This study investigates the potential of bacterial metalloprotease genes as therapeutic agents for Alzheimer's Disease (AD). The research focuses on Pseudomonas 20ei1 (P. 20ei1), a bacterial species isolated from lagoon water, which contains pitrilysin genes that share similarities with human insulysin. Six pqqL-related genes were identified in P. 20ei1 for this study. The research methodology involved bioinformatic studies, bacterial cell culture in various media, RNA extraction, purification, and cDNA synthesis. Quantitative PCR analysis was performed to analyze gene expression levels in different cDNA samples. Enzyme-mediated lipid oxidation is an important regulatory event in cell signaling, with oxidized lipids being potent signaling molecules that can illicit dramatic changes in cell behavior. For example, peroxidation of an arachidonoyl poly-unsaturated fatty acid by the human enzyme 15-lipoxygenase-2 (15-LOX-2) has been associated with formation of atherosclerotic plaques. Previous work on synthetically oxidized membranes has shown that oxidized lipid tails will change their conformation to facilitate interactions between the peroxide group and the lipid headgroups. However, this phenomenon has not been directly observed for a lipid membrane that has undergone enzyme-catalyzed oxidation. In this study, we report on the …

The purpose of this study was to examine how secondary humanities teachers modify Open Educational Resources (OERs) to align with the Universal Design for Learning (UDL) framework. Originally developed to democratize access to educational content, OERs have become widely used across K–12 settings in the U.S., particularly following initiatives such as the Common Core standards and the U.S. Department of Education’s #GoOpen Project. Kindergarten teachers and support professionals in the social-emotional community within a local school district experienced changes in student behaviors and social skills since returning to in-person schooling following the pandemic. After COVID-19, teachers found themselves engaging students who presented a lack of social skill development, contributing to the behaviors within the classroom microsystem. This study used Urie Bronfenbrenner’s ecological systems theory to support research efforts to examine how learning environments and interactions contributed to students’ preparation for Kindergarten. Women-owned small and medium-sized enterprises (SMEs) often face significant challenges in securing funding due to a lack of networks and resources, which limits their access to both traditional and nontraditional financing options. Grounded in the social feminist theory, the purpose of this qualitative pragmatic inquiry was to explore effective strategies women-owned SMEs in the northeastern United States can use to secure financing for startup and daily operations. Participants were seven women who successfully operated small service-based businesses in the northeastern United States, implementing strategies that sustained their businesses for at least two years. This paper examines the impact of utilizing technology for mathematics education. It reviews multiple categories of technological tools available for classroom use, including hardware, software, and online resources, and discusses their potential benefits and drawbacks. Positive aspects of technology integration include personalized learning experiences, increased engagement and collaboration, and enhanced visualization and conceptual understanding. Staphylococcus aureus is a highly significant pathogen with several well studied and defined virulence factors. However, the metabolic pathways that are required to facilitate infection are not well described. Previous data have documented that S. aureus requires glucose catabolism during initial stages of infection. Therefore, certain nutrients whose biosynthetic pathway is under carbon catabolite repression and CcpA, including arginine, must be acquired from the host. However, even though S. aureus encodes pathways to synthesize arginine, biosynthesis of arginine is repressed even in the absence of glucose. Why is S. aureus a functional arginine auxotroph? Background/aim: Mucinous breast carcinoma (MC) is thought to be a favorable-differentiated form of invasive breast cancer and is rarely preferred for neoadjuvant chemotherapy (NAC). The study aimed to define the response of MCs to NAC.Materials and methods: The study reviewed the demographic, clinicopathologic characteristics, management, and follow-up data of 70 patients diagnosed with MC between May 2010 to December 2020 by examining patients’ historical files and oncology records.Results: The median age, tumor size, and follow-up period of patients were respectively 52.9 (range 32 to 87) years, 25.8 (range 8 to 88) mm, and 61.5 (range 18 to 143) months. Background/aim: Differentiating multisystem inflammatory syndrome in children (MIS-C) from adenovirus infection (AI) can be challenging due to similar clinical and laboratory findings. This study aimed to identify distinguishing characteristics and develop a scoring system to facilitate accurate diagnosis.Materials and methods: A comprehensive review of medical records was undertaken for 108 MIS-C cases and 259 patients with confirmed AI. A comparison of laboratory data and clinical findings was conducted across patient groups through appropriate statistical tests.Results: MIS-C patients were significantly older than AI patients (p<0.001). Diarrhea, rash, abdominal pain, vomiting, non-exudative conjunctivitis, lymphadenopathy tachycardia, bradycardia, hypotension, hypoxia seizures, agitation, headache, and altered consciousness symptoms were more frequently associated with MIS-C (p<0.001), while cough and runny nose were significantly more common in AI (p<0.001). Lymphopenia and thrombocytopenia were more prevalent in MIS-C patients (p<0.001). AI and MIS-C were scored by giving 1 point each to the parameters that created the difference. For AI, ≤60 months, presence of cough, runny nose and absence of diarrhea, rash, abdominal pain, vomiting, non-exudative conjunctivitis, lymphadenopathy, tachycardia, bradycardia, hypotension, hypoxia seizures, agitation, headache, and altered consciousness, lymphopenia, thrombocytopenia and CRP <124.5 mg/L were determined as each parameter plus one point. A total score above 14 could predict AI with a high degree of accuracy sensitivity values stood at around 97.5% and specificity at 92.6%.Conclusion: The proposed inpatient scoring system, when used in conjunction with polymerase chain reaction testing, may improve the early differentiation of AI and MIS-C. This approach could help reduce unnecessary testing and optimize resource allocation. Further research with larger samples should investigate this novel scoring system to establish its reliability and generalizability.

Background/Aim: This study aims to compare the success rates of rigid registration (RR) and elastic registration (ER)systems in diagnosing all cancers and clinically significant prostate cancer (csPC) in software-based targeted prostate biopsies (TPBs) by performing matching analysis.Materials and Methods: The data of 2,061 patients from six centers where software-based TPB was performed were used. Background/aim: The transition from pediatric to adult-oriented care for individuals with juvenile-onset systemic lupus erythematosus (SLE) poses significant challenges. This study aims to assess the outcomes of transitioning patients with juvenile-onset SLE from pediatric to adult-oriented care.Materials and Methods: Patients with juvenile-onset SLE were included. They were transferred in face-to-face meetings where at least one pediatric rheumatologist and one adult rheumatologist were present (the transition time: October-December 2020).Results: The median (25th-75th percentiles) age at diagnosis and the time of the first examination at an adult-oriented rheumatology department of 65 SLE patients were 14.3 (10.9-15.1) years and 19.2 (18.5-20.4) years, respectively …

Bacterial membranes are complex and dynamic, arising from an array of evolutionary pressures. One enzyme that alters membrane compositions through covalent lipid modification is MprF. We recently identified that Streptococcus agalactiae MprF synthesizes lysyl-phosphatidylglycerol (Lys-PG) from anionic PG, and a novel cationic lipid, lysyl-glucosyldiacylglycerol (Lys-Glc-DAG), from neutral glycolipid Glc-DAG. This unexpected result prompted us to investigate whether Lys-Glc-DAG occurs in other MprF-containing bacteria, and whether other novel MprF products exist. This project seeks to understand the ways in which Queer Theory can be applied to secondary language arts education to enhance students' understanding of identity and gender in literature. For the purposes of this project, Shakespeare’s Twelfth Night is being used as a case study text. This project includes a thorough analysis of Twelfth Night through a queer lens, a literature review of Queer Theory and educational pedagogy, a block plan and related instructional and assessment materials, a rationale for the block plan, and project limitations and implications for future research. BACKGROUND: Significant gap remains in the implementation of guideline-directed medical therapy (GDMT) in patients with heart failure after a hospitalization. We aimed to evaluate the use and titration of GDMT at discharge and over a 12-month period after hospital discharge and to identify factors associated with GDMT use and titration.
METHODS AND RESULTS: The CONNECT-HF (Care Optimization Through Patient and Hospital Engagement Clinical Trial for Heart Failure) trial evaluated the effect of a hospital and postdischarge quality improvement intervention in participants with heart failure with reduced ejection fraction.
