermann

This study highlights distinct patterns in help-seeking behavior among college students with narcissistic traits, focusing on grandiose and vulnerable manifestations of these traits. Two undergraduate samples (N₁ = 398, N₂ = 424) completed measures of narcissistic traits, stigma, psychological distress, and help-seeking intentions. Higher levels of vulnerable narcissistic traits were positively linked to help-seeking willingness, with indirect effects through heightened distress (positive) and stigma (negative). In contrast, grandiose narcissistic traits showed no consistent direct or indirect effects on help-seeking behavior. Genome-wide association studies (GWAS) on the Middle Eastern population, including the United Arab Emirates (UAE), have been relatively limited. The present study aims to investigate genotype-face morphology associations in the UAE population through Genome Wide Association Studies (GWAS). Phenotypic data (44 face measurements) from 172 Emiratis was obtained through three-dimensional (3D) scanning technology and an automatic face landmarking technique. GWAS analysis revealed associations of 19 genetic loci with six face features, 14 of which are novel. Background: Alzheimer’s disease (AD) is a progressive neurodegenerative disorder with poorly understood pathology. Elevated tau, phospho-tau and mitochondrial dysfunction are significantly correlated with an increased risk of AD and are therefore targets for disease-modifying therapy. In this study, we examined the effects of polyphenolic extracts from six different varieties of sorghum: Shawaya short black-1 (Black), IS1311C (Brown), QL33/QL36 (Red), B923296 (Red), QL12 (White), and QL33 (Red) on the attenuation of beta amyloid-induced phospho-tau levels, total tau levels, and mitochondrial dysfunction in neuronal cells.
