The mutational landscape of phylogenetically ultraconserved elements (UCEs), especially those in noncoding DNAs (ncUCEs), and their functional relevance in cancers remain poorly characterized. Here, we perform a systematic analysis of whole-genome and in-house targeted UCE sequencing datasets from more than 3000 patients with cancer of 13,736 UCEs and demonstrate that ncUCE somatic alterations are common. Using a multiplexed CRISPR knockout screen in colorectal cancer cells, we show that the loss of several altered ncUCEs significantly affects cell proliferation. In-depth functional studies in vitro and in vivo further reveal that specific ncUCEs can be enhancers of tumor suppressors (such as ARID1B) …

Brain metastases (BrMs) are the most common brain tumors in patients and are associated with poor prognosis. Investigating the systemic and environmental factors regulating BrM biology represents an important strategy to develop effective treatments. Toward this goal, we explored the contribution of the gut microbiome to BrM development by using in vivo breast-BrM models under germ-free conditions or antibiotic treatment. This revealed a detrimental role of gut microbiota in fostering BrM initiation. We thus evaluated the impact of antibiotics and BrM outgrowth on the gut-brain axis. We found the bacterial genus Alistipes was differentially present under antibiotic treatment and BrM …

Surgical removal of primary tumors reverses tumor-mediated immune suppression in pre-clinical models with metastatic disease. However, how cytoreductive surgery in the metastatic setting modulates the immune responses in patients, especially in the context of immune checkpoint therapy (ICT), is not understood. We report the first prospective, pilot, non-comparative clinical trial (NCT02210117) to evaluate the feasibility, clinical benefits, and immunologic changes of combining three different ICT-containing strategies with cytoreductive surgery or biopsy for patients with metastatic clear cell renal cell carcinoma. Primary safety endpoint of this trial has been met, with 43 patients completing cytoreductive surgery, 36 patients undergoing …

Nuclease-deactivated Cas (dCas) proteins can be used to recruit epigenetic effectors, and this class of epigenetic editing technologies has revolutionized the ability to synthetically control the mammalian epigenome and transcriptome. DNA methylation is one of the most important and well-characterized epigenetic modifications in mammals, and while many different forms of dCas-based DNA methyltransferases (dCas-DNMTs) have been developed for programmable DNA methylation, these tools are frequently poorly tolerated and/or lowly expressed in mammalian cell types. Further, the use of dCas-DNMTs has largely been restricted to cell lines, which limits mechanistic insights in karyotypically normal contexts and hampers translational utility in the …

Overview Of Chromatographic Methods Of Analysis Of Penicillins In Food Matrices Of Animal Origin, Tarekegn Tadesse Unade, Gudeta Duga Geresu, Pawar A. Krishnamanjari
International Journal of Health and Allied Sciences
Penicillins are β-lactam antibiotics indicated for preventing and treating diseases in veterinary medical practice. Improper use of these antibiotics in animals may result in residues in foodstuffs such as milk, egg, and meat, which can cause unwanted effects like allergy, and development of bacterial resistance. So, it is important to regulate the use of penicillins in foodstuffs of animal origin. Regulatory bodies in different countries established maximum residue levels to be allowed in food matrices. To address the regulatory requirements, food matrices need to be investigated with proper methods of analysis. Several analytical methods were developed and validated to determine …

Antimicrobial resistance (AMR) is a major global health problem. The growing number of antibiotic-resistant microbes needs the development of novel antibiotics. Nanotechnology shows potential for overcoming bacterial resistance. Nanoparticles, particularly those around 100 nm, display higher antibacterial activity by using methods that differ from traditional antibiotics. When particle size reduces, materials such as zinc and silver improve their antibacterial action. There are several techniques to evaluate nanoparticle antibacterial activity, each having merits and downsides. This study critically evaluates selected literature, identifies bactericidal mechanisms, and discusses assessment procedures, emphasizing the importance of innovative approaches to bacterial infections.

Evaluation Of Physical And Chemical Condition Of A Group Of Waterloggedarchaeological Woods From The Yenikapı 16 Shipwreck, Sedef Hetto, Namik Kiliç, Asli Gökçe Kiliç
Turkish Journal of Earth Sciences
This study examined the physical and chemical condition of waterlogged archaeological wood (WAW) samples from the Yenikapı 16 shipwreck of plane (Platanus L.), elm (Ulmus L.), pine (Pinus L.), and fir (Abies L.), representing species with different wood properties. The maximum water content (MWC) and basic density (BD) values of 74 WAW samples from the shipwreck were calculated, and 7 of 74 samples were selected for analysis using scanning electron microscopy (SEM), X-ray fluorescence (XRF), and Fourier transform infrared spectroscopy (FTIR). The availability of samples, appropriate size, the species of the wood, and the wood’s degradation were all considered. The …

The Neolithic lifestyle, which began in the 12th millennium BP with the emergence of villages in the core region extendingfrom Iran to central and southeastern Anatolia and Syria, began to spread to neighboring geographies, such as the Marmara region,in the 9th millennium BP. During the 8.2 ka event that occurred between 8300 and 7900 BP, cold and dry conditions prevailed in thenorthern hemisphere. During the winter months, the humidity decreased and cooling of about 6 ± 2 °C was observed. In order totest whether the 8.2 ka event was also effective in the İstanbul region, a set of inorganic geochemical …

In this work, we report the observation of uncatalyzed ene reactions between several phenylpropanoid compounds and diethyl azodicarboxylate (DEAD). For allylbenzene, the reaction produces the ene product at molar ratios of up to 1:2 of allylbenzene to DEAD. At higher levels of DEAD, more complex reactions are observed. For the reaction between methyl eugenol and DEAD, similar ene reaction products have been found. However, the reaction of eugenol with DEAD is more complex; in addition to the ene reaction, other reactions happen at the same time. Most of the structures of the resulting products have been elucidated using NMR spectroscopy …

The Central Plains of China is one of the earliest domestication hubs for broomcorn and foxtail millet. This region nurtured rich archaeological cultures and experienced extensive interactions with neighboring populations during the Neolithic period. However, limited genomic research has hindered a clear understanding of the maternal genetic structure and population expansion history in this era. Here, we report 12 complete mitochondrial genomes from individuals of the Yanzhai site population from Henan province of China during the Longshan Culture period, revealing a high level of maternal genetic diversity. The close genetic distance between these individuals and those of ancient populations from …
