Neurotensin (NTS), a small neuropeptide, was recently established as a key paracrine mediator of ovulation. NTS mRNA is highly expressed by granulosa cells in response to the luteinizing hormone (LH) surge, and multiple NTS receptors are expressed by cells of the ovulatory follicle. To identify the role of NTS receptors NTSR1 and SORT1 in ovulation in vivo, the dominant follicle of cynomolgus macaques (Macaca fascicularis) was injected with either vehicle control, the general NTS receptor antagonist SR142948, the NTSR1-selective antagonist SR48692, or the SORT1-selective antagonist AF38469. Human chorionic gonadotropin (hCG) was then administered to initiate ovulatory events. …

Every heartbeat depends on cyclical contraction-relaxation produced by the interactions between myosin-containing thick and actin-based thin filaments (TFs) arranged into a crystalline-like lattice in the cardiac sarcomere. Therefore, the maintenance of thin filament length is crucial for myocardium function. The thin filament is comprised of an actin backbone, the regulatory troponin complex and tropomyosin that controls interactions between thick and thin filaments. Thin filament length is controlled by the tropomodulin family of proteins; tropomodulin caps pointed ends of thin filaments, and leiomodin (Lmod) promotes elongation of thin filaments by a “leaky-cap” mechanism. Breast cancer is one of the most common cancers globally. Unfortunately, many patients with breast cancer develop resistance to chemotherapy and tumor recurrence, which is primarily driven by breast cancer stem cells (BCSCs). BCSCs behave like stem cells and can self-renew and differentiate into mature tumor cells, enabling the cancer to regrow and metastasize. Key markers like CD44 and aldehyde dehydrogenase-1 (ALDH1), along with pathways like Wingless-related integration site (Wnt), Notch, and Hedgehog, are critical to regulating this stem-like behavior of BCSCs and, thus, are being investigated as targets for various new therapies. Antibiotic resistance (AR) has become a critical global health crisis, threatening the efficacy of antibiotics and increasing healthcare costs, morbidity, and mortality. The One Health approach, which emphasizes the interconnectedness of human, animal, and environmental health, is increasingly recognized as an essential framework for addressing the complex drivers of AR. This review explores the mechanisms of resistance, such as genetic mutations, horizontal gene transfer, and biofilm formation, along with contributing factors such as the overuse of antibiotics in healthcare and agriculture. Transcription factors are frequent cancer driver genes, exhibiting noted specificity based on the precise cell of origin. We demonstrate that ZIC1 exhibits loss-of-function (LOF) somatic events in group 4 (G4) medulloblastoma through recurrent point mutations, subchromosomal deletions and mono-allelic epigenetic repression (60% of G4 medulloblastoma). In contrast, highly similar SHH medulloblastoma exhibits distinct and diametrically opposed gain-of-function mutations and copy number gains (20% of SHH medulloblastoma). Overexpression of ZIC1 suppresses the growth of group 3 medulloblastoma models, whereas it promotes the proliferation of SHH medulloblastoma precursor cells. Procedural terrain generation, the algorithmic creation of digital terrain, finds use in multiple types of digital media. As the capabilities of modern computation increase, the ability to create more and more realistic terrains fully procedurally at scale improves. Modern methods of procedural generation have also overlapped with these advances, most notably advances in hardware. To account for this, a survey was done of modern methods for procedural terrain generation. Smooth procedural noise functions are one of the backbones of procedural terrain generation. Bone remodeling is a dynamic process that requires a balance between bone resorption by osteoclasts and bone formation by osteoblasts to maintain skeletal integrity. Disruptions to this equilibrium can lead to pathological conditions such as osteoporosis. Osteoclasts originate from macrophages, and their differentiation is regulated by several signaling pathways, notably the notch signaling pathway. Gamma-secretase, an enzyme crucial for initiating notch signaling, can be inhibited by DAPT. However, DAPT also targets non-Notch substrates, complicating the identification of genes specifically regulated by Notch. Inland Pacific Northwest prairie is an endangered ecosystem that has been reduced by conversion to agriculture. There is growing interest in the role of soil microorganisms in prairie ecosystems, which may be crucial for restoration of the plant community through interspecies interactions within the rhizosphere. One method for improving soil microorganism community composition is compost tea: enriched liquid compost extract. Compost tea is used in regenerative agriculture to improve crop yields and protect against pathogens. Little is known about the mechanisms that generate epileptic spasms following perinatal brain injury. Recent studies have implicated reduced levels of Insulin-like Growth Factor 1 (IGF-1) in these patients' brains. Other studies have reported low levels of the inhibitory neurotransmitter, GABA. In the TTX brain injury model of epileptic spasms, we undertook experiments to evaluate the impact of IGF-1 deficiencies on neocortical interneurons and their role in spasms. Quantitative immunohistochemical analyses revealed that neocortical interneurons that express glutamic acid decarboxylase, parvalbumin, or synaptotagmin 2 co-express IGF-1. In epileptic rats, expression of these three interneuron markers were reduced in the neocortex. …

Multimodal neural interfaces open new opportunities in brain research by enabling more sophisticated and systematic neural circuit dissection. Integrating complementary features across distinct functional domains, these multifunctional neural probes have greatly advanced the interrogation of complex neural circuitry. However, introducing multiple functionalities into a compact form factor for freely behaving animals presents substantial design hurdles that complicate the device or require more than one device. Moreover, fixed functionality poses challenges in meeting the dynamic needs of chronic neuroscience inquiry, such as replacing consumable parts like batteries or drugs. The mutation in Gsα-coding GNAS exons, popular as gsp oncogene, is the most frequent mutation across all heterotrimeric G proteins involved in oncogenesis. GNAS R201, the most frequently mutated, followed by Q227, are found predominantly across various neoplasms and cancers such as IPMN, pituitary, thyroid, appendiceal, colorectal, etc. This review emphasizes the pivotal significance of the gsp oncogene and its ramifications underpinning the sustained addiction to GNAS mutation. Parents in Australia today have to make decisions about what counts as appropriate digital media use for their young children in a cultural context characterised by conflicting advice, impractical screen use recommendations and risk-focussed media messaging. These conditions make it challenging for parents to purposefully approach and plan not only their children's current digital engagements but also their futures as digital citizens. How parents negotiate these tensions in managing their young children's digital presents, and how they approach planning for their digital futures, remains underexplored. With the increasing complexity of modern power systems, effective control of DC–DC converters has become crucial to ensure stability and efficiency. This paper focuses on optimizing the parameters of a known fractional-order proportional–integral–derivative (FOPID) controller for the control of a DC–DC buck–boost converter. The control of a DC–DC buck–boost converter is achieved using aFOPID approach. The gains of this technique have been enhanced utilizing the snake optimization (SO) algorithm. This converter exhibits unfavourable behaviour due to its non-minimum structure, necessitating a well-regulated controller to guarantee stability. Almost everyone is affected by loneliness at some stage in their lives, and the long-term effects are dire, leading to depression, addiction, and suicide. To date, interventions have focused on external treatments with limited success. In this study, it is argued that underlying antecedents of social loneliness maintain negative behaviour, whereby it becomes chronic and problematic. Four intrinsic antecedents of social loneliness are examined (Secure and Insecure Attachment, Emotional Regulation Problems – Expressive Suppression, Unclear Self-Concept, and Neuroticism) along with negative (Ignoring and Withdrawing) behavioural coping styles. Ninety-six, mainly female (78%), psychology undergraduates were administered a battery of six questionnaires. …

BACKGROUND: The antigen Na-GST-1, expressed by the hookworm Necator americanus, plays crucial biochemical roles in parasite survival. This study explores the development of mRNA vaccine candidates based on Na-GST-1, building on the success of recombinant Na-GST-1 (rNa-GST-1) protein, currently assessed as a subunit vaccine candidate, which has shown promise in preclinical and clinical studies.
METHODOLOGY/FINDINGS: By leveraging the flexible design of RNA vaccines and protein intracellular trafficking signal sequences, we developed three variants of Na-GST-1 as native (cytosolic), secretory, and plasma membrane-anchored (PM) antigens. Objectives: This meta-analysis investigated whether multi-ingredient protein supplements (MIPS) combined with exercise improve body composition and muscle fitness in women. It also examined how participant characteristics, training protocols, and supplementation strategies might influence these outcomes. Methods: A systematic search of five electronic databases was conducted through February 2025 to identify randomized controlled trials evaluating the effects of MIPS combined with exercise training on body composition and muscle-related outcomes in women. Management of cultural landscapes has struggled to understand and adapt to Indigenous ways of managing heritage through a reciprocal duty of care. We argue that barriers to Indigenous cultural landscapes are threefold. First, conceptual barriers exist due to the prevalence of inappropriate heritage concepts and frameworks from the Global North. Second are relational barriers in the form of asymmetrical relationships of power between non-Indigenous heritage ‘experts’ and Indigenous individuals and communities. Third, political barriers present in neo-colonial politics are at odds with the goals of Indigenous movements. This paper integrates insights from affect theory into Foucault’s governmentality framework to analyse school markets. Using a Deleuzian approach to affect, we theorise school markets as an affective milieu that composes the field of competition. Drawing on interviews with staff at two rival high schools, we highlight how an affective milieu that included threat, anxiety, insecurity, resentment and confidence shaped market dynamics. Our analysis shows that affective milieus reflect and reinforce power asymmetries in local school markets and are a condition for generating competitive practices to enhance schools’ capacities to affect and be affected. Purpose: This paper aims to examine why many producers of origin-bound products struggle to collectively drive increases in market value, despite shared interests. The authors apply a collaborative market driving (CMD) lens to the early stages of collective efforts, to identify the key motivators, visioning processes and explore why CMD can fail to initiate. This chapter presents an overview of physical and acoustic behavioral responses of marine mammals to underwater sounds. A literature review was undertaken, and data on received levels at the animal when certain types of responses were observed were compiled in an online supplementary spreadsheet. Based on this, an overview of responses was written, organized first by species and then by sound type. In-air and underwater sound sources were considered. The most studied sound types were mid-frequency sonar and acoustic deterrent devices and then impact pile driving and vessels. This feature article highlights the importance of such research in academia with examples drawn from the tourism literature. It also critically discusses issues related to publishing conceptual research in premier journals. Objectives: The aim of this project was to evaluate the feasibility and acceptability of virtual reality (VR) as a leisure activity for people living in residential aged care. Virtual reality experiences may offer alternatives for residents with limited mobility, or during times when older people are unable to leave residential care. Methods: The intervention used VR videos delivered via a head-mounted device. Pilgrimage Studies is a growing field across the arts and humanities, yet pilgrimage writing is not established as a distinct genre. In this article I share my exploration of pilgrimage as a creative writing process and potential genre. Background Modifiable risk factors have been linked to 45% of dementia cases. Mobile health (mHealth) interventions targeting lifestyle-related risk factors with remote coaching have the potential to reach underserved high-risk populations globally. To date, little is known about the implementation of such interventions in China. Methods Fifty semi-structured interviews were conducted with 14 participants and 11 health coaches involved in the PRODEMOS trial. This trial investigated whether a coach-supported mHealth application intervention can reduce dementia risk in people aged 55–75 years with multiple risk factors. Cockburn Sound, a temperate embayment on the lower west coast of Western Australia, is immensely important for the State and adjacent capital city of Perth. Aim: We decipher global patterns in seagrass growth and structure to advance seagrass science, facilitate multidisciplinary research, and promote effective management strategies and policy development for seagrass habitats. Location: Global. Time period: 1975 to 2024. Major taxa studied: Seagrasses—marine angiosperms. Methods: A global data set on seagrass total biomass, total net primary productivity (NPP), shoot density, and percent cover was compiled across 11 genera.
