Adolescent alcohol use is a strong predictor for the subsequent development of alcohol use disorders later in life. Additionally, adolescence is a critical period for the onset of affective disorders, which can contribute to problematic drinking behaviours and relapse, particularly in females. Previous studies from our laboratory have shown that exposure to adolescent intermittent ethanol (AIE) vapour alters glutamatergic transmission in the bed nucleus of the stria terminalis (BNST) and, when combined with adult stress, elicits sex-specific changes in glutamatergic plasticity and negative affect-like behaviours in mice. Building on these findings, the current work investigated whether BNST stimulation could substitute …

The heterogeneity of the whole-exome sequencing (WES) data generation methods present a challenge to a joint analysis. Here we present a bioinformatics strategy for joint-calling 20,504 WES samples collected across nine studies and sequenced using ten capture kits in fourteen sequencing centers in the Alzheimer's Disease Sequencing Project. The joint-genotype called variant-called format (VCF) file contains only positions within the union of capture kits. The VCF was then processed specifically to account for the batch effects arising from the use of different capture kits from different studies. We identified 8.2 million autosomal variants. 96.82% of the variants are high-quality, and …

There is an urgent need for new antibiotics given the rise of antibiotic resistance, and succinyl-diaminopimelate desuccinylase (DapE, E.C. 3.5.1.18) has emerged as a promising bacterial enzyme target. DapE from Haemophilus influenzae (HiDapE) has been studied and inhibitors identified, but it is essential to explore DapE from different species to assess selective versus broad-spectrum therapeutics. We have determined the structure of DapE from the ESKAPE pathogen Acinetobacter baumannii (AbDapE) and studied inhibition by known inhibitors of HiDapE. AbDapE is inhibited by captopril and sulfate comparable to HiDapE, but AbDapE was not significantly …

Integrative Genomic Analyses Reveal Putative Cell Type-Specific Targets Of The Drosophila Ets Transcription Factor Pointed, Komal Kumar Bollepogu Raja, Kelvin Yeung, Yoon-Kyung Shim, Graeme Mardon

Faculty, Staff and Students Publications

The Ets domain transcription factors direct diverse biological processes throughout all metazoans and are implicated in development as well as in tumor initiation, progression and metastasis. The Drosophila Ets transcription factor Pointed (Pnt) is the downstream effector of the Epidermal growth factor receptor (Egfr) pathway and is required for cell cycle progression, specification, and differentiation of most cell types in the larval eye disc. Despite its critical role in development, very few targets of Pnt have been reported previously. Here, we employed an integrated approach by combining genome-wide single cell and bulk data to identify putative cell type-specific Pnt targets. …

In the United States (US), biosafety and biosecurity oversight of research on viruses is being reappraised. Safety in virology research is paramount and oversight frameworks should be reviewed periodically. Changes should be made with care, however, to avoid impeding science that is essential for rapidly reducing and responding to pandemic threats as well as addressing more common challenges caused by infectious diseases. Decades of research uniquely positioned the US to be able to respond to the COVID-19 crisis with astounding speed, delivering life-saving vaccines within a year of identifying the virus. We should embolden and empower this strength, which is …

Trex2 Deficiency Suppresses Spontaneous And Genotoxin-Associated Mutagenesis, Teresa Marple, Mi Young Son, Xiaodong Cheng, Jun Ho Ko, Patrick Sung, Paul Hasty

Faculty, Staff and Student Publications

TREX2, a 3'-5' exonuclease, is a part of the DNA damage tolerance (DDT) pathway that stabilizes replication forks (RFs) by ubiquitinating PCNA along with the ubiquitin E3 ligase RAD18 and other DDT factors. Mismatch repair (MMR) corrects DNA polymerase errors, including base mismatches and slippage. Here we demonstrate that TREX2 deletion reduces mutations in cells upon exposure to genotoxins, including those that cause base lesions and DNA polymerase slippage. Importantly, we show that TREX2 generates most of the spontaneous mutations in MMR-mutant cells derived from mice and people. TREX2-induced mutagenesis is dependent on the nuclease and DNA-binding attributes of TREX2. …

Patients with cholangiocarcinoma have poor clinical outcomes due to late diagnoses, poor prognoses, and limited treatment strategies. To identify drug combinations for this disease, we have conducted a genome-wide CRISPR screen anchored on the bromodomain and extraterminal domain (BET) PROTAC degrader ARV825, from which we identified anticancer synergy when combined with genetic ablation of members of the mTOR pathway. This combination effect was validated using multiple pharmacological BET and mTOR inhibitors, accompanied by increased levels of apoptosis and cell cycle arrest. In a xenograft model, combined BET degradation and mTOR inhibition induced tumor regression. Mechanistically, the 2 inhibitor classes converged …

The Comparative Impact Of Acesulfame-Potassium, Stevia Extract, And Monk Fruit On The Lifespan Of Drosophila Melanogaster, Beverly Holton Jan 2024

The Comparative Impact Of Acesulfame-Potassium, Stevia Extract, And Monk Fruit On The Lifespan Of Drosophila Melanogaster, Beverly Holton

Journal of the South Carolina Academy of Science

Non-nutritive sweeteners (NNS), non-caloric sweetening devices, have been widely commercialized to reduce sugar consumption. This intent has associated NNS with health benefits, though reports have correlated the consumption of these substitutes with noncommunicable diseases. The interaction between NNS and distinct physiological processes has been examined, though research into the broader implications of these interactions, toward longevity, for instance, is lacking. The purpose of this study was to compare the impact of three recently FDA-approved NNS - acesulfame-Potassium (ace-K), stevia, and monk fruit - on the survival of Drosophila melanogaster, a model species for longevity research, to discern the possible impacts …

The Effect Of Crystalline Phase On The Toxicity Of Tio2 Nanoparticles To Chlorella, Jisoo Lee Jan 2024

The Effect Of Crystalline Phase On The Toxicity Of Tio2 Nanoparticles To Chlorella, Jisoo Lee

Journal of the South Carolina Academy of Science

Nanoparticles (NPs), or particles with dimensions that go below 100 nanometers, can be released into the environment in various ways, such as through cosmetic products like sunscreen. Titanium dioxide (TiO2) is a commonly used nanoparticle in sunscreen, and it has the potential to be harmful to various aquatic organisms including algae. The purpose of this study was to examine how the crystalline phase of TiO2 NPs (brookite or rutile) influenced their toxicity to the microalgae species Chlorella. It was hypothesized that brookite phase TiO2 NPs would inhibit the growth of Chlorella more than rutile phase TiO …

The Effect Of The Addition Of Carrot Seed Oil And Raspberry Seed Oil On The Uv Intensity Of A Sunscreen, Isra Siddique

Journal of the South Carolina Academy of Science

With continuous developments in research indicating the potential health risks of chemical sunscreen filters, the novel approach of incorporating natural oils with UV light blocking properties has been explored. Natural oils extracted from fruit and vegetables have been proven to possess photoprotective properties, but their effectiveness in combination with synthetic sunscreen filters has not been as thoroughly investigated. The purpose of this study was to determine whether the addition of carrot and raspberry seed oil to an SPF 30 sunscreen would decrease the UV intensity of UVB light passing through the sunscreen and oil mixture. It was hypothesized that if …

Simple Summary

Most neurofibromatosis type 1 (NF1) patients present with neurological issues in parallel with abnormal brain white matter and myelin. Although links for NF1 neuropathophysiology and abnormal myelin were proposed long ago, no current data can openly support or refute this idea. Various studies suggest that Nf1 mutations affect myelin biology and function, but any impact on learning/memory remains unclear. Here, we show that mice with an Nf1 mutation induced in adult myelinating cells present learning, but not memory, issues in a voluntary fine motor skill test, the complex wheel (CW). The specific parameters shaping CW learning curves are …

The investigation of the effects of prenatal cocaine exposure (PCE) on offspring has been inconsistent, with few studies investigating biological outcomes in humans. We profiled genome-wide DNA methylation (DNAm) of umbilical cord blood (UCB) from newborns with (n = 35) and without (n = 47) PCE. We used DNAm data to (1) assess pediatric epigenetic clocks at birth and (2) to estimate epigenetic scores (ES) for lifetime disorders. We generated gestational epigenetic age estimates (DNAmGA) based on Knight and Bohlin epigenetic clocks. We also investigated the association between DNAmGA and UCB serum brain-derived neurotrophic factor (BDNF) levels. Considering the large-scale …

Eucalypts (Myrtaceae tribe Eucalypteae) are currently placed in seven genera. Traditionally, Eucalyptus was defined by its operculum, but when phylogenies placed Angophora, with free sepals and petals, as sister to the operculate bloodwood eucalypts, the latter were segregated into a new genus, Corymbia. Yet, generic delimitation in the tribe Eucalypteae remains uncertain. Here, we address these problems using phylogenetic analysis with the largest molecular data set to date. We captured 101 low-copy nuclear exons from 392 samples representing 266 species. Our phylogenetic analysis used maximum likelihood (IQtree) and multispecies coalescent (Astral). At two nodes critical to generic delimitation, we tested …

Acetic Acid: An Underestimated Metabolite In Ethanol-Induced Changes In Regulating Cardiovascular Function, Andrew D. Chapp, Zhiying Shan, Qing Hui Chen

Michigan Tech Publications

Acetic acid is a bioactive short-chain fatty acid produced in large quantities from ethanol metabolism. In this review, we describe how acetic acid/acetate generates oxidative stress, alters the function of pre-sympathetic neurons, and can potentially influence cardiovascular function in both humans and rodents after ethanol consumption. Our recent findings from in vivo and in vitro studies support the notion that administration of acetic acid/acetate generates oxidative stress and increases sympathetic outflow, leading to alterations in arterial blood pressure. Real-time investigation of how ethanol and acetic acid/acetate modulate neural control of cardiovascular function can be conducted by microinjecting compounds into autonomic …

Microbial species capable of co-existing with healthy individuals, such as the commensal fungus Candida albicans, exploit multifarious strategies to evade our immune defenses. These strategies include the masking of immunoinflammatory pathogen-associated molecular patterns (PAMPs) at their cell surface. We reported previously that C. albicans actively reduces the exposure of the proinflammatory PAMP, β-1,3-glucan, at its cell surface in response to host-related signals such as lactate and hypoxia. Here, we show that clinical isolates of C. albicans display phenotypic variability with respect to their lactate- and hypoxia-induced β-1,3-glucan masking. We have exploited this variability to identify responsive and non-responsive clinical isolates. …

Htra1 Prevents And Reverses Α-Synuclein Aggregation, Rendering It Non-Toxic And Seeding Incompetent, Sheng Chen

Arts & Sciences Graduate Student Theses and Dissertations

Protein misfolding and accumulation of misfolded species are associated with multiple neurodegenerative diseases, including Parkinson’s disease (PD), amyotrophic lateral sclerosis (ALS), and frontotemporal dementia (FTD). α-Synuclein (α-Syn) misfolding and accumulation into Lewy bodies is closely linked to Parkinson’s disease. In ALS and FTD, TDP-43 and FUS are found to be aggregated in disease pathology. These proteins aggregate and associate with both a toxic gain of function of misfolded form and a loss of function due to misfolding. Therefore, it is critical to understand molecular mechanisms by which cells can prevent protein misfolding and how the cell can reverse the aggregation …

The role of hematopoietic Hedgehog signaling in myeloproliferative neoplasms (MPNs) remains incompletely understood despite data suggesting that Hedgehog (Hh) pathway inhibitors have therapeutic activity in patients. We aim to systematically interrogate the role of canonical vs. non-canonical Hh signaling in MPNs. We show that Gli1 protein levels in patient peripheral blood mononuclear cells (PBMCs) mark fibrotic progression and that, in murine MPN models, absence of hematopoietic Gli1, but not Gli2 or Smo, significantly reduces MPN phenotype and fibrosis, indicating that GLI1 in the MPN clone can be activated in a non-canonical fashion. Additionally, we establish that hematopoietic Gli1 has a …

The resting zone of the postnatal growth plate is organized by slow-cycling chondrocytes expressing parathyroid hormone-related protein (PTHrP), which include a subgroup of skeletal stem cells that contribute to the formation of columnar chondrocytes. The PTHrP-Indian hedgehog feedback regulation is essential for sustaining growth plate activities; however, molecular mechanisms regulating cell fates of PTHrP+ resting chondrocytes and their eventual transformation into osteoblasts remain largely undefined. Here, in a mouse model, we specifically activated Hedgehog signaling in PTHrP+ resting chondrocytes and traced the fate of their descendants using a tamoxifen-inducible Pthrp-creER line with patched-1-floxed and tdTomato reporter alleles. Hedgehog-activated PTHrP+ chondrocytes …

Multivalent Epigraph Hemagglutinin Vaccine Protects Against Influenza B Virus In Mice, Erika M. Petro-Turnquist, Brigette Corder Kampfe, Amber Gadeken, Matthew J. Pekarek, Eric A. Weaver

Nebraska Center for Virology: Faculty Publications

Influenza B virus is a respiratory pathogen that contributes to seasonal epidemics, accounts for approximately 25% of global influenza infections, and can induce severe disease in young children. While vaccination is the most commonly used method of preventing influenza infections, current vaccines only induce strain-specific responses and have suboptimal efficacy when mismatched from circulating strains. Further, two influenza B virus lineages have been described, B/Yamagatalike and B/Victoria-like, and the limited cross-reactivity between the two lineages provides an additional barrier in developing a universal influenza B virus vaccine. Climate change is increasing the frequency and severity of short-term (~1 y) drought events-the most common duration of drought-globally. Yet the impact of this intensification of drought on ecosystem functioning remains poorly resolved. This is due in part to the widely disparate approaches ecologists have employed to study drought, variation in the severity and duration of drought studied, and differences among ecosystems in vegetation, edaphic and climatic attributes that can mediate drought impacts. To overcome these problems and better identify the factors that modulate drought responses, we used a coordinated distributed experiment to quantify the impact of short-term drought on …

BACKGROUND: Antibody-dependent cell-mediated cytotoxic (ADCC) response mediated by natural killer (NK) cells correlates with decreased infection risk in studies involving simian immunodeficiency virus (SIV)/simian-human immunodeficiency virus (SHIV), and human immunodeficiency virus (HIV) vaccine candidates. Currently, the heterogeneities of the functional subset of rhesus macaque natural killer (RMNK) cells are under-characterized.

METHOD: We engaged the RMNK cells with ADCC-mediating anti-HIV-1 monoclonal antibodies (ADCCAbs) or anti-CD16 antibodies and used CD107a expression as the surrogate marker for RMNK cells actively involved in ADCC. CD107a+ and CD107a- populations were analyzed individually using single-cell RNA sequencing.

RESULTS: Subsets of CD107a+ RMNK cells produced …

The epidermal growth factor receptor (EGFR) is a receptor tyrosine kinase with important roles in many cellular processes as well as in cancer and other diseases. EGF binding promotes EGFR dimerization and autophosphorylation through interactions that are well understood structurally. How these dimers relate to higher-order EGFR oligomers seen in cell membranes, however, remains unclear. Here, we used single-particle tracking (SPT) and Förster resonance energy transfer imaging to examine how each domain of EGFR contributes to receptor oligomerization and the rate of receptor diffusion in the cell membrane. Although the extracellular region of EGFR is sufficient to drive receptor dimerization, …

BACKGROUND: It is unknown whether dietary intake of polyunsaturated fatty acids (PUFA) modifies the cardiovascular disease (CVD) risk associated with a family history of CVD. We assessed interactions between biomarkers of low PUFA intake and a family history in relation to long-term CVD risk in a large consortium.

METHODS: Blood and tissue PUFA data from 40 885 CVD-free adults were assessed. PUFA levels ≤25th percentile were considered to reflect low intake of linoleic, alpha-linolenic, and eicosapentaenoic/docosahexaenoic acids (EPA/DHA). Family history was defined as having ≥1 first-degree relative who experienced a CVD event. Relative risks with 95% CI of CVD were …

Human Whole-Exome Genotype Data For Alzheimer's Disease, Yuk Yee Leung, Carlos Cruchaga, Et Al.

2020-Current year OA Pubs

Acculturation And Intimate Partner Violence Among Kenyans In The United States, Max J. Stein, Peter Ndiang’Ui, Eunice Menja Jan 2024

Acculturation And Intimate Partner Violence Among Kenyans In The United States, Max J. Stein, Peter Ndiang’Ui, Eunice Menja

Journal of Social, Behavioral, and Health Sciences

Intimate partner violence (IPV) is abuse by a partner or spouse. This study focused on IPV among Kenyan immigrants to the United States. Several studies reported connections between IPV and cultural tensions experienced during the acculturation process. Scholars disagree whether acculturation buffers against IPV by exposing immigrants to adaptive social norms or heightens risk factors among those facing challenges acclimating to new settings. Whereas this association has been researched among Latinx and Asian communities in the United States, it is understudied among African and especially Kenyan diasporas. This descriptive study explored how acculturation and IPV among U.S. Kenyans were experienced …

Nanoparticle delivery to solid tumors is a prime challenge in nanomedicine. Here, we approach this challenge through the lens of biogeochemistry, the field that studies the flow of chemical elements within ecosystems as manipulated by living cellular organisms and their environments. We leverage biogeochemistry concepts related to gold cycling against pancreatic cancer, considering mammalian organisms as drivers for gold nanoparticle biosynthesis. Sequestration of gold nanoparticles within tumors has been demonstrated as an effective strategy to enhance radiotherapy; however, the desmoplasia of pancreatic cancer impedes nanoparticle delivery. Our strategy overcomes this barrier by applying an atomic-scale agent, ionic gold, for intratumoral …

Aquaculture is crucial for global food and nutrition security due to the inability of wild harvests to meet increasing demand. Africa’s contribution to aquaculture is generally low, despite its potential for economic development, food security, and reduced unemployment. The study focuses on the African catfish (Clarias gariepinus) a freshwater fish species that is widely farmed for food in Africa and other parts of the world. Proper nutrition is essential for the growth and development of the fish, and understanding their nutritional requirements is critical for producing healthy and high-quality fish. This review article provides an overview of the knowledge on …

Impact Of Type Of K Fertilizer On Dcad Of Fen Grassland In Northeast Germany, A. Behrendt, J. Pickert, F. Eulenstein, B. Kroschewski

IGC Proceedings (1977-2023)

A lower DCAD is required in the diet fed during the prepartum period, but increasing the DCAD of diets fed during lactation increased milk yield and dry matter intake (Overton 2020). We conducted 3 cutting date/type of fertilizer trials, harvested with 5, 4 and 3 cuts per year over 3 years. Compared to the chloride based K fertilizer, the application of a sulphate based K fertilizer did not change dry matter yield and fodder quality (NEL-, XP-content) but led to significant higher DCAD in the grass in cut 1 - 3 of all trials over 3 years. The increase of …
