Lupins are a promising source of protein that can complement soybean-based proteins in food and animal feed. Narrow-leafed lupin (Lupinus angustifolius) is the predominant lupin species cultivated in Australia and Europe. Consumption of lupin products can lead to toxicity in both humans and animals, primarily due to the presence of naturally occurring alkaloid compounds. The aim of the present research was to examine the alkaloid content of commercial narrow-leafed lupin grown in Australia and compare this to wild accessions. Mouse mammary tumor virus (MMTV) is a betaretrovirus that induces breast cancer and sporadically, T-cell lymphomas in mice. MMTV serves as a pertinent model for elucidating the mechanism(s) of oncogenesis and the genetics associated with the development of mammary tumors. Being a rodent retrovirus, MMTV is phylogenetically distant from human and other primate retroviruses; thus, positioning it ideally as a potential vector of choice for human gene therapy. This may circumvent safety concerns such as recombination with other related endogenous human retroviruses. Vertebrate pollination has evolved in many plant lineages, with pollination by birds the most prevalent strategy. However, in Banksia (Proteaceae) many species are visited by a combination of nectarivorous birds, non-flying mammals (NFMs), and insects, though the relative importance of these pollen vectors for reproduction remains poorly understood. Here, we investigate the contribution to fruit set of birds, NFMs, and insects in Banksia catoglypta—a heathland shrub from south-west Australia. This species flowers in winter and produces large, golden-yellow inflorescences that are typically presented near the ground and consist of rigid, strongly scented flowers with a long stigma-nectary distance—suggesting pollination by …

Background: The role of the glymphatic system in multiple sclerosis (MS)-related disability remains underexplored. Diffusion-tensor image analysis along the perivascular space (DTI-ALPS) offers a non-invasive method to assess glymphatic function.
Objective: To evaluate glymphatic function in MS patients with lower and higher disability.
Methods: This study included 118 MS patients who underwent structural, diffusion-weighted imaging, and clinical assessment. The participants were divided into lower (MS-L, n = 57) and higher disability (MS-H, n = 61) subgroups. Brain parenchymal fraction (BPF), lesion load (LL), and DTI-ALPS index were measured. The ability to accelerate and attain high velocities is essential for both individual and team sport athletes. The purpose of this explorative study was to retrospectively analyze the ground reaction force using Statistical Parametric Mapping (SPM) vector field analysis, as traditional scalar analyses often fail to acknowledge the interdependence of force vector components. The ground reaction force vector and the scalar components (i.e., vertical and horizontal force) were analyzed for 28 male sprinters and 24 male soccer players at 8.0 and 8.5 m·s−1, and between 85%, 90%, 95%, and 100% of their maximum velocity. Understanding the mechanisms involved in whole body glucose regulation is key for the discovery of new treatments for type 2 diabetes (T2D). Historically, glucose regulation was largely focused on responses to insulin and glucagon. Impacts of incretin-based therapies, and importance of muscle mass, are also highly relevant. Recently, bone was recognized as an endocrine organ, with several bone proteins, known as osteokines, implicated in glucose metabolism through their effects on the liver, skeletal muscle, and adipose tissue. Drones have emerged as a powerful tool in animal detection, significantly advancing wildlife monitoring, conservation, and management by capturing high-resolution, real-time imagery over areas often inaccessible or challenging for human observers to reach. However, manual analysis of drone imagery for animal detection is labour-intensive and time-consuming. The application of deep learning methods, particularly convolutional neural networks, in automating animal detection from drone imagery has the potential to revolutionise wildlife monitoring, conservation, and management protocols. Foodborne pathogens of Bacillus cereus (B. cereus), non-STEC Escherichia coli (non-STEC E. coli) and Staphylococcus aureus (S. aureus) are currently non-notifiable in Australia unless attributed to a food poisoning outbreak. Due to the lack of data around individual cases and isolations in foods, any changes in prevalence may go undetected. This report explores the dynamics of community philanthropy and family philanthropy in Mexico in order to inform efforts to strengthen the connection between the two. In terms of community philanthropy, our primary focus is upon Mexico’s community foundation model, which is the most institutionalized form of community philanthropy and a uniquely important actor in the philanthropic ecosystem. Wealthy families and family businesses in Mexico are known to have strong local ties and to prioritize place-based development, and Mexico has more community foundations than any other Latin American country. There is growing evidence that reef-building corals can acclimate to novel and challenging thermal conditions. However, potential trade-offs that accompany acclimation remain largely unexplored. We investigated physiological trade-offs in colonies of a globally abundant coral species (Pocillopora acuta) that were acclimated ex situ to an elevated temperature of 31 °C (i.e., 1 °C above their bleaching threshold) for six years. By comparing them to conspecifics maintained at a cooler temperature, we found that the energy storage of corals was prioritized over skeletal growth at the elevated temperature. Since the onset of the COVID-19 pandemic, a variety of diagnostic approaches, including RT-qPCR, RAPID, and LFA, have been adopted, with RT-qPCR emerging as the gold standard. However, a significant challenge in COVID-19 diagnostics is the wide range of symptoms presented by patients, necessitating early and accurate diagnosis for effective management. Although RT-qPCR is a precise molecular technique, it is not immune to false-negative results. In contrast, CRISPR-based detection methods for SARS-CoV-2 offer several advantages: they are cost-effective, time-efficient, highly sensitive, and specific, and they do not require sophisticated instruments. This qualitative study explored the experiences of African American educational leaders in navigating racial microaggressions in the workplace, specifically within K-12 educational settings. Using Taxonomy of Racial Microaggressions and emotion regulation theory as frameworks, this research examines how these leaders manage, respond to, and cope with various forms of racial bias while maintaining their professional identities. Data were collected through in-depth interviews with African American leaders, and thematic analysis was used to identify recurring patterns of microaggressions and corresponding emotion regulation strategies. Dispositional optimism has been shown to be associated with better sleep. However, most existing studies rely on subjective assessments of sleep, which may not align with objective assessments of sleep. Additionally, research investigating the mechanisms underlying the association between optimism and sleep is lacking. Moreover, the confounding role of possible content overlap in measures relevant to the constructs of interest has been neglected. This paper reviews, compares, and critically evaluates two broad groups of sustainable agriculture models: “sustainable agriculture” and “sustainable agrifood systems”. The “sustainable agriculture” models—comprising organic farming, regenerative agriculture, climate-smart agriculture, carbon-capture agriculture, and nature-based solutions—focus primarily on improving ecological sustainability through farm-level practices. These models emphasize reducing external, industrial inputs, enhancing biodiversity, and promoting climate resilience, relying on technological and market-based solutions to address environmental concerns. On the other hand, the “sustainable agrifood systems” models—agroecology, alternative food networks, and permaculture—offer more ambitious visions of systemic transformation. Pathogenic bacteria infection, especially Clostridium perfringens (C. perfringens), markedly threatened the health of animals, and further caused huge economic loss. In this study, Bacillus licheniformis HJ0135 (BL) was used. Oxford cup bacteriostatic test and inhibitory rate test were conducted to evaluate the antibacterial ability of BL. Results showed the strongest inhibitory role of BL on C. perfringens (P < 0.05). Afterwards, 540 one-day-old yellow-feather broilers (32.7 ± 0.2 g) were randomly allocated into 3 groups, including CON group (basal diet), CP group (basal diet + 1 × 109 CFU C. perfringens in gavage), and BL + CP group (basal diet containing 7.5 × 106 CFU/g BL + 1 × 109 CFU C. perfringens in gavage). At d 70, broilers in the CP and BL + CP groups were treated with C. perfringens by continuously oral administration for 5 d. The experiment lasted for 75 d. The serum, immune organs, jejunal mucosa, and cecal contents were collected for analysis. In vivo experiment showed that BL supplementation markedly improved (P < 0.05) BW, ADG, thymus index, serum immunoglobins and antioxidases, reduced feed conversion ratio (FCR) and serum pro-inflammatory cytokines of C. perfringens-infected broilers. Furthermore, the increased jejunal injury and levels of pro-inflammatory cytokines, decreased gene expressions of tight junction proteins in the jejunal mucosa were significantly alleviated (P < 0.05) by BL. More importantly, the activation of NOD-, LRR- and pyrin domain-containing protein 3 (NLRP3) inflammasome was inhibited (P < 0.05) by BL to further attenuate jejunal damage. Besides, BL supplementation markedly increased (P < 0.05) the cecal isobutyric acid and isovaleric acid. Microbial analysis showed that BL changed the composition and relative abundances of microbiota in the cecal contents (P < 0.05), especially the short chain fatty acids (SCFAs)-producing bacteria including Eubacterium_coprostanoligenes_group, Megamonas, Faecalibacterium, and Lactobacillus, which further protected against C. perfringens-induced jejunal inflammation in broilers. Our study laid a theoretical basis for the application of probiotics in lessening C. perfringens-related diseases in poultry farming.

Background aims: Hu8F4 is a T-cell receptor-like antibody with high affinity for the leukemia-associated antigen PR1/HLA-A2 epitope. Adapted into a chimeric antigen receptor (CAR) format, Hu8F4-CAR is composed of the Hu8F4 single-chain variable fragment, the human IgG1 CH2CH3 extracellular spacer domain, a human CD28 costimulatory domain and the human CD3ζ signaling domain. We have demonstrated high efficacy of Hu8F4-CAR-T cells against PR1/HLA-A2-expressing cell lines and leukemic blasts from patients with acute myeloid leukemia in vitro. Purpose of review: This review aims to synthesize available literature on uterine-conserving treatment options for atypical endometrial hyperplasia and grade 1 endometrial carcinoma while highlighting remaining unanswered questions.
Recent findings: The need for uterine-conserving treatment options for atypical endometrial hyperplasia and grade 1 endometrial carcinoma is growing with the increasing number of cases in younger patients or those who cannot undergo surgery. Calcineurin is a highly conserved calcium/calmodulin-dependent serine/threonine protein phosphatase with diverse functions. Inhibition of calcineurin is known to enhance the lifespan of Caenorhabditis elegans through multiple signaling pathways. Aiming to study the role of calcineurin in regulating innate immunity, we discover that calcineurin is required for the rhythmic defecation motor program (DMP) in C. elegans. Calcineurin inhibition leads to defects in the DMP, resulting in intestinal bloating, rapid colonization of the gut by bacteria, and increased susceptibility to bacterial infection. We demonstrate that intestinal bloating caused by calcineurin inhibition mimics the effects of calorie restriction, resulting in enhanced lifespan. …

Acute respiratory distress syndrome (ARDS) is characterized by bilateral chest infiltration and acute hypoxic respiratory failure. ARDS carries significant morbidity and mortality despite advancements in medical management, calling for the development of novel therapeutic targets. Hypoxia-inducible factor (HIF) is a heterodimeric protein involved in various essential pathways, including metabolic reprogramming, immune modulation, angiogenesis and cell cycle regulation. HIF is routinely degraded in homeostasis conditions via the prolyl hydroxylase domain/von Hippel-Lindau protein pathway. However, HIF is stabilized in ARDS via various mechanisms (oxygen-dependent and independent) as an endogenous protective pathway and plays multifaceted roles in different cell populations. BACKGROUND: Motor mapping allows for determining the macroscopic organization of motor circuits and corresponding motor movement representations on the cortex. Techniques such as intracortical microstimulation (ICMS) are robust, but can be time consuming and invasive, making them non-ideal for cortex-wide mapping or longitudinal studies. In contrast, optogenetic motor mapping offers a rapid and minimally invasive technique, enabling mapping with high spatiotemporal resolution. However, motor mapping has seen limited use in tracking 3-dimensonal, multi-limb movements in awake animals. This gap has left open questions regarding the underlying organizational principles of motor control of coordinated, ethologically-relevant movements involving multiple limbs.

Bourbon virus is a tickborne virus that can cause human disease. Cases have been reported in Kansas, Oklahoma, and Missouri, USA. We identified Bourbon virus-specific neutralizing antibodies in patients from North Carolina. Bourbon virus infections are likely more common than previously thought, highlighting the need for improved diagnostics and surveillance.

Countless pages have been devoted to the question of why everyone should pay tax, yet its opposite has gone largely unnoticed: why should some people and organizations not pay tax? Our tax system exempts from ordinary income taxation a wide and diverse array of people and organizations engaged in significant economic activity—from parents providing childcare services for their family to consular activities and charities operating animal shelters—seemingly without a convincing explanation. Immunotherapy has developed into an important modality of modern cancer treatment. Unfortunately, checkpoint inhibitor immunotherapies are currently delivered systemically and require frequent administration, which can result in toxicity and severe, sometimes fatal, adverse events. Localized delivery of immunomodulators for oral cancer and oral potentially malignant disorders offers the promise of maximum therapeutic potential and reduced systemic adverse effects. This review will discuss the limitations of current standard-of-care systemic therapies and highlight research advances in localized, intratumoral delivery platforms for immunotherapy for oral cancer and oral potentially malignant disorders.

Plant-waste-derived bioactive compounds have generated growing interest recently. In this context, this project examined the phenolic content, antioxidant capacity, antimicrobial activity, and antibiofilm capability of the seed extract of Phoenix dactylifera. Six date seed varieties (Fardh, Sukarri, Negal, Majdool, Zahidi, and Khadrawy) were studied. In this study, a novel ultrasound-assisted extraction technique is compared with the conventional extraction method (maceration using 80% ethanol), taking into account both the quality and quantity of polyphenols. All agricultural plant production requires investment of water, energy, and land, especially in the Gulf Region, where environmental conditions are not ideally suitable for crop farming. Currently, less than 40% of agricultural plant biomass production constitutes food, which greatly limits the overall feasibility of food production systems worldwide. Crop residues constitute over half of global agricultural biomass, and food waste and biomass from public parks and gardens further add to the vast pool of non-edible plant material. This study proposes a novel approach to crop yield prediction and risk assessment using neutrosophic logic and least squares regression. By integrating these methods, we aim to improve accuracy in predicting crop losses under uncertain conditions.
