OBJECTIVE: The diversity of nomenclature and naming strategies makes therapeutic terminology difficult to manage and harmonize. As the number and complexity of available therapeutic ontologies continues to increase, the need for harmonized cross-resource mappings is becoming increasingly apparent. This study creates harmonized concept mappings that enable the linking together of like-concepts despite source-dependent differences in data structure or semantic representation.
MATERIALS AND METHODS: For this study, we created Thera-Py, a Python package and web API that constructs searchable concepts for drugs and therapeutic terminologies using 9 public resources and thesauri. By using a directed graph approach, Thera-Py captures commonly used …

Objective: Blood-based biomarkers provide a crucial information in the progress of neurodegenerative diseases with a minimally invasive sampling method. Validated blood-based biomarker application in people with amyotrophic lateral sclerosis would derive numerous benefits. Canine degenerative myelopathy is a naturally occurring animal disease model to study the biology of human amyotrophic lateral sclerosis. Serum derived exosomes are potential carriers for cell-specific cargoes making them ideal venue to study biomarkers for a variety of diseases and biological processes. This study assessed the exosomal proteins that may be assigned as surrogate biomarker that may reflect biochemical changes in the central nervous system.

Greater Socioenvironmental Risk Factors And Higher Chronic Pain Stage Are Associated With Thinner Bilateral Temporal Lobes, Lisa H Antoine, Burel R Goodin, Et Al.
2020-Current year OA Pubs
INTRODUCTION: Previous research indicates ethnic/race group differences in pain and neurodegenerative diseases. Accounting for socioenvironmental factors reduces ethnic/race group differences in clinical and experimental pain. In the current study sample, we previously reported that in individuals with knee pain, ethnic/race group differences were observed in bilateral temporal lobe thickness, areas of the brain associated with risk for Alzheimer's disease, and related dementias. The purpose of the study was to determine if socioenvironmental factors reduce or account for previously observed ethnic/race group differences and explore if a combined effect of socioenvironmental risk and chronic pain severity on temporal lobe cortices is …

Anchorage-independent survival after intravasation of cancer cells from the primary tumor site represents a critical step in metastasis. Here, we reveal new insights into how MUC13-mediated anoikis resistance, coupled with survival of colorectal tumor cells, leads to distant metastasis. We found that MUC13 targets a potent transcriptional coactivator, YAP1, and drives its nuclear translocation via forming a novel survival complex, which in turn augments the levels of pro-survival and metastasis-associated genes. High expression of MUC13 is correlated well with extensive macrometastasis of colon cancer cells with elevated nuclear YAP1 in physiologically relevant whole animal model systems. Interestingly, a positive correlation …

An Exploration Of Two Information Literacy Open Learning Object Repositories: Value, Content, And Engagement, Tiffanie Lynn Ford-Baxter
Communications in Information Literacy
Information literacy (IL) open learning object repositories (LORs) provide a space for librarians to find and exchange instruction resources and lessons. Given many librarians enter the workforce with little or no formal training or educational opportunities to learn about pedagogy, these repositories are indispensable resources to the Library and Information Science field. This study explored the contents of two popular IL LORs, Project Cora and the Association of College and Research Libraries Framework for Information Literacy Sandbox, to uncover how users engage with the resources and how the repositories differ. This study's findings suggest that while resources within the LORs …

Heart failure (HF) is a chronic and progressive clinical disorder characterized by an inability to pump sufficient blood to meet metabolic demands. It poses a substantial global healthcare burden, leading to high morbidity, mortality, and economic impact. Current treatments for HF include lifestyle modifications, guideline-directed medical therapies (GDMT), and device interventions, but the need for novel therapeutic approaches remains significant. The introduction of vericiguat, a soluble guanylate cyclase stimulator, has shown promise in improving outcomes for heart failure patients. Vericiguat addresses the underlying pathophysiological mechanisms of heart failure by augmenting the cyclic guanosine monophosphate (cGMP) pathway, leading to enhanced cardiac …

Theses and Dissertations
Every year, over 20 million people globally are affected by bone tissue loss due to disease or trauma. Bone tissue engineering (BTE) is touted as a promising approach to promote bone tissue repair and regeneration. In this realm, development of collagen-based scaffolds incorporated with a bioceramic is often attempted to generate tissue scaffolds that resemble the organic and inorganic composition of native bone. In addition, a variety of different collagen processing techniques have been employed to produce anisotropically aligned scaffolds that can provide the essential biochemical and structural cues to guide cell response. While there have been numerous studies on …

Low Reynolds Number Locomotion Near Interfaces In Two-Fluid Media, Avriel Rowena Mae Cartwright
Low Reynolds Number Locomotion Near Interfaces In Two-Fluid Media, Avriel Rowena Mae Cartwright
Theses and Dissertations
Microorganisms often swim within complex fluid environments composed of multiple materials with very different properties. Biological locomotion, including swimming speed, is significantly impacted by the physical composition and rheology of the surrounding fluid environment, as well as the presence of phase boundaries and free interfaces, across which physical properties of the fluid media may vary greatly. Through computational simulations, we first investigate the classical Taylor’s swimming sheet problem near interfaces within multi-fluid environments using a two-fluid immersed boundary method. The accuracy of the methodology is illustrated through comparisons with analytical solutions. Our simulation results indicate that the interface dynamics and …
Effects Of Perforations On Internal Cathodic Protection And Recruitment Of Marine Organisms To Steel Pipes, Alexander John Paluzzi
Effects Of Perforations On Internal Cathodic Protection And Recruitment Of Marine Organisms To Steel Pipes, Alexander John Paluzzi
Theses and Dissertations
Steel monopile support structures for offshore wind turbines require protection from corrosion on their external and internal surfaces. Cathodic protection (CP) works effectively to protect the external surfaces of monopiles, but internally, byproducts from aluminum sacrificial anode CP (SACP) and impressed current CP (ICCP) induce acidification that accelerates steel corrosion. This project investigated the effects of perforations on the performance of aluminum SACP and ICCP when used inside of steel pipes. Additionally, marine growth on the external and internal surfaces of the pipes was assessed to determine if CP byproducts affect marine organism development.
Two sealed and two perforated steel …

C-Day Computing Showcase
Cardiovascular diseases account for nearly a third of global deaths, posing a challenge that machine learning can help address. However, data privacy concerns hinder the direct application of conventional machine learning in this sensitive area. This paper explores Federated Learning (FL) as a decentralized strategy to mitigate these concerns by allowing for local data processing. FL's design ensures that only processed updates, not raw data, are shared with a central server, maintaining individual privacy. Our research assesses FL's practicality and effectiveness in predicting heart disease while adhering to ethical and legal norms. We build upon previous studies, such as Wanyong …

Gr-504 Synthetic Dna Sequence Generation And Classification For Species Discrimination, Nishat Tasnim
C-Day Computing Showcase
The two main goals of this research are to apply machine learning models in computational biology to classify DNA sequences from different species and to create synthetic DNA sequences using GANs. Generative Adversarial Networks (GANs) synthesize DNA sequences while preserving key characteristics like sequence length and GC content. The dataset is enhanced by these artificial sequences, which makes classification jobs better. The classification accuracy of black rat and human genome sequences is evaluated using machine learning models, including Random Forest, SVM, and Logistic Regression. Notably, when trained with synthetic data, all models perform better.

Background: N7-methylguanosine (m7G) is an important posttranscriptional modification affecting mRNA and tRNA functions and stability. The genes regulating the m7G process have been previously found involved in the carcinogenesis process. We aimed to analyze the role of m7G-related genes as potential prognostic markers for lung squamous cell carcinoma (LSCC).
Methods: Twenty-nine m7G-related genes were selected for the analysis in the LSCC cohort of the Cancer Genome Atlas (TCGA). Univariate, multivariate, and Kaplan-Meier analyses were used to evaluate the predictive value of risk model developed with m7G signature for overall survival (OS). The Gene Ontology (GO) and Kyoto Encyclopedia of Genes …

Cognitive stress affects humans mentally and physically, and specifically may impact emotional recognition. Previous studies have focused on the perceived emotional valence of nouns vs. adjectives and the emotional recognition of photographs. While this has led to the creation of an emotional valence database, there has yet to be a study to explore how stress can change perceived emotional valence. Using data from the Affective Norms for English Words (ANEW), the Portland Arithmetic Stress Test (PAST), and the Montreal Imaging Stress Task (MIST), a study was created to explore how participants emotional valence of words would change after being placed …

Lysosomes play important roles in catabolism, nutrient sensing, metabolic signaling, and homeostasis. NPC1 deficiency disrupts lysosomal function by inducing cholesterol accumulation that leads to early neurodegeneration in Niemann-Pick type C (NPC) disease. Mitochondria pathology and deficits in NPC1 deficient cells are associated with impaired lysosomal proteolysis and metabolic signaling. It is thought that activation of the transcription factor TFEB, an inducer of lysosome biogenesis, restores lysosomal-autophagy activity in lysosomal storage disorders. Here, we investigated the effect of trehalose, a TFEB activator, in the mitochondria pathology of NPC1 mutant fibroblasts in vitro and in mouse developmental Purkinje cells ex vivo. We …

This work reports a novel synthesis, characterization, and performance of antimicrobial polymeric thin films utilizing unreported ternary blend of Chitosan, PLA, and PCL. For binary systems, the selection of matching polymers has improved the material’s structures and properties; however, the transition to using a ternary system is not well-established yet. The ability to homogeneously blend 3 biopolymers would expand the potential to prepare new materials with tailored properties. Herein, unary, binary, and ternary films were produced by physical blending using the casting and solvent evaporation method. The films were then characterized using different techniques for comparing the properties of 3 …

The Role of MYC-EGR1 Pathway in Colorectal Cancer Development and its Relationship to Wnt Signaling
Colorectal tumors show high levels of Wnt pathway activity, most often due to an initiating mutation in critical Wnt pathway components. Wnt pathway activation leads to b-catenin-driven expression of genes that generally stimulate cell cycle progression. One such Wnt pathway target gene is MYC, a well described oncogene involved in many cancers, including colorectal. MYC is known to stimulate proliferation but in certain cellular contexts, can stimulate apoptosis by stimulating the expression of EGR1. This is termed a …
