Homozygous familial hypercholesterolemia (HoFH) is an ultra-rare inherited condition that affects approximately one in 300,000 people. The disorder is characterized by extremely high, life-threatening levels of low-density lipoprotein (LDL) cholesterol from birth, leading to significant premature cardiovascular morbidity and mortality, if left untreated.

Homozygous familial hypercholesterolemia is severely underdiagnosed and undertreated in the United States (US), despite guidelines recommendations for universal pediatric lipid screening in children aged 9–11. Early diagnosis and adequate treatment are critical in averting premature cardiovascular disease in individuals affected by HoFH. Yet, an unacceptably high number of people living with HoFH remain undiagnosed, misdiagnosed, and/or receive …

Purpose: Structured HCT models addressing planning, transfer, and integration into adult care for adolescents and young adults with childhood-acquired chronic conditions are becoming more prevalent. However, consensus on outcome measures to assess health care transition (HCT) interventions particularly for intellectual and developmental disabilities (IDD) population is lacking. This scoping review identified potential HCT outcome measures for young adults (aged 18-26) with IDD using the Quadruple Aim Framework.

Methods: On August 6, 2021 and April 27, 2023, Medline Ovid, Embase, Web of Science, PsycINFO, and Cochrane databases were searched using the terms "young adult," "intellectual disability," "developmental disability, "cognitive dysfunction," "autism," …

Background: Idiopathic pulmonary fibrosis (IPF) is the result of multiple cycles of epithelial cell injury and fibroblast activation; currently, there is no clear etiology. Increasing evidence suggests that protein metabolism and amino acids play a crucial role in IPF, but the role of D-amino acids is not yet clear. The aim of this study was to identify novel mediators in order to test the hypothesis that D-amino acid oxidase (DAO) plays a significant role in the pathogenesis of IPF.

Methods: We analyzed DAO gene expression in patients with IPF and mice with bleomycin (BLM)-induced lung fibrosis. We performed in vitro …

Background

Despite that stakeholder participation in evidence synthesis could result in more useful outcomes, there are few examples of processes that actively involve them in synthesis work. Techniques are needed that engage diverse stakeholders as equal partners in knowledge co-production. The aims of this paper are to describe an innovative participatory process of synthesising a large body of academic research products and compare the findings of the participatory process against two traditional approaches to synthesis: a rapid review and a structured review.

Methods

First, a rapid synthesis of all research outputs (n = 86) was conducted by researchers with …

Background: As the number of thrombectomy passes increases during endovascular therapy (EVT) for acute stroke, neurointerventionalists must weigh the risks of potential complications against the risk of failing to recanalize the affected vessel. Thus, we investigated the effectiveness and safety of EVT for patients with acute stroke in which ≥5 EVT passes were performed.

Methods: This retrospective cohort study from January 2013 to December 2022 included 31 thrombectomy-capable centers. Patients in whom ≥5 EVT passes were made were included in the analysis. The patients with successful recanalization defined as modified Thrombolysis in Cerebral Infarction ≥2b at 5, 6, 7, or …

Gut Pathobiont Enrichment Observed In A Population Predisposed To Dementia, Type 2 Diabetics Of Mexican Descent Living In South Texas, Lisa M Matz, Nisarg S Shah, Laura Porterfield, Olivia M Stuyck, Michael D Jochum, Rakez Kayed, Giulio Taglialatela, Randall J Urban, Shelly A Buffington Jan 2024

Type 2 diabetes (T2D) is a common forerunner of neurodegeneration and accompanying dementia, including Alzheimer's Disease (AD), yet the mechanisms underlying this comorbidity remain unresolved. Individuals of Mexican descent living in South Texas have increased prevalence of comorbid T2D and early onset AD, despite low incidence of the APOE-ε4 risk variant among the population and an absence of a similar predisposition among relatives residing in Mexico - suggesting a role for environmental factors in coincident T2D and AD susceptibility. We therefore sought to test if differences in gut community structure could be observed in this population prior to any AD …

One-Carbon Metabolites, Fetal Growth, Maternal Oxidative Stress, And Fetal Programming During Early Gestation, Layla King Jan 2024

One-Carbon Metabolites, Fetal Growth, Maternal Oxidative Stress, And Fetal Programming During Early Gestation, Layla King

Animal Sciences

In ruminant production systems, maternal nutrient intake and supplementation practices vary widely throughout the industry. Adequate maternal nutrient supply is critical for fetal development and subsequent offspring performance and health. Therefore, determining strategic supplementation of specific nutrients during critical developmental windows is crucial to fetal growth during early gestation will positively impact beef cattle production systems. In this dissertation, two experiments using beef heifers were conducted to test the hypothesis that differing maternal rate of gain (achieved through differing intake) during the first 63 d of pregnancy would compromise indices of maternal and fetal development; and that strategic nutrient supplementation …

Dietary Impacts On Mitochondrial Respiration Of Gestating Heifers And Fetal Offspring, Kathryn Slavick Jan 2024

Dietary Impacts On Mitochondrial Respiration Of Gestating Heifers And Fetal Offspring, Kathryn Slavick

Animal Sciences

Two projects were conducted to evaluate dietary impacts on mitochondrial respiration of gestating heifers and fetal offspring. Our first hypothesis was that maternal vitamin and mineral supplementation during gestation improves the liver, muscle, and jejunum mitochondrial function of F1 and F2 offspring. In project 1, vitamin and mineral supplementation was provided to the F0 generation of dams from breeding to calving to isolate the effects of nutrition during pregnancy on future generations. Our second hypothesis was that altering limit-fed diets' forage: concentrate ratio would influence mitochondrial respiration in maternal and fetal jejunum and liver. In project 2, replacement heifers were …
