Pancreatic ductal adenocarcinoma (PDAC) tumours carry multiple gene mutations and respond poorly to treatments. There is currently an unmet need for drug carriers that can deliver multiple gene cargoes to target high solid tumour burden like PDAC. Here, we report a dual targeted extracellular vesicle (dtEV) carrying high loads of therapeutic RNA that effectively suppresses large PDAC tumours in mice. The EV surface contains a CD64 protein that has a tissue targeting peptide and a humanized monoclonal antibody. Cells sequentially transfected with plasmid DNAs encoding for the RNA and protein of interest by Transwell®-based asymmetric cell electroporation release abundant targeted …

Is A Psychotic Anthropology Possible? Or How To Have Inclusive Anthropologies Of Subjectivity And Personhood, John Marlovits, Matthew J. Wolf-Meyer
Faculty Research, Scholarly, and Creative Activity
Dominant anthropological theories of mind, cognition, and consciousness reify particular ways of being in the world as "normal," which marginalizes the experiences of people who do not meet normative expectations of personhood or exhibit nonnormative subjectivities. By focusing on atypical forms of communication and self-representation in the ethnographic record, which draws from work in the anthropology of disability and psychological anthropology, we argue for the need to attend to interactions and behavior as the necessary basis for anthropological studies of personhood and subjectivity. These foci, which build on a foundation provided by affect theory and disability studies, stand to open …

Appropriate And Optimal Classifier For Beef Quality Discrimination By A Low-Cost Optical Apparatus, Said E. Abdallah, Wael M. Elmessery, Waleed Z. Hassan, Nora S. Al-Sattary, Amr A. Abohany, Ahmed Elashry
Information Sciences Letters
In this paper, we present an optimal classifier for beef quality discrimination by a low-cost optical apparatus. Detecting beef spoilage in beef factories is a sophisticated process because beef spoilage is a mixture of physical and chemical changes. A low-cost Light-Dependent Resistor (LDR), and a light source were used to collect reflection spectra during the analysis of beef. The LabVIEW platform was programmed to acquire the obtained data from the microcontroller (Arduino) to predict beef quality. For the beef quality discrimination process, un-supervising machine learning called Principal Components Analysis (PCA) was used, and the score plot percentage was of the …

This paper reflects on previous work using QE to examine patterns of discourse of adolescent learners in a virtual, global, collaborative informal learning setting. The collective impact of involvement in the project on participants’ experiences was observed in various reflective interviews over the last five years. The deep reflection of this work resulted in a research shift from the general impact on the participants to a shift towards examining how such experiences shape self-identity, such as recognizing identity congruence, relational self, and overcoming negative identity fostered by master narrative frameworks. An initial examination of pre-assessment interviews indicates that younger students …

Developing A Point-Of-Care Detection Kit For Parkinson’S Disease Using Gold Nanoparticle-Based Lateral Flow Assay, Ali Aljassabi
USF Tampa Graduate Theses and Dissertations
Parkinson’s disease (PD) is the second most common neurodegenerative disease after Alzheimer’s disease and the fastest-growing neurodegenerative disorder in the world. The major issue with PD is that it takes several years of biological changes until the patient starts to experience physiological symptoms that could affect their daily lives. The major hallmark of PD is the appearance of Lewy body inclusions in the neuron’s cytoplasm, which is rich in a protein called α-Synuclein (α-Syn). Several studies have shown that the levels of α-Syn in blood increase because the excess α-Syn is released from neurons into the bloodstream through extracellular vesicles. …

Background and Objective: The blood-brain barrier (BBB) serves as a dynamic, selective shield, safeguarding the central nervous system (CNS) by separating the brain from circulating blood, preserving its microenvironment, and ensuring stability. However, in the presence of brain pathology, drug delivery across the BBB and blood-tumor barrier (BTB) becomes challenging, hindering effective treatments. Borneol exhibits promise in bidirectionally modulating the BBB under pathological conditions, suggesting at potential clinical applications for related diseases. Our primary goal in this review is to investigate borneol’s potential clinical utility in bidirectionally regulating the BBB under pathological conditions.

Spatial Structure Of Germination And Seed-Associated Fungi In Tropical Pioneer Seeds, Daniela Varon Garcia
Spatial Structure Of Germination And Seed-Associated Fungi In Tropical Pioneer Seeds, Daniela Varon Garcia
USF Tampa Graduate Theses and Dissertations
Negative density-dependent regulation is the process by which host-specific natural enemies reduce the survival of seeds and seedlings of conspecifics near adult trees, favoring heterospecific recruitment, and ultimately increasing local diversity. Negative density-dependent regulation by fungal pathogens has been consistently reported to promote plant diversity in the tropics, even though they tend to be multi-host and host-specificity is rarely reported. Nevertheless, evidence suggests that host-generalist fungi can have host-specific effects that produce disproportionate negative effects on some plant species but not others, promoting regional diversity. However, such studies have not tested for both host-specificity and distance-dependent effects in fully replicated …
Entanglements Of Teenage Food Security Within High School Pantries In Pinellas County, Florida, Karen T. Díaz Serrano
Entanglements Of Teenage Food Security Within High School Pantries In Pinellas County, Florida, Karen T. Díaz Serrano
USF Tampa Graduate Theses and Dissertations
Food insecurity has the ability to shape an individual’s or a family’s everyday life and take emotional, psychological, and physical tolls. Among adolescents, not having access to nutritious food could lead to physical effects during growth and development. Moreover, the stress and anxiety of not knowing where their next meal is coming from and the social stigma associated with being food insecure can have negative effects on mental health. A recent solution to these issues is locating food pantries in high schools. Purpose: This exploratory study examined how high school students in Pinellas County, Florida perceive the use of a …
Substrate Specificity In Abc Transporters Using The E. Coli Methionine Import System, John H. Guardado
Substrate Specificity In Abc Transporters Using The E. Coli Methionine Import System, John H. Guardado
Featured Student Work
ATP-binding cassette (ABC) transporters use the energy of ATP to move substrates across membranes against a concentration gradient. The role of ABC transporters is crucial in several essential cellular functions and mutations in ABC transporters in humans have been linked to several conditions, including cystic fibrosis, liver disease, and diabetes. Despite their central roles in homeostasis, the mechanism of ABC transporters remains poorly understood. Our research is focused on studying an ABC importer in E. coli, as a model system, to examine the mechanism of substrate specificity and transport. The bacterial methionine import system consists of a membrane-embedded transporter, MetNI, …

BACKGROUND: Among the three NTRK genes, NTRK2 possesses a tremendous structural complexity and involves tumorigenesis of several types of tumors. To date, only STRN and RBPMS are identified in the fusion with NTRK2 in adult soft tissue tumors. More recently, the highly selective Trk tyrosine kinases inhibitors, including larotrectinib and entrectinib, have shown significant efficacy for treating tumors harboring NTRK fusions and were approved by FDA.
CASE PRESENTATION: We report a case of sarcoma in a 35-year-old female harboring two STRN-NTRK2 gene fusions, with a good clinical response to first-line larotrectinib treatment. Core biopsy of the 16.5 cm gluteal mass …

The aim of this inquiry is to inform institutional practices within museums on queer inclusivity and further scholarship on this topic by engaging with the queer art community and public during an exhibition held in …
The Involvement Of Caspases In The Process Of Nuclear Removal During Lens Fiber Cell Differentiation, Rifah Gheyas, A. Sue Menko
The Involvement Of Caspases In The Process Of Nuclear Removal During Lens Fiber Cell Differentiation, Rifah Gheyas, A. Sue Menko
Department of Pathology, Anatomy, and Cell Biology Faculty Papers
The terminal differentiation of lens fiber cells involves elimination of their organelles, which must occur while still maintaining their functionality throughout a lifetime. Removal of non-nuclear organelles is accomplished through induction of autophagy following the spatiotemporal suppression of the PI3K/Akt signaling axis. However, blocking this pathway is not alone sufficient to induce removal of fiber cell nuclei. While the final steps in fiber cell nuclear elimination are highlighted by the appearance of TUNEL-positive nuclei, which are associated with activation of the lens-specific DNaseIIβ, there are many steps in the process that precede the appearance of double stranded DNA breaks. We …

Heterozygous SNVs or CNV deletions involving the FOXF1 gene, or its distant enhancer, are causative for 80-90% of cases of alveolar capillary dysplasia with misalignment of pulmonary veins. Recently, we proposed bimodal structure and parental functional dimorphism of the lung-specific FOXF1 enhancer, with Unit 1 having higher activity on the paternal chr16 and Unit 2 on the maternal chr16. Here, we describe a novel unusually sized pathogenic de novo copy-number variant deletion involving a portion of the FOXF1 enhancer on maternal chr16 that implies narrowing Unit 2 to an essential ~ 9-kb segment. Using a restrictase-based assay, we found that …
Negative Estrangement: Fantasy And Race In The Drow And Drizzt Do’Urden, Steven Holmes
Negative Estrangement: Fantasy And Race In The Drow And Drizzt Do’Urden, Steven Holmes
Mythlore: A Journal of J.R.R. Tolkien, C.S. Lewis, Charles Williams, and Mythopoeic Literature
This essay introduces the concept of negative estrangement to help understand current cultural interventions into the norms of depicting fantasy races. First, this essay builds on Shklovsky’s concept of estrangement to describe the literary practice of negative estrangement, wherein artists craft “more evil” foes based on hybridized amalgamations of stereotypes to create antipathy toward a subject, be it monster or fantasy race. This practice is sometimes used in service of confronting the issue of race and racism, despite seeming to reify or rearticulate racist stereotypes.
This essay builds on Tolkien’s argument in favor of creating “more evil” foes to exemplify …

"A Bleak, Barren Land": Women And Fertility In The Lord Of The Rings, Dylan L. Henderson
Mythlore: A Journal of J.R.R. Tolkien, C.S. Lewis, Charles Williams, and Mythopoeic Literature
Critics, and even the general public, have noted the absence of women in The Lord of the Rings, an absence so glaring that it could hardly be overlooked. Many feminist scholars have, as a result of this deficiency, denounced J.R.R. Tolkien, interpreting this lack of female characters as indicative of repressed misogyny. Others, however, have defended the author, pointing out that the female characters that do exist could be considered role models. This essay offers an alternative interpretation and contends that the absence of women in the novel, though potentially reducing its appeal to modern readers, reinforces one of …

Photocurable Ionized Vinylimidazole Liquid Salt For Antibacterial Application, Gina Park, Lisa Ahlberg, Robert Zdor
Celebration of Research and Creative Scholarship
There has been an increasing demand for antibacterial dental composite as it can prevent the biofilm formation and the adhesion of bacteria after the treatment causing the consequential cavities. Previous study explored the antibacterial activity of the heterogenous mixture of heterocyclic polymer and resin. It showed limitations as the mixture did not show antibacterial activity since the mixture could only contain a small amount of heterocyclic polymer within the resin due to its inconvenience of shaping and low miscibility. To overcome this limitation, this research focused on synthesizing heterocyclic antibacterial monomer liquid salt and curing it with UV light to …

The thermal activation process by which a system passes from one local energy minimum to another is a recurring motif in physics, chemistry, and biology. For instance, biopolymer chains are typically modeled in terms of energy landscapes, with folded and unfolded conformations represented by two distinct wells separated by a barrier. The rate of transfer between wells depends primarily on the height of the barrier, but it also depends on the details of the shape of the landscape along the trajectory. We consider the case of bias due to an external force, analogous to the pulling force applied in optical …

Recent developments in digital light processing (DLP) can advance the structural and biochemical complexity of perfusable in vitro models of the blood–brain barrier. Here, we describe a strategy to functionalize complex, DLP-printed vascular models with multiple peptide motifs in a single hydrogel. Different peptides can be clicked into the walls of distinct topologies, or the peptide motifs lining channel walls can differ from those in the bulk of the hydrogel. The flexibility of this approach is used to both characterize the effects of various bioactive domains on endothelial coverage and tight junction formation, in addition to facilitating astrocyte attachment in …
