Spinal motor neuron (MN) persistent inward currents (PIC) increase and sustain the MN output. PIC is amplified by descending monoamines and reduced by afferent inhibition. There is strong coupling between plantar cutaneous sensory input and lower limb MNs. Given that monoaminergic drive is altered with postures and motor tasks, our purpose was to assess posture-dependent plantar cutaneous sensory modulation of tibialis anterior (TA) MNs. We hypothesize that: 1) a supine position will suppress PIC, 2) removing the load from the bottom of the heel will inflate PIC, and 3) cutaneous stimulation of the plantar aspect of the heel will reduce …

Bacterial human pathogens are among the leading causes of death around the world, especially in low income and developing countries. One important element in a bacterium’s ability to cause disease are genes that directly contribute to pathogenicity called virulence factors. A second significant aspect are antimicrobial resistance genes which allow microorganisms to persist in the presence of antimicrobial agents. In this project I aimed to determine if Salmonella isolated from different sources differed in pathogenicity profiles based on the complement of genes identified through genomic analysis. Gadolinium (Gd) is a rare earth element (REE), primarily used in contrast agents for magnetic resonance imaging (MRI), with additional applications in metallurgy, phosphors, neutron shielding, and other niche applications. Both Gd and Gd-based contrast agents (GBCAs, the form used for MRIs) have been detected downstream of wastewater effluent discharges and there is currently little research into the risk this poses to aquatic ecosystems. Located on both sides of the Polish-Belarusian border in north-eastern Poland, the Białowieża forest is the largest remaining remnant of the vast primeval forest that once covered most of Europe. It is also home to Europe’s biggest land animal—the European bison. Over the past century Polish governments, as well as modern-day Polish and European environmental groups, have promoted the forest as the “last European wilderness.” Even today, the public regularly protests and decries news of human encroachment on the forest. In Canada, annual trout production is valued at over $60M with approximately 10,000 tonnes of trout produced per year. Rainbow trout (Oncorhynchus mykiss) are one of the most commonly farmed fresh-water fish in Canada. The high-density farming practices involved in aquaculture increase rainbow trout susceptibility to aquatic viruses such as infectious pancreatic necrosis virus (IPNV). The innate antiviral immune response in rainbow trout is poorly characterized, and the functions of numerous interferon stimulated genes (ISG) in rainbow trout remain unclear. Invasive sea lamprey (Petromyzon marinus) contributed to massive declines in fisheries in the Laurentian Great Lakes due to their parasitism of large-bodied salmonid fishes in the 20th century. Chemical control measures, which involve the application of 3-trifluoromethyl-4-nitrophenol (TFM) into rivers containing larval sea lamprey, were successfully implemented approximately 60 years ago to control populations. Recently, however, it was discovered that TFM tolerance increases in sea lamprey with a rise in temperature for reasons which are incompletely understood. Nor is it understood how sea lamprey will respond to warmer temperatures due to climate change, which could affect their …

Abstract
Ionoregulation and nitrogenous waste (N-waste) metabolism in fishes has been extensively documented in teleosts and elasmobranchs, but less so in lampreys (Petromizontiformes). All lamprey species undergo metamorphosis which drastically changes their physiology and modes of feeding. However, differences in ionoregulatory mechanisms due to anadromous versus non anadromous life history strategies emerge following metamorphosis in different lamprey species. Furthermore, some species, such as anadromous juvenile sea lamprey (Petromyzon marinus), are parasitic and migrate to seawater (SW) to ingest blood protein from host fishes, while juveniles of freshwater (FW), non-parasitic species, such as the American brook lamprey …

Fire blight is known as a disease of Rosaceae and it displays infection symptoms in about 200 species in 40 rosaceous genera. The most economically important crops that are affected by fire blight are apples and pears. Fire blight can cause severe outbreaks and a single fire blight outbreak can disrupt orchard production for years. The bacterium that causes this disease is indigenous in North America, and according to Statistics Canada, economic losses in apples and pears were valued at an estimated $4 million in 2005, which is approximately 5% of the total production. Independent, online microcelebrity influencers are increasingly many people’s source of news. A popular group of these influencers, called the Alternative Influence Network (AIN) has come under scrutiny in recent years for collaborating with radical, far-right political actors to advance particular viewpoints (Lewis, 2018). While those within the AIN claim these outputs represent their freedom of speech, some researchers have voiced concern around the possibility of these influencers proliferating or even popularizing radical political ideals. Snakes are often considered non-social animals. However, this is mostly an untested assumption and the limited research that has been done has shown that snakes interact socially when given the opportunity. Here, the complexity of snake social interactions was explored through a social cognition and personality lens. Group and individual behavioural assays were used to explore flexibility and consistency in social interactions. The results provide evidence that elucidate the function, development, and evolution of these behaviours in snakes. The aquaculture industry is a big part of the global economy, supplying food and supporting many livelihoods, yet it faces formidable challenges from diverse aquatic pathogens, especially viruses. Effective antiviral treatments are lacking for both humans and economically significant farmed fish like rainbow trout (Oncorhynchus mykiss). When viruses invade host cells, pattern recognition receptors detect viral components, initiating intracellular signaling and type I interferon (IFN-I) production. IFN-I triggers interferon-stimulated genes (ISGs) that establish an antiviral state in infected and nearby cells, curbing viral spread. Indigenous people hold a respectful and reciprocal relationship with the land, which is central to their cultures and societies. However, the appropriation of traditional lands lies at the heart of settler colonialism. This has resulted in a prolonged struggle by Indigenous people and their allies against land dispossession for settler occupation and resource extraction that has shaped historical and contemporary Indigenous land defence movements. This research explores the experiences and challenges faced by Indigenous people who engage in land defence in Canada, focusing on their strategies, resilience and motivations in protecting the land. The settler-caused climate crisis poses severe global threats, driving action from youth and emerging adults. Prior to COVID-19 lockdowns, youth-led climate organizations were highly active, but social distancing has hindered youth engagement and exacerbated psychological distress among marginalized emerging adult climate justice activists. Previous research indicates that well-being and social participation are interrelated. Guided by the critical transformative paradigm, this qualitative research examines the effects of care webs on the well-being and sustained engagement of marginalized emerging adults in climate justice activism. The global economy is increasingly being weaponized. Citing security concerns from traditional defence to economic competitiveness, health emergency, and climate crisis, states are turning to sanctions, tariffs, export controls, investment screening, and subsidies. But while economic statecraft is becoming common, rules remain scarce. Questions about notice, duration, proportionality, harm minimization, compensation, retaliation, and/or rebalancing lack clear answers and seem almost theoretical. Once, we might have hoped the World Trade Organization (WTO) would play a role in developing such rules. They say a picture is worth a thousand words—and charts, drawings, diagrams, computer animations, and even tangible items are utilized at trial in virtually every case tried in the federal and state court systems. Litigants have come to depend heavily upon such aids to engage visual learners in the jury box and to present a compelling narrative. And the creative use of trial aids has only increased with the rapid technological advancements of recent decades. Prevalence And Determinants Of Diarrhoea And Acute Respiratory Infections Among Children Aged Under Five Years In West Africa: Evidence From Demographic And Health Surveys, Derrick Nyantakyi Owusu, Henry Ofori Duah, Duah Dwomoh, Yakubu Alhassan

Background Diarrhoea and pneumonia are the leading causes of morbidity and mortality in children aged < 5 y (under five) globally. This study sought to investigate the prevalence and determinants of diarrhoea and acute respiratory infections (ARIs) among children under five in West Africa. Methods The most recent demographic and health survey (DHS) standard for 13 West African countries was used in the study. We calculated the prevalence of diarrhoea and ARIs (2 wk prior to the survey) and performed multivariable complex logistic regression analysis to identify possible predictors of diarrhoea and ARIs. Results The weighted prevalence of diarrhoea and ARI was 13.7% and 15.9%, respectively. The prevalence of comorbid diarrhoea and ARI was 4.4%. Children aged < 2 y (p< 0.001), mothers aged < 30 y (p< 0.003), mothers without formal education (p< 0.001), poor households (p< 0.001) and poor nutritional status, wasting (p=0.005) and underweight (p< 0.001), were the independent predictors of diarrhoea. The independent predictors of ARIs were children with no childhood vaccinations (p=0.002), use of solid fuel in the household (p=0.007), being underweight (p=0.05) and diarrhoea (p< 0.001). Conclusions The findings imply the need for holistic public health interventions such as increased vaccination coverage, population-based nutritional programmes and campaigns on the use of cleaner cooking fuel targeted at high-risk subgroups in the population to reduce the burden and adverse effects of diarrhoea and ARIs in the West African region.

Immune dysfunction is one of the central components in the development and progression of endometriosis by establishing a chronic inflammatory environment. Western-style high-fat diets (HFD) have been linked to greater systemic inflammation to cause metabolic and chronic inflammatory diseases, and are also considered an environmental risk factor for gynecologic diseases. Here, we aimed to examine how HFD cause an inflammatory environment in endometriosis and discern their contribution to endometriotic-associated hyperalgesia. Our results showed that HFD-induced obesity enhanced abdominal hyperalgesia that was induced by endometriotic lesions. The MAP2K7 signaling pathway activates the c-Jun NH2-terminal protein kinase (JNK) in response to stress signals, such as inflammatory cytokines, osmotic stress, or genomic damage. While there has been interest in inhibiting JNK due to its involvement in inflammatory processes and cancer, there is increasing focus on developing MAP2K7 inhibitors to enhance specificity when MAP2K7 activation is associated with disease progression. Generation And Characterization Of In Vitro Transcribed Mrna, Cristina Poveda, Yi-Lin Chen, Ulrich Strych

Here we describe the in vitro preparation of mRNA from DNA templates, including setting up the transcription reaction, mRNA capping, and mRNA labeling. We then describe methods used for mRNA characterization, including UV and fluorescence spectrophotometry, as well as gel electrophoresis. Moreover, characterization of the in vitro transcribed RNA using the Bioanalyzer instrument is described, allowing a higher resolution analysis of the target molecules. For the in vitro testing of the mRNA molecules, we include protocols for the transfection of various primary cell cultures and the confirmation of translation by intracellular staining and western blotting.

Coral capacity to tolerate low pH affects coral community composition and, ultimately, reef ecosystem function. Low pH submarine discharges (‘Ojo’; Yucatán, México) represent a natural laboratory to study plasticity and acclimatization to low pH in relation to ocean acidification. A previous >2‐year coral transplant experiment to ambient and low pH common garden sites revealed differential survivorship across species and sites, providing a framework to compare mechanistic responses to differential pH exposures. The cellular junctional architecture remodeling by Listeria adhesion protein-heat shock protein 60 (LAP-Hsp60) interaction for Listeria monocytogenes (Lm) passage through the epithelial barrier is incompletely understood. Here, using the gerbil model, permissive to internalin (Inl) A/B-mediated pathways like in humans, we demonstrate that Lm crosses the intestinal villi at 48 h post-infection. In contrast, the single isogenic (lap− or ΔinlA) or double (lap−ΔinlA) mutant strains show significant defects. LAP promotes Lm translocation via endocytosis of cell-cell junctional complex in enterocytes that do not display luminal E-cadherin. In comparison, InlA facilitates …

Background
Tick-borne diseases are a growing public health threat in the United States. Despite the prevalence and rising burden of tick-borne diseases, there are major gaps in baseline knowledge and surveillance efforts for tick vectors, even among vector control districts and public health agencies. To address this issue, an online tick training course (OTTC) was developed through the Southeastern Center of Excellence in Vector-Borne Diseases (SECOEVBD) to provide a comprehensive knowledge base on ticks, tick-borne diseases, and their management.

Seasonal reproduction can provide species with fitness advantages by allowing the birth of young to coincide with favorable environmental conditions, particularly in environments with high seasonal and intra-annual variation in these conditions. Seven of the eight species in the bear family (Ursidae) reproduce seasonally in both managed care and the wild; however, data for the eighth species, the sun bear (Helarctos malayanus), is unclear. Offspring that delay dispersal in cooperatively breeding species have been hypothesized to gain direct fitness benefits via parental facilitation—being passively tolerated on their natal territory by their parents—thereby enjoying enhanced survival and increased probability of acquiring a breeding position in the population. Here we describe active facilitation in the acorn woodpecker (Melanerpes formicivorus) by parents and siblings assisting the dispersal of helpers in their social group. Helpers in this species compete for reproductive opportunities in “power struggles” that take place when all breeders of one sex die or disappear, creating a reproductive vacancy. Turning is an important aspect of life underwater, playing integral roles in predator avoidance, prey capture, and communication. While turning abilities have been explored in a diversity of adult nekton, little is currently known about turning in early ontogeny, especially for cephalopods. In this study, we investigated the turning abilities of hatchling common cuttlefish (Sepia officinalis, n = 49) and dwarf cuttlefish (Sepia bandensis, n = 30), using both kinematic and wake-based analyses. Using body tracking software and particle image velocimetry (PIV), we found that S. officinalis turned faster than S. bandensis, but both species …

Atlantic reef-building corals and coral reefs continue to experience extensive decline due to increased stressors related to climate change, disease, pollution, and numerous anthropogenic threats. To understand the impact of ocean warming and reef loss on the estimated extinction risk of shallow water Atlantic reef-building scleractinians and milleporids, all 85 valid species were reassessed under the IUCN Red List Categories and Criteria, updating the previous Red List assessment of Atlantic corals published in 2008. For the present assessment, individual species declines were estimated based on the modeled coral cover loss (1989–2019) and projected onset of annual severe bleaching events (2020–2050) …
