Determining The Impact Of Lethal Temperature And Relative Humidity When Combined With Various Insecticide Classes Against A Bed Bug (Cimex Lectularius) Population., Bandana Shrestha

LSU Master's Theses

Common bed bugs (Cimex lectularius) are obligatory blood-feeding pests that have adapted well to survive in challenging environmental conditions. Known for their high tolerance to dehydration, low water loss, and high critical thermal maximum, they have become difficult to manage. Environmental factors, particularly temperature and humidity, play a significant role in their survival. However, with growing resistance to insecticides, integrated pest management (IPM) strategies are now essential for effective bed bug control. A key strategy in IPM involves combining insecticides with heat treatments. This method aims to overcome the limitations of heat treatment alone, as bed bugs may …

Rabbit Control: Warren And Harbourage Destruction, Department Of Primary Industries And Regional Development, Western Australia Nov 2024

Rabbit Control: Warren And Harbourage Destruction, Department Of Primary Industries And Regional Development, Western Australia

Biosecurity factsheets

If correctly and appropriately undertaken, the destruction of rabbit warrens, or modification of the habitat so it becomes less favourable to rabbits, can be an important and effective part of reducing the impacts of rabbits.

Destruction of warrens can improve the overall effectiveness of rabbit control programs, even though many rabbits live and shelter above-ground under bushes, and in scrub). If undertaken properly, the ripping of rabbit warrens can often provide a relatively permanent and long-term benefit. Similarly, removing or modifying other forms of rabbit-harbourage (for example, removing rock piles) can help, particularly in semi-urban areas or on smallholdings.

Characterization Of The Ubiquitin‐Modified Proteome Of Recombinant Chinese Hamster Ovary Cells In Response To Endoplasmic Reticulum Stress, Karuppuchamy Selvaprakash, Christiana‐Kondylo Sideri, Michael Henry, Esen Efeoglu, David Ryan, Paula Meleady

Research Outputs: 2025-Present

Chinese hamster ovary (CHO) cells remain the most widely used host cell line for biotherapeutics production. Despite their widespread use, understanding endoplasmic reticulum (ER) stress conditions in recombinant protein production remains limited, often creating bottlenecks preventing improved production titers and product quality. Ubiquitination not only targets substrates (e.g., misfolded proteins) for proteasome degradation but also has important regulatory control functions including cell cycle regulation, translation, apoptosis, autophagy, etc. and hence is likely to be central to understanding and controlling the productivity of recombinant biotherapeutics. This study aimed to uncover differentially expressed ubiquitinated proteins following artificial induction of ER-stress in recombinant …

Trade-Offs In A Reef-Building Coral After Six Years Of Thermal Acclimation, Anna Roik, Marlene Wall, Melina Dobelmann, Samuel Nietzer, Anna Fiesinger, Miriam Reverter, David Brefeld, Peter J Schupp, Matthew Jackson, Marie Rutsch, Julia Strahl

School of Biological and Marine Sciences

There is growing evidence that reef-building corals can acclimate to novel and challenging thermal conditions. However, potential trade-offs that accompany acclimation remain largely unexplored. We investigated physiological trade-offs in colonies of a globally abundant coral species (Pocillopora acuta) that were acclimated ex situ to an elevated temperature of 31 °C (i.e., 1 °C above their bleaching threshold) for six years. By comparing them to conspecifics maintained at a cooler temperature, we found that the energy storage of corals was prioritized over skeletal growth at the elevated temperature. This was associated with the formation of higher density skeletons, lower calcification rates …

Severe joint infections, such as septic arthritis, require rapid diagnostic testing of the synovial fluid aspirated from joints level so that a surgical team can be assembled quickly. We present a diffuse reflectance spectroscopy (DRS) system for noncontact determination of infection. Using a light-tight syringe holder and fiber optic probe, diffusely reflected light from 475 to 655 nm was acquired from 18 patient samples through the wall of a syringe in a noncontact and sterile manner. We determined the reflectance ratios at two different wavelengths—R 490/R 600 and R 580/R 600 and found statistically significant …

Novel biomaterials that bridge the knowledge gap in coupling molecular/protein signatures of disease/stress with rapid readouts are a critical need of society. One such scenario is an imbalance between bodily heat production and heat dissipation which leads to heat stress in organisms. In addition to diminished animal well-being, heat stress is detrimental to the poultry industry as poultry entails fast growth and high yields, resulting in greater metabolic activity and higher body heat production. When stressed, cells overexpress heat shock proteins (such as HSP70, a well-established intracellular stress indicator) and may undergo changes in their mechanical properties. Liquid crystals (LCs, …

Epithelial to mesenchymal transition (EMT) is believed to be a principal factor contributing to cancer metastasis. The post-transcriptional and post-translational mechanisms underlying EMT are comparatively underexplored. We previously demonstrated that the CELF1 RNA binding protein is necessary and sufficient to drive the EMT of breast epithelial cells, and that the relative protein expression of CELF1 in this context was dictated at the post-translational level. Here, we elucidate the mechanism of this regulation. Mass spectrometric analysis of CELF1 isolated from mesenchymal MCF-10A cells identified multiple sites of serine and threonine phosphorylation on the protein, correlating with the increased stability of this …

Co-occurring mutations in KEAP1 in STK11/LKB1-mutant NSCLC activate NFE2L2/NRF2 to compensate for the loss of STK11-AMPK activity during metabolic adaptation. Characterizing the regulatory crosstalk between the STK11-AMPK and KEAP1-NFE2L2 pathways during metabolic stress is crucial for understanding the implications of co-occurring mutations. Here, we found that metabolic stress increased the expression and phosphorylation of SQSTM1/p62, which is essential for the activation of NFE2L2 and AMPK, synergizing antioxidant defense and tumor growth. The SQSTM1-driven dual activation of NFE2L2 and AMPK was achieved by inducing macroautophagic/autophagic degradation of KEAP1 and facilitating the AXIN-STK11-AMPK complex formation on the lysosomal membrane, respectively. In contrast, …

Prolonged antibiotic exposure causes dangerous hematologic side effects, including neutropenia, in up to 34% of patients. Murine studies established a link between the intestinal microbiota and hematopoiesis. To identify factors that predispose to neutropenia in pediatric patients, we evaluated changes in microbiota‐derived metabolites and intestinal microbiota composition after prolonged courses of antibiotics. In this multi‐center study, patients with infections requiring anticipated antibiotic treatment of two or more weeks were enrolled. Stool samples were obtained at the start and completion of antibiotics or at neutropenia onset (prospective arm). Some patients were enrolled in a retrospective arm in which a stool sample …

TFEB, a bHLH-leucine zipper transcription factor belonging to the MiT/TFE family, globally modulates cell metabolism by regulating autophagy and lysosomal functions. Remarkably, loss of TFEB in mice causes embryonic lethality due to severe defects in placentation associated with aberrant vascularization and resulting hypoxia. However, the molecular mechanism underlying this phenotype has remained elusive. By integrating in vivo analyses with multi-omics approaches and functional assays, we have uncovered an unprecedented function for TFEB in promoting the formation of a functional syncytiotrophoblast in the placenta. Our findings demonstrate that constitutive loss of TFEB in knock-out mice is associated with defective formation of …

METHODS: We examined the cross-sectional association of vital exhaustion (assessed using the Maastricht Vital Exhaustion Questionnaire [MVEQ]) with cardiac biomarker (high-sensitivity troponin T [hs-TnT], N-terminal pro-B-type natriuretic peptide [NT-proBNP]) and high-sensitivity C-reactive protein (hs-CRP) levels in 11,542 ARIC study participants without CVD at ARIC visit 2 using multivariable logistic and linear …

BACKGROUND: Spontaneous coronary artery dissection (SCAD) is a rare and often underdiagnosed cause of acute coronary syndrome (ACS), predominantly affecting younger women without traditional cardiovascular risk factors. The management of SCAD remains a subject of debate, likely secondary to inconclusive evidence. This study aims to compare the clinical outcomes of SCAD patients treated with optimal medical therapy (OMT) versus those who underwent percutaneous coronary intervention (PCI) using a national population-based cohort.

METHODS: We conducted a retrospective analysis using the National Inpatient Sample (NIS) database from 2016 to 2020. The study included patients identified with SCAD using the ICD-10-CM (the International …
