Introduction: Despite recent advances, triple-negative breast cancer (TNBC) patients remain at high risk for recurrence and metastasis, which creates the need for innovative therapeutic approaches to improve patient outcomes. Cryoablation is a promising, less invasive alternative to surgical resection, capable of inducing tumor necrosis via freeze/thaw cycles. Necrotic cell death results in increased inflammatory signals and release of preserved tumor antigens, which have the potential to boost the local and systemic anti-tumor immune response. Recent increases in community-associated Clostridioides difficile infections have highlighted the importance of monitoring toxigenic C. difficile from animal and environmental sources. We provide the complete circularized genomes of two toxigenic C. difficile strains isolated from feral horse faeces. Genome N64 (sequence type 964) consists of a single chromosome of 4,078,791 bp, while genome H251 (sequence type 963) comprises one chromosome (4,304,722 bp) and three plasmids (150,942 bp, 11,534 bp, and 9,074 bp).

The controlled release of silver as a biocide from Ag-coated medical implants is desirable. However, the biocompatibility of Ag leachates is poorly understood. This study investigated the toxicity of silver released from the silver plated titanium implants to human primary osteoblast cells; and the effect of cell culture medium on the silver speciation and bioavailability.
METHODS: Ti6Al4V discs were coated with Ag nanoparticles (NPs), silver plus hydroxyapatite (HA) nanoparticles (Ag+nHA), or Ag NPs plus microparticles (Ag+mHA). Primary human osteoblast cells were exposed to the leachates from the various discs for up to 7 days.

Unsupervised Environment Design (UED) formalizes the problem of autocurricula through interactive training between a teacher agent and a student agent. The teacher generates new training environments with high learning potential, curating an adaptive curriculum that strengthens the student’s ability to handle unseen scenarios. Existing UED methods mainly rely on regret, a metric that measures the difference between the agent’s optimal and actual performance, to guide curriculum design. Regret-driven methods generate curricula that progressively increase environment complexity for the student but overlook environment novelty–a critical element for enhancing an agent’s generalizability. Australian Aboriginal people experience stressors from inequalities across crucial social determinants, including deep and entrenched disadvantage and exclusion. The impact of unaddressed historical issues is pervasive and intergenerational. The disproportionate rates of Aboriginal youth suicide, juvenile detention and imprisonment highlight the inadequacy of existing social and emotional wellbeing programs and services for Aboriginal children and young people. There is increasing recognition in Australia that aligning social and emotional wellbeing interventions with Western values and conceptions of mental health is one of the main barriers to service uptake among Aboriginal people. The history of exercise science research has undergone relatively little examination, and exercise science students receive little education about the field’s history. Photographs are tools that lecturers and writers can use to educate audiences about history. Yet, no resource exists that tells educators where they can find photographs of individuals participating in early studies about exercise or related topics (e.g., physical education, physical medicine). Here, the aim was to identify papers published before 1980 that contain photographs of boys and men participating in studies about exercise or related topics, aggregate the papers into a bibliometric list, and describe the photographs. …

Cognitive processes such as learning, memory, and decision-making are integral to an organism’s fitness, allowing to integrate cues from its environment and adjust its behavior accordingly. Historically, cognitive abilities were thought to have evolved according to a Scala Naturae design—an idea that has since been disproven—which suggested that complex cognitive abilities were exclusive to “higher” taxa. This perspective has created significant phylogenetic gaps in our understanding of cognitive evolution. This paper introduces a novel approach for variable selection in survival analysis by integrating neutrosophic logic into the Cox Proportional Hazards (Cox PH) model to address the limitations of recent studies related to high dimensionality. Neutrosophic logic, is a mathematical framework that allows for uncertainty, indeterminacy, and inconsistency, and particularly well suited for handling the complexity and often-ambiguous nature of biological data. In general topology the notion of pds-irresolute and aδs-irresolute functions were introduced by Beceren and Noiri. In the present paper, these concepts of pδs-irresolute (briefly, Npδs-irresolute) and aδs-irresolute (briefly, Naδs-irresolute) functions are explored for the first time in neutrosophic topological spaces (NTS) as generalized version. We proved that every Naδs-irresolute function is Npδs-irresolute function but not conversely. Some characterizations, counter examples, and fundamental features are also presented. By neutrosophic pre-open, neutrosophic δ-open, and neutrosophic δ-semi-open sets, some new fundamental properties of such functions are provided. Objectives
Sorhgum-sudangrass (SS) and cowpea (CW) can increase cattle gains when sod-seeded to mixed bermudagrass (MB); however, establishment time and cost may not be recouped by improved animal production. Additionally, valuing weight gain at varying production stages, without a market transaction, prompted an evaluation of different pricing methods.Materials and Methods
Cattle gains on MB, SS, and a 50:50 mix of SS+CW (SSCW) pastures were evaluated. One pricing method linearly interpolated 10-yr-average prices across weight categories. The “end of affirmative action” is the beginning of this story. In Students for Fair Admissions v. Harvard (SFFA), the Supreme Court struck a near fatal blow to race-consciousness. Many institutions have since pivoted to “race neutral alternatives.” This is a natural turn. But one that faces immediate headwinds.
The same entities that demanded Harvard pursue racial diversity through colorblind means have sued public high schools for doing just that. These litigants assert a “right to inequality”—a theory that would pit the equal protection clause against equality itself. Even if normatively jarring, a right to inequality might seem a …

The celebration of the anniversary of the Center for Health Law, Ethics & Human Rights (the “Center”) provides an opportunity to reflect on what defines the field of health law, as well as its conjoined twins of bioethics and human rights. The related fields are vast, and the subjects they encompass are ever-expanding. It is probably impossible to lay out a summary that does justice to their expansive, interdisciplinary scope. BACKGROUND: Recombinant Necator americanus Glutathione-S-Transferase-1 (Na-GST-1) formulated on Alhydrogel (Na-GST-1/Alhydrogel) is being developed to prevent anemia and other complications of N. americanus infection. Antibodies induced by vaccination with recombinant Na-GST-1 are hypothesized to interfere with the blood digestion pathway of adult hookworms in the host. Phase 1 trials have demonstrated the safety of Na-GST-1 formulated on Alhydrogel, but further optimization of the vaccine adjuvant formulation may improve humoral immune responses, thereby increasing the likelihood of vaccine efficacy.

Alzheimer's disease is one of at least 26 diseases characterized by tau-positive accumulation in neurons, glia or both. However, it is still unclear what modifications cause soluble tau to transform into insoluble aggregates. We previously performed genetic screens that identified tyrosine kinase 2 (TYK2) as a candidate regulator of tau levels. Here we verified this finding and found that TYK2 phosphorylates tau at tyrosine 29 (Tyr29) leading to its stabilization and promoting its aggregation in human cells. We discovered that TYK2-mediated Tyr29 phosphorylation interferes with autophagic clearance of tau. We also show that TYK2-mediated phosphorylation of Tyr29 facilitates pathological tau …

Access to genomic sequencing (GS) and resulting recommendations have not been well described in pediatric oncology. GS results may provide a cancer predisposition syndrome (CPS) diagnosis that warrants screening and specialist visits beyond cancer treatment, including testing or surveillance for family members. The Texas KidsCanSeq (KCS) Study evaluated implementation of GS in a diverse pediatric oncology population. We conducted semi-structured interviews (n = 20) to explore experiences of KCS patients' families around learning about a CPS diagnosis and following up on recommended care. Precision oncology is driven by biomarkers. For glioblastoma multiforme (GBM), the most common malignant adult primary brain tumor, O6-methylguanine-DNA methyltransferase (MGMT) gene promoter methylation is an important prognostic and treatment clinical biomarker. Time consuming pre-analytical steps such as biospecimen storage, fixation, sampling, and processing are sources of data irreproducibility, and all these pre-analytical variables are confounded by intratumor heterogeneity of MGMT promoter methylation. Background: Respiratory syncytial virus (RSV) causes significant morbidity and mortality, especially in young children. Why RSV infection in children is more severe compared to healthy adults is not fully understood.
Methods: We used ex-vivo human nasal organoid platforms from infants and adults to investigate the underlying mechanism of this disease disparity at the initial site of RSV replication, the nasal epithelium.
Results: Infant-derived human nasal organoid-air liquid interface (HNO-ALIs) lines were more susceptible to early RSV replication. Moreover, infant-derived HNO-ALIs elicited a statistically significant greater overall cytokine response, enhanced mucous production, and greater cellular damage compared to their adult counterparts. …

Androgens are classically thought to act through intracellular androgen receptors (AR/NR3C4), but they can also trigger non-genomic effects via membrane proteins. Although several membrane androgen receptors have been characterized in vitro, their functions in vivo remain unclear. Using a chemical-genetic screen in zebrafish, we found that GPRC6A, a G-protein-coupled receptor, mediates non-genomic androgen actions during embryonic development. Exposure to androgens (androstanedione, DHT and testosterone) caused cardiac edema or tail curvature in wild-type embryos, as well as in ar mutants, suggesting AR-independent pathways. The innate immune system plays an important role in the early defense against pathogens. To better understand the chicken’s innate inflammatory responses, we concurrently examined the cellular responses at the injection site and in the blood following intradermal (i.d.) injections of lipopolysaccharide (LPS) or peptidoglycan (PGN) into the pulp of growing feathers (GFs). Both LPS from Gram-negative bacteria and PGN from Gram-positive bacteria are potent stimulators of innate inflammatory responses. The time-course study conducted revealed similarly elevated levels of heterophils and macrophages in injected GF-pulps during the first 24 h for both LPS and PGN. However, PGN also stimulated rapid …

Human respiratory syncytial virus (hRSV) is a major cause of lower respiratory tract infections in young children, the elderly, and immunocompromised. Despite its discovery over 60 years ago and the global impact on human health, limited effective prophylactic or therapeutic options have been available for hRSV infections. This has largely been attributed to the legacy of vaccine failure in the 1960s using a formalin-inactivated RSV , which led to enhancement of disease post exposure to hRSV infection and hampered subsequent development of vaccine candidates. Controllable and reproducible animal models of aneurysmal subarachnoid hemorrhage (SAH) are crucial for the systematic study of the pathophysiology and treatment of this debilitating condition. However, current animal models have not been successful in replicating the pathology and disabilities seen in SAH patients, especially the long-term neurocognitive deficits that affect the survivor's quality of life. Therefore, there is an unmet need to develop experimental models that reliably replicate the long-term clinical ramifications of SAH - especially in mice where genetic manipulations are straightforward and readily available. In the last three decades, ablation of atrial fibrillation (AF) has become an evidence-based safe and efficacious treatment for managing the most common cardiac arrhythmia. Alzheimer's disease is one of at least 26 diseases characterized by tau-positive accumulation in neurons, glia or both. However, it is still unclear what modifications cause soluble tau to transform into insoluble aggregates. We previously performed genetic screens that identified tyrosine kinase 2 (TYK2) as a candidate regulator of tau levels. Here we verified this finding and found that TYK2 phosphorylates tau at tyrosine 29 (Tyr29) leading to its stabilization and promoting its aggregation in human cells. We discovered that TYK2-mediated Tyr29 phosphorylation interferes with autophagic clearance of tau. We also show that TYK2-mediated phosphorylation of Tyr29 facilitates pathological tau …

This chapter analyses the emerging data realms in China and the European Union against the backdrop of the Comprehensive Agreement on Investment (CAI). It examines the link between data protection and investment flows and undertakes a comparative analysis of the PIPL and GDPR. Moreover, this chapter will also analyse the Digital Silk Road in promoting China’s model of data sovereignty. While early trade and investment agreements did not consider data relevant for the regulation of cross-border commerce, modern treaties are increasingly cognisant of rules for data protection.
