Dynamically evolving adhesions between cells and extracellular matrix (ECM) transmit time-varying signals that control cytoskeletal dynamics and cell fate. Dynamic cell adhesion and ECM stiffness regulate cellular mechanosensing cooperatively, but it has not previously been possible to characterize their individual effects because of challenges with controlling these factors independently. Therefore, a DNA-driven molecular system is developed wherein the integrin-binding ligand RGD can be reversibly presented and removed to achieve cyclic cell attachment/detachment on substrates of defined stiffness. Problematic alcohol use (PAU), a trait that combines alcohol use disorder and alcohol-related problems assessed with a questionnaire, is a leading cause of death and morbidity worldwide. Here we conducted a large cross-ancestry meta-analysis of PAU in 1,079,947 individuals (European, N = 903,147; African, N = 122,571; Latin American, N = 38,962; East Asian, N = 13,551; and South Asian, N = 1,716 ancestries). We observed a high degree of cross-ancestral similarity in the genetic architecture of PAU and identified 110 independent risk variants in within- and cross-ancestry analyses. Neurofibromatosis type 1 (NF1) is an autosomal dominant condition caused by germline mutations in the neurofibromin 1 (NF1) gene. Children with NF1 are prone to the development of multiple nervous system abnormalities, including autism and brain tumors, which could reflect the effect of NF1 mutation on microglia function. Using heterozygous Nf1-mutant mice, we previously demonstrated that impaired purinergic signaling underlies deficits in microglia process extension and phagocytosis in situ. To determine whether these abnormalities are also observed in human microglia in the setting of NF1, we leveraged an engineered isogenic series of human induced pluripotent stem cells to generate human …

The principal limitations of the terms NAFLD and NASH are the reliance on exclusionary confounder terms and the use of potentially stigmatising language. This study set out to determine if content experts and patient advocates were in favor of a change in nomenclature and/or definition. A modified Delphi process was led by three large pan-national liver associations. The consensus was defined a priori as a supermajority (67%) vote. An independent committee of experts external to the nomenclature process made the final recommendation on the acronym and its diagnostic criteria. IMPORTANCE: Apolipoprotein E (APOE)*2 and APOE*4 are, respectively, the strongest protective and risk-increasing, common genetic variants for late-onset Alzheimer disease (AD), making APOE status highly relevant toward clinical trial design and AD research broadly. The associations of APOE genotypes with AD are modulated by age, sex, race and ethnicity, and ancestry, but these associations remain unclear, particularly among racial and ethnic groups understudied in the AD and genetics research fields.

Multi-omics approaches are being used increasingly to study physiological and pathophysiologic processes. Proteomics specifically focuses on the study of proteins as functional elements and key contributors to, and markers of the phenotype, as well as targets for diagnostic and therapeutic approaches. Depending on the condition, the plasma proteome can mirror the platelet proteome, and hence play an important role in elucidating both physiologic and pathologic processes. This experimental study aims to examine the influence of many crucial parameters on the workability and compressive strength of Ready-Mix Concrete (RMC). The study utilized two distinct varieties of superplasticizers obtained from the local market. The fine aggregates utilized in this study were sourced from Sylhet sand, whereas the coarse aggregates were comprised of boulder crushed stone chips. To prevent early bridge failures, effective Structural Health Monitoring (SHM) is vital. Vibration-based damage assessment is a powerful tool in this regard, as it relies on changes in a structure’s dynamic characteristics as it degrades. By measuring the vibration response of a bridge due to passing vehicles, this approach can identify potential structural damage. Broad mite, (Polyphagotarsonemus latus Banks) (Acari: Tarsonemidae), is an economically important pest of blackberry production across the world. Since it was first reported on primocane-fruiting blackberries in 2010, little work has gone into developing IPM tools and understanding impact on floricane-fruiting cultivars. Affiliation is important for women during times of stress. Although, affiliation has been linked to women’s hormones across the menstrual cycle, many women use hormonal contraceptives which disrupt the links between hormones, psychology, and behavior. This study examined how hormonal contraceptives (hormonal IUD vs. the oral contraceptive pill) and menstrual cycle phase (follicular vs. luteal) are associated with affiliative motivations following the Trier Social Stress Test (TSST). Further, this study examined women’s cortisol reactivity and whether cortisol reactivity mediated affiliative motivation following the stressor. The formation of Mississippi Valley-type (MVT) lead-zinc (Pb-Zn) ores is complex, involving an interaction of tectonic compression, hydrothermal fluid migration, and geochemical processes within the subsurface. Fundamental questions regarding this interaction remain unclear, including the source(s) of the Pb and Zn ions that make up the ores. Fibroblast growth factors (FGFs) are a family of cell signaling proteins conserved across multiple species. Each individual FGF elicits different cellular functions including, but not limited to, proliferation, migration, differentiation, angiogenesis, and wound healing. One of the most studied members, fibroblast growth factor 2 (FGF2), has demonstrated substantial wound healing capacity in a wide range of tissues including skeletal, muscular, neural, respiratory, epithelial, and cardiovascular. Mid-southern U.S. rice (Oryza sativa L.) producers are challenged every year to control problematic barnyardgrass [Echinochloa crus-galli (L.) Beauv.] and weedy rice (Oryza sativa L.) because they have evolved resistance to most labeled herbicides. Oxyfluorfen is a Herbicide Resistance Action Committee (HRAC)/Weed Science Society of America (WSSA) Group 14 herbicide that inhibits protoporphyrinogen oxidase within the plant. Currently, there are no HRAC/WSSA Group 14 herbicides labeled for barnyardgrass or weedy rice control in midsouthern rice production. A robust finding in creativity research is that during idea generation, later (vs. earlier) responses on a divergent thinking task tend to be more creative, while the number of responses start off quickly, and then slow down over time, termed the serial order effect. Black African American Men (BAAM) suffer disproportionately from trauma related challenges and have a higher risk of encountering trauma across the lifespan. The negative impact of trauma is a major public health concern in the United States, evidence suggests trauma negatively impacts the physical well-being, mental health, and mortality rate. Eimeria species oocysts cause coccidiosis, one of the costliest diseases in the poultry industry. Coccidiosis infections cause significant economic losses due to reduced performance in chickens and turkeys. The objective of this thesis was to determine fecal bacterial recovery from chlorhexidine (CHX) salt suspensions containing Eimeria maxima M6 oocysts and evaluate the effect of CHX salts on oocysts during long-term cold storage. For bacterial recovery experiments, CHX digluconate and CHX gluconate were tested at different concentrations on poultry fecal slurries during oocyst shedding. The objective of this study of the architectural limestone of the 1891 Powerhouse/Icehouse in Eureka Springs, Arkansas was to analyze the environmental factors that influenced the deterioration and surface recession of this historical stone structure. The focus of this research was to examine and establish a baseline study that may assist in future monitoring and documentation of this historic limestone structure on stone weathering. A replicable methodology was specifically created toward this baseline research in hopes that it represented a solid baseline study.
