Ataxia telangiectasia and Rad3-related protein (ATR) is a key DNA damage response protein that facilitates DNA damage repair and regulates cell cycle progression. As such, ATR is an important component of the cellular response to radiation, particularly in cancer cells, which show altered DNA damage response and aberrant cell cycle checkpoints. Therefore, ATR's pharmacological inhibition could be an effective radiosensitization strategy to improve radiotherapy. We assessed the ability of an ATR inhibitor, AZD6738, to sensitize cancer cell lines of various histologic types to photon and proton radiotherapy. We found that radiosensitization took place through persistent DNA damage and abrogated G2 …

Staphylococcus aureus remains a leading global cause of bacterial infection-associated mortality and has eluded prior vaccine development efforts. S. aureus α-toxin (Hla) is an essential virulence factor in disease, impairing the T cell response to infection. The anti-Hla antibody response is a correlate of human protective immunity. Here we observe that this response is limited early in human life and design a vaccine strategy to elicit immune protection against Hla in a neonatal mice. By targeted disruption of the interaction of Hla with its receptor ADAM10, we identify a vaccine antigen (Hla

Hypoxia-Inducible Factor-1Α Inhibition In Renal Cell Carcinoma, Kinsey Morey, Santosh Nimkar, Samir Dalia Oct 2024

Hypoxia-Inducible Factor-1Α Inhibition In Renal Cell Carcinoma, Kinsey Morey, Santosh Nimkar, Samir Dalia

Student Publications

The tumorigenesis of clear cell renal cell carcinoma (ccRCC), an aggressive variant of renal cell carcinoma (RCC), is primarily attributable to the mutational inactivation of the Von Hippel–Lindau (VHL) gene. This mutation causes VHL syndrome, which is associated with tumor growth in various body parts, including the brain, spinal cord, eyes, adrenal glands, pancreas, kidney, and reproductive tract. RCC is the leading cause of death in patients with VHL syndrome. The VHL gene acts as a tumor suppressor that prevents the proliferation of various oncogenes by controlling the hypoxia-inducible factor (HIF). The HIF pathway is directly linked to the control …

Assessing Benthic Recovery Below The United Kingdom's First Large-Scale, Offshore, Longline Mussel Farm, Llucia Mascorda-Cabre, Philip Hosegood, Martin Attrill, Emma Sheehan Oct 2024

Assessing Benthic Recovery Below The United Kingdom's First Large-Scale, Offshore, Longline Mussel Farm, Llucia Mascorda-Cabre, Philip Hosegood, Martin Attrill, Emma Sheehan

School of Biological and Marine Sciences

The recent expansion of the aquaculture industry into the offshore marine environment is perceived to have a comparatively low environmental impact coupled with a high growth potential as a sustainable, large-scale source of healthy protein. This study assessed changes in sediment characteristics and infauna assemblages following the development of the first large-scale offshore longline mussel farm in the United Kingdom. By evaluating the effects of the farm on the sediment ecosystem and by studying infaunal sediment environmental parameters along a distance gradient away from the farm, the first before-after control-impact (BACI) study of an offshore shellfish farm is presented. Sediment …

The Role Of Group 2 Innate Lymphoid Cells In Restoring Lung Health And Chitinase Function After Epithelial Injury, Haerin Jung Oct 2024

The Role Of Group 2 Innate Lymphoid Cells In Restoring Lung Health And Chitinase Function After Epithelial Injury, Haerin Jung

Arts & Sciences Graduate Student Theses and Dissertations

Alveolar epithelial cells provide oxygen transport and act as a first line of defense against pathogens and environmental triggers that disturb tissue homeostasis. However, in the context of epithelial injury, fibrotic and inflammatory lung diseases develop where regenerative responses may become dysregulated or recurrently engaged. One cause of lung injury is infection with cytopathic respiratory viruses such as influenza and SARS-CoV-2 viruses, which induce lung inflammation, tissue damage, and epithelial cell injury and loss. Restoration of barrier integrity after lung injury is critical for re-establishing healthy lung function but the underlying mechanisms are unclear. In the steady state, lung epithelial …

Il-7rα On Cd4+ T Cells Is Required For Their Survival And The Pathogenesis Of Experimental Autoimmune Encephalomyelitis, Gholamreza Azizi, Bram Van Den Broek, Larissa Ishikawa, Hamed Naziri, Reza Yazdani, Guang-Xian Zhang, Bogoljub Ciric, Mohamad Rostami Oct 2024

Il-7rα On Cd4+ T Cells Is Required For Their Survival And The Pathogenesis Of Experimental Autoimmune Encephalomyelitis, Gholamreza Azizi, Bram Van Den Broek, Larissa Ishikawa, Hamed Naziri, Reza Yazdani, Guang-Xian Zhang, Bogoljub Ciric, Mohamad Rostami

Department of Neurology Faculty Papers

BACKGROUND: The IL-7 receptor alpha (IL-7Rα) binds both IL-7 and thymic stromal lymphopoietin (TSLP). IL-7Rα is essential for the development and survival of naive CD4+ T cells and their differentiation to effector/memory CD4+ T cells. Mice lacking IL-7Rα have severe lymphopenia and are resistant to experimental autoimmune encephalomyelitis (EAE), a model for multiple sclerosis. However, it has been reported that IL-7Rα on peripheral CD4+ T cells is disposable for their maintenance and EAE pathogenesis, which does not align with the body of knowledge on the role of IL-7Rα in the biology of CD4+ T cells. Given that a definitive study …

Despite showing the greatest primate diversity on the planet, genomic studies on Amazonian primates show very little representation in the literature. With 48 geolocalized high coverage whole genomes from wild uakari monkeys, we present the first population-level study on platyrrhines using whole genome data. In a very restricted range of the Amazon rainforest, eight uakari species (Cacajao genus) have been described and categorized into the bald and black uakari groups, based on phenotypic and ecological differences. Despite a slight habitat overlap, we show that posterior to their split 0.92 Mya, bald and black uakaris have remained independent, without gene flow. …

Aspect-Ratio-Dependent Heat Transport By Baroclinic Acoustic Streaming, Jacques Abdul Massih, Remil Mushthaq, Guillaume Michel, Gregory P. Chini Oct 2024

Aspect-Ratio-Dependent Heat Transport By Baroclinic Acoustic Streaming, Jacques Abdul Massih, Remil Mushthaq, Guillaume Michel, Gregory P. Chini

Faculty Publications

Standing acoustic waves have been known to generate Eulerian time-mean ‘streaming’ flows at least since the seminal investigation of Lord Rayleigh in the 1880s. Nevertheless, a recent body of numerical and experimental evidence has shown that inhomogeneities in the ambient density distribution lead to much faster flows than arise in classical Rayleigh streaming. The emergence of these unusually strong flows creates new opportunities to enhance heat transfer in systems in which convective cooling cannot otherwise be easily achieved. To assess this possibility, a theoretical study of acoustic streaming in an ideal gas confined in a rectangular channel with top and …

Biogenic Reef Creation And Biodiversity Enhancement By An Offshore Longline Mussel Farm, Llucia Mascorda-Cabre, Philip Hosegood, Martin Attrill, Emma Sheehan Oct 2024

Biogenic Reef Creation And Biodiversity Enhancement By An Offshore Longline Mussel Farm, Llucia Mascorda-Cabre, Philip Hosegood, Martin Attrill, Emma Sheehan

School of Biological and Marine Sciences

As the mussel farming industry moves offshore, there is a need to fill evidence gaps on its ecological impacts. Over eight years, this research presents the results of a long-term monitoring study of the UK’s first large scale offshore mussel farm following a BACI (Before-After Control-Impact) approach to evaluate the temporal and spatial effects of the farm on benthic habitat and epibenthic species assemblages. Situated in historical fishing ground, the farm is located on a degraded seabed ecosystem dominated by soft sediment. After seven years of farming, damaged seabed increased its complexity, with biogenic reef being created (30 % increase). …

2020-Current year OA Pubs

In the Spring of 2020, the United States of America (USA) deployed COVID-19 convalescent plasma (CCP) to treat hospitalized patients. Over 500,000 patients were treated with CCP during the first year of the pandemic. In this study, we estimated the number of actual inpatient lives saved by CCP treatment in the United States of America based on CCP weekly use, weekly national mortality data, and CCP mortality reduction data from meta-analyses of randomized controlled trials and real-world data. We also estimate the potential number of lives saved if CCP had been deployed for 100% of hospitalized patients or used in …

PANDION: The Osprey Journal of Research and Ideas

Artist Statement

Latticed Stinkhorn is a watercolor piece originally created for the special gallery of the 2023 Scientific Illustration course at the University of North Florida. It was inspired by my enduring fascination with the world of fungi, particularly specimens of the family Phallaceae. This watercolor piece offers a simple perspective of Clathrus ruber, a species commonly referred to as Latticed Stinkhorn. Like many members of the family, the reproductive fruiting body first surfaces encased in a cream-colored soft shell with a texture comparable to a leathery reptile egg. As the body ages, the initial shell disintegrates to reveal a …

Artist Statement

The Moon Jelly is a piece I created as a part of the Scientific Illustration course I participated in during the summer semester of 2023. This piece was inspired by the livestreams of the Monterey Bay Aquarium that film these moon jellies (Aurelia aurita) in their tank. The Moon Jelly is an ink drawing created as part of a collection focused on Monterey Bay Aquarium animals. Through this artwork I hoped to draw attention to smaller marine animals that may otherwise be overlooked even though they provide a vital role in the ecosystem. Moon jellyfish are important marine …

Hepatic Targets To Control Blood Glucose And Potentially Aid In The Treatment Of Diabetes, Rena A. Velissarios Oct 2024

Hepatic Targets To Control Blood Glucose And Potentially Aid In The Treatment Of Diabetes, Rena A. Velissarios

PANDION: The Osprey Journal of Research and Ideas

Maintaining glucose homeostasis is extremely important to remain in good health and prevent disease. This homeostasis is achieved via the regulation of hepatic metabolic pathways like gluconeogenesis and glycogenesis by insulin, which is expressed during high blood glucose. Insulin is responsible for facilitating the uptake of glucose into other tissues from the blood, as well as signaling whether to stimulate or slow glucose metabolism in the liver. Hyperglycemia is one of the main effects of diabetes, a disease characterized by the body’s inability to produce or respond to insulin. Without insulin, hepatic glucose output continues and blood glucose accumulates, causing …

The Short Allele Of The Serotonin Transporter Gene (Slc6a4) Increases Predisposition To Anxiety And Negative Emotional States, Gabriella Hitti Oct 2024

The Short Allele Of The Serotonin Transporter Gene (Slc6a4) Increases Predisposition To Anxiety And Negative Emotional States, Gabriella Hitti

PANDION: The Osprey Journal of Research and Ideas

Mental disorders, such as anxiety and mood disorders, have long been a focus of psychological research, and increasing evidence points to a genetic basis for their occurrence. Polymorphisms of the serotonin transporter (5-HTT) gene (SLC6A4) may predispose an individual to anxiety-related symptoms. SLC6A4 has two common alleles: the short (s) allele, which results in less 5-HTT protein production, and the long (l) allele, which results in more 5-HTT. Integrated findings from psychometric evaluations, behavioral animal models, and biological assessments establish a link between the s allele of SLC6A4 and heightened anxiety phenotypes. The allele’s influence on serotonin levels and brain …

A Behavioral Analysis Of Brisby, The Jacksonville Zoo And Gardens’ Double Wattled Southern Cassowary, Gabriel J. Springer Oct 2024

A Behavioral Analysis Of Brisby, The Jacksonville Zoo And Gardens’ Double Wattled Southern Cassowary, Gabriel J. Springer

PANDION: The Osprey Journal of Research and Ideas

Previous studies have demonstrated the uniqueness and importance of wild type Southern Cassowary behaviors in relation to their rainforest ecosystem. The primary goal of this observational study was to determine if the behavior exhibited by a Southern Cassowary residing at the Jacksonville Zoo and Gardens was comparative to the observed behavior of wild Southern Cassowaries. The secondary goal was to observe individual behavioral traits of the focus individual, Brisby. It was hypothesized that Southern Cassowaries residing in captivity would still exhibit stereotypical behaviors similar to those residing in native rainforest habitats and the documented foraging behavior that supports the ecological …

Indigenous Peoples And Environmental Justice: Recognition, Rights, And Representation Of Indigenous Peoples In Bangladesh's Environmental Regulatory Framework, Nasrin Akter Oct 2024

Indigenous Peoples And Environmental Justice: Recognition, Rights, And Representation Of Indigenous Peoples In Bangladesh's Environmental Regulatory Framework, Nasrin Akter

LLM Theses

Using an Environmental Justice theoretical lens, this thesis explores the extent to which the environmental legislative landscape of Bangladesh has ensured environmental justice for Indigenous peoples. In doing so, the thesis concentrates on three main areas: recognition, rights of Indigenous people under the environmental legal framework as well as their representation in the environmental decision making bodies. The research evaluates Bangladesh environmental laws against standards elaborated in the environmental justice literature on each of these three areas. The research employs a combination of legal and policy analysis, as well as a critical analytical approach as part of its methodology. This …
