Gestational Weight Gain And Epilepsy In The Offspring: A Population-Based Retrospective Cohort Study, Jiawen Chen
Gestational Weight Gain And Epilepsy In The Offspring: A Population-Based Retrospective Cohort Study, Jiawen Chen
Public Health Theses
AbstractBackground and aims: Epilepsy is one of the most common neurologic disorders in childhood, but the etiology for the majority of the cases remains unknown. As a modifiable trait in pregnancy, gestational weight gain (GWG) has been shown to be a risk factor for multiple adverse birth outcomes. This study aims to investigate whether non-optimal GWG is associated with the risk of epilepsy in the offspring using a linked database in California. Methods: A population-based cohort study was conducted using the linked database from the Birth Certificate Record in California (2007-2015) for GWG, and the diagnostic records of epilepsy from …

Development And Optimization Of A Novel Eeev Microsphere Immunoassay: Using Xeno-Surveillance Methodology To Characterize Avian Herd Immunity, Kaitlyn Gabriele Maurais
Public Health Theses
Introduction: Eastern equine encephalitis virus (EEEV), a mosquito-borne alphavirus, is detected throughout the eastern United States. The primary enzootic vector, Culiseta melanura (Blacktailed mosquito), allows transmission among 50+ avian host species resulting in complex disease patterns. Spillover to dead-end host mammals, such as equines and humans, rarely occurs, however, human cases can cause severe neurological deficits and have a 30% mortality rate. Eco-epidemiology and avian host immunity patterns are not well characterized for EEEV and require ongoing surveillance to develop more targeted interventions. Methods: Xeno-surveillance is an approach to survey EEEV through hematophagous arthropods, instead of collecting samples from a …

Obesity is an epidemic that affects over 40% of adults in America. Obesity can be divided into metabolically healthy and metabolically unhealthy obesity, with the latter being characterized by symptoms closely associated with metabolic syndrome, type 2 diabetes, and metabolic dysfunction-associated steatotic liver disease (MASLD). What causes the transition from metabolically healthy to metabolically unhealthy obesity is not fully known. Further, it is unclear how this transition correlates with the onset of co-morbid conditions. We propose that Ubiquitin D (FAT10) may play a role in transitioning into a metabolically unhealthy state in response to obesity. This dissertation evaluated two main …

Lake Tanganyika, in central Africa, contains a diverse and endemic fauna under threat from global climate change, overfishing, and nearshore sediment pollution. Previous studies of sediment pollution focused justifiably on impacts along rocky shorelines where diversity is high, but Lake Tanganyika also contains widespread shelly accumulations (shell beds) unprecedented in the modern East African lakes, but where impacts are less con- strained. Here we integrate multiple datasets from three sites along the Tanzanian shoreline to explore how variation in sedimentation rates and sediment quality impacts shell-bed substrate and diversity and abundance of ostracodes and sponges across sites that exhibit varying …

The Loss Of Βι Spectrin Alters Synaptic Size And Composition In The, Michael C Stankewich, Luanne L. Peters, Jon S Morrow
Faculty Research 2024
Introduction: Deletion or mutation of members of the spectrin gene family contributes to many neurologic and neuropsychiatric disorders. While each spectrinopathy may generate distinct neuropathology, the study of βΙ spectrin’s role (Sptb) in the brain has been hampered by the hematologic consequences of its loss.
Methods: Jaundiced mice (ja/ja) that lack βΙ spectrin suffer a rapidly fatal hemolytic anemia. We have used exchange transfusion of newborn ja/ja mice to blunt their hemolytic pathology, enabling an examination of βΙ spectrin deficiency in the mature mouse brain by ultrastructural and biochemical analysis.
Results: βΙ spectrin is widely utilized throughout the brain as …

The increase in the global atmospheric CO2 concentration is expected to increase the productivity of forests, but the dynamic processes of such increased productivity in the forest canopy remain unclear. In this study, diurnal and seasonal variations and vertical changes in photosynthetic rates were investigated in Camphor tree (Cinnamomum camphora) forests in subtropical China. The effect of photosynthetically active radiation (PAR) and CO2 concentrations on photosynthetic rates were also examined in the studied forests. Results showed the diurnal patterns of photosynthesis exhibited two peaks on sunny days, but only one peak on cloudy days. The daily average photosynthetic rate on …

Though the portfolio of medicines that are extending and improving the lives of patients continues to grow, drug discovery and development remains a challenging business on its best day. Safety liabilities are a significant contributor to development attrition where the costliest liabilities to both drug developers and patients emerge in late development or post-marketing. Animal studies are an important and influential contributor to the current drug discovery and development paradigm intending to provide evidence that a novel drug candidate can be used safely and effectively in human volunteers and patients. However, translational gaps-such as toxicity in patients not predicted by …

A number of studies have reported an association between phosphorus, red blood cell (RBC) production, and iron metabolism. However, it is difficult to distinguish whether the effect of phosphorus is direct or through the actions of FGF23, and it is not clear whether phosphorus is positively or negatively associated with RBC production. In the present study, we investigated the effects of a) increased phosphorus load and b) phosphorus deficiency on erythropoiesis and iron metabolism in association with FGF23. Mice were fed either a 1.2% or 1.65% phosphorus diet and compared to mice fed a control diet containing 0.6% of phosphorus. …

The greenhouse to icehouse transition at the Eocene-Oligocene boundary (34 Ma) marked the appearance of continental-scale glaciation in Antarctica. The material recovered from Ocean Drilling Program Site 696 is the only record spanning this major climatic shift in the Weddell Sea region. Using scanning electron microscopy, quartz microtextures in 13 samples across the Eocene-Oligocene transition were analyzed to understand the degree of glacial modification and the transportation history of the >150 µm material. The quartz grains were visually grouped, characterized, and interpreted by grain outline, relief, and surface microtextures. Glacial textures are present throughout the entire interval (34.4–33.2 Ma), with …

Apurinic/apyrimidinic endonuclease 1 (APE1) is involved in DNA repair and transcriptional regulation mechanisms. This multifunctional activity of APE1 should be supported by specific structural properties of APE1 that have not yet been elucidated. Herein, we applied atomic force microscopy (AFM) to characterize the interactions of APE1 with DNA containing two well-separated G-rich segments. Complexes of APE1 with DNA containing G-rich segments were visualized, and analysis of the complexes revealed the affinity of APE1 to G-rich DNA sequences, and their yield was as high as 53%. Furthermore, APE1 is capable of binding two DNA segments leading to the formation of loops …

A series of 7-substituted coumarin derivatives have been characterized as pan-aldo-keto reductase family 1C (AKR1C) inhibitors. The AKR1C family of enzymes are overexpressed in numerous cancers where they are involved in drug resistance development. 7-hydroxy coumarin ethyl esters and their corresponding amides have high potency for AKR1C3 and AKR1C2 inhibition. Coumarin amide 3 a possessed IC50 values of 50 nM and 90 nM for AKR1C3 and AKR1C2, respectively, and exhibits 'drug-like' metabolic stability and half-life in human and mouse liver microsomes and plasma. Compound 3 a was employed as a chemical tool to determine pan-AKR1C2/3 inhibition effects both as a …

Aldo-keto reductase 1C3 (AKR1C3) is a protein upregulated in prostate cancer, hematological malignancies, and other cancers where it contributes to proliferation and chemotherapeutic resistance. Androgen receptor splice variant 7 (ARv7) is the most common mutation of the AR receptor that confers resistance to clinical androgen receptor signalling inhibitors in castration-resistant prostate cancer. AKR1C3 interacts with ARv7 promoting stabilization. Herein we report the discovery of the first-in-class AKR1C3 Proteolysis-Targeting Chimera (PROTAC) degrader. This first-generation degrader potently reduced AKR1C3 expression in 22Rv1 prostate cancer cells with a half-maximal degradation concentration (DC50) of 52 nM. Gratifyingly, concomitant degradation of ARv7 was …
