Invasive Macroalgae In Native Seagrass Beds: Vectors Of Spread And Impacts, Louise Firth, Andy Foggo, Thomas Watts, Antony Knights, Stacey Deamicis

School of Biological and Marine Sciences

Background and AimsWorldwide, invasive species are spreading through marine systems at an unprecedented rate with both positive and negative consequences for ecosystems and the biological functioning of organisms. Human activities from shipping to habitat damage and modification are known vectors of spread, although biological interactions including epibiosis are increasingly recognized as potentially important to introduction into susceptible habitats.MethodsWe assessed a novel mechanism of spread – limpets as transporters of an invasive alga, Sargassum muticum, into beds of the seagrass Zostera marina – and the physiological impact of its invasion. The association of S. muticum with three limpet species and other …

The challenge of managing aquatic connectivity in a changing climate is exacerbated in the presence of additional anthropogenic stressors, social factors, and economic drivers. Here we discuss these issues in the context of structural and functional connectivity for aquatic biodiversity, specifically fish, in both the freshwater and marine realms. We posit that adaptive management strategies that consider shifting baselines and the socio-ecological implications of climate change will be required to achieve management objectives. The role of renewable energy expansion, particularly hydropower, is critically examined for its impact on connectivity. We advocate for strategic spatial planning that incorporates nature-positive solutions, ensuring …

Amplifying Pcr Productivity And Environmental Sustainability Through Shortened Cycling Protocols, Matthew Pedlar, Matthew J. Emery, Philip J. Warburton

School of Biological and Marine Sciences

Since its inception in the 1980s, advancements in PCR technology using improved thermal cyclers, engineered DNA polymerases and commercial master mixes, have led to increased PCR productivity. Despite these advancements, PCR cycling protocols have largely remained unchanged over the same period. This study aimed to systemically evaluate the effect of reduced PCR cycling parameters on amplicon production. The 1466bp fragment from the 16S rRNA gene present in low-, medium- and high-CG bacteria was amplified using three commercially available PCR master mixes. The shortest cycling parameters required to successfully amplify the 16S fragment from all bacteria and master mixes comprised 30-cycles …

Shifting The Focus From Species To Individuals In Invasion Biology: Individual Differences In Jumping Behaviour, Aaron Jessop, Miguel Barbosa, Michael Morrissey

School of Biological and Marine Sciences

Dispersal is critical to population persistence, colonization and connectivity which are all critical components of invasive success. While individual propensity to disperse varies within populations, the underlying mechanisms promoting individual dispersal remain unclear. Collectively, dispersal is influenced by the environment and individual phenotype. Here we investigated individual dispersal propensity in the Trinidadian guppy, Poecilia reticulata, and related individual variation in dispersal behaviour to phenotype and social/environmental conditions. Namely, we addressed the effect of sex ratio (social/environmental factor) on the tendency for individuals to disperse across physical barriers via jumping behaviour. Jumping is vital for the dispersal of many aquatic species …

School of Geography, Earth and Environmental Sciences

A novel method for determining the yield of bacterial growth using square wave voltammetry and screen printed electrodes is described here. This procedure was shown to be a reliable and accurate way to quickly determine the level of bacterial growth within culturing media and these results suggest that the combination of such sensors and the application of square wave voltammetry is suitable for applications such as within a bioreactor where the rapid determination of bacterial growth may be necessary.

Background: Ambergris is a coprolith produced by some sperm whales (∼1 %). It has been hypothesised that the metals in ambergris derive from the haemolymph of squid in the whale diet (mainly copper). However, few data exist. We report analysis of 10 trace metals in each of 50 ambergris samples by inductively coupled plasma-mass spectrometry. Main Findings: Acid digests of the samples contained ∼5 to >6880 µg g−1 air-dried weight total metals; mainly of iron (present in ∼90 % of samples, 38), copper (∼95 % of samples, 40), zinc (93 % of samples, 39) and cadmium (∼95 % of …

Guidelines For Protecting And Promoting Decapod Crustacean Welfare In Research, Mark Briffa Jan 2024

Guidelines For Protecting And Promoting Decapod Crustacean Welfare In Research, Mark Briffa

School of Biological and Marine Sciences

The decapod order contains ~15,000 species, including crabs, lobsters, crayfish, prawns, and shrimps. The idea that these animals might experience pain was long dismissed because they were thought to respond to noxious stimuli and tissue damage purely by nociceptive reflex—a position questioned by Sherwin (2001). Later experimental studies demonstrated that their responses were complex, long-lasting, and largely consistent with criteria for pain that had been produced initially for vertebrates (Elwood 2019). While the nature and extent of potential sentience in decapods remains an active area of research, these animals were recently recognized as sentient for the purposes of UK law, …

Low-fishmeal and protein-saving diets are two prominent nutritional strategies utilized to address challenges related to the scarcity and sustainability of protein sources in aquaculture. However, these diets have been associated with adverse effects on the growth performance, feed utilization, and disease resistance of aquatic animals. To mitigate these challenges, exogenous protease has been applied to enhance the quality of diets with lower protein contents or fishmeal alternatives, thereby improving the bioavailability of nutritional ingredients. Additionally, protease preparations were also used to enzymatically hydrolyze fishmeal alternatives, thus enhancing their nutritional utilization. The present review aims to consolidate recent research progress on …

Phenomics is the acquisition of high-dimensional data on an individual-wide scale and is proving transformational in areas of biological research related to human health including medicine and the crop sciences. However, more broadly, a lack of accessible transferrable technologies and research approaches is significantly hindering the uptake of phenomics, in contrast to molecular-omics for which transferrable technologies have been a significant enabler. Aquatic embryos are natural models for phenomics, due to their small size, taxonomic diversity, ecological relevance, and high levels of temporal, spatial and functional change. Here, we present LabEmbryoCam, an autonomous phenotyping platform for timelapse imaging of developing …

Sediment Sorting Within A Relatively Wave-Exposed And Sandy Subtidal Seagrass (Zostera Marina) Meadow, Gerd Masselink, Tom Brown, Tim Scott, Liane Brodie

School of Biological and Marine Sciences

Seagrasses impact on sedimentary intertidal and subtidal ecosystems by affecting local hydrodynamics, geomorphology and sediment properties. Their influence on hydrodynamics is to reduce flow velocities in their canopies, and this leads to increased net sedimentation rates and reduction of the grain size. Most investigations of the seagrass-hydrodynamics-sediment feedback system has been carried out over silt and fine-sand beds under tide-dominated conditions, mostly in the intertidal zone. Here, we use sedimentological data from a relatively wave-exposed and subtidal seagrass (Zostera marina) meadow in the Isles of Scilly with a fine-to-medium sand bed and show that the sand within the seagrass meadow …

Introduction: Vestigial-like 1 (VGLL1) is a co-transcriptional activator that binds to TEA domain-containing transcription factors (TEADs). Its expression is upregulated in a variety of aggressive cancer types, including pancreatic and basal-like breast cancer, and increased transcription of VGLL1 is strongly correlated with poor prognosis and decreased overall patient survival. In normal tissues, VGLL1 is most highly expressed within placental trophoblast cells, which share the common attributes of rapid cellular proliferation and invasion with tumor cells. The impact of VGLL1 in cancer has not been fully elucidated and no VGLL1-targeted therapy currently exists.

Methods: The aim of this study was to …

Breast cancer is the most frequent malignancy in women, constituting 15.2% of all new cancers diagnosed in the United States. Distant breast cancer metastasis accounts for the majority of breast cancer-related deaths; brain metastasis is the third most common site for metastatic breast cancer but is associated with worst prognosis of approximately eight months of survival. Current treatment options for breast cancer brain metastasis (BCBM) are limited and ineffective. To help identify new and effective therapies for BCBM, it is important to investigate the mechanisms by which breast cancer cells metastasize to the brain and thrive in the brain microenvironment. …

Purpose: Sperm DNA fragmentation (SDF) is a functional sperm abnormality that can impact reproductive potential, for which four assays have been described in the recently published sixth edition of the WHO laboratory manual for the examination and processing of human semen. The purpose of this study was to examine the global practices related to the use of SDF assays and investigate the barriers and limitations that clinicians face in incorporating these tests into their practice.

Materials and methods: Clinicians managing male infertility were invited to complete an online survey on practices related to SDF diagnostic and treatment approaches. Their responses …

Purpose: Due to various physical degradation factors and limited counts received, PET image quality needs further improvements. The denoising diffusion probabilistic model (DDPM) was a distribution learning-based model, which tried to transform a normal distribution into a specific data distribution based on iterative refinements. In this work, we proposed and evaluated different DDPM-based methods for PET image denoising.

Methods: Under the DDPM framework, one way to perform PET image denoising was to provide the PET image and/or the prior image as the input. Another way was to supply the prior image as the network input with the PET image included …

Introduction: The impact of spaceflight on the immune system and mitochondria has been investigated for decades. However, the molecular mechanisms underlying spaceflight-induced immune dysregulations are still unclear.

Methods: In this study, blood from eleven crewmembers was collected before and during International Space Station (ISS) missions. Transcriptomic analysis was performed in isolated peripheral blood mononuclear cells (PBMCs) using RNA-sequencing. Differentially expresses genes (DEG) in space were determined by comparing of the inflight to the preflight samples. Pathways and statistical analyses of these DEG were performed using the Ingenuity Pathway Analysis (IPA) tool.

Results: In comparison to pre-flight, a total of 2030 …

Hyperbaric oxygen (HBO) therapy is a well-established method for improving tissue oxygenation and is typically used for the treatment of various inflammatory conditions, including infectious diseases. However, its effect on the intestinal mucosa, a microenvironment known to be physiologically hypoxic, remains unclear. Here, we demonstrated that daily treatment with hyperbaric oxygen affects gut microbiome composition, worsening antibiotic-induced dysbiosis. Accordingly, HBO-treated mice were more susceptible to

Hypoxia-inducible factors (HIFs) are essential to the homeostasis of hypoxic tissues. Although HIF-2α, is expressed in nucleus pulposus (NP) cells, consequences of elevated HIF-2 activity on disc health remains unknown. We expressed HIF-2α with proline to alanine substitutions (P405A; P531A) in the Oxygen-dependent degradation domain (HIF-2αdPA) in the NP tissue using an inducible, nucleus pulposus-specific K19

INTRODUCTION: Bone metastases are associated with increased morbidity and decreased quality of life in patients with solid tumors. Identifying patients at increased risk of bone metastases at diagnosis could lead to earlier interventions. We sought to retrospectively identify the incidence and predictive factors for bone metastases at initial diagnosis in a large population-based dataset.

METHODS: The Surveillance, Epidemiology, and End Results (SEER) database was used to identify patients 18 years-old or older diagnosed with solid cancers from 2010 to 2019. Patients with hematologic malignancies and primary tumors of the bone were excluded. We calculated the incidence and predictive factors for …

Cognitive And Functional Performance And Plasma Biomarkers Of Early Alzheimer's Disease In Down Syndrome, Emily K Schworer, Beau M Ances, Et Al.

2020-Current year OA Pubs

INTRODUCTION: People with Down syndrome (DS) have a 75% to 90% lifetime risk of Alzheimer's disease (AD). AD pathology begins a decade or more prior to onset of clinical AD dementia in people with DS. It is not clear if plasma biomarkers of AD pathology are correlated with early cognitive and functional impairments in DS, and if these biomarkers could be used to track the early stages of AD in DS or to inform inclusion criteria for clinical AD treatment trials.

METHODS: This large cross-sectional cohort study investigated the associations between plasma biomarkers of amyloid beta (Aβ)42/40, total tau, and …
