Using Living Labs To Engage Communities And Stakeholders In The Development And Knowledge Exchange Of Urban Health And Sanitation Solutions In The Global South, Emmanuel Tsekleves, Roger Pickup, Manoj Roy

DRS Biennial Conference Series

Engaging communities and stakeholders in developing user-centred urban health solutions, whilst linking the research to their own development, pose major challenges for design researchers working in the Global South. In a number of circular sanitation projects in a community school in Ghana, we co-designed and installed an anaerobic digester delivering electricity and sanitation improvements. To enhance impact we developed and pilot-tested a Living and physical Lab design approach. One project focussed on hand hygiene. We introduced students to ‘making the invisible visible’ by visualising microbes from their hands and assessing handwashing effects. Our findings suggest that visualisation of microbes not …

Reimagining Antibiotic Delivery At Home: Designing A Wearable Cool-Infuser, Geoffrey Thompson, Rowan Page, Benjamin Rogers Jun 2024

Reimagining Antibiotic Delivery At Home: Designing A Wearable Cool-Infuser, Geoffrey Thompson, Rowan Page, Benjamin Rogers

DRS Biennial Conference Series

As antimicrobial resistance (AMR) intensifies, hospitals face increased pressure due to the high demand for bed spaces and the associated risks of hospital-acquired infections. Outpatient Antimicrobial Therapy (OPAT) offers a solution by enabling patients to receive life-saving intravenous antibiotic treatments in the community rather than in hospitals. However, temperature-sensitive antibiotics can degrade when exposed to heat, creating design challenges in OPAT settings. Current antibiotic infusion devices focus on the clinical need and outcomes of treatment, leaving room to improve the holistic patient experience of living with the device. Through a human-centred design approach, this project develops a Wearable cool-infuser that …

A Participatory Approach To Healthcare Service Improvement Focused On Staff Behavior Change, Fernando Carvalho, Val Mitchell, Gyuchan Thomas Jun Jun 2024

A Participatory Approach To Healthcare Service Improvement Focused On Staff Behavior Change, Fernando Carvalho, Val Mitchell, Gyuchan Thomas Jun

DRS Biennial Conference Series

Healthcare systems are complex social systems wherein improvements related to staff practice and behavior can be difficult to implement. Knowledge and practice can be uneven between various specializations, hierarchical imbalances limit the degree of agency of different professionals, and evidence-based guidelines may be interpreted or implemented according to context-specific factors. When changes are imposed from a top-down perspective, invaluable insight and know-how from frontline staff are usually left out, making it further difficult (if not altogether impractical) to implement interventions. The current paper presents a case study focused on improving urinary tract infection diagnosis and treatment, in the emergency department …

Pilgrimage In The Covid-19 Era: Uncovering Supply Side Challenges And Opportunities In Media Representations, Reni Polus, Ismail Shaheer, Naresh P. Nayak

International Journal of Religious Tourism and Pilgrimage

Pilgrimage travel, a common practice across major religions, has historically exhibited remarkable resilience to global crises, owing to the unwavering conviction of its followers. However, the COVID-19 pandemic marked a significant departure from this trend, disrupting pilgrimage tourism, unlike any event since World War II. Despite extensive research on the pandemic’s impact on tourism, limited attention has been directed towards understanding its specific effects on pilgrimage travel. This study seeks to address this gap by examining the challenges and opportunities faced by pilgrimage tourism suppliers in the aftermath of COVID-19. To obtain a comprehensive global perspective, we collected and analysed …

The Camino de Santiago is one of the most recognised cultural pilgrimage routes in the world. Notably, its current influence transcends the strictly inspirational and religious aspects, constituting also a socio-economic, cultural, historical, heritage and tourism cornerstone. This paper analyses, from an innovative bibliometric perspective, the influence of the Camino de Santiago on the academic literature in the period from 1980 to the present. For this purpose, two data sources have been used comparatively: manuscripts indexed in Scopus and doctoral theses. The results reveal a growth of its academic relevance in the last decade, especially in indexed publications. The analysis …

Novel Adaptations In Iron Regulation Acquired During Chronic Fungal Cf Infections, Daniel R. Murante Jun 2024

Novel Adaptations In Iron Regulation Acquired During Chronic Fungal Cf Infections, Daniel R. Murante

Dartmouth College Ph.D Dissertations

Chronic fungal infections are highly recalcitrant to treatment; we postulated that as populations persist, increasing genetic diversity is reflected in phenotypic heterogeneity, contributing to treatment inefficacies. The study of evolutionary patterns is underrepresented in chronic fungal infections, and to supplement this body of knowledge, we leveraged isolates acquired from four individuals with chronic fungal-dominated cystic fibrosis infections. We evaluated in-host evolution through a whole-genome sequencing approach, comparing multiple isolates obtained from each subject's sputum and bronchoalveolar lavage fluid. Our analysis found non-synonymous mutations that arose in parallel across the independent infections in the gene MRS4, which encodes a mitochondrial …

Application And Development Of Ceragenins In Medical Device Coatings For Clinical Settings, Elliot E. Sherren Jun 2024

Application And Development Of Ceragenins In Medical Device Coatings For Clinical Settings, Elliot E. Sherren

Theses and Dissertations

Hospital-acquired infections (HAIs) pose a significant and increasing threat to global health. One primary cause of this threat is increasing antibiotic resistance. As traditional antibiotics continue to grow less effective, there is an urgent need for novel antimicrobial strategies. This work explores the potential of ceragenins, also known as cationic steroid antimicrobials (CSAs), as a promising alternative to combat HAIs. Specifically, we investigated potential roles that CSAs can play in the context of multiple medical device coatings in healthcare settings. Ceragenins are synthetic mimic of antimicrobial peptides (AMPs) which exhibit broad-spectrum antimicrobial activity against many common pathogens that have been …

Recent developments in immunotherapy, including immune checkpoint blockade (ICB) and adoptive cell therapy (ACT), have encountered challenges such as immune-related adverse events and resistance, especially in solid tumors. To advance the field, a deeper understanding of the molecular mechanisms behind treatment responses and resistance is essential. However, the lack of functionally characterized immune-related gene sets has limited data-driven immunological research. To address this gap, we adopted non-negative matrix factorization on 83 human bulk RNA sequencing (RNA-seq) datasets and constructed 28 immune-specific gene sets. After rigorous immunologist-led manual annotations and orthogonal validations across immunological contexts and functional omics data, we demonstrated …

Advancing Clinical Microbiology: Applications And Future Of Next-Generation Sequencing, Rob E. Carpenter Jun 2024

Advancing Clinical Microbiology: Applications And Future Of Next-Generation Sequencing, Rob E. Carpenter

Human Resource Development Faculty Publications and Presentations

Next-generation sequencing (NGS) has reformed infectious disease management, including COVID-19. While real-time polymerase chain reaction (PCR) is widely used for rapid pathogen detection, it requires predefined targets. NGS offers an unbiased approach, detecting multiple pathogens simultaneously without prior knowledge. Despite its potential, NGS implementation in clinical settings faces challenges like high costs and technical complexity. NGS platforms like Illumina, Ion Torrent, and Nanopore provide high-throughput sequencing, identifying pathogens and resistance markers. Applications include whole genome sequencing (WGS), metagenomic NGS (mNGS), and targeted NGS (tNGS). Integrating NGS with conventional methods could improve diagnostics, but current evidence is mixed for supporting its …

Individuals with uncontrolled diabetes are highly susceptible to tuberculosis (TB) caused by Mycobacterium tuberculosis (M. tb) infection. Novel treatments for TB are needed to address the increased antibiotic resistance and hepatoxicity. Previous studies showed that the administration of liposomal glutathione (L-GSH) can mitigate oxidative stress, bolster a granulomatous response, and diminish the M. tb burden in the lungs of M. tb-infected mice. Nonetheless, the impact of combining L-GSH with conventional TB treatment (RIF) on the cytokine levels and granuloma formation in the livers of diabetic mice remains unexplored. In this study, we evaluated hepatic cytokine profiles, GSH, and tissue pathologies …
