Every heartbeat depends on cyclical contraction-relaxation produced by the interactions between myosin-containing thick and actin-based thin filaments (TFs) arranged into a crystalline-like lattice in the cardiac sarcomere. Therefore, the maintenance of thin filament length is crucial for myocardium function. The thin filament is comprised of an actin backbone, the regulatory troponin complex and tropomyosin that controls interactions between thick and thin filaments. Thin filament length is controlled by the tropomodulin family of proteins; tropomodulin caps pointed ends of thin filaments, and leiomodin (Lmod) promotes elongation of thin filaments by a “leaky-cap” mechanism. In the United States, medical educators have expressed concern that an integrated and condensed pre-clerkship curriculum risks leaving gaps in student pharmacology knowledge. This study evaluates a remote-learning clinical pharmacology elective that integrates fundamental pharmacology concepts and principles of medication safety and explores how the course impacts senior medical students' self-reported readiness for residency. Clinical Pharmacology and Medication Safety is a remote elective course for senior medical students. Troponin C (TnC) is the Ca²⁺-sensing subunit of troponin that is responsible for activating thin filaments in striated muscle, and, in turn, for regulating the systolic and diastolic contractile function of cardiac muscle. The secondary structure of vertebrate TnC is mainly composed of α-helices, with nine helices named sequentially, starting from the amino terminus, from N to A–H. The N-helix is a 12-residue-long α-helix located at the extreme amino terminus of the protein and is the only helical structure that does not participate in forming Ca²⁺-binding EF-hands. Breast cancer is one of the most common cancers globally. Unfortunately, many patients with breast cancer develop resistance to chemotherapy and tumor recurrence, which is primarily driven by breast cancer stem cells (BCSCs). BCSCs behave like stem cells and can self-renew and differentiate into mature tumor cells, enabling the cancer to regrow and metastasize. Key markers like CD44 and aldehyde dehydrogenase-1 (ALDH1), along with pathways like Wingless-related integration site (Wnt), Notch, and Hedgehog, are critical to regulating this stem-like behavior of BCSCs and, thus, are being investigated as targets for various new therapies. Background

Chronic insomnia and obstructive sleep apnea commonly co-occur. Few studies have explored the neurophysiological and neurocognitive characteristics of COMISA, which could help guide improving treatment diagnostic tools and determining novel therapeutic targets. This study aims to explore the neurophysiological and neurocognitive characteristics of COMISA using electroencephalographic (EEG) spectral analysis and subjective and objective neurocognitive measurements.

Methods

Participants were from our community recruited OSA-insomnia-COMISA cohort with 206 included for our current analysis including 74 chronic insomniacs (CIs), 55 OSA patients and 77 COMISA patients. Codon Composition In Human Oocyters Reveals Age-Associated Defects In Mrna Decay, Pavla Brachova, Lane K. Christenson, Nehemiah S. Alvarez

Oocytes from women of advanced reproductive age exhibit diminished developmental potential, but the underlying mechanisms remain incompletely defined. Oocyte maturation depends on translational control of maternal mRNA synthesized during growth. We performed a computational analysis on human oocytes from women < 30 versus ≥40 years and observed that mRNA GC content correlates negatively with half-life in oocytes from young (< 30 yr) but positively with oocytes from aged (>40 yr) women. In young oocytes, longer mRNA half-life is associated with lower protein abundance, whereas in aged oocytes GC content correlates positively with protein abundance. During the GV-to-MII transition, codon composition stratifies stability: codons that support rapid translation (optimal) stabilize mRNA, while slow-translating codons (non-optimal) promote decay. Antibiotic resistance (AR) has become a critical global health crisis, threatening the efficacy of antibiotics and increasing healthcare costs, morbidity, and mortality. The One Health approach, which emphasizes the interconnectedness of human, animal, and environmental health, is increasingly recognized as an essential framework for addressing the complex drivers of AR. This review explores the mechanisms of resistance, such as genetic mutations, horizontal gene transfer, and biofilm formation, along with contributing factors such as the overuse of antibiotics in healthcare and agriculture. Introduction

Cardiovascular complications are the leading cause of morbidity and mortality in patients with type 2 diabetes (T2D), which is recognized as a major independent risk factor for coronary artery disease, stroke, peripheral vascular disease, and heart failure. The interrelationship between metabolic dysfunction and cardiovascular disease is complex and multifactorial, involving hyperglycemia, insulin resistance, inflammation, and oxidative stress. Myosins produce force and movement in cells through interactions with F-actin1. Generation of movement is thought to arise through actin-catalysed conversion of myosin from an ATP-generated primed (pre-powerstroke) state to a post-powerstroke state, accompanied by myosin lever swing2,3. However, the initial, primed actomyosin state has never been observed, and the mechanism by which actin catalyses myosin ATPase activity is unclear. Here, to address these issues, we performed time-resolved cryogenic electron microscopy (cryo-EM)4 of a myosin-5 mutant having slow hydrolysis product release5,6. [Summary] Cardiomyocytes endure physical stress from the myocardium environment while generating their own mechanical strains. The force generated by sarcomeres is transmitted both longitudinally to adjacent sarcomeres and laterally to the cytoskeleton via intermediate filaments. This mechanical stimulus impacts other organelles, including the nucleus, thus playing a vital role in sensing and signaling nuclear adaptations. Purpose

The research aims to understand the stories of women leaders who have demonstrated bravery in leadership. By analyzing their lived experiences through storytelling and narratives, it seeks to shed light on the challenges and motivations behind their brave actions, contributing to a deeper understanding of bravery in leadership within gender and organizational contexts.

In 2017, Finland began levying tuition fees on non-European Union (EU)/European Economic Area (EEA) international students. Using data from multiple Finnish government sources, we study the impact of tuition fee reform at various stages of the higher education enrollment selection process. Outcomes examined include how the policy change affects student application behavior, admissions and acceptances by institutions, and student enrollments. To identify the effects of the policy change, we employ a difference-in-differences approach using national register data and program-level tuition variations. Over the last few years, there has been a surge in anti-LGBTQIA+ and, specifically, anti-transgender, nonbinary, and gender non-conforming legislation. These laws target the rights of LGBTQIA+ students in K–12 schools, leading to policies censoring gender-inclusive curricula, mandating the outing of students, and restricting transgender students’ access to sports and facilities. The rapid emergence of these bills has outpaced research on how leaders—particularly justice-centred leaders—navigate these hostile policy contexts. In 2018, the U.S. government levied tariffs affecting more than $250 billion of Chinese products. As a result, many American buyers began turning to alternative suppliers to avoid the significant new costs associated with purchasing from China. Recent research found that transactions between U.S. buyers and Chinese suppliers fell by more than 18% immediately following the start of the trade war. However, the study also found that not all companies were affected equally: Chinese firms that were more innovative or more socially responsible were more likely to retain their U.S. buyers, while buyers were more likely to distance themselves from …

The Demographic Profiles of San Diego and Imperial Counties provides a snapshot of the two counties' vulnerable populations, including low-income communities, youth, immigrants, refugees, individuals experiencing homelessness, and retirement-age adults. While these counties differ in population size by more than an order of magnitude, the backdrop of California's rising cost of living, shifting immigration policies, income and health disparities, and the projections for an increasing aging population present compounding barriers to financial stability and access to services for all ages. Background: Rehabilitation methods that include anodal transcranial direct current stimulation (atDCS) and extended reality (XR) exercises have been used to enhance neural networks and improve functional performance in stroke patients, but the neuromuscular and neurophysiological mechanisms underlying these improvements are not fully understood. The purpose of this study was to examine the effects of atDCS during XR rehabilitation exercises on cortical, neuromuscular, and clinical outcomes of stroke survivors. Methods: Nineteen chronic stroke survivors were placed into either a transcranial direct current stimulation (tDCS) or a Sham group, without significant (p > 0.73) differences in the baseline levels of disability between groups. …

This study investigates how data privacy concerns influence user trust in TikTok across two major regions: the European Union (EU) and the United States (US). With over one billion active users worldwide, TikTok has faced intense scrutiny due to its Chinese ownership and concerns over data governance. Using the privacy calculus theory and the Stimulus–Organism–Response (SOR) model as theoretical foundations, the research explores the relationship between privacy concerns, trust, and continued use of TikTok. The persistent health disparities faced by Black, Indigenous, and People of Color (BIPOC) in the United States are deeply rooted in systemic racism embedded within medical education and clinical practice. Shaping Leadership: An Autoethnographic Exploration Of Inspirational Moments, Sabine Mathilde Maiberger

Leadership is a complex construct shaped by the interplay of theoretical knowledge and meaningful personal experiences that define us as people. This autoethnographic exploration investigates the question: How have inspirational moments shaped the leader I am today? Understanding how personal experiences influence leaders’ development is critical to leadership studies. To address this, nine of my pivotal life moments were selected and validated through personal artifacts, such as journals and letters, ensuring analytical depth. These moments were analyzed through servant, justice-oriented, and transformational leadership theoretical lenses. The Lived Experiences Of Trauma In Autistic Adults: A Thematic Analysis, Briana Trageser

A qualitative research study focused on Autistic adults explored the lived experiences of trauma. Participants included Autistic adults who have either been professionally diagnosed with Autism Spectrum Disorder or who self-identify as Autistic. Data was collected through a survey with 12 open-ended questions. A thematic analysis revealed six themes: (a) adverse childhood experiences, (b) exposure to violence and loss, (c) difficulty relating to others, (d) complex mind and sensory experiences, (e) medical and mental health challenges, and (f) autistic identity. This research is a qualitative phenomenological inquiry into reconnecting Indigenous participants’ experiences in how they navigate and integrate their cultural heritage, identity, and intergenerational experiences to shape their present and future lives. Previous literature examines characterizing Indigeneity, navigating multiple spaces, and culture as wellness. Rooted in Tribal Critical Race Theory, semi-structured interviews were conducted with nine participants who self-identified as Indigenous. A thematic analysis highlighted six themes of reconnection: a) oppression, b) ancestors, c) belonging, d) barriers to reconnection, e) learning, and f) connection with community. This qualitative hermeneutic phenomenological study explores the lived experiences of transracial adoptees of the global majority (BIPOC), focusing on their bicultural identity integration and its impact on mental health. During semi-structured interviews, nine participants shared their stories of navigating cultural and racial dynamics as adoptees in predominantly White adoptive families and communities. Storytelling, when using the National Storytelling Network’s definition, “the interactive art of using words and actions to reveal the elements and images of a story while encouraging the listener’s imagination,” can be found in most classrooms around the world. If we agree with Walter Fisher and see stories as content with “coherence” and “fidelity” (Fisher, 1987), then much of the communication in classrooms could be considered a form of storytelling. This dissertation examines the roles, challenges, and contributions of a Black Gay father, a demographic largely absent from academic discourse. While literature explores themes of the church, LGBTQ+ identities, and social justice, there remains a critical gap addressing the lived experiences of a Black Gay father. This study fills that void by elevating narratives as community activists, family leaders, and clergy members, acknowledging their transformative impact within familial and societal structures.
