Synaptotagmin-9 In Mouse Retina, Chris S. Mesnard, Cassandra L. Hays, Lou E. Townsend, Cody L. Barta, Channabasavaiah B. Gurumurthy, Wallace B. Whoreson

Neuroscience Faculty Publications

Synaptotagmin-9 (Syt9) is a Ca2+ sensor mediating fast synaptic release expressed in various parts of the brain. The presence and role of Syt9 in retina is unknown. We found evidence for Syt9 expression throughout the retina and created mice to conditionally eliminate Syt9 in a cre-dependent manner. We crossed Syt9fl/fl mice with Rho-iCre, HRGP-Cre, and CMV-Cre mice to generate mice in which Syt9 was eliminated from rods (rodSyt9CKO), cones (coneSyt9CKO), or whole animals (CMVSyt9). CMVSyt9 mice showed an increase in scotopic electroretinogram (ERG) b-waves evoked by bright flashes with no change in a-waves. Cone-driven photopic ERG b-waves were not significantly …

Emerging evidence implicates common genetic variation - aggregated into polygenic scores (PGS) - in the onset and phenotypic presentation of rare diseases. Here, we comprehensively map individual polygenic liability for 1102 open-source PGS in a cohort of 3059 probands enrolled in the Genomic Answers for Kids (GA4K) rare disease study, revealing widespread associations between rare disease phenotypes and PGSs for common complex diseases and traits, blood protein levels, and brain and other organ morphological measurements. Using this resource, we demonstrate increased polygenic liability in probands with an inherited candidate disease variant (VUS) compared to unaffected carrier parents. Further, we show …

Nanofiber-Hydrogel Composite Scaffold Fabrication Methods For Peripheral Nerve Regeneration, Jacob Lee Carter Sep 2024

Nanofiber-Hydrogel Composite Scaffold Fabrication Methods For Peripheral Nerve Regeneration, Jacob Lee Carter

Theses and Dissertations

Nerve graft conduits (NGCs) are a rapidly advancing field for treating peripheral nerve injuries that aim to guide the growth of regenerating axons across damaged gaps. A more effective NGC can be manufactured by combining the use of nanofibers to act as guidewires within a 3-D hydrogel that imitates the extracellular matrix of nerve tissue. Several methods have been developed to embed aligned nanofibers in an ordered architecture within a hydrogel matrix. The first method involves layer-by-layer additive manufacturing to create NGCs with rows of polycaprolactone (PCL) nanofibers surrounded by a gelatin methacrylate (GelMe) hydrogel in a 3-D structure. A …

Novice teachers benefit from individualized and cohort mentoring to help them successfully navigate student teaching and the first years of independent teaching. However, schools are experiencing a rise in attrition rates post-pandemic, especially in math and science. Mentoring, goal setting, and community-building are deemed important for retaining novices and providing them with individualized support as they navigate instructional challenges and professional responsibilities. This self-study investigates the experiences of a mentor while leading a new Professional Development & Mentoring Program for math and science novices. Educative mentoring and Hegarty’s Reflective framework helped guide the reflections of the mentor as mentoring strategies …

Tensions Between Catholic Identity And Academic Achievement: Roles Of Catholic School Leaders In Southeastern Nigeria, Augustina Ngozi Mbata Sep 2024

Tensions Between Catholic Identity And Academic Achievement: Roles Of Catholic School Leaders In Southeastern Nigeria, Augustina Ngozi Mbata

Dissertations

Despite the series of documents by the sacred ecumenical council and replete research enumerating the two-dimensional focus of Catholic schools: faith and academics and its collaborative nature, Nigerian (southeast) Catholic primary and secondary schools are continually experiencing activities of disharmony between Catholic identity and students' academic achievements. To better understand the nature of disharmony and its causes from the perspectives of leaders, teachers, and parents, this study examines the major challenges that face the representation and teaching of Catholic identity and the students’ academic achievement in the Catholic schools of southeastern Nigeria. While the purpose is critical to the student’s …

A Preliminary Fuzzy Inference System For Predicting Atmospheric Ozone In An Intermountain Basin, John R. Lawson, Seth N. Lyman Sep 2024

A Preliminary Fuzzy Inference System For Predicting Atmospheric Ozone In An Intermountain Basin, John R. Lawson, Seth N. Lyman

Mathematics and Statistics Faculty Publications

High concentrations of ozone in the Uinta Basin, Utah, can occur after sufficient snowfall and a strong atmospheric anticyclone creates a persistent cold pool that traps emissions from oil and gas operations, where sustained photolysis of the precursors builds ozone to unhealthy concentrations. The basin's winter-ozone system is well understood by domain experts and supported by archives of atmospheric observations. Rules of the system can be formulated in natural language ("sufficient snowfall and high pressure leads to high ozone"), lending itself to analysis with a fuzzy-logic inference system. This method encodes human expertise as machine intelligence in a more prescribed …

The Individual’S Perception Of Ai And Its Effects On The Organization’S Ai Initiatives, Catherine Petersen

JWU Dissertations & Theses

Investment in Artificial Intelligence (AI) does not result in returns for business despite improving technology (Ransbotham et al., 2020; Storm & Borgman, 2020). This study investigated one potential reason for the failure of AI when implemented within an organization with the intent to drive success with this technology. AI is unique amongst technologies due to its ability to inspire emotions from potential users and these emotions can affect the individual’s willingness to adopt the technology. The research questions were:

1. Does an individual’s perception of AI have a moderating effect on their behavioral intention to use a product that includes …

Aging Drives Cd8 T Cells To Functional Extremes: From Non-Specific Cytotoxicity To Tissue-Mediated Exhaustion, Irina Shchukina Sep 2024

Aging Drives Cd8 T Cells To Functional Extremes: From Non-Specific Cytotoxicity To Tissue-Mediated Exhaustion, Irina Shchukina

Arts & Sciences Graduate Student Theses and Dissertations

CD8 T cells are a crucial component of the immune system, primarily responsible for executing the effector response against viral infections and cancer. Aging significantly impacts most immune cell types, with particularly dramatic changes observed in CD8 T cells. One major age-related difference is the marked decline in thymus activity, which leads to a substantial reduction in naive CD8 T cells and an increased abundance of memory CD8 T cells. Furthermore, recent work from our lab demonstrated that while young memory CD8 T cells are relatively homogeneous, memory CD8 T cells diverge into two subpopulations: PD1- memory CD8 T cells …

Influences Of Quality Of Maternal Care And Environmental Enrichment On Associative Memory Function In Rats With Early Life Lead Exposure, Jay S. Schneider, Courtney Williams, Shamaila Zafar, Jaehyun Joo, Blanca E. Himes

Department of Pathology, Anatomy, and Cell Biology Faculty Papers

INTRODUCTION: Children in low socioeconomic status (SES) communities are at higher risk of exposure to lead (Pb) and potentially more severe adverse outcomes from Pb exposures. While the factors encompassing SES are complex, low SES households often have less enriching home environments and parent-child interactions. This study investigated the extent to which environmental/behavioral factors (quality of maternal care and richness of the postnatal environment) may modify adverse effects from Pb exposure.

METHODS: Long-Evans female rats were randomly assigned to Control (no Pb), Early Postnatal (EPN: birth through weaning), or Perinatal (PERI: 14 days pre-mating through weaning) Pb exposure groups. From …

Mako: NSU Undergraduate Student Journal

Biosynthetic gene clusters (BGCs) are groups of microbial genes that play a critical role in producing secondary metabolites with varied functions. Secondary metabolites include compounds such as siderophores, antibiotics, antifungal compounds, toxins, and pigments. The BGCs are modular units that usually contain a transcription factor, core biosynthetic gene, other genes involved in modifications and addition of side chains as well as genes needed for extracellular export. In specific, the metabolites produced by BGCs have diverse biological activities and usually display antimicrobial properties. The genome of Streptomyces murinus strain SPC1 revealed a complete BGC involved in the biosynthesis of the antifungal …

Exploring Genetic Elements Related To The Co-Occurrence Of Crohn’S Disease And Rheumatoid Arthritis, Vamsi Korisapati, Karam Asad, Andrew T. Ozga

Mako: NSU Undergraduate Student Journal

Crohn’s disease (CD) is a rare inflammatory bowel disease that negatively impacts the lining of the digestive tract, while Rheumatoid Arthritis (RA) is an autoimmune disorder wherein the body attacks healthy cells, also causing inflammation. The primary cause of inflammation in both diseases is an overactive immune response, but patients who are diagnosed with RA are typically prescribed nonsteroidal anti-inflammatory drugs, which is not a suggested method for treating CD. Here we do a literature survey through PubMed to uncover genetic variants common to both diseases and highlighted four shared genes: HLA-DRB1, NOD2, CARD9, and CXCL10. We then examined these …

Cytotoxic Potential Of Fungi In The Genus Ganoderma, Abril Chao, Omobolanle A. Ayangade, Braham Dhillon, Navi Gill

Mako: NSU Undergraduate Student Journal

Ganoderma lucidum is a mushroom forming fungus that has been used in traditional Chinese medicine for over 2,000 years. G. lucidum, also known as lingzhi or reishi, is consumed in several Asian cultures to promote health and longevity. Bioactive molecules with anticancer properties have been isolated and characterized primarily from the species Ganoderma lucidum, and the cytotoxic potential of other species in the genus Ganoderma remains relatively unclear. Bioactive molecules isolated from mushroom fruiting bodies are demonstrated to have cytotoxicity against different cancer cell lines. Triterpenes and polysaccharides are the two major groups of compounds that exhibit antitumor and anti-inflammatory …

Drivers Of Coral Reef Benthic Changes And Implications On Ecosystem Functioning And Services, Miriam Reverter Sep 2024

Drivers Of Coral Reef Benthic Changes And Implications On Ecosystem Functioning And Services, Miriam Reverter

School of Biological and Marine Sciences

Increasing anthropogenic stressors will likely result in many coral reefs undergoing compositional and functional benthic changes, including towards non-hard coral dominated communities. The decline of reef building species and the increase of alternative (i.e., non reef-building) organisms profoundly affect the functioning of coral reefs and the services provided, but this remains poorly understood, especially for non-algal alternative communities. Here, we propose a standardised framework to study coral reef benthic changes and review the knowledge on coral reef shifts and their implications on coral reef functioning and service provisioning using this framework, which contains the five following elements: 1) contextual information, …

Body, Ritual, And Place: Advancing Their Chronotopic Interrelationship In Design Praxis And Pedagogy Through An Exploration Of Juxtaposed Surfaces, Threshold Sites Of Dialogic Encounter, And Collage, Robert Brown, Zoe Latham Sep 2024

Body, Ritual, And Place: Advancing Their Chronotopic Interrelationship In Design Praxis And Pedagogy Through An Exploration Of Juxtaposed Surfaces, Threshold Sites Of Dialogic Encounter, And Collage, Robert Brown, Zoe Latham

School of Art, Design and Architecture

Echoing philosopher Mikhail Bakhtin’s chronotope, an ‘intrinsic connectedness of temporal and spatial relationships,’ our work explores the interrelation of the body, ritual, and place as a chrontopic site of design and pedagogy. This exploration reaffirms architecture’s grounding in people, their inhabitation of place, and spatial form, an interrelatedness too often underexamined withinarchitectural education. First positioning ritual thematically, our discussion addresses three interrelated issues: the role of juxtaposed ritual surfaces in activating the body, ritual and place; the potential of ritual surfaces as threshold sites of dialogic encounter; and collage as a medium to investigate and communicate the interrelationship of body, …

Lamellipodia-Mediated Osteoblast Haptotaxis Guided By Fibronectin Ligand Concentrations On A Multiplex Chip, Xiaotian Feng, Seoyoung Jeong, Melissa L. Carr, Radhika P. Atit, Samuel E. Senyo

Faculty Scholarship

Skull morphogenesis is a complex, dynamic process involving two different germ layers and progressing to the coordinated, directional growth of individual bones. The mechanisms underlying directional growth toward the apex are not completely understood. Here, a microfluidic chip-based approach is utilized to test whether calvarial osteoblast progenitors undergo haptotaxis on a gradient of Fibronectin1 (FN1) via lamellipodia. Mimicking the embryonic cranial mesenchyme's FN1 pattern, FN1 gradients is established in the chip using computer modeling and fluorescent labeling. Primary mouse calvarial osteoblast progenitors are plated in the chip along an array of segmented gradients of adsorbed FN1. The study performs single-cell …

As we face multiple environmental crises (e.g. climate change, nutrient pollution, freshwater scarcity), there is often a disconnect between an entity’s activities and the pollution resulting from that activity. Footprint tools help address this disconnect. They are effective in educating people, institutions and communities on how their resource use results in environmental pollution and what we can do to moderate that pollution. These tools connect an entity’s activities with the associated pollution. Footprint tools are also constantly evolving. As we better understand how to best estimate emissions—especially from sources further removed like those from the supply chain—the guidance and methods …
