The coqui frog (Eleutherodactylus coqui) was introduced to the island of Hawai'i in the 1980s, and has spread across much of the island. There is concern they will invade higher elevation areas where negative impacts on native species are expected. It is not known if coqui change behavior and baseline physiology in ways that allow them to invade higher elevations. We investigated where coqui are found across the island and whether that includes recent invasion into higher elevations. Ribosome biogenesis is an energetically costly process, and tight regulation is required for stoichiometric balance between components. This requires coordination of RNA polymerases I, II, and III. Lack of nutrients or the presence of stress leads to downregulation of ribosome biogenesis, a process for which mechanistic target of rapamycin complex I (mTORC1) is key. mTORC1 activity is communicated by means of specific transcription factors, and in yeast, which is a primary model system in which transcriptional coordination has been delineated, transcription factors involved in regulation of ribosomal protein genes include Fhl1p and its cofactors, Ifh1p and Crf1p. Resident female wood ducks (Aix sponsa) in the southeastern United States are managed through nest box programs, with the goal of enhancing local hunting opportunities. Historically, hooded mergansers (Lophodytes cucullatus) were the only other cavity-nesting waterfowl species in the southeastern United States known to use these nest boxes. However, the recent northward range expansion of black-bellied whistling-ducks (Dendrocygna autumnalis) has extended into the range of wood ducks and mergansers. All three species are brood parasites, potentially using one another as hosts for their parasitic eggs and impacting their reproductive output. My research evaluated the …

UNLABELLED: Lipid droplets (LD) are dynamic organelles that serve as hubs of cellular metabolic processes. Emerging evidence shows that LDs also play a critical role in maintaining redox homeostasis and can mitigate lipid oxidative stress. In multiple cancers, including prostate cancer, LD accumulation is associated with cancer aggressiveness, therapy resistance, and poor clinical outcome. Prostate cancer arises as an androgen receptor (AR)-driven disease. Among its myriad roles, AR mediates the biosynthesis of LDs, induces autophagy, and modulates cellular oxidative stress in a tightly regulated cycle that promotes cell proliferation. In previous work, my research group and I reported on the synthesis of Anaephenes A and B and confirmed their biological activities. We also found that these compounds were active against drug resistant bacteria such as methicillin-resistant Staphylococcus aureus (MRSA). This finding lead to a series of structure activity relationship studies (SAR) to develop analogs with improved biological activities. Near the start of my graduate studies, my team and I reported an SAR where we generated eighteen analogs. At the heart of the DNA/ALVAC/gp120/alum vaccine’s efficacy in the absence of neutralizing antibodies is a delicate balance of pro- and anti-inflammatory immune responses that effectively decreases the risk of SIVmac251 acquisition in macaques. Vaccine efficacy is linked to antibodies recognizing the V2 helical conformation, DC-10 tolerogenic dendritic cells eliciting the clearance of apoptotic cells via efferocytosis, and CCR5 downregulation on vaccine-induced gut homing CD4+ cells. RAS activation is also linked to vaccine efficacy, which prompted the testing of IGF-1, a potent inducer of RAS activation with vaccination. Rheumatic illnesses are chronic diseases that cause the immune system to attack the bodies joints, muscles, bones, and organs (Mayo Clinic Health System, 2022). In many cases, rheumatic illnesses can cause severe disability and end organ damage leading to mortality if left untreated. Many rheumatological patients are on biologic disease modifying anti-rheumatic drugs (bDMARDs) and this has significantly improved the outcome for patients with rheumatic diseases (Humphreys et al., 2016). Objective: This study aimed to explore the composition and the functional bioactivity of tea polyphenols from cocoa tea. Methods: Chemical components of crude green tea polyphenol extracts from cocoa tea were analyzed and identified using high resolution mass spectrometry. Additionally, their potential functional effects were predicted by network pharmacological analysis. Results: The in vitro hyperglycemic potential of crude green tea polyphenol from cocoa tea was preliminarily confirmed based on their strong inhibition on α-glucosidase and α-amylase. Suicidal ideation disproportionally affects the LGBTQ+ community. Community connectedness and mindfulness can be protective against suicidal ideation, whereas emotion regulation difficulty is associated with increased suicidal ideation. Community connectedness, emotion regulation difficulty and mindfulness have demonstrated relationships to each other in the LGBTQ+ community and while they are each independently associated with suicidal ideation, their combined impact on suicidal ideation has not yet been examined. Introduction—Approximately 1% of the world’s population is impacted by epilepsy, a chronic neurological disorder characterized by seizures. One-third of epileptic patients are resistant to AEDs, or have medically refractory epilepsy (MRE). One non-invasive treatment that exists for MRE includes the ketogenic diet, a high-fat, low-carbohydrate diet. Despite the KD’s success in seizure attenuation, it has a few risks and its mechanisms remain poorly understood. The KD has been shown to improve metabolism and mitochondrial function in epileptic phenotypes. Potassium channels have implications in epileptic conditions as they have dual roles as metabolic sensors and control neuronal excitation.

Genomic abnormalities are strongly associated with cancer and infertility. In this study, we develop a simple and efficient method - multiple genetic abnormality sequencing (MGA-Seq) - to simultaneously detect structural variation, copy number variation, single-nucleotide polymorphism, homogeneously staining regions, and extrachromosomal DNA (ecDNA) from a single tube. MGA-Seq directly sequences proximity-ligated genomic fragments, yielding a dataset with concurrent genome three-dimensional and whole-genome sequencing information, enabling approximate localization of genomic structural variations and facilitating breakpoint identification. Additionally, by utilizing MGA-Seq, we map focal amplification and oncogene coamplification, thus facilitating the exploration of ecDNA's transcriptional regulatory function.

Glioblastoma multiforme (GBM) is the most deadly brain tumor, effective treatment options for which still remain elusive. The current treatment procedure of maximal resection followed by chemotherapy has proved to be grossly insufficient to prevent disease progression and death. Despite best efforts, the maximum survival post-diagnosis is a mere 1.5 years. Therefore, there is a huge unmet clinical need to find effective therapeutic procedures to prevent the pathogenesis and relapse of GBM. Small-molecule inhibitors of signaling pathways are an attractive option to prevent various types of tumors. Selenium is an essential trace element in living organisms, and is present in selenoenzymes with antioxidant activity, like glutathione peroxidase (GPx) and thioredoxin reductase (TrxR). The search for small selenium-containing molecules that mimic selenoenzymes is a strong field of research in organic and medicinal chemistry. In this review, we review the synthesis and bioassays of new and known organoselenium compounds with antioxidant activity, covering the last five years. A detailed description of the synthetic procedures and the performed in vitro and in vivo bioassays is presented, highlighting the most active compounds in each series.

Coffee drinks are sugar-sweetened beverages made from coffee powder, sugar, and water through heating. High consumption of coffee drinks is associated with sugar content, which can increase the risk of obesity. Overconsumption of UPF can increase the risk of overweight and obesity. In this thesis, the challenge of dynamic spectrum management is treated following an economic-based perspective. In particular, a novel bioinspired spectrum allocation from the Network Service Providers (NSPs) to the users is introduced, exploiting the theory of symbiosis under the free competition. The overall objective is to determine the NSPs optimal bandwidth prices to maximize their profit, while satisfying the users bandwidth needs. The aforementioned spectrum management framework is analyzed while exploiting a novel 3D network architecture consisting of High Altitude Platforms (HAPs), Unmanned Aerial Vehicles (UAVs), and ground gNBs NSPs, each one of them presenting different operational characteristics and …

OBJECTIVE: To examine the association between size and margin status of ductal carcinoma in situ (DCIS) and risk of developing ipsilateral invasive breast cancer and ipsilateral DCIS after treatment, and stage and subtype of ipsilateral invasive breast cancer.
DESIGN: Multinational, pooled cohort study.
SETTING: Four large international cohorts.
PARTICIPANTS: Patient level data on 47 695 women with a diagnosis of pure, primary DCIS between 1999 and 2017 in the Netherlands, UK, and US who underwent surgery, either breast conserving or mastectomy, often followed by radiotherapy or endocrine treatment, or both.

Inter-organelle contact and communication between mitochondria and sarco/endoplasmic reticulum (SR/ER) maintain cellular homeostasis and are profoundly disturbed during tissue ischemia. We tested the hypothesis that the formin Diaphanous-1 (DIAPH1), which regulates actin dynamics, signal transduction and metabolic functions, contributes to these processes. We demonstrate that DIAPH1 interacts directly with Mitofusin-2 (MFN2) to shorten mitochondria-SR/ER distance, thereby enhancing mitochondria-ER contact in cells including cardiomyocytes, endothelial cells and macrophages. Solution structure studies affirm the interaction between the Diaphanous Inhibitory Domain and the cytosolic GTPase domain of MFN2. In male rodent and human cardiomyocytes, DIAPH1-MFN2 interaction regulates mitochondrial turnover, mitophagy, and oxidative stress. …

Emotional health problems in the workplace often hinder the integration and retention of autistic employees (AE), a challenge identified in many sectors. Recent literature highlights the consequences of these problems, such as burnout leading to reduced productivity and resignation. Previous research supports the effectiveness of virtual reality (VR) for training a variety of specific skills (e.g. riding a bus or plane travel), as well as more complex social skills, such as emotion recognition and functional communication. In addition, existing studies on using physiological self-monitoring in AE training offer a promising approach to promoting improved emotional health. The present paper reports …

We document rare finds of fossil bats from two localities representing the Pleistocene epoch in southern and central Nebraska, Albert Ahrens locality (No-104, late Irvingtonian age, Middle Pleistocene), Nuckolls County, and Litchfield (Sm-102, late Rancholabrean age, latest Pleistocene), Sherman County. The Albert Ahrens local fauna with strong boreal influence has produced two bats, Lasiurus cf. borealis and Cf. Myotis sp. The Litchfield local fauna, also showing a strong boreal influence, has yielded two bats, Eptesicus fuscus and Cf. Myotis, among a diverse Pleistocene fauna of small vertebrates and a pollen record indicating a boreal mixed conifer and deciduous woodland, …

Objectives: To highlight the clinical characteristics of paediatric patients presenting with non-Hodgkin's lymphoma, treatment toxicities, and outcome.
Methods: The retrospective study was conducted at the Aga Khan University Hospital, Karachi, and comprised data of all paediatric patients aged 0-18 years diagnosed with non-Hodgkin's lymphoma from 2010 to 2020. Demographic characteristics, presentation, treatment provided, complications, if any, and treatment outcome were recorded. Data was analysed using SPSS 21.

The discharge summary is a resume of all patient care and treatment periods that must be filled in entirely by the doctor in charge within 1x24 hours after the patient is pronounced discharged. This study examines the completeness and timeliness of filling in discharge summaries by using the system approach. This research is a qualitative study to obtain input and transformation through in-depth interviews and review of hospital documents, then reinforced with quantitative research to get an overview of the output through observation and study of discharge summary forms. The lesser grain borer, Rhyzopertha dominica, and the rice weevil, Sitophilus oryzae, are two of the most destructive pests of stored rice. Their management has relied heavily upon fumigant insecticides. However, amid growing concerns that fumigant-resistant pests are becoming widespread, there is an urgent need for diversified management techniques and integrated pest management. Here, we explored management techniques and the behavior of these beetles. Host plant resistance toward both beetles was evaluated for 13 varieties of rice. Risk matrices are used in water safety planning to prioritize improvements to drinking water systems. While water safety plans (WSPs) are promoted globally, no study has evaluated whether risk matrices are comprehensively constructed to accurately assess risk. We used risk matrix criteria adapted from previous risk matrix research to evaluate risk matrices found in twelve templates across global jurisdictions. WSP templates were found using the WSPortal website and definitions of likelihood and impact were extracted from each template to assist in the evaluation of WSP risk matrices.
