Oncogenic KRAS (KRAS*) contributes to many cancer hallmarks. In colorectal cancer, KRAS* suppresses antitumor immunity to promote tumor invasion and metastasis. Here, we uncovered that KRAS* transforms the phenotype of carcinoma-associated fibroblasts (CAF) into lipid-laden CAFs, promoting angiogenesis and tumor progression. Mechanistically, KRAS* activates the transcription factor CP2 (TFCP2) that upregulates the expression of the proadipogenic factors BMP4 and WNT5B, triggering the transformation of CAFs into lipid-rich CAFs. These lipid-rich CAFs, in turn, produce VEGFA to spur angiogenesis. In KRAS*-driven colorectal cancer mouse models, genetic or pharmacologic neutralization of TFCP2 reduced lipid-rich CAFs, lessened tumor angiogenesis, and improved overall survival. …

UNLABELLED: The tumor microenvironment (TME) restricts antitumor CD8+ T-cell function and immunotherapy responses. Cancer cells compromise the metabolic fitness of CD8+ T cells within the TME, but the mechanisms are largely unknown. Here we demonstrate that one-carbon (1C) metabolism is enhanced in T cells in an antigen-specific manner. Therapeutic supplementation of 1C metabolism using formate enhances CD8+ T-cell fitness and antitumor efficacy of PD-1 blockade in B16-OVA tumors. Formate supplementation drives transcriptional alterations in CD8+ T-cell metabolism and increases gene signatures for cellular proliferation and activation. Combined formate and anti-PD-1 therapy increases tumor-infiltrating CD8+ T cells, which are essential for …

Paleoclimate Of The Peruvian Andean Highlands During The Late Holocene, Jarunetr Sae-Lim
Arts & Sciences Graduate Student Theses and Dissertations
The tropical and central Andean highlands (>4,000 m above sea level) supply critical freshwater resources to the Amazon Basin, supporting large communities in mountain regions and lowlands. Due to the scarcity of historical weather data spatially and temporally, it is challenging to quantify hydroclimate and environmental responses to anthropogenic climate change. Paleoclimate reconstruction and modeling can provide critical insights into key hydroclimate processes and how hydroclimate in this region has responded to climate change and variability through time. However, biomarker-based paleoclimate proxy reconstructions are still very rare in this region. This dissertation quantifies local-scale and regional-scale hydroclimate and environmental …

INTRODUCTION: The clinical benefit of the anti-CTLA-4 monoclonal antibody (mAb) ipilimumab has been well established but limited by immune-related adverse events, especially when ipilimumab is used in combination with anti-PD-(L)1 mAb therapy. To overcome these limitations, we have developed XTX101, a tumor-activated, Fc-enhanced anti-CTLA-4 mAb.
METHODS: XTX101 consists of an anti-human CTLA-4 mAb covalently linked to masking peptides that block the complementarity-determining regions, thereby minimizing the mAb binding to CTLA-4. The masking peptides are designed to be released by proteases that are typically dysregulated within the tumor microenvironment (TME), resulting in activation of XTX101 intratumorally. Mutations within the Fc region …

The impact of monthly temperature on child anthropometric outcomes varies across countries, regions, and cultural practices. Economic status alone is …
Hybrid Test-Smell Based Approach For Prediction Of Flaky Tests, Saurabh Bodke
Hybrid Test-Smell Based Approach For Prediction Of Flaky Tests, Saurabh Bodke
Theses
Regression testing is an essential component in software development, aimed at verifying that recent code changes do not negatively impact existing functionalities. A prevalent challenge in this area is the occurrence of flaky tests, which exhibit inconsistent outcomes—passing or failing without any code alterations. Such tests erode the reliability of automated testing frameworks, leading to increased debugging efforts and diminished confidence in software stability. This research addresses the challenge of flaky tests by exploring machine learning and prediction models, specifically focusing on test smells and four principal root causes of flakiness: Async Wait, Concurrency, Test Order Dependency, and Resource Leak. …

Improving The Test Smell-Based Detection Of Flaky Tests, Shubham Kumar Karum
Theses
Regression testing, a critical process in software development, ensures that the recent code changes have not adversely affected existing functionality. A significant challenge in this domain is the existence of flaky tests—tests that inconsistently pass or fail without any changes to the code. These flaky tests undermine the reliability of automated testing and can lead to increased debugging and decreased confidence in software stability. To tackle the issue of flaky tests, there exists approaches that make use of machine learning and prediction models. However, approaches that rely on analyzing test case vocabulary typically face challenges like higher costs to run …

Myopia is becoming increasingly common in young generations all over the world, and it is predicted to become the most common cause of blindness and visual impairment in later life in the near future. Because myopia can cause serious complications and vision loss, it is critical to create and prescribe effective myopia treatment solutions that can help prevent or delay the onset and progression of myopia. The scientific understanding of myopia's causes, genetic background, environmental conditions, and various management techniques, including therapies to prevent or postpone its development and slow its progression, is rapidly expanding. However, some significant information gaps …

Significant amounts of hydrocarbon resources are left behind after primary and secondary recovery processes, necessitating the application of enhanced oil recovery (EOR) techniques for improving the recovery of trapped oil from subsurface formations. In this respect, the wettability of the rock is crucial in assessing the recovery and sweep efficiency of trapped oil. The subsurface reservoirs are inherently contaminated with organic acids, which renders them hydrophobic. Recent research has revealed the significant impacts of nanofluids, surfactants, and methyl orange on altering the wettability of organic-acid-contaminated subsurface formations into the water-wet state. This suggests that the toxic dye methylene blue (MB), …

Neurodegenerative diseases (ND) are characterized by progressive loss of neuronal function. Mechanisms of ND pathogenesis are incompletely understood, hampering the development of effective therapies. Langerhans cell histiocytosis (LCH) is an inflammatory neoplastic disorder caused by hematopoietic progenitors expressing mitogen-activated protein kinase (MAPK)-activating mutations that differentiate into senescent myeloid cells that drive lesion formation. Some individuals with LCH subsequently develop progressive and incurable neurodegeneration (LCH-ND). Here, we showed that LCH-ND was caused by myeloid cells that were clonal with peripheral LCH cells. Circulating BRAFV600E

Background: Children with heart disease frequently require anticoagulation for thromboprophylaxis. Current standard of care (SOC), vitamin K antagonists or low-molecular-weight heparin, has significant disadvantages.
Objectives: The authors sought to describe safety, pharmacokinetics (PK), pharmacodynamics, and efficacy of apixaban, an oral, direct factor Xa inhibitor, for prevention of thromboembolism in children with congenital or acquired heart disease.
Methods: Phase 2, open-label trial in children (ages, 28 days to < 18 years) with heart disease requiring thromboprophylaxis. Randomization 2:1 apixaban or SOC for 1 year with intention-to-treat analysis.
Primary endpoint: a composite of adjudicated major or clinically relevant nonmajor bleeding. Secondary endpoints: PK, pharmacodynamics, quality of life, and exploration of efficacy.
Results: From 2017 to 2021, 192 participants were randomized, 129 …
