This dissertation discusses research that focuses on understanding transneptunian objects (TNOs) using a variety of techniques and approaches. In Chapter 1, I introduce the main concepts used throughout this dissertation and discuss the current understanding of the transneptunian region. In Chapter 2, I discuss my efforts to understand how Neptune's late stages of migration affect the Haumea family, the only known collisional family in the transneptunian region. Using advanced simulations of Neptune migration, I find that the Haumea family can plausibly form before the termination of giant planet migration and show that this extensively mixes the family. This document demonstrates the design process of an Augmented-Reality visual interaction design for improving and enhancing the astronomical observing experience for amateurs. Astronomical observation can be fun and romantic when sharing the experience with friends, family, or someone special. Sharing those magnificent moments with friends and family when astronomical events happen could be joyful. However, there are some inevitable barriers. In order to have a successful meteor shower observation, observers must follow a series of strict and professional rules that make the experience usually unaffordable for amateurs. Epidermal growth factor receptor variant III (EGFRvIII, the deletion of exons 2-7) is a recurrent intragenic EGFR::EGFR.E1E8 fusion that occurs in high-grade gliomas. The presence of EGFRvIII in other solid tumors has not been well characterized. We retrospectively reviewed advanced malignant solid tumor cases tested by a custom hybrid capture 610-gene next-generation sequencing platform from 2021 to 2022. EGFRvIII was identified in 17 of 4331 (0.4%) cases, including 16 of 238 (7%) brain tumors and 1/301 (0.3%) breast tumors. Photolabeling of intracellular molecules is an invaluable approach to studying various dynamic processes in living cells with high spatiotemporal precision. Among fluorescent proteins, photoconvertible mechanisms and their products are in the visible spectrum (400-650 nm), limiting their in vivo and multiplexed applications. Here we report the phenomenon of near-infrared to far-red photoconversion in the miRFP family of near infrared fluorescent proteins engineered from bacterial phytochromes. This photoconversion is induced by near-infrared light through a non-linear process, further allowing optical sectioning. Photoconverted miRFP species emit fluorescence at 650 nm enabling photolabeling entirely performed in the near-infrared range. We use miRFPs as …

Upon T-cell activation, the levels of the secondary messenger diacylglycerol (DAG) at the plasma membrane need to be controlled to ensure appropriate T-cell receptor signaling and T-cell functions. Extended-Synaptotagmins (E-Syts) are a family of inter-organelle lipid transport proteins that bridge the endoplasmic reticulum and the plasma membrane. In this study, we identify a novel regulatory mechanism of DAG-mediated signaling for T-cell effector functions based on E-Syt proteins. We demonstrate that E-Syts downmodulate T-cell receptor signaling, T-cell-mediated cytotoxicity, degranulation, and cytokine production by reducing plasma membrane levels of DAG. ncreased breast cancer (BC) mortality risk posed by delayed surgical resection of tumor after diagnosis is a growing concern, yet the underlying mechanisms remain unknown. Our cohort analyses of early-stage BC patients reveal the emergence of a significantly rising mortality risk when the biopsy-to-surgery interval was extended beyond 53 days. Additionally, histology of post-biopsy tumors shows prolonged retention of a metastasis-permissive wound stroma dominated by M2-like macrophages capable of promoting cancer cell epithelial-to-mesenchymal transition and angiogenesis. This quantitative study aims to determine whether food-sharing (gathering to eat) can be considered a suitable medium to facilitate increased cohesion within African American families. This study focuses on informal food-sharing as an extension of formal meal-sharing. Previous research links consistent formal meal-sharing to positive and secure identity development, better familial attachment, and improved family cohesion. These practices leave a lasting, detrimental impact on the African American family model. Additional barriers to some African American family meal-sharing practices include working extended and atypical work hours, food insecurity, and poor familial communication. Teachers and students alike suffer from anxiety, stress, and depression. Mindfulness-based strategies have been indicated to help teachers and students deal with stress, anxiety, and depression. Teachers often report not wishing to add more learning strategies to their packed schedules and workload. Education in mindfulness is not easily accessible due to the time constraints of this overworked population. The criteria for participation in the study required the college students in preservice education classes to be over 18 years old, enrolled in the teacher education program, and not have participated in any mindfulness in the last six months. Catholics who are faithful to the teachings of the Catholic Church on the indissolubility of marriage and who experience a civil divorce are at high risk for moral injury. This study was anchored on a character domain theoretical framework, and it investigated the impact of moral injury on character identity (measured by self-discrepancy) through the mediation of the perception of suffering. Religiosity was explored as a moderator for the relationships of moral injury to perception of suffering, perception of suffering to character identity, and moral injury to character identity. This qualitative method research paper with survey design identifies the need to implement an associate in fine arts degree in music at community colleges located in the rural northwestern and northern regions of North Carolina. A music degree would provide early college students with music courses comparable to those offered in traditional area high schools and greater accessibility for a diverse student population of community college students. The purpose of this hermeneutical phenomenological qualitative study was to describe African-American clergy’s lived experiences with mental health and mental health’s influence on the construction of their teachings, sermons, and church practice. The purpose of this mixed-methods study was to discover the knowledge of human trafficking of the pastors in the Baptist Convention of Maryland/Delaware churches and their responses to this crime. The guiding theory was the words of Jesus in Exodus: Chapter 3 concerning slavery and the tribulations of Israelites. A questionnaire provided enlightenment regarding the levels of knowledge in the church communities, how pastors viewed it, the methods by which they received the information, and how they responded to it. The decline of the youth attendance is evident in many Protestant churches. This research paper examined forty-three young believers from three Haitian Baptist churches, respectively, located in Brooklyn, Queens, and the Bronx. These data results help develop a suitable spiritual program that includes the six key influencing factors for spiritual growth: discipleship, mentoring, parental influence, church attendance, personal devotion, and ministerial involvement. This spiritual program was tested on a small group of young people from Great Commission Church in Queens.
