Per- And Polyfluoroalkyl Substances (Pfas) Effects On Tissue Disposition, Proteome, And Health Effects, Juliana Agudelo Areiza

Open Access Dissertations

Per- and polyfluoroalkyl substances are a class of nonbiodegradable synthetic aliphatic environmental toxicants that are ubiquitous in the environment. A small subset, specifically perfluorooctanesulfonic acid (PFOS) and perfluorooctanoic acid (PFOA), have been designated as highly hazardous to human health by the Environmental Protection Agency (EPA) under the Comprehensive Environmental Response, Compensation, and Liability Act (CERCLA). PFOA and PFOS-associated adverse health effects include, but are not limited to, immune suppression, thyroid dysfunction, increased serum cholesterol, liver damage (i.e., alanine aminotransferase (ALT)), and developmental toxicity (i.e., low fetal birth weight and neurotoxicity). In this doctoral dissertation, it was hypothesized that environmental PFAS …

A Socio-Ecological Investigation Of Ulithian Spearfishing, Joshua Pi Jan 2025

A Socio-Ecological Investigation Of Ulithian Spearfishing, Joshua Pi

Open Access Dissertations

Coral reef socio-ecological systems (SESs) characterize the interconnected relationships between coral reef ecosystems and reef-dependent communities. One such SES exists in Ulithi Atoll, Federated States of Micronesia, where Ulithians fish, steward, and govern their reefs through customary marine tenure, traditional ecological knowledge, and cultural taboos. In recent decades, rapid modernization in Ulithi has introduced new fishing technologies, transformed fishing practices, and ultimately reshaped the functioning of Ulithi’s traditional SES. One significant change was the introduction of spearfishing during the mid-20th century, which is more selective and individualistic compared to other community-centric fishing methods that have existed for generations. This …

“What’S Supp” Developing An In Vitro Model For Healthy Oral Biofilms, Alex Labossiere Jan 2025

“What’S Supp” Developing An In Vitro Model For Healthy Oral Biofilms, Alex Labossiere

Open Access Dissertations

The study of oral plaque has largely been influenced by the study of pathogens. Here in Chapter 1, I will discuss how this pathogenic focus has been dominant until the onset of microbiome research and begin to explain microbiomes in the context of supragingival plaque in homeostatic communities. From there we will then discuss the possible roots where dysbiosis may arise from plaque to cause disease and ways in which we can study healthy supragingival plaque by in vitro models.

In chapter 2, I will discuss molecular commensalism and how it can be viewed as a tool to further understand …

Open Access Dissertations

The present study examined the impact of The Greatest 8Ô text messaging service on parental involvement in promoting children’s mental health and wellness. A quasi-experimental design compared 63 subscribers and 115 non-subscribers of the service. Parents completed an online survey assessing confidence, engagement, beliefs about eight core mental health skills, and perceived child improvement, along with exploratory measures regarding information sources, preferred communication methods, and barriers to accessing resources. It was hypothesized that subscribers would report higher confidence, engagement, and beliefs than non-subscribers, that duration in the program would predict these outcomes, and that engagement would moderate the relationship between …

Ultra-Rapid Electrophilic Cysteine Arylation Using Pyridinium Salts, Bradley M. Lipka Jan 2025

Ultra-Rapid Electrophilic Cysteine Arylation Using Pyridinium Salts, Bradley M. Lipka

Open Access Dissertations

Small molecule-based bioconjugation enables in vitro and in vivo functionalization of proteins and peptides, significantly expanding the chemical space of biomacromolecules. Among these transformations, nucleophilic aromatic substitution (SNAr) of cysteine residues is a widely employed strategy for protein bioconjugation.

Although many reagents facilitate cysteine arylation, they are disadvantageous in two crucial aspects - limited synthetic modularity and undesired hydrophobicity. First, conventional cysteine arylating reagents are frequently activated by multiple electron-withdrawing groups (EWGs) on a single aromatic ring, leaving few synthetic handles to derivatize the reagents for advanced applications. Second, these labeling reagents are generally hydrophobic in nature and …

Introduction: Preeclampsia (PE) is a complex multisystem disorder of pregnancy associated with abnormal placentation, vascular anomalies, and systemic inflammation and hypertension. Previous research assessing inflammatory triggers of the condition used plasma, amniotic fluid, or explant samples. Studies using placental tissue from either vaginal or cesarean deliveries are confined to semiquantitative analysis using subjective scoring methods and generally involve a small sample size.

Methods: In this study, we have quantified the expression of inflammatory mediators by immunohistochemical image analysis of archived placental tissues obtained from cesarean delivery of preeclamptic, chorioamnionitic, and normal pregnancies.

Results: Among the inflammatory mediators, we found a …

The activity of nucleoside and nucleotide analogs as antiviral agents requires phosphorylation by intracellular enzymes. Phosphate-substituted analogs have low bioavailability due to the presence of ionizable negatively-charged groups. To circumvent this limitation, several prodrug approaches have been proposed and developed. Herein, we hypothesized that the conjugation of anti-HIV fatty acids with the nucleotide tenofovir alafenamide (TAF) (1) could improve the anti-HIV activity of the nucleotide. Several fatty acyl amide conjugates of TAF were synthesized and evaluated in comparative studies with TAF. The synthesized compounds were evaluated as racemic mixtures for anti-HIV activity in vitro in a single-round HIV-1 infection assay …

Radio frequency identification system (RFID) tags are tiny electronic devices that include a distinctive code that can be detected by a reader that supports RFID. The string of characters is used for recognizing and monitoring the object to which the tag is connected. RFID tags interact with RFID readers using radio frequency. While the tag comes into close proximity to the scanner, that device sends a radio signal that triggers the tag. The tag then responds by sending its unique identity. RFID pinpointing ability is verified experimentally in two different techniques, one utilizing radar and the other a photosensitive transition. …

Making Headway On Sports Concussion: Sport-Specific Strategies For Concussion Prevention, Marcus Moore Jan 2025

Making Headway On Sports Concussion: Sport-Specific Strategies For Concussion Prevention, Marcus Moore

All Faculty Publications

Concussions in sports are a significant public health concern worldwide. Concussion is a traumatic brain injury (‘TBI’) resulting from biomechanical forces transmitted to the head that disrupt normal neurological function. Globally, an estimated 69 million TBIs occur annually. Concussive effects can persist as long-term syndromes (post-concussion syndrome), or compound themselves into life-threatening emergencies (second-impact syndrome). Chronic, even subconcussive, impacts can ultimately lead to degenerative neurological conditions (chronic traumatic encephalopathy). Troublingly, to date, there remains no medical cure for concussions. Prevention is thus paramount, as well as management, to avoid the compounding of injury or dangerous complications, as noted. Sports and …

Environmentally Friendly Chelation For Enhanced Algal Biomass Deashing, Agyare Asante, George Daramola, Ryan W. Davis, Sandeep Kumar

Civil & Environmental Engineering Faculty Publications

High ash content in algal biomass limits its suitability for biofuel production by reducing combustion efficiency and increasing fouling. This study presents a green deashing strategy using nitrilotriacetic acid (NTA) and deionized (DI) water to purify Scenedesmus algae, which was selected for its high ash removal potential. The optimized sequential treatment (DI, NTA chelation, and DI+NTA treatment at 90–130 °C) achieved up to 83.07% ash removal, reducing ash content from 15.2% to 3.8%. Elevated temperatures enhanced the removal of calcium, magnesium, and potassium, while heavy metals like lead and copper were reduced below detection limits. CHN analysis confirmed minimal …

Laboratory Investigation Of Shape And Initial Orientation Effects On Surf Zone Object Migration, Temitope E. Idowu, Jack A. Puleo

Civil & Environmental Engineering Faculty Publications

Discarded objects like munitions in marine environments pose public safety risks. The behavior of various density objects deployed at four cross-shore positions in the surf zone of a large-scale 120 m x 5 m x 5 m wave flume were observed under different forcing conditions. Net migration was predominantly directed offshore, with approximately 70 % offshore migration observed near the outer surf zone. Density, shape, and initial orientation were identified as important to object behavior, with density acting as the dominant driver in 67 % of the object pairing scenarios. The influence of shape and initial orientation on net migration …

Streamlining Wildlife Regulation, Robert L. Fischman Jan 2025

Streamlining Wildlife Regulation, Robert L. Fischman

Articles by Maurer Faculty

A recent burst of revisions in the bedrock regulations governing projects that may adversely affect wildlife represents a generational shift in policy. Streamlining federal decision -making drove much of this reform. Streamlining offers a path to address climate change without abandoning a longstanding commitment to wildlife conservation. The conservation community recognizes the need to build new infrastructure to reduce the rate of climate change through air emissions. The most urgent priority is decarbonizing the electrical ~rid through renewable energy generation and a better transmission network. Adaptation to the "new abnormal" of climate-driven environmental disruption demands federal approvals for a host …

Data Injustice In Global Justice, Asaf Lubin, Cherry Tang

Articles by Maurer Faculty

In May 2020, the United Nations Secretary-General unveiled a sweeping “Data Strategy for Action by Everyone, Everywhere,” seeking to unlock the UN’s “full data potential.” The International Criminal Court’s Office of the Prosecutor followed suit, declaring in 2023 its intent to acquire advanced cyber forensic tools so as to hold the “widest range of digital evidence globally.” Across international institutions, data-driven governance has become the norm, with humanitarian agencies and tribunals transforming into “data hubs and information clearinghouses.” This Article critiques the unfettered datafication of global justice by international courts and organizations. These entities have aggressively expanded their data-driven operations …

Climate Prosecution As Climate Regulation, Cindy J. Cho Jan 2025

Climate Prosecution As Climate Regulation, Cindy J. Cho

Articles by Maurer Faculty

Last term, the Supreme Court weakened the federal government’s ability to regulate. Three new decisions—and one landmark case from the previous term—will affect everything from drug approvals to overtime pay to national security. One issue stands out as a political flashpoint that will prove particularly difficult to address via regulation in the wake of this term: climate change. Given the unlikelihood of congressional climate action any time soon, those concerned about the risks of a warming planet will no doubt be looking for alternatives to regulation. Private and state civil lawsuits against polluters reflect an important legal tradition that must …

Reconstituting Corporate Power & Accountability, Cindy J. Cho, Donald Braman, Theresa Gabaldon

Articles by Maurer Faculty

Modern society faces a growing crisis of corporate impunity. While corporations generate immense value, they increasingly inflict harm at scales that dwarf those of traditional street crime. Decades of deregulation, unchecked corporate lobbying, and a judiciary actively dismantling the administrative state have created a dangerous accountability vacuum. And just as federal oversight is collapsing, the Supreme Court's expanding preemption doctrine is handcuffing state regulators. This Article proposes a paradigm shift: the revitalization of state criminal authority, a power largely shielded from federal interference. Upon securing a conviction or deferred prosecution agreement, states should mandate that the offending corporation conduct all …

Tuberculosis is one of the deadliest infectious diseases and continues to be a major health risk in many parts of the world. Even today, the century-old Bacillus Calmette-Guerin (BCG) vaccine is the only formulation on the market and is ineffective for several sections of the global population responsible for transmission. In the search for antigens that can mount a robust immune response, we have reported the recombinant expression and purification of two novel membrane proteins, the Cation transporter protein V (CtpV) and the Mycobacterial copper transporter B (MctB) present on the membrane surface of Mycobacterium tuberculosis. CtpV was tested …

Limbic predominant age-related TDP-43 encephalopathy neuropathologic change (LATE-NC) is highly prevalent in late life and a common co-pathology with Alzheimer's disease neuropathologic change (ADNC). LATE-NC is a slowly progressive, amnestic clinical syndrome. Alternatively, when present with ADNC, LATE-NC is associated with a more rapid course. With the emergence of anti-amyloid therapeutics, discrimination of LATE-NC from ADNC is critical and will lead to greater clinical recognition of amnestic patients without ADNC. Furthermore, co-pathology with LATE-NC may influence outcomes of these therapeutics. Thus there is a need to identify patients during life with likely LATE-NC. We propose criteria for clinical diagnosis of …

Background

Prehospital identification of large‐vessel occlusion (LVO) stroke facilitates the development of a regional triage protocol that could lead to an improvement in intrahospital and interhospital workflows. This requires stroke education of emergency medical services (EMS) personnel to improve stroke recognition, severity assessment, and prenotification. However, the current state of EMS personnel's knowledge of LVO stroke, their training for stroke severity assessment, and their preferences for educational methods are unknown. We conducted a nationwide EMS survey across the United States to glean insights that can inform and customize future EMS educational efforts.
