Metabolic imbalance leading to inflammatory hypoxia and stabilization of hypoxia-inducible transcription factors (HIFs) is a hallmark of inflammatory bowel diseases. We hypothesize that HIF could be stabilized in CD4+ T cells during intestinal inflammation and alter the functional responses of T cells via regulation of microRNAs. Our assays reveal markedly increased T cell-intrinsic hypoxia and stabilization of HIF protein during experimental colitis. microRNA screen in primary CD4+ T cells points us towards miR-29a and our subsequent studies identify a selective role for HIF-2α in CD4-cell-intrinsic induction of miR-29a during hypoxia. Mice with T cell-intrinsic HIF-2α deletion display elevated T-bet (target …

Journal of Food and Drug Analysis
We employed a straightforward microwave-assisted extraction technique to investigate the presence of carbon dots (CDs) in Chinese herbal extracts derived from fructus gardeniae and gardenia charcoal, designated as CDs-1 and CDs-2, respectively. The found CDs exhibited unique photoluminescence with quantum yields of 0.95% for CDs-1 and 1.81% for CDs-2, indicating significant potential for bioimaging applications. Both CD types maintain approximately 80% of their fluorescence intensity after 120 minutes of continuous 365 nm UV exposure, underscoring their stability and suitability for prolonged biological studies. Moreover, antioxidant activity tests showed that CDs-2 have a higher scavenging capacity, with an SC-50 value of …

Optimization, Physicochemical Stability And In Vivo Study Of Alginate-Chitosan Composites As Nanocarriers For Low Molecular Weight Angiotensin I-Converting Enzyme (Ace)-Inhibitory Peptide, Shehu Muhammad Auwal, Siti Balqis Muhammad Ghanisma, Nazamid Saari
Journal of Food and Drug Analysis
Chitosan and alginate, are non-toxic and biodegradable polymers used to enhance the stability of biotherapeutics by loading them into nanocarriers. In this study, the stone fish-derived low molecular weight peptide (Ala-Leu-Gly-Pro-Gln-Phe-Tyr), exhibited an in vitro ACE-inhibitory activity of 94.43 ± 2.05% and an IC50 of 0.012 ± 0.001 mM. The peptide was encapsulated via ionic gelation with alginate followed by polyelectrolyte complexation with chitosan. The resulting ACE-inhibitory peptide-loaded alginate-chitosan nanoparticles (ACE-I-ALG-CS NPs) were optimized to achieve small particle size (212.60 nm) and high encapsulation efficiency (EE, 74.48%). This was based on an optimum chitosan concentration (0.420%w/v), homogenization speed (6000 …

Production And Characterization Of Encapsulated Thymoquinone Loaded Bacterial Cellulose/Chitosan Composite Nano Film And Investigation Of Its In Vitro Antibacterial Activity, Aytul Bayraktar, Tunc Tuba
Journal of Food and Drug Analysis
In this study, nanostructured thin films were produced from thymoquinone (TQ)-loaded composites encapsulated with different ratios of bacterial nanocellulose (BNC) to chitosan (CS). The study aimed to investigate their characterization, antibacterial effects, and potential as drug film coating materials. Chemical features and morphological characteristics were determined, and release tests and antibacterial assays were conducted on the thin film layers. As a result, BNC/CS-thymoquinone composite films obtained at different ratios were found to exhibit the highest antibacterial effect, particularly films with a 70% BNC/CS to 30% thymoquinone ratio, against Salmonella typhimurium bacteria.

Alzheimer's disease (AD) is a chronic and progressive neurodegenerative disorder that affects millions of individuals worldwide. Researchers have conducted numerous studies to find accurate biomarkers for early AD diagnosis and develop more effective treatments. The main pathological hallmarks of AD are amyloid beta and Tau proteins. Other biomarkers, such as DNA, RNA, and proteins, can also be helpful in early AD diagnosis. To diagnose and treat AD promptly, it is essential to accurately measure the concentration of biomarkers in the cerebrospinal fluid or blood. However, due to the low concentrations of these biomarkers in the body, highly sensitive analytical techniques …

Understanding the causes of the ~90 ppmv atmospheric CO2 swings between glacial and interglacial climates is an important open challenge in paleoclimate research. Although the regularity of the glacial-interglacial cycles hints at a single driving mechanism, Earth System models require many independent physical and biological processes to explain the full observed CO2 signal. Here we show that biologically sequestered carbon in the ocean can explain an atmospheric CO2 change of 75 ± 40 ppmv, based on a mass balance calculation using published carbon isotopic measurements. An analysis of the carbon isotopic signatures of different water masses indicates similar regenerated carbon …

Remote sensing tools, along with Global Navigation Satellite System cattle collars and digital soil maps, may help elucidate spatiotemporal relationships among soils, terrain, forages, and animals. However, standard computational procedures preclude systems-level evaluations across this continuum due to data inoperability and processing limitations. Deep learning, a subset of neural network, may elucidate efficiency of livestock production and linkages within the livestock-grazing environment. Consequently, we applied deep learning to environmental remote sensing data to (1) develop predictive models for yield and forage nutrition based on vegetation indices and (2) at a pixel-level (per 55 m2), identify how grazing is …
