Tree growth and longevity trade-offs fundamentally shape the terrestrial carbon balance. Yet, we lack a unified understanding of how such trade-offs vary across the world's forests. By mapping life history traits for a wide range of species across the Americas, we reveal considerable variation in life expectancies from 10 centimeters in diameter (ranging from 1.3 to 3195 years) and show that the pace of life for trees can be accurately classified into four demographic functional types. We found emergent patterns in the strength of trade-offs between growth and longevity across a temperature gradient. Furthermore, we show that the diversity of …

Optimizing doses for multiple indications is challenging. The pooled approach of finding a single optimal biological dose (OBD) for all indications ignores that dose-response or dose-toxicity curves may differ between indications, resulting in varying OBDs. Conversely, indication-specific dose optimization often requires a large sample size. To address this challenge, we propose a Randomized two-stage basket trial design that Optimizes doses in Multiple Indications (ROMI). The 2016 and 2021 World Health Organization 2021 Classification of central nervous system tumors have resulted in a major improvement in the classification of isocitrate dehydrogenase (IDH)-mutant gliomas. With more effective treatments many patients experience prolonged survival. However, treatment guidelines are often still based on information from historical series comprising both patients with IDH wild-type and IDH-mutant tumors. They provide recommendations for radiotherapy and chemotherapy for so-called high-risk patients, usually based on residual tumor after surgery and age over 40. Meningiomas are the most common primary intracranial tumors in adults and are increasing in incidence due to the aging population and increased access to neuroimaging. While most exhibit nonmalignant behavior, a subset of meningiomas are biologically aggressive and are associated with treatment resistance, resulting in significant neurologic morbidity and even mortality. In recent years, meaningful advances in our understanding of the biology of these tumors have led to the incorporation of molecular biomarkers into their grading and prognostication. Despite the numerous sequencing methods available, the diversity in RNA size and chemical modification makes it difficult to capture all RNAs in a cell. We developed a method that combines quasi-random priming with template switching to construct sequencing libraries from RNA molecules of any length and with any type of 3' modifications, allowing for the sequencing of virtually all RNA species. Our ligation-independent detection of all types of RNA (LIDAR) is a simple, effective tool to identify and quantify all classes of coding and non-coding RNAs. Background: Brain metastases (BM) are a common, severe complication in patients with non-small cell lung cancer (NSCLC) and are difficult to treat due to their complex tumor biology and the intricate microenvironment of the brain.

Objectives: This review examines the current role of immune checkpoint inhibitors (ICIs) in treating NSCLC with BM, focusing on the latest clinical trials, emerging strategies, current guidelines, and future directions. We highlight the efficacy of ICIs as monotherapy and in combination with other treatments such as radiotherapy, stereotactic radiosurgery, chemotherapy, and anti-VEGF agents.

Leptomeningeal metastases (LM) are increasingly becoming recognized as a treatable, yet generally incurable, complication of advanced cancer. As modern cancer therapeutics have prolonged the lives of patients with metastatic cancer, specifically in patients with parenchymal brain metastases, treatment options, and clinical research protocols for patients with LM from solid tumors have similarly evolved to improve survival within specific populations. It remains unknown whether, and how intestinal stem cells (ISC) adapt to inflammatory exposure, and if the adaptation leaves scars will affect their subsequent regeneration. We investigated the consequences of inflammation on Lgr5+ISCs in well-defined clinically relevant models of gastrointestinal acute graft-versus-host disease (GI GVHD). Utilizing single cell transcriptomics, organoid, metabolic, epigenomic and in vivo models we found that Lgr5+ISCs undergo metabolic changes that lead to accumulation of succinate, which reprograms its epigenome. These changes reduced the ability of ISCs to differentiate and regenerate ex vivo in serial organoid cultures and also in vivo following serial transplantation. Furthermore, ISCs demonstrated …

This study compiled data on the distribution of sponge species on the coasts of Türkiye. The checklist consists of 183 speciesbelonging to 52 families, 16 orders, and 3 classes. The orders Haplosclerida (32 species), Poecilosclerida (28 species), Tetractinellida (26species), Dictyoceratida (21 species), and Suberitida (18 species) have the highest numbers of species found in the region. The Aegeancoast has the highest number of sponge species (155 species), followed by the Sea of Marmara (84 species), the Levantine coast (50species), and the Black Sea coast (24 species). The compilation of papers on the phyla Cnidaria and Ctenophora inhabiting the coasts of Türkiye (Black Sea, Sea of Marmara,Aegean Sea, and Levantine Sea) together with the new records of cnidarian species presented in this study yielded 296 cnidarian and8 ctenophore species. Among the cnidarian groups, Hydrozoa were represented by 160 species, Scyphozoa by 16 species, Anthozoa by87 species, and Myxozoa by 31 species. The compilation of scientific papers dealing with marine annelids (including Sipuncula) along the coasts of Türkiye (Black Sea, Sea of Marmara, Aegean Sea and Levantine Sea) together with the new records of polychaete species (22 species) presented in this study resulted in 827 species belonging to 63 families. Polychaeta were represented by 782 species, Sipuncula by 21 species, Oligochaeta by 18 species, Hirudinea by 5 species and Echiura by 1 species. Syllidae (127 species), Spionidae (57 species) and Serpulidae (56 species) were the most species-rich families and their numbers varied among the seas. The Turkish marine fish diversity was revised and updated in this study based on data published in the last decade. There arecurrently 552 species in the inventory, reflecting an increase of 7.8% since the previous checklist and belonging to the classes of Teleostei(479 spp.), Elasmobranchii (68 spp.), Chondrostei (3 spp.), Petromyzonti (1 sp.), and Holocephali (1 sp.). The current status of bryozoan species distributed along the coast of Türkiye were revised and an updated checklist including 185 species (24 cyclostomatids, 20 ctenostomatids and 141 cheilostomatids) was presented. Cheilostomes largely prevail (141 species, 87 genera and 50 families) followed by cyclostomes (24 species, 15 genera and 9 families) and ctenostomes (20 species, 9 genera and 8 families) in bryozoan fauna of Türkiye. Facial expressions are critical for non-verbal communication. The Canis genus epitomizes the interplay between behaviour and morphology in the evolution of non-verbal communication. Recent work suggests that the levator anguli oculi medialis (LAOM) muscle is unique to dogs (Canis familiaris) within the Canis genus and evolved due to domestication. The LAOM raises the inner eyebrows, resulting in the ‘puppy dog eyes’ expression. Here, we test whether the LAOM is a derived trait in dogs by (i) examining the facial expression muscles of a closely related and ancestral wild Canis species, the coyote (C. latrans) and (ii) comparing our results with …

The world is undergoing a biodiversity crisis in which hundreds of species have gone extinct within the last few centuries, rivaling previous mass extinction events that have only occurred five times in the history of the planet. Anthropogenic stressors such as the climate crisis, over exploitation of natural resources, deforestation, and pollution have created and fostered a mass extinction event that is taking place in every corner of the planet. Antiretroviral therapy (ART) has significantly improved the clinical outcomes of patients infected with human immunodeficiency virus (HIV). However, long-term use of certain antiretroviral drugs has been associated with renal mitochondrial toxicity (MtT), leading to potentially severe renal dysfunction. To date, there is no available test for renal MtT. Urine-derived stem cells (USCs) have emerged as a valuable non-invasive tool for studying renal pathology. In this study, we examine the effects of antiretroviral drugs on mitochondrial function in USCs from HIV patients as a first step towards establishing USCs as a tool for renal mitochondrial toxicity testing.
