Buffer Vs. Swabs: Efficacy Of Carrier Nucleic Acids In Robotic Extractions, Shawn Cecil

This study compared the efficiency of two carrier nucleic acid methods, specifically using salmon sperm DNA (spDNA), in robotic solid-phase extractions of cotton swab samples. Robotic extractions reduce processing time and workload but result in substantially lower DNA recovery than manual organic extraction. This research tested 10 µg of spDNA being added to the lysis buffer or pre-treating the swabs with it before DNA was added. Both methods showed a significant increase in recovery compared to extractions with no carrier. In the buffer extractions, recovery ranged from 69.4% to 117.5% and the swabs being pre-treated ranged from 84.2% to 125.8% …

The prevalence of type 2 diabetes mellitus (T2DM) in children and adolescents has increased substantially over recent decades, paralleling rising rates of pediatric obesity and metabolic risk. Youth-onset T2DM is characterized by significant insulin resistance accompanied by an inadequate compensatory insulin response, resulting in progressive hyperglycemia and early beta-cell dysfunction. Incidence varies widely among racial and ethnic groups, with the highest rates observed in non-Hispanic Black, Hispanic, Native American, Asian American, and Pacific Islander youth. Risk factors include obesity, family history, high-risk ethnicity, pubertal insulin resistance, prenatal metabolic exposures, and comorbid conditions such as polycystic ovary syndrome. Objectives Executive function deficits in bipolar disorder (BD) are major contributors to disability in older age BD (OABD). We investigated the difference between OABD and age-equated healthy comparators (HC); and, in the OABD group, the associations of executive function with age, symptom severity, global cognition, and daily functioning. Design Cross-sectional analysis of executive function in OABD versus HC. Setting Analysis of large archival dataset harmonized from 12 international OABD studies. The main purpose of this study is to determine if there is a correlation between pollen counts and total canine IgE antibody levels of dogs living in Louisville, KY. As universities grapple with funding cuts and heightened social and political pressure, the emerging field of Discipline-Based Education Research (DBER) aims to improve student learning outcomes through experimental instruction. While using instruction as an experiment, with hypotheses and experimental tests, DBER research may affect student learning and experience in unintended ways for students involved in these classes. BACKGROUND: Previous clinical, radiological and machine learning studies have predicted 90-day outcomes following subarachnoid hemorrhage (SAH). The present study was designed to determine whether early changes in mRNA expression of immune, clotting and other genes expressed in peripheral blood can predict patient outcomes at 90 days after SAH and possibly provide insights into the molecular factors that promote good versus poor outcomes.

METHODS: Peripheral blood was drawn after SAH and from vascular risk factor controls (VRFC) and RNAseq performed to measure mRNA expression. A mixed effects regression model identified potential predictors and machine learning algorithms derived the best predictors of …

Purpose: Radiation-induced heart damage is a significant concern in the treatment of non-small cell lung cancer (NSCLC) that can have debilitating or life-threatening consequences. Current strategies focus on minimizing heart exposure, but individual susceptibility varies. Existing evidence also suggests that a uniform "one-size-fits-all" dosimetric constraint for the heart may not be optimal for all patients.

This comprehensive literature review synthesizes the most recent evidence and evolving clinical guidelines concerning percutaneous coronary intervention (PCI) from 2015 to 2025. The study integrates findings from 42 peer-reviewed trials, meta-analyses, registries, and international guideline statements issued by the American College of Cardiology/American Heart Association (ACC/AHA), European Society of Cardiology (ESC), and related societies. Results reveal a decade-long evolution in PCI practice marked by a shift from anatomy-driven to physiology and imaging-guided decision-making, supported by tools such as fractional flow reserve (FFR), instantaneous wave-free ratio (iFR), intravascular ultrasound (IVUS), and optical coherence tomography (OCT). Persistent postoperative opioid use (PPOU) is an emerging challenge in pediatric perioperative care, with rates as high as 4.7% in opioid-naive adolescents. Despite advances in multimodal analgesia, current protocols often fail to prevent long-term opioid exposure, particularly after high-risk surgeries such as spinal fusions. While multiple strategies exist to reduce PPOU in children, including regional anesthesia and non-opioid analgesics, this review specifically focuses on methadone and pharmacogenomic-guided opioid prescribing as promising approaches. Emerging single-photon camera (SPC) technologies have unique challenges in data acquisition and processing. Unlike conventional sensors that produce a single 8- to 16-bit brightness value per pixel, SPCs record photon arrivals with many more samples per pixel, using high floating-point precision for each photon collected. This means that they must handle potentially millions of timestamps, especially at higher spatial resolutions and in the presence of ambient light, creating bottlenecks within the pixel circuitry. The fabrication of a new class of biomimetic biomaterials is reported using nanostructured microgel pastes formed from “overpacked” assemblies of ultrasoft poly(N-isopropyl acrylamide-co-acrylic acid) microgels and their composites with collagen. Despite the solid-like nature of microgel pastes, collagen fibrillogenesis is robust and rapid, with a 3D collagen network forming throughout the paste volume. Structural organization within the composite is interrogated via a suite of microscopy methods, while rheological characterization provides insight into the static and dynamic mechanical properties of the materials. The parent–child relationship has been found to be a predictor of children’s academic and socio-emotional outcomes, with parent-child closeness linked to motivation and persistence (Mo & Singh, 2008; Silinskas & Kikas, 2022) and parent-child conflict associated with emotional and behavioral difficulties (Dotterer et al., 2008; Pinquart, 2017). There is insufficient understanding of the molecular basis of prostate cancer (PCa) across different populations. We perform a large-scale proteome-wide association study (PWAS) to identify proteins with genetically regulated expression in plasma to be associated with PCa risk across populations. We develop genetic prediction models for expression of 1578, 1993, 1218, and 1390 proteins for African (n = 450), European (n = 758), Asian (n = 289), and Hispanic/Latino (n = 474) males, respectively, and evaluate associations of genetically regulated protein expression with PCa risk in 19,391 PCa cases and 61,608 controls of African population, 122,188 cases and 604,640 controls …

The Crane Trust is a regional nonprofit conservation organization that has protected land in the biologically important Central Platte River Valley for over four decades. A mammal inventory was conducted on the Crane Trust’s main habitat complex in 1980. Since, a variety of environmental conditions have shifted locally, regionally, and globally. Microfluidic oxygenators promise to advance extracorporeal membrane oxygenation (ECMO) devices with enhanced hemodynamics and low prime volume. We are developing a silicon-based membrane oxygenator that will offer improved gas transfer and fluid flow control. Polyethylene glycol (PEG) has been used to improve hemocompatibility by providing excellent resistance to protein adsorption. Here, we characterized a polyethylene glycol surface modification of composite silicon–PDMS membranes to evaluate their effects on microfluidic oxygenator properties. X-ray photoelectron spectroscopy (XPS) and water contact angle goniometry confirmed successful PEG attachment, evidenced by the presence of characteristic C-O bonds and increased hydrophilicity, which was stable for 2 weeks. …

Backpropagation (BP) relies on biologically implausible mechanisms like global error transport. This project investigates two alternatives: Error-Driven Local Representation Alignment (LRA), which utilizes local error loops, and Predictive Forward-Forward (PFF), which employs forward-forward algorithm and "goodness" optimization to eliminate backward passes. We quantify the trade-off between biological plausibility and efficiency by measuring the convergence lag inherent to localized updates. Theoretically, we analyze how PFF and LRA resolve the weight transport problem and enable local credit assignment, positioning these frameworks as viable candidates for energy-efficient "mortal computation" on neuromorphic hardware.

Protein function prediction is of high importance when it comes to applications such as bioinformatics, of which drug discovery and disease research, in particular, are key components. Experimental methods for determining protein function are accurate but typically cumbersome and expensive, and this is why computational approaches are needed for this. This study specifically explores, through a research-based approach, different deep learning methods to predict protein functions using amino acid sequences, structural information, and protein interaction data. This Article advocates for a fundamental shift in the way artificial intelligence (“AI”) consciousness is evaluated, moving beyond anthropocentric models that rely on human cognitive benchmarks. Traditional tests, such as the Turing Test, assume that human-like behavior is the ultimate criterion for consciousness, overlooking the possibility of novel, non-human modes of consciousness. Individuals And Couples With Infertility: Quality Of Life, Relationship Satisfaction, Resilience, Depression, And Shame, Niko C. Wilson, Sejal Barden

Journal of Counseling Sexology & Sexual Wellness: Research, Practice, and Education

This study examined the relationship between fertility-related quality of life, relationship satisfaction, resilience, depression, and shame in individuals and couples with infertility (N = 556). Using structural equation modeling (SEM) and the actor-partner interdependence model (APIM), the study assessed gender covariates and dyadic influences in couples (n = 52). SEM results showed fertility-related quality of life impacted resilience, depression, and shame, while its relational aspect influenced relationship satisfaction. APIM identified partner effects on relationship satisfaction and shame. Findings provide insights for counseling practices, promoting relationship well-being in couples facing infertility.

Journal of Counseling Sexology & Sexual Wellness: Research, Practice, and Education

The purpose of the study was to clarify the definition of sex positivity and identify sex-positive attitudes, knowledge, and skills through the development and evaluation of the Sex Positivity Inventory for Counselors (SPI-C). The exploratory factor analysis supported a six-factor, 34-item scale, accounting for 32.9% of the variance within the study. The SPI-C had an excellent internal consistency of 0.96 and consisted of the following factors: 1) Procedural Sex Positivity, 2) Comfortability with Sexuality, 3) Perceived Sex Positivity, 4) Fundamental Sex Education, 5) Sexological Worldview, and 6) Unconditional Positive Sexual Regard. Implications for practice, counselor education, and supervision are discussed.

We describe a single Math Teachers’ Circle session during which a group of teachers collectively engaged in a “mini mathematics research experience.” This provides a model for providing research experiences for mathematics teachers through content-based professional development programs. We conjecture that these experiences will have many of the same benefits as research experiences for teachers in lab sciences.

This project investigates the primary anthropogenic drivers of wildlife admissions at Lake Erie Nature and Science Center using a quantitative analysis of 10,172 intake records from the WILD-ONe database. Window collisions were by far the leading cause of admission, accounting for 54% of all intakes. Orphaning represented 12%, followed by vehicle collisions (5%) and domestic cat attacks (4%). By integrating findings from current wildlife rehabilitation research, this study identifies evidence-based strategies, such as bird-safe window treatments, public education on natural animal behavior, and targeted urban planning, that can substantially reduce preventable injuries. When rivers achieve a great enough flow rate, sediment along their riverbed becomes mobilized and transported downstream. This process is known as bedload transport. As bedload transport initiates, micro-collisions of various sediments and gravel generate underwater acoustic signals that can be detected by hydrophones.
