As global temperatures rise due to climate change, nature-based solutions are increasingly being used to restore nature and to remove carbon from the atmosphere. Nature-based solutions draw on methodologies such as ecological forestry and improved forest management to reduce emissions and maximize resilience. These protocols are still in their infancy and additional measurements and case studies are important to optimizing these protocols. Herein this dissertation we describe multiple measurement methodologies for both greenhouse gas (GHG) emissions and for ecosystem services. In this dissertation, artificial intelligence (AI) models are designed and used to accelerate inverse design of composites and manufacturing processes. The critical bottlenecks in machine learning (ML) including data availability, data quality, model generalization and adaptation, interpretability, physical consistency, and the ’black box’ nature of models for the inverse design are addressed. And the proposed AI models are tested under different engineering scenarios. Firstly, a fast deep neural operator (DNO) structure was developed to significantly reduce training time. Glioblastoma (GBM) accounts for over 66% of all malignant central nervous system and brain tumors with a 5-year survival rate below 5%, which has remained relatively unchanged over the last four decades. To bypass the most challenging barrier to treatment, the blood-brain barrier (BBB), one intracranial implant is clinically available for GBM treatment. However, this treatment produces severe adverse effects with no significant improvements in patient outcomes. This research examines the impact of food law politics on community empowerment and sustainable agriculture in Indonesia, focusing on the Sustainable Food Garden Program (P2L). Utilizing a normative juridical analysis, it explores how legal frameworks can support or obstruct community-led agricultural initiatives. The study highlights the empowering effect of laws aligned with community goals and the challenges arising from legislation favoring industrial agriculture. The incidence rate of autism spectrum condition (ASC) has increased significantly in recent decades, as awareness of the condition and its impacts increases amongst clinicians, parents, and the general population. Medical literature has proposed that there may be a relationship between ASC and participation in the computing field. This study tests for the prevalence of autism spectrum condition traits measured by delivering the Autism Spectrum Quotient (AQ) to a population of undergraduate computer science students. In this paper, we study the dynamical analysis of a stochastic Leslie–Gower biological predator– prey model. Earlier, the Leslie–Gower model was studied in the context of biological systems, including cases involving cannibalism. In our model, we investigate the dynamic properties of a stochastic Leslie–Gower predator–prey ecological system using the stability of invariant measures on invariant sets, where the invariant measures are shown to be ergodic. We also conduct a threshold analysis to study the stochastic persistence and extinction of species. Stochastic bifurcation is also examined. Death didn’t used to be so taboo. As late as the 18th century, death was a prominent aspect of literature, and people of all ages were invited to the deathbeds of family members and friends alike. In the Victorian era, death was so much a culture as was living. Introduction: Despite recent advances, triple-negative breast cancer (TNBC) patients remain at high risk for recurrence and metastasis, which creates the need for innovative therapeutic approaches to improve patient outcomes. Cryoablation is a promising, less invasive alternative to surgical resection, capable of inducing tumor necrosis via freeze/thaw cycles. Necrotic cell death results in increased inflammatory signals and release of preserved tumor antigens, which have the potential to boost the local and systemic anti-tumor immune response. National and international biobanking efforts led to the collection of large and inclusive imaging genetics datasets that enable examination of the contribution of genetic and environmental factors to human brains in illness and health. High-resolution neuroimaging (~104–6 voxels) and genetic (106–8 single nucleotide polymorphic [SNP] variants) data are available in statistically powerful (N = 103–5) epidemiological and disorder-focused samples. Performing imaging genetics analyses at full resolution afforded in these datasets is a formidable computational task even under the assumption of unrelatedness among the subjects. Niclosamide, an FDA-approved anti-parasitic drug, has demonstrated significant potential as a repurposed anti-cancer agent due to its ability to interfere with multiple oncogenic pathways. However, its clinical application has been hindered by poor solubility and bioavailability. Lipid-based nanocarrier systems such as liposomes, solid lipid nanoparticles (SLNs), nanostructured lipid carriers (NLCs), and lipid nanoemulsions (LNE), along with lipid prodrugs, have successfully been employed by researchers to overcome these limitations and improve niclosamide’s pharmacokinetic profile. Genome-wide association studies (GWAS) have become well-powered to detect loci associated with telomere length. However, no prior work has validated genes nominated by GWAS to examine their role in telomere length regulation. We conducted a multi-ancestry meta-analysis of 211,369 individuals and identified five novel association signals. Enrichment analyses of chromatin state and cell-type heritability suggested that blood/immune cells are the most relevant cell type to examine telomere length association signals. We validated specific GWAS associations by overexpressing KBTBD6 or POP5 and demonstrated that both lengthened telomeres. CRISPR/Cas9 deletion of the predicted causal regions in K562 blood cells reduced expression of …

Quadriceps weakness after anterior cruciate ligament reconstruction (ACLR) is a well-known phenomenon, with more persistent quadriceps weakness observed after ACLR with a bone-patellar tendon-bone or quadriceps tendon autograft than with a hamstring tendon autograft. Longstanding quadriceps weakness after ACLR has been associated with suboptimal postoperative outcomes and the progression of radiographic knee osteoarthritis, making the recovery of quadriceps size and strength a key component of ACLR rehabilitation. This research explores the intersection of race, gender, and the litigation of domestic violence cases in Memphis, Tennessee. This is explored through qualitative research that combines a theoretical framework with structured interviews of criminal defense attorneys. Utilizing a sample of 5 attorneys with recent exposure to domestic violence cases, this study aims to reveal whether or not intimate partner violence is litigated differently across racial lines. This is done through analysis of the treatment of victims depending on race, sentencing rates, and perceptions of credibility within the justice system. This thesis examines the long-term effects of parenting styles and attachment experiences on adult psychological well-being, relational competence, academic aspirations, and self-concept. Through qualitative analysis of semi-structured interviews with three participants from the Chattanooga, Tennessee area, the study investigates how family dynamics—framed by Baumrind’s parenting models and Bowlby’s attachment theory—shape developmental trajectories across the lifespan. Findings indicate that authoritative parenting fosters resilience, empathy, and secure attachments, while authoritarian and avoidant parenting styles often lead to relational hesitancy and diminished self-worth. Emphasizing the value of responsive, balanced parenting, this study underscores its role in promoting emotional security and adaptive coping, with …

Background: More than 50% of patients with recurrent pregnancy loss (RPL) are left without explanations for their losses. However, current research suggests that up to 90% of these individuals could receive explanations through evidence-based laboratory testing. Professional guidelines vary in their recommendations for laboratory testing and RPL evaluation, which can contribute to delayed or fragmented care and untreated maternal disease. Inequities in access to genetic testing further complicate the diagnostic landscape of RPL since payor policies often determine which tests are covered, leaving many patients without access due to financial constraints.

The relationship between adverse childhood experiences (ACEs) and mental health later in life has been well-established. However, first-person qualitative narratives describing how ACEs negatively impact mental health of young adults are rare.Objective

Flat-lying carbonate deposits in the Lower Jurassic Navajo Sandstone have long been interpreted as interdune lacustrine deposits. This dissertation critically assesses such inference through the study of an exceptionally well-preserved carbonate deposit in the Trough Canyon area, northwest of Moab, Utah. Thickness measurements along multiple cross-sectional exposures were used to construct an isopach map, approximating the original size and lenticular geometry of the deposit. Dip directions were recorded from foreset strata in cross-bed sets stratigraphically overlying the deposit to infer the approximately coeval direction of siliciclastic transport across the carbonate basin. This thesis investigates the environmental degradation of plastic materials, with a focus on polyethylene terephthalate (PET), through a multi-phase study conducted at the Salton Sea, a closed-basin lake in a semi-arid region of California. The research addresses three core objectives: (1) developing a microplastic (MP) extraction method tailored for clay-rich sediments, (2) characterizing surface degradation of various plastic types, including PET, BoPET, HDPE, LDPE, and EPS, using advanced microscopy and spectroscopy, and (3) examining the long-term weathering of PET through samples collected from dated lake strandlines.

Increased absorption of anthropogenic carbon dioxide (CO2) into the world’s oceans has resulted in ocean acidification (OA). Acidification induces physiological and behavioral responses from marine invertebrates that vary by taxa. Resilience to OA has been noted in crustaceans, though negative impacts are still reported. It has been shown that the American lobster (Homarus americanus) can be physiologically impacted by acidified conditions. The study of iron within cells has advanced significantly since the Fenton reaction was proposed as a mechanism for ferroptosis. Various chemical probes have been developed for examining iron and reactive oxygen species (ROS), both of which are involved in the Fenton reaction. Iron, the most abundant transition metal in the human body, primarily exists as Fe(II) and Fe(III) in the cellular environment. The labile iron pool (LIP) is suspected to predominantly exist as Fe(II) due to endogenous reductants. Venezuelan equine encephalitis virus (VEEV) is an enveloped positive-sense RNA virus in the alphavirus genus of the Togaviridea family, that causes encephalitis in humans and equids. Currently no FDA-approved vaccine or treatment is available for human use, emphasizing the importance of studying host-VEEV interaction. lncRNAs are regulatory RNAs involved in modulation of antiviral pathways during viral infections. Here we report differential lncRNAs expression in primary mouse astrocytes and neurons during non-pathogenic (TC-83) but not pathogenic (TrD) VEEV infection. Accumulating evidence has suggested that the tumor immune microenvironment (TIME) drastically impacts cancer patients’ clinical outcomes, including prognosis and immunotherapy response. However, understanding TIME remains challenging due to its complexity and heterogeneity. In this dissertation, we introduce TimiGP (Tumor Immune Microenvironment Illustration based on Gene Pairs), a computational framework designed to address this challenge. Leveraging single-cell RNA-seq (scRNA-seq) and bulk gene expression data alongside clinical information, TimiGP constructs a cell-cell interaction network that elucidates the relationship between immune cell function and relevant clinical outcomes, such as prognosis and treatment response. WT1 is a zinc finger transcription factor widely expressed in the urogenital system. Human mutations of WT1 lead to pediatric nephroblastoma as well as frequent differences of sex development (DSDs). Previous studies have suggested that WT1 acts as an activator for Amhr2, a necessary component of typical male differentiation.
