Paediatrics and Child Health, East Africa
Introduction: Children with moderate or severe wasting are at particularly high risk of recurrent or persistent diarrhoea, nutritional deterioration and death following a diarrhoeal episode. Lactoferrin and lysozyme are nutritional supplements that may reduce the risk of recurrent diarrhoeal episodes and accelerate nutritional recovery by treating or preventing underlying enteric infections and/or improving enteric function.
Methods and analysis: In this factorial, blinded, placebo-controlled randomised trial, we aim to determine the efficacy of lactoferrin and lysozyme supplementation in decreasing diarrhoea incidence and improving nutritional recovery in Kenyan children convalescing from comorbid diarrhoea and wasting. Six hundred children aged 6–24 months with …

Leveraging Machine Learning And Stochastic Programming To Address Vaccine Hesitancy In Public Health Resource Allocation, Hieu Trung Bui
Graduate Theses and Dissertations
Infectious disease outbreaks highlight the urgent need for effective strategies to distribute vaccines and allocate critical healthcare resources to contain the disease and reduce its negative impacts on the population. Managing these allocations is a significant challenge, especially in marginalized communities facing uncertainty in healthcare demand and logistical constraints. This dissertation addresses these challenges by investigating factors that influence dynamic changes in vaccine hesitancy (VH) and its implications for disease spread and healthcare resource demand. It develops optimization models for vaccine distribution and resource allocation under uncertainty, validated with data from the COVID-19 pandemic in the U.S. The first study …
Uncovering The Hidden Diversity Of Antibody Heavy Chains And Their Implications For Autoantibody Mediated Disease., Easton Earl Ford
Uncovering The Hidden Diversity Of Antibody Heavy Chains And Their Implications For Autoantibody Mediated Disease., Easton Earl Ford
Electronic Theses and Dissertations
Understanding the diversity of antibody (IG) molecules at the DNA and RNA level is imperative for understanding immunological processes and disease. Much of the work to uncover IG diversity has been focused on diversity in the variable region of the IG molecule which is crucial for antigen binding. However, the diversity of the constant region responsible for the functions of IG has largely been ignored in the field of immunogenetics. The work presented in this thesis challenges the dogma that the constant region is invariant in terms of genetic diversity. In this thesis we present the development of a long-read …
Molecular Mechanisms Of Exercise-Stimulated Resolution Of Inflammation And Its Dietary Dependence., Ernesto H. Pena Calderin
Molecular Mechanisms Of Exercise-Stimulated Resolution Of Inflammation And Its Dietary Dependence., Ernesto H. Pena Calderin
Electronic Theses and Dissertations
Diet-induced obesity and adipose tissue (AT) inflammation that ensues sets the stage for systemic metabolic inflammation, disrupted insulin signaling, and eventual type 2 diabetes (T2D) development. At the center of this pathology lies a disruption in endogenous inflammation resolution pathways characterized by an imbalance in lipid mediator production (greater abundance of pro-inflammatory to pro-resolving lipids). Additionally, obese states are characterized by adrenergic deficiency, especially in response to physical activity. Inflammatory processes have been experimentally mitigated by either the pharmacological inhibition of pro-inflammatory signals (i.e., Leukotriene B4 (LTB4) and its receptor BLT1, which establishes a potent chemotactic stimulus …

COVID-19 Papers, Posters, and Presentations
Cytotoxic T lymphocytes (CTLs) destroy virally infected cells and are critical for the elimination of viral infections such as those caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Delayed and dysfunctional adaptive immune responses to SARS-CoV-2 are associated with poor outcomes. Treatment with allogeneic SARS-CoV-2-specific CTLs may enhance cellular immunity in high-risk patients providing a safe, direct mechanism of treatment. Thirty high-risk ambulatory patients with COVID-19 were enrolled in a phase 1 trial assessing the safety of third party, SARS-CoV-2-specific CTLs. Twelve interventional patients, 6 of whom were immunocompromised, matched the HLA-A∗02:01 restriction of the CTLs and received …

Characterizing Oxylipins In Human Milk Extracellular Vesicles, Priscila Herrera
Food Science (MS) Theses
Extracellular vesicles (EVs) are non-replicating lipid bilayer membrane-bound particle formed by a parent cell and secreted into the extracellular environment. EVs in human milk are bioavailable and influence physiologic pathways that may contribute to newborn growth and development. There is interest in the oxylipin content of EVs that may contribute to the bioavailability of EVs. This research aims to characterize the oxylipin content of human milk EVs. Centrifugation and precipitation were used to isolate EVs from human milk (n = 20). The Folch method was used to extract lipids, and liquid chromatography-mass spectrometry (LC– MS) was used to qualify and …
Furthering Ascidian Taxonomy Using Molecular Biology, Nicholas Gulnick
Furthering Ascidian Taxonomy Using Molecular Biology, Nicholas Gulnick
Electronic Theses and Dissertations
Ascidians are our closest invertebrate relatives and comprise nearly 3,000 species separated into three orders: Aplousobranchia (most speciose), Stolidobranchia, and Phlebobranchia (least speciose). Ascidians can be classified as either solitary or colonial organisms. Species delimitation using morphological characters alone has had varied results. Well known, widely distributed, morphological species have turned out to be catch all species comprised of several cryptic species. Molecular markers can help mitigate some of the issues presented by strictly using morphological observations, including resolving the status of cryptic species, and accessing the expert knowledge required to identify a species. By incorporating molecular markers and pairing …

Engaging Telepsychology As A Culturally Appropriate Communicative Approach For Mental Health Intervention For Women In Ondo State, Nigeria, Toyosi Motilola Olola
Theses and Dissertations
Background: Assessing the mental health issues among Nigerian women is complex due to cultural norms, stigmatization, and traditional beliefs, which often lead to the concealment of mental health problems. Mental health issues are frequently seen through a spiritual or supernatural lens, exacerbating stigma and preventing proper medical care. There is also a lack of comprehensive data on help-seeking behaviors, with many women relying on religious leaders and traditional healers for support. Telepsychology can solve these communication challenges by offering a confidential and accessible platform that bridges traditional beliefs and modern mental health care. This study explores cultural influences on help-seeking …

Manipulation Of Cellular Proliferating Cell Nuclear Antigen (Pcna) During Herpes Simplex Virus Type 1 Infection, Jessica Packard, Jill A. Dembowski
Electronic Theses and Dissertations
Herpes simplex virus type 1 (HSV-1) is a double stranded DNA virus that replicates within the nucleus of host cells. Using accelerated native isolation of proteins on nascent DNA (aniPOND), cellular proteins were found to associate with HSV-1 replication forks. One host factor of interest is proliferating cell nuclear antigen (PCNA), the DNA sliding clamp that tethers DNA polymerases and DNA damage response proteins to replicating cellular DNA. Although HSV-1 encodes its own DNA polymerase processivity factor (UL42), PCNA plays a key role in HSV-1 infection. We used commercially available PCNA inhibitors and short hairpin RNAs (shRNA) to investigate how …

Electronic Theses and Dissertations
The research described in this dissertation focuses on the investigation of the structure and dynamics of regions of RNA implicated in two diseases: COVID-19 and amyotrophic lateral sclerosis/frontotemporal dementia (ALS/FTD). Both studies sought to create experimentally corroborated models that were used to characterize the structure and dynamics of the C9orf72 repeat expansion and the s2m region in coronaviruses, respectively and provided the foundation for future work. In our work involving the s2m region of SARS-CoV-2, the virus responsible for COVID-19, we determined that the homology modeling approach commonly used to derive atomistic structures when no coordinates are available yields a …
Partial Immunity To Mycoplasma Gallisepticum After An Initial Infection Is Long-Lasting And Reduces Transmission In Flocks Of Canaries, Madeline Sudnick
Partial Immunity To Mycoplasma Gallisepticum After An Initial Infection Is Long-Lasting And Reduces Transmission In Flocks Of Canaries, Madeline Sudnick
Graduate Theses and Dissertations
Pathology during a second infection can be influenced by immunity and an increase in resistance or tolerance of individuals. Population-level differences in the ability to maintain immunity can lead to contrasting survival outcomes between groups and determine how future outbreaks will spread. However, long term wildlife disease surveillance can be challenging due to lack of infrastructure and host-pathogen specific tests. Calculating disease transmission likelihood requires understanding individual-level heterogeneity in pathology and behavior, and there is limited knowledge in understanding how populations in different stages of an epidemic will transmit disease. In Chapter I, we aim to help fill a gap …

The Use Of Fourier-Transformed Infrared Spectroscopy With An Infrared Biotyper To Establish Classifiers For Major Serogroups Of Moraxella Bovis Based On Spectral Profiles, Jordyn Bailey Schommer
Honors Program: Senior Projects (Public)
A classifier tool was created to identify major serogroups of Moraxella bovis based on spectral profiles collected from bacterial cells through the use of Fourier transformed infrared spectroscopy. M. bovis strains were selected from a group of previously sequenced and genotyped isolates, which provided the pilin sequence type for each isolate. The obtained spectroscopic profiles were analyzed through artificial neural networks to create a classifier tool that could identify the PilA type for unknown strains. Once a sufficient spectral library was obtained, the isolates were split into a training and a validation group, each containing spectra from isolates of five …
Does Early Life History Influence A Novel Biomarker For Stress In Dogs And Predict Their Attachment To Their Owners?, Kylie Hughes-Duvall
Does Early Life History Influence A Novel Biomarker For Stress In Dogs And Predict Their Attachment To Their Owners?, Kylie Hughes-Duvall
Psychology Theses, Dissertations, and Student Creative Activity
Previous research suggests dogs with adverse life experiences (e.g., abuse, neglect) have altered HPA-axis activity and attachment styles. Although research on stress biomarkers in dogs has primarily focused on the HPA-axis and cortisol, human research suggests the sympathoadrenal medullary (SAM) pathway is also altered by acute stress and associated with changes in salivary α-amylase. Though α-amylase has been identified in canine saliva, it has yet to be studied as a biomarker for acute stress. Furthermore, limited research exists on the influence of life adversity on dogs’ attachment styles. This study investigated salivary α-amylase as a biomarker of acute stress in …

Antimicrobial And Chemical Characterization Of Cold Atmospheric-Pressure Plasmas, Dalton Albert Miller
Boise State University Theses and Dissertations
Microbial contamination of food processing facilities is a major cause of human disease and economic loss. There are approximately 48,000,000 cases of foodborne illness in the United States annually, leading to 128,000 hospitalizations and 1,300 deaths. The total economic damages are estimated at $36 billion each year, including productive hours, medical expenses, and the disposal of contaminated foodstuffs. Current methods to reduce contamination on food processing surfaces utilize steam or concentrated chemicals (hydrogen peroxide, hypochlorite, strong acids, strong bases) that are hazardous to workers and required large volumes of water to remove prior to resuming contact with food. Ongoing outbreaks …

Canine Parasites In Southern Nevada Urban Dog Parks: Paravec Study, Miklo Azrael A. Alcala
Canine Parasites In Southern Nevada Urban Dog Parks: Paravec Study, Miklo Azrael A. Alcala
UNLV Theses, Dissertations, Professional Papers, and Capstones
While dogs are one of humans’ greatest companions, they also serve as hosts for a multitude of parasites. Urban dog parks serve as environments where the transmission of diseases is likely to occur. Exposure to infective agents, contained within the soil, intermediate hosts (e.g., rodents, rabbits), and feces, can directly affect the health of dogs and their owners. Other factors that can further influence canine health include age, previous diagnoses of parasites/diseases, and migration. Many dog owners visit dog parks to encourage socialization and exercise. Such activities are meant to improve health, but unawareness of microbiological organisms can lead to …

Optimization Of Mass Spectrometry-Based Methods For Low-Input And Spatial Proteomics, Andikan Jones Nwosu
Theses and Dissertations
Eukaryotic cells are highly heterogeneous. These cells are arranged into different compartments, carrying out separate functions and facilitating biological processes. Proteins are the effector biomolecules targeted to subcellular locations that help fulfill specific tasks in living organisms. Spatial proteomics can help unravel molecularly how protein abundance and localization are altered in cells, which is not feasible in traditional bulk-scale proteomics. To achieve this, our lab has developed a miniaturized sample processing platform called nanoPOTS, reduced separation columns' inner diameter to increase ionization efficiency and concentrate analytes for mass spectrometers and optimized data acquisition modes for increasing proteome coverage in spatial …

Neutrophil extracellular traps (NETs) function to control infectious agents as well as to propagate inflammatory response in a variety of disease conditions. DNA damage associated with chromatin decondensation and NACHT domain-leucine-rich repeat-and pyrin domain-containing protein 3 (NLRP3) inflammasome activation have emerged as crucial events in NET formation, but the link between the two processes is unknown. In this study, we demonstrate that poly(ADP-ribose) polymerase-1 (PARP-1), a key DNA repair enzyme, regulates NET formation triggered by NLRP3 inflammasome activation in neutrophils. Activation of mouse neutrophils with canonical NLRP3 stimulants LPS and nigericin induced NET formation, which was significantly abrogated by pharmacological …

OBJECTIVES: Cachexia is a metabolic disorder and comorbidity with cancer and heart failure. The syndrome impacts more than thirty million people worldwide, accounting for 20% of all cancer deaths. In acute myeloid leukemia, somatic mutations of the metabolic enzyme isocitrate dehydrogenase 1 and 2 cause the production of the oncometabolite D2-hydroxyglutarate (D2-HG). Increased production of D2-HG is associated with heart and skeletal muscle atrophy, but the mechanistic links between metabolic and proteomic remodeling remain poorly understood. Therefore, we assessed how oncometabolic stress by D2-HG activates autophagy and drives skeletal muscle loss.
METHODS: We quantified genomic, metabolomic, and proteomic changes in …

Extending and safeguarding tropical forest ecosystems is critical for combating climate change and biodiversity loss. One of its constituents, lianas, is spreading and increasing in abundance on a global scale. This is particularly concerning as lianas negatively impact forests’ carbon fluxes, dynamics, and overall resilience, potentially exacerbating both crises. While possibly linked to climate-change-induced atmospheric CO2 elevation and drought intensification, the reasons behind their increasing abundance remain elusive. Prior research shows distinct physiological differences between lianas and trees, but it is unclear whether these differences confer a demographic advantage to lianas with climate change. Guided by extensive datasets collected …
